Marine Bacteria as a Source of Antibiotics Against Staphylococcus aureus: Natural Compounds, Mechanisms of Action, and Discovery Strategies.

Staphylococcus aureus is a major opportunistic pathogen responsible for a wide spectrum of human infections, including severe and difficult-to-treat cases. The emergence of multidrug-resistant strains limits the efficacy of conventional antibiotic therapies and poses a significant global public health challenge. In this context, the search for novel antibiotics has intensified, with increasing interest in marine resources, an ecosystem still largely underexplored. Marine bacteria produce a vast array of secondary metabolites with unique structures and potentially novel modes of antibacterial a

Asymmetric Cu─N─Ru Bridgedsite Nanozyme-Loaded Injectable Thermogel Boosts Cuproptosis-Like Death for Multidrug-Resistant Urinary Tract Infections.

Multidrug-resistant urinary tract infections (MDR-UTIs) remain one of the most challenging clinical infections globally owing to the paucity of effective therapeutic agents. Nanozymes-triggered cuproptosis-like bacterial death has emerged as a promising bactericidal strategy. However, its efficacy is compromised by insufficient catalytic sites and suboptimal synergy among active centers. Consequently, the development of nanozymes capable of efficiently inducing cuproptosis-like death holds considerable potential for treating MDR-UTIs. Herein, the Cu-ZIF8-Ru nanozyme was engineered that leverag

Self-healing quaternized chitosan-phenylboronic hydrogel with ceria nanozymes and stem-cell exosomes simultaneously eradicates MRSA biofilm and regenerates diabetic wounds.

Diabetic wounds represent a major clinical challenge due to their susceptibility to bacterial biofilm infections, persistent oxidative stress, and impaired tissue regeneration. Current therapeutic approaches typically address these pathological factors individually, limiting their overall efficacy in achieving complete wound closure. There is thus an urgent need for integrated platforms capable of simultaneously targeting multiple barriers to healing. Here we show that a self-healing hydrogel composed of quaternized chitosan-phenylboronic acid (QCSP), ceria nanoparticles (CeNPs), and embryonic

Effective treatment of multidrug-resistant Riemerella anatipestifer infection in ducks using a compound Chinese herbal medicine in drinking water.

Riemerella anatipestifer (R. anatipestifer), a pathogenic bacterium, causes systemic infection in poultry. Although antibiotics can alleviate R. anatipestifer infection, their irrational use has worsened antibiotic resistance, rendering existing drugs less effective against the growing number of multidrug-resistant strains. Treatment with compound Chinese herbal medicines (CCHMs), which show the ability to eliminate pathogens and reduce inflammation in the body, offers a promising alternative for the treatment of bacterial infections. Herein, we characterized a multidrug-resistant R. anatipest

Heterogeneous multicopy of blaCTX-M variants on the same plasmid enhances evolutionary adaptability in clinical Klebsiella pneumoniae.

Pathogenic bacteria continually evolve under antimicrobial pressure through acquired resistance genes, making it crucial to understand their evolutionary strategies. We identify a clinical Klebsiella pneumoniae isolate resistant to ceftazidime/avibactam (CZA), harboring heterogeneous multicopy blaCTX-M, among which a blaCTX-M-249 variant mediates CZA resistance. Both blaCTX-M-249 and its closely related allele blaCTX-M-65 are dominant within the clonal population and are located at two loci on the same plasmid, with their proportions shifting under antibiotic pressure. Using experimental and m

Combatting antibiotic resistance: challenges and emerging therapeutic strategies.

The discovery of antibiotics marked a crucial milestone in medical history, widely credited for its pivotal role in saving countless lives. However, the escalating resistance of microbes to traditional antimicrobial drugs (antibiotics) now jeopardizes the effectiveness of these life-saving drugs. The World Health Organization adopted a Global Action Plan (GAP) in 2015, recognizing the looming threat to human health posed by antimicrobial resistance (AMR). As of 2019, 4.95 million global deaths are attributed to AMR, with 1.27 million deaths recorded in that year alone. In this review, a thorou

A macrophage-mimetic nanocarrier co-loaded with geraniol and FK13-a1 for MRSA-induced acute lung injury.

Current clinical strategies for Methicillin-resistant Staphylococcus aureus (MRSA)-induced acute lung injury (ALI) predominantly focus on single-approach interventions such as anti-inflammatory therapy. However, due to the complex, multi-pathway pathological network underlying the disease, targeting a single pathway often yields suboptimal therapeutic outcomes. Consequently, there is a pressing need to develop innovative drug delivery systems capable of systematically addressing this intricate pathological process. Geraniol, a naturally derived monoterpene alcohol, exhibits multiple pharmacolo

In vivo selection of CMY-219 conferring resistance to ceftazidime-avibactam in an OXA-484-producing E. coli ST410.

Resistance to ceftazidime-avibactam (CAZ-AVI) is a growing problem. This study describes the selection of CMY-219, a CMY-42 variant (G156D), conferring resistance to CAZ-AVI in an OXA-484-producing Escherichia coli ST410 after treatment. It raises concern about the risk of selection of CMY variants under CAZ-AVI exposure in ST410 and related clones, which commonly carry CMY-42, are prone to carbapenemase acquisition, and harbor modified PBP3.

Synthesis of Lignin-Derived Hierarchical Porous Carbon via Hydrothermal-Phosphoric Acid Synergistic Activation for Enhanced Adsorption of Tetracycline.

Tetracycline is a low-cost broad-spectrum antibiotic and widely used in medicine and aquaculture. Its residues are usually released into the environment through wastewater, which may lead to the spread of antibiotic resistance genes and pose ecological risks. To address this environmental issue, a hierarchical lignin-derived porous carbon (LPHC) was synthesized using renewable biomass lignin as the precursor through a combined phosphoric acid-activated hydrothermal pretreatment. The resulting LPHC was used to effectively remove tetracycline from aqueous solutions. Characterization results indi
