M.pneumoniae is a major cause of human respiratory infection whose pathogenicity relies on adhesion proteins, secreted virulence factors, and immune evasion mechanisms. This review contextualizes these strategies within the broader phylogeny of clinically relevant Mycoplasma species while addressing the diverse extrapulmonary manifestations of M. pneumoniae. Current challenges in managing M. pneumoniae infections are multifaceted. The post-pandemic resurgence reflects a population-level immune debt resulting from prolonged non-pharmaceutical interventions, with test positivity rising sharply a

During the 2010s, Escherichia coli ST131 clade A tested sensitive to most antibiotics. In the 2020s, Europe experienced outbreaks of fluoroquinolone-resistant ST131-A that contained CTX-M β-lactamases and OXA-48-like carbapenemases. We investigated the acquisition of different antimicrobial resistance determinants in the emergence of multidrug-resistant (MDR) ST131-A. Phylogenetic analysis and Bayesian inference were utilized to define subclades and clusters (within subclades) and investigate the evolutionary history of ST131-A, using sequenced genomes and publicly available genomic data (n=2,

Salmonella spp. are major foodborne pathogens transmitted through contaminated poultry products. The emergence of antibiotic-resistant strains complicates treatment and increases the risk of multidrug-resistant (MDR). In Morocco, data on antimicrobial resistance (AMR) profiles in poultry-associated Salmonella remain limited. This study aimed to assess the prevalence and AMR patterns of Salmonella spp. isolated from poultry samples collected in Morocco in 2023. A total of 1339 poultry samples (ceca, turkey, and chicken meat) were collected using a representative stratified sampling approach fro

OBJECTIVES: Antimicrobial resistance (AMR) is a serious threat to human and animal health; the emergence of 3rd-generation cephalosporin- and fluoroquinolone-resistant Enterobacterales is particularly problematic. Because Escherichia coli is the most common cause of urinary tract infections in dogs, AMR has real-world impact. The objectives of this study were to evaluate the antimicrobial susceptibility of canine urinary isolates of E. coli and to determine if AMR is increasing in Saskatchewan.
ANIMALS AND PROCEDURE: A total of 576 diagnostic isolates of canine urinary E. coli from individual

BACKGROUND: Tuberculosis (TB) preventive treatment (TPT) is a core component of TB control. Levofloxacin was recently recommended by the World Health Organization as TPT in children exposed to multidrug-resistant tuberculosis (MDR-TB). However, the affordability and feasibility of TPT provision for governments, health services, individual households and global donors is uncertain.
METHODS: This costing analysis used empirical data collected alongside the TB-CHAMP clinical trial which investigated levofloxacin TPT in children exposed to MDR-TB in South Africa. We modelled the societal (health

New pyrrolo[1,2-a]quinoxaline-2-carboxylic acid hydrazone derivatives were synthesized from nitroaniline using a multi-step pathway. Structure characterization of these novel substituted pyrrolo[1,2-a]quinoxaline-carboxylic acid hydrazones was achieved by using FT-IR, 1H NMR, 13C NMR, X-Ray and HRMS spectral analysis. These pyrroloquinoxaline hydrazone compounds were then evaluated in vitro for their potential on the human acute myeloid leukemia cell line MV4-11, in which (N'-pyridin-2-ylmethylene)pyrrolo[1,2-a]quinoxaline-2-carboxylic acid hydrazone 1a showed an IC50 value of 15.51 µM. Emergence of drug-resistant Mycobacterium tuberculosis necessitates the development of new drugs. Here, we studied the activity of new piperazine nitrofuranylamides against a collection of clinical strains from an multidrug-resistant tuberculosis (MDR-TB) high-burden country. A series of nitrofuranylamides 9-15 was synthesized and evaluated using a collection of diverse clinical strains representing globally spread genotypes (Beijing, LAM, and Haarlem; susceptible, MDR, and pre-extensively drug resistant). Minimal inhibitory concentrations (MIC) were determined using a resazurin assay. Whole-g

Quinolone group antibiotics have proven to be an effective drug against Mycobacterium tuberculosis. However, point mutation in DNA gyrase, one of the primary targets for quinolone antibiotics, has led to conformational alterations in the binding sites, leading to a change in drug pharmacodynamics. This has contributed to the development of drug-resistant strains such as multidrug-resistant strains (MDR) and an extreme drug-resistant strains (XDR). In this study, our objective was to find the impact of mutation A90V and A90E on DNA Gyrase A of Mycobacterium tuberculosis. Ten quinolone antibioti

Antibiotic residues are commonly found in food. The effect of dietary exposure to veterinary antibiotics on the transmission of antibiotic-resistant bacteria and antibiotic resistance genes (ARGs) from food to humans is unknown. We found that dietary exposure to enrofloxacin reduced microbial diversity, interactions, and the immune responses; weakened the colonization resistance of the resident microbiota; and promoted the colonization of exogenous Escherichia coli K-12 MG1655 in the simulated human intestine both in vitro and in vivo experiments in mice. In addition to the growth advantages f

BACKGROUND: Staphylococcus aureus infections have become a significant public health issue due to increasing the resistance against known antibiotics, especially by Methicillin-Resistant Staphylococcus aureus (MRSA). Fluoroquinolones are broad- -spectrum class of antibiotics mostly utilized in treating various bacterial infections and those caused by S. aureus. Reported data indicated that mutations of Ser84 to Leu, Ser85 to Pro and Glu88 to Lys in DNA gyrase A enzyme are the major cause of fluoroquinolone resistance against S. aureus. Therefore, the development of a novel targeted molecule wi

Quinolones, discovered in the 1970s, have played a critical role in revolutionizing the treatment of bacterial infections due to their broad-spectrum antimicrobial activity. Over the decades, these compounds have been extensively studied, resulting in the development of numerous new derivatives. This review explores the history and development of quinolones, focusing on their Structure-Activity Relationship (SAR), mechanisms of action, and the challenges posed by antimicrobial resistance. The key resistance mechanisms include mutations in DNA gyrase and topoisomerase IV, which reduce drug bind
