Enzymes-enhanced antibiotic therapy reduces biofilms to undetectable levels in an implant-associated infection model.

Implant-associated infections caused by biofilm-forming bacteria, such as Staphylococcus aureus, remain a major clinical challenge due to their high tolerance to conventional antibiotic therapies. We report a dual-targeted therapeutic strategy that combines a tri-enzymatic cocktail designed to degrade key components of the biofilm matrix (TEC; comprising a DNA/RNA endonuclease, an endo-1,4-β-D-glucanase, and a β-N-acetylhexosaminidase), with vancomycin, both delivered via a thermosensitive poloxamer 407 hydrogel, for localized treatment of S. aureus biofilms. The formulation was evaluated both

Genomic and functional insights into a novel Lactococcus sp. AK05 from Cirrhinus mrigala with potent antagonism against Aeromonas hydrophila.

In aquaculture, addressing antibiotic-resistant fish pathogens underscores the immediate necessity for the identification of candidate probiotic strains with favorable safety and functional attributes as alternatives to antibiotics. We isolated and characterized a new strain of Lactococcus sp., called AK05, from the gut of Cirrhinus mrigala. The phenotypic tests showed that it can survive and grow in harsh conditions, including a pH of 4, and 0.8% bile salts. At 8% NaCl, it survived and exhibited merely a one-log rise in CFU·mL⁻¹, while NaCl levels ≤ 5% facilitated over a 4-log increase in 24

Could oritavancin be a promising alternative treatment for staphylococcal bone and joint infections? Insights from the determination of oritavancin minimum inhibitory concentrations in a collection of clinical isolates from the French National reference centre for staphylococci.

Oritavancin is a long-acting lipoglycopeptide with excellent bone penetration and activity against biofilm-embedded bacteria, making it a promising candidate for the treatment of bone and joint infections (BJIs). To explore its potential in this clinical context, we assessed the in vitro activity of oritavancin in comparison with other glyco-, lipo- and glycolipo-peptides against a panel of 148 multidrug-resistant staphylococcal clinical isolates, mainly collected from BJI cases. Although oritavancin showed lower overall activity than dalbavancin, resistance to oritavancin could not be reliabl

Oral vancomycin for primary sclerosing cholangitis and associated inflammatory bowel disease - paving a path forward.

INTRODUCTION: Primary sclerosing cholangitis (PSC) is a fibro-inflammatory cholangiopathy strongly associated with inflammatory bowel disease (PSC-IBD). With no approved PSC therapy, clinicians face uncertainty about oral vancomycin (OV) as a therapeutic option. This review synthesizes clinical effectiveness evidence alongside mechanistic data. AREAS COVERED: We searched PubMed/Google Scholar for studies of vancomycin in PSC from 1998 through November, 2025. OV shows consistent IBD benefits and variable liver responses. In PSC-IBD, clinical and endoscopic remission occurred in 60% at 6 months

Staphylococcus aureus resistance to non-β-lactam antibiotics: global genomic epidemiology and machine learning feasibility assessment.

Staphylococcus aureus is increasingly resistant to β-lactam antibiotics, making non-β-lactam cell-wall-targeting drugs crucial alternatives. Growing resistance to these agents highlights the need to identify genomic factors influencing susceptibility. Machine learning can integrate genomic and phenotypic data to predict minimum inhibitory concentrations (MICs) and uncover resistance mechanisms across time and regions. We obtained 112,360 S. aureus genomes from NCBI GenBank (March 2024), applying quality filters and standardizing metadata. Resistance genes and mutations were identified using AM

Production of Multiple Variants of the Antimicrobial Sactipeptide Gnavucin D by the Human Gut Isolate Mediterraneibacter gnavus HB038.

While several bacteriocins have been identified from gut-isolated cultures, there remains a need for the discovery of bacteriocins with varying inhibition spectra for strain applications such as microbiome editing, pathogen elimination and colonization resistance. With this in mind, here we describe a new antibacterial sactipeptide gnavucin D, produced by Mediterraneibacter gnavus HB038 isolated from a healthy 2-year-old child. Gnavucin D has activity against the pathogens Clostridium perfringens, Streptococcus agalactiae, Bacillus cereus, and vancomycin-resistant Enterococcus. The gene cluste

Contribution of viable but non culturable cells and small colony variants in antibiotic insusceptibility and therapeutic failure against S. aureus and P. aeruginosa biofilm co-infections.

P. aeruginosa and S. aureus are often co-isolated from biofilm-associated infections, such as those afflicting cystic fibrosis (CF) patients. Biofilms, along with the interspecies interactions, play a significant role in fostering antibiotic insusceptibility, contributing to infection chronicity. Previously, we showed that S. aureus adopts a viable but non-culturable (VBNC) state in biofilms with P. aeruginosa. Here, we aimed to gain insight into the impact of VBNC and phenomena such as phenotypic switching on antimicrobial treatment and vice-versa. Single- and dual-species biofilms of two iso

Host genetics shapes the recovery of the gut microbiome after antibiotic treatment: the role of the blood group related B4galnt2 gene.

UNLABELLED: The intestinal microbiota is integral to host health, metabolism, and colonization resistance. Antibiotics can disrupt microbial homeostasis, leading to dysbiosis and altered colonization resistance. While antibiotic-induced microbiota disruption is well-documented, less is known about how host genetics shapes post-antibiotic recovery. Here, we investigate the impact of B4galnt2, a blood-group-related glycosyltransferase gene, on microbiota recovery following antibiotic treatment. Using a longitudinal, multi-omic approach-including 16S rRNA gene sequencing, metagenomics, and metatr

Argyrin B exhibits potent therapeutic efficacy in a Clostridioides difficile-infection mouse model while preserving microbiota functionality.

Clostridioides difficile infection (CDI) remains a leading cause of antibiotic-associated diarrhoea, with high recurrence rates and limited treatment options that preserve gut microbiota. Current treatments, including vancomycin, further disrupt the already compromised gut microbiota, often prolonging dysbiosis and increasing the risk of colonization by resistant pathogens. This study demonstrates that argyrin B exhibits potent activity against C. difficile in vitro and significantly reduces bacterial burden in a mouse model of infection. Argyrin B displayed a narrow antimicrobial spectrum, su

Between-centre variation in antibiotic therapeutic drug monitoring and obtaining surveillance cultures in Dutch ICUs: A retrospective observational study.

PURPOSE: Antimicrobial resistance is a major concern in intensive care units (ICUs), where infections and antibiotic use are common. Antimicrobial stewardship programs (ASPs) promote optimal infectious disease management through interventions such as Therapeutic Drug Monitoring (TDM) and selective digestive/oropharyngeal decontamination (SDD/SOD), but data on ICU-specific quality indicators (QIs) and between-centre variation are limited. This study aimed to evaluate infectious disease management in Dutch ICUs by assessing adherence to, and variation in, TDM and SDD/SOD practices. METHODS: In

Meta-analysis of global prevalence, antibiotic susceptibility, and virulence genes of Vibrio spp. in aquatic products.

Vibrio-associated foodborne diseases constitute a substantial global health burden, and aquatic products are frequently contaminated with antibiotic-resistant and potentially virulent Vibrio. This meta-analysis aimed to quantify the global prevalence, antibiotic resistance, and virulence genes of Vibrio in aquatic products. From 108 eligible studies, pooled prevalence of Vibrio reached 20.99% (95% CI: 18.04-24.09%), exhibiting a decreasing trend over time. Contamination rates were higher in shellfish (25.43%) than in shrimp (21.69%) and fish (17.63%), and in farm samples (26.31%) compared to r
