INTRODUCTION: The KRAS G12D inhibitor MRTX1133 represents a major advance in targeting oncogenic KRAS, but adaptive resistance driven by ERK reactivation, dependent on wild-type (WT) RAS membrane localization, limits its efficacy.
RESULTS: Here, we identify a rational strategy to overcome this resistance mechanism using FGTI-2734, a dual farnesyltransferase (FT) and geranylgeranyltransferase-1 (GGT-1) inhibitor that disrupts WT RAS membrane localization. FGTI-2734 blocks MRTX1133-induced ERK feedback reactivation and synergizes with MRTX1133 to inhibit proliferation and induce apoptosis in KR

In view of the worldwide expansion of life-threatening invasive fungal diseases (IFDs) and fungal drug resistance, new antifungal drugs are urgently needed. To guide research and public health policies, the World Health Organization (WHO) specified 19 priority-concern human mold and yeast pathogens associated with serious risk of mortality or morbidity. We assessed the in vitro susceptibility of twenty-three fungal pathogens, 13 of them included in the WHO priority concern list, to a set of 24 polyamine derivatives generated by linking either alkylated ethylenediamine or a polyamine macrocycle

A series of 24 novel fluoroquinolone-uracil hybrids via various lengths linkers were designed, synthesized and evaluated for their antibacterial activities against both Gram-negative and Gram-positive bacteria, including resistant strains. All target compounds were fully characterized by 1H NMR, 13C NMR and high-resolution mass spectrometry (HRMS). Most of the tested FQs-AU hybrids displayed a significant antipathogenic activity. For instance, A3, A4, A5, A9, A10, A11, B16, C19 and C22 demonstrated broad-spectrum antibacterial activity against the 6 laboratory strains and 9 clinically isolated

There is a growing need to understand ciprofloxacin (CIP) resistance in less prevalent Salmonella serovars like Salmonella Dublin, which causes life-threatening conditions in both humans and animals. This study investigated potential factors contributing to CIP-resistance in a Salmonella Dublin isolate. The isolate was detected from an initial screening of 17 biobanked Salmonella isolates using the Kirby-Bauer disk diffusion (KBDF) method. The minimum inhibitory concentration (MIC) values of the identified CIP-resistant Salmonella Dublin isolate and a CIP-susceptible isolate of the same serova

Pseudomonas aeruginosa is considered a priority pathogen by the World Health Organization due to its resistance to antibiotics. Isolates resistant to ciprofloxacin (CPFX), a bactericide commonly used against P. aeruginosa, usually carry the mutations T83I or D87N in the GyrA subunit of the DNA gyrase. Yet, the molecular mechanisms by which these mutations confer CPFX-resistance to P. aeruginosa are unknown. Here we solved the crystal structure of the P. aeruginosa gyrase catalytic cleavage core and used it to carry out molecular dynamic (MD) simulations of CPFX-gyrase binding in the wild-type

Fluoroquinolones (FQLs) are synthetic derivatives of the quinolone class, originally developed from the nalidixic acid scaffold. FQLs are widely used in human and animal medicine due to their broad spectrum of activity against Gram-positive and Gram-negative bacteria. Their strong bactericidal properties result from the inhibition of bacterial DNA transcription and replication. However, inappropriate use of these antibiotics often leads to toxic side effects, environmental pollution, and the development of antibiotic resistance in bacteria. Recently, numerous methods for determination of FQLs

Antimicrobial resistance exacerbates the difficulty of clinical bacterial infection treatment, as single-target antibiotics rapidly lose efficacy shortly post-clinical use. Such resistance highlights an urgent need for multitargeted therapeutics. Metalloantibiotics, combining metal ions with antimicrobials to disrupt diverse bacterial pathways, represent a promising strategy to circumvent resistance. Here, we engineer a sideromycin-bismuth molecular nanoassembly for treating ciprofloxacin-resistant Pseudomonas aeruginosa. Using phylogenomics-driven methods, we identify four hydroxamate siderop

Persistent fluoroquinolone (FQ) antibiotics in water bodies boost bacterial resistance and endanger aquatic ecosystems, necessitating efficient removal. In this work, a natural-biopolymer sodium alginate-based adsorbent (SA@Fe3O4-SPABS) was prepared via cross-linking and amidation to introduce multiple functional groups onto the adsorbent framework. Gatifloxacin (GAT) was selected as a representative fluoroquinolone to evaluate the adsorption performance. The kinetic data were well described by the pseudo-first-order model. Based on the Langmuir model, SA@Fe3O4-SPABS exhibited an adsorption ca

Stenotrophomonas maltophilia is an emerging multidrug-resistant (MDR) pathogen that primarily causes healthcare-associated infections. This bacterium employs two key resistance mechanisms-intrinsic and acquired-to withstand antimicrobial toxicity, facilitating its spread and persistence within healthcare settings. This review focuses on acquired resistance mechanisms in S. maltophilia, highlighting genetic mutations and gene acquisition through horizontal gene transfer (HGT). Mutations that confer antimicrobial resistance commonly occur in drug targets (e.g., gyrA and parC, which encode DNA gy

BACKGROUND: Carbapenem-resistant Enterobacterales (CRE) pose a critical global threat. However, the genomic epidemiology, transmission dynamics (clonal vs. horizontal gene transfer), and mechanisms driving co-resistance in Southern Vietnam remain poorly understood. This study aimed to use Whole-Genome Sequencing (WGS) to characterize the molecular epidemiology, transmission mechanisms, and co-resistance patterns of CRE from a major referral center in Southern Vietnam.
METHODOLOGY: We performed a cross-sectional study using whole-genome sequencing on 189 CRE isolates (K. pneumoniae, E. coli, E

Biofilm accumulation in recirculating cooling-water systems (RCSs) contributes significantly to microbial corrosion and heat transfer impairment. However, traditional chlorine-based bactericides require frequent replenishment for sustained performance and chlorine ions exacerbate pipeline corrosion. This study employs controlled-release borates (CRBs) to ensure continuous boron release and thorough biofilm contact. The release rate of CRBs in the first 10 days was 39.2 mg/(L·d), following zero-order release kinetics. At dosages above 0.4 g-B/L, CRBs reduced total heterotrophic bacteria counts

The earliest responses of pathogenic bacteria to antibiotics can affect the outcome of an infection. While long-term adaptations have been extensively studied, the immediate transcriptional changes that unfold immediately following antibiotic exposure remain poorly understood. Here, we applied iModulon analysis to time-resolved transcriptomic data from Escherichia coli exposed to subinhibitory concentrations of two antibiotics (ampicillin and ciprofloxacin), capturing transcriptional regulatory changes occurring within the first 30 min of exposure. This analysis proposes an integrated, three-p
