Discovery of triazole derivatives for biofilm disruption, anti-inflammation and metal ion chelation.

In the face of bacterial hazards to human health and resistance to multiple antibiotics, there is an urgent need to develop new antibiotics to meet the challenge. In this paper, the triazolyl heterocyclic (3-amino-1,2,4-triazole, D) was synthesised efficiently using thiourea as starting material. Finally, the end product E was obtained by aldehyde-amine condensation reaction and the structures of all compounds were determined by spectral analysis. In vitro antimicrobial activity showed that E10 had a MIC of 32 μg/mL against the tested Escherichia coli and 16 μg/mL against the tested Staphyloco

The role of antibacterial prophylaxis in high-risk neutropenia: benefits, risks, and current perspectives.

PURPOSE OF REVIEW: To review the benefits, risks and specific considerations surrounding antibacterial prophylaxis (ABP) in adults with neutropenia, focusing primarily on high-risk patients with hematologic malignancies (HM) and/or hematopoietic cell transplantation (HCT). RECENT FINDINGS: There has been an overall reduction in benefit of fluoroquinolone prophylaxis (FQP) observed in recent studies, with a lack of overall mortality benefit and less efficacy in reducing Gram-negative bloodstream infections (BSI) rates, which may be explained by increasing rates of fluoroquinolone resistance (b

Therapeutic approaches for septicemia induced by multidrug-resistant bacteria using desert-adapted plants.

AIM: Septicemia, a life-threatening condition, can arise when bacterial infections are left untreated, allowing the pathogens to spread into the bloodstream. Moreover, infections caused by MDR bacteria are particularly challenging, as they can persist and lead to septicemia even when treated with conventional antibiotics. This study aimed to address this crisis by investigating combination therapies using desert-adapted medicinal plant extracts, including Jasonia candicans (J. candicans), Cistanche tubulosa, Moltkiopsis ciliata, and Thymelea hirsuta, as alternative treatments. The goal was to

Comparison of augmented prophylaxis using intra-prostatic antibiotic injection with standard oral prophylaxis in preventing the infectious complications following trans-rectal ultrasound (TRUS)-guided prostatic biopsy: a randomized controlled study.

BACKGROUND: Globally, infective complications following TRUS-guided biopsies along with emerging fluoroquinolone-resistant bacterial strains are a growing concern. Lack of specific guidelines on this subject necessitates the need to explore potential strategies to prevent such complications. Intra-prostatic antibiotic administered during TRUS is evaluated as an effective strategy to minimize infections with minimal systemic adverse effects. OBJECTIVE: To compare the efficacy of Augmented Prophylaxis using combined single dose intra-prostatic antibiotic injection and oral prophylactic antibiot

Piezoelectric Energy Harvesting from the Thorax Vibration of Freely Flying Bees.

Periodontitis is a chronic inflammatory disease mediated by the immune system. Its pathogenesis involves the interaction of multiple factors, among which the accumulation of dental plaque is considered the initial key factor in the onset of the disease. As the pathogenic bacteria in plaque proliferate and metabolites are released, the host's immune system produces a strong response, leading to an inflammatory response and structural destruction of local tissues. Traditional treatment relies on mechanical scraping and antibiotics but suffers from tissue damage, difficulty in removing deep-seate

Synthesis, anti-biofilm and molecular docking of amino-substituted chalcones targeting Staphylococcus aureus sortase A.

Antibiotic resistance is an urgent global health challenge that requires the development of new antibacterial agents. In this study, 14 aminochalcones bearing electron-withdrawing groups were synthesized and evaluated for antibacterial activity. Chalcone C5, with an ortho-chlorine on ring B, demonstrated the most potent effect, notably against methicillin-susceptible and methicillin-resistant Staphylococcus aureus (MICs of 1.9 and 3.9 µg/mL, respectively), comparable to vancomycin. C5 showed synergistic interaction with vancomycin, reducing its MIC tenfold. Time-kill assays confirmed C5's bact

New dimension of ciprofloxacin hybrids: Synthesis, characterization and anticancer evaluation of newly C3-modified ciprofloxacin derivatives.

The strategic application of drug repurposing and repositioning in the design of hybrid molecules represents a promising alternative in contemporary drug discovery. Recent studies have highlighted the anticancer activity of ciprofloxacin derivatives, attributing this effect primary to their capacity to inhibit DNA topoisomerase II. In silico ADME profiling of C3-modyfied ciprofloxacin analogues, have further identified their role as potential P-glycoprotein inhibitors. Building upon these findings, we proposed the synthesis of a series of ciprofloxacin-peptidomimetic hybrid compounds to explor

Efficient Synthesis of Clovibactin and Its Analogues: Promoting the Discovery of More Potent Antibiotics via Multiple Screenings.

Increasingly, antimicrobial resistance has become a major concern for public health. The recently discovered cyclic depsipeptide, clovibactin, is a potential antibiotic candidate with potent activities against multidrug-resistant pathogens. It contains three d-amino acids, including a rare d-hydroxyasparagine residue. Herein, we present the efficient synthesis of the suitably protected d-hydroxyasparagine building block and the natural product clovibactin. Efforts have also been made by us to establish the structure-activity relationship (SAR) of clovibactin and to hunt for more active analogu

Efflux pump modulation by Montelukast and its roles in restoring antibiotic susceptibility in multidrug-resistant Staphylococcus aureus.

BACKGROUND: Staphylococcus aureus and its drug-resistant mutants are mentioned among the WHO's high-priority list of pathogens. Antibiotics like fluoroquinolones and cephalosporins are used to treat multidrug-resistant S. aureus infections. However, a higher expression of efflux pumps (NorA, NorB, and AbcA) induces multidrug resistance. The master regulator, MgrA, regulates the expression of most of these efflux pumps in S. aureus. The phosphorylation status of MgrA is determined by the cellular PknB/RsbU ratio, where PknB, a serine-threonine kinase, and RsbU, a serine-threonine phosphatase, a

The antibacterial efficacy of nitroxoline against multidrug resistant Escherichia coli associated with copper binding.

Nitroxoline (NIT) has been approved for the treatment of uncomplicated urinary tract infections (UTIs) for more than half a century, yet its antimicrobial properties remain incompletely understood. Here, we determined the intricate connections between NIT's metal-chelating capabilities and its antibacterial activity. Metal ion binding characteristics were measured by Ultraviolet-visible (UV-vis) spectroscopy. Biochemical assays and molecular docking studies were performed to elucidate the underlying mechanism. We found that NIT could interact with a diverse array of metal ions, including Cu2+,

FS536, a novel nitric oxide-releasing doxorubicin hybrid, reverts multidrug resistance in lung cancer cells.

Источник пометил публикацию как отозванную

The design of molecular hybrids that chemically conjugate nitric oxide (NO)-donors with anticancer drugs, offering site-specific and time-controlled properties, is a promising strategy in cancer therapy. In this work, we designed, synthesized, and characterized a novel doxorubicin (DOXO)-NO-donor hybrid, named FS536, by chemically conjugating DOXO with a diazeniumdiolate moiety. Upon incubation in human serum, FS536 simultaneously released both DOXO and NO through enzymatic hydrolysis. FS536 significantly inhibited the proliferation of the DOXO-resistant A549 lung cancer cell line (A549-DR), o
