Isolation of Bacteriophage: An Antibiotic Alternative for the Control of Salmonella Infections.

The rise of multidrug resistance (MDR) poses a significant challenge to global health, reducing the effectiveness of conventional antibiotics. The World Health Organization (WHO) classifies Salmonella as a priority pathogen due to its global burden in foodborne disease and the escalating threat of MDR strains. Bacteriophages (phages) are gaining attention as alternatives to traditional antibiotics. Their strict host specificity has potential applications in both therapeutic and food safety contexts. However, the first step toward practical application is the successful isolation of phages from

4-Hydroxycoumarin inhibits biofilm formation and enhances antibiotic efficacy against Escherichia coli O157:H7.

BACKGROUND: Conventional bactericidal antibiotics often fail to eradicate Escherichia coli O157:H7 (E. coli O157) infections due to biofilm-mediated antimicrobial tolerance, and monotherapy can inadvertently drive resistance selection. This study aimed to identify compounds capable of modulating LuxS/AI-2-related phenotypes and to evaluate their potential to disrupt biofilms and enhance antibiotic efficacy against E. coli O157. METHODS: The effects of 4-HC on biofilm formation, extracellular polymeric substance (EPS) biosynthesis, and flagella-mediated motility were assessed in vitro. Antibio

Long-term spatiotemporal dynamics of β-lactamase resistome in the Han River, South Korea.

The global dissemination of antimicrobial resistance, particularly resistance to widely prescribed β-lactam antibiotics, is a critical public health threat worldwide. Aquatic ecosystems act as key reservoirs where antibiotic resistance genes accumulate and are transmitted between environmental bacteria and human pathogens within a One Health framework. To move beyond short-term observations and better understand these cross-sectoral dynamics, we conducted 5-year longitudinal surveillance using shotgun metagenomics across different seasons along the Han River, South Korea. Our analysis revealed

Single-entity tracking of intracellular resistance gene degradation by lattice-engineered nFe0 in anoxic groundwater.

Resolving how reactive materials interact with antibiotic-resistant bacteria at the single-entity level is essential for understanding intracellular resistance gene (iARGs) removal in anoxic groundwater, yet such processes remain inaccessible to ensemble-averaged methods. Here, a single-particle-single-cell analytical framework was established using time-of-flight mass spectrometry in combination with exogenous membrane labeling and endogenous elemental fingerprinting. This enabled the identification of single-entity material-membrane association, along with effective assessments of membrane d

Identification and whole-genome sequencing of blood-brain barrier-penetrating Riemerella anatipestifer isolated from chickens and ducks.

Riemerella anatipestifer (RA) is a major cause of bacterial meningitis, which poses a serious threat to the poultry industry. To identify RA strains with a strong ability to cross the blood-brain barrier (BBB) and analyze their genomic characteristics, a co-infection model was established using multiple RA strains with different serotypes in this study. We found that during co-infection with multiple RA strains, certain RA strains rapidly penetrated the BBB and were recovered from brain tissue as early as 0.25 h post-infection. The pioneer strains were selected to further evaluate their BBB-pe

Combined Residues of Frequently Detected Herbicides are Stronger Predictors of Soil Resistome and Virulence Potential than Single Residues.

Overuse of herbicides has resulted in a global epidemic of evolved resistance in soil ecosystems. While single herbicide risk has been documented, the impact of multiple herbicide residues on the soil resistomes remains poorly understood. Our study quantified the residues of 24 herbicides, profiled antibiotic resistance genes (ARGs) and virulence factor genes (VFGs), and reconstructed 1086 metagenome-assembled genomes (MAGs) from 93 agricultural soils across China. The analytical results demonstrate that the frequently detected herbicide residues (e.g., 60-90% detection frequency) are stronger

Microplastics and nanoplastics as potential ecological oncogenic co-drivers: rewiring host-microbe interactions in cancer.

Microplastics and nanoplastics (MPs/NPs) are increasingly recognized as biologically active interfaces that may reshape host-microbe interactions in carcinogenesis. This review proposes a hypothesis-generating mechanistic framework in which MPs/NPs may influence cancer-related processes through the integration of environmental exposure, epithelial barrier disruption, immune dysregulation, and tumor microenvironment remodeling. Although applicable to multiple epithelial interfaces, the framework primarily focuses on the intestinal mucosa, where the most substantial evidence exists. Through plas

Differential impacts of early, extended-spectrum, and last-resort β-lactams on manure-derived microbiomes and resistomes in an in vitro model.

Understanding the environmental dissemination of antimicrobial resistance (AMR) is crucial for mitigating its spread and reducing the clinical burden of resistant infections. In a previous study, we demonstrated that copiotrophic enrichment in a medium that stimulates the growth of gut-associated bacteria can reveal clinically associated bacterial populations and antibiotic resistance genes (ARGs) that are not detected through direct environmental sampling. In the present study, we applied this enrichment approach to poultry manure to examine how β-lactamase-resistant antibiotics (cefotaxime a

Solving the Staphylococcus aureus puzzle: lessons to guide future MRSA vaccine development.

SUMMARYThe broad-spectrum use of antibiotics allows for the development of resistance in symptomatically and asymptomatically carried pathogenic bacteria. Antibiotic-resistant bacteria, with this review focusing on Staphylococcus aureus, cause significant morbidity and mortality and put considerable strain on the global healthcare system. In a vaccinated individual, bacteria are less likely to develop resistance mechanisms compared to an individual treated with antibiotics, highlighting a potential method for conserving antibiotic use. However, despite over a century of research, a S. aureus v

Inoculum Effect in Multidrug-Resistant Gram-Negative Isolates: Impact on Ceftazidime and Cefepime Susceptibility.

Multidrug- or extensively drug-resistant (MDR or XDR) Gram-negative infections remain a global challenge, further complicated by the inoculum effect (IE), which can significantly alter the in vitro activity of β-lactam antibiotics and may contribute to treatment failure in infections with a high bacterial burden. In this study, we evaluated the IE of ceftazidime (CAZ) and cefepime (FEP) against multidrug-resistant Gram-negative clinical isolates. Fifty isolates, including Escherichia coli (E. coli), Klebsiella pneumoniae (K. pneumoniae), Pseudomonas aeruginosa (P. aeruginosa), and Proteus mira

Spatially Controlled Dual-Functionalization of a Titanium Keratoprosthesis: A Proof-of-Concept In Vivo Study.

Keratoprosthesis (KPro), an artificial corneal implant, is an established surgical option for restoring vision in severe corneal blindness; however, long-term outcomes remain limited by complications such as fibrosis and infection. Current biomaterial strategies generally lack the ability to address these distinct pathological processes at spatially defined implant-tissue interfaces. Here, we present a modified titanium KPro backplate with spatially controlled dual-functionalization, enabling spatially decoupled functionalization of opposing implant surfaces to address different biological cha

D-enantiomeric β-casein 197 disrupts biofilm-associated infection of MDR Pseudomonas aeruginosa and Acinetobacter baumannii through superior LPS binding and host cell protection.

Biofilms formed by multidrug-resistant (MDR) Pseudomonas aeruginosa and Acinetobacter baumannii are a major cause of chronic nosocomial infections and therapeutic failure. L-form antimicrobial peptides (AMPs) are rapidly degraded by proteases, whereas D-enantiomeric peptides offer enhanced resistance to proteases. Here, we synthesized L- and D-β-casein 197, a milk-derived AMP, and systematically compared their antibiofilm activities against eight clinical isolates of P. aeruginosa and A. baumannii. The D-form reduced biofilm biomass by up to 83% during formation and by up to 88% in preformed b

Performance evaluation of a polydopamine-modified carbon cloth bioanode for concurrent bioelectricity generation and ciprofloxacin transformation within a microbial fuel cell.

Ciprofloxacin (CIP) residues are difficult to eliminate using conventional biological treatment, whereas advanced oxidation processes commonly require substantial chemical or energy input. In this study, a dual-chamber microbial fuel cell equipped with a polydopamine-modified carbon cloth bioanode was evaluated for concurrent bioelectricity generation and ciprofloxacin removal. Polydopamine was deposited on carbon cloth by cyclic voltammetry electropolymerization. Scanning electron microscopy confirmed the formation of a conformal nanostructured coating on the carbon fibers. The microbial fuel

Synthesis and biological evaluation of 2-substituted heterocyclic quinolones as inhibitors of Vibrio cholerae sodium pumping NADH-ubiquinone oxidoreductase (NQR).

The rise of multi-drug-resistant bacteria highlights the need for antibiotics that act through novel mechanisms of action. The Na+-pumping NADH-Ubiquinone (UQ) oxidoreductase (NQR) is a respiratory enzyme foundin many pathogenic bacteria including Vibrio cholerae and plays a central role in energy production and redox balance. This enzyme is essential to the virulence of V. cholerae and structural motifs that are unique to NQR are not found in the humans, making it an excellent target for antibiotic drug design. Herein we report the synthesis and biological evaluation of 2-substituted heterocy
