Genome-Driven Discovery of Anti-MDR Bacterial Heptapeptides from a Cold-Seep-Derived Bacillus Strain.

With the increasing emergence of multidrug-resistant (MDR) bacteria, there is an urgent need to discover new antibiotics. In this study, genome mining coupled with anti-bacterial assay guided the targeted isolation of two new heptapeptides nobilamide Q3 (1) and R3 (2). These compounds were identified as new stereoisomers of the known scaffold A-3302-B (3). The structures of these compounds were elucidated through a combination of MS, NMR spectroscopy and Marfey's analysis. Anti-MDR bacterial assays showed that compounds 2 and 3 exhibited effective growth inhibition against the Gram-positive MD

Encapsulated probiotics as antimicrobial agents: mechanisms and delivery strategies against multidrug-resistant pathogens.

The rapid escalation of antimicrobial resistance (AMR) has rendered many conventional antibiotics ineffective, emphasizing the need for alternative therapeutic approaches. Probiotics have emerged as promising biotherapeutic agents capable of inhibiting multidrug-resistant (MDR) pathogens through diverse mechanisms, including secretion of antimicrobial metabolites (bacteriocins, organic acids, short-chain fatty acids, and hydrogen peroxide), competitive exclusion, quorum-sensing interference, and immune modulation. However, their clinical application is limited by poor stability under environme

Mapping antibiotic use and calf-rearing practices on Brazilian dairy farms.

Antimicrobial use in preweaning dairy calves is largely driven by the high morbidity and mortality observed in early life and by on-farm management practices. However, such use also contributes to the emergence and dissemination of antimicrobial-resistant bacteria, raising important public and environmental health concerns. This study aimed to characterize antimicrobial use patterns and management practices in Brazilian dairy herds. An online questionnaire was distributed between June and November 2020 via social media, e-mail, and WhatsApp messages to reach dairy farm managers, including prod

Reining in Multidrug Resistance Protein 1 via Binding Its Flexible Interdomain Linker with Sequence-Selective Peptide-Binding Nanoparticles.

Overexpressed ATP-binding cassette (ABC) transporters, such as multidrug resistance protein 1 (MRP1) encoded by ABCC1, are responsible for multidrug resistance in anticancer treatment due to their abilities to prevent drugs from reaching their lethal intracellular concentrations. Similar overexpression of drug efflux pumps is a major contributor to antimicrobial resistance seen in bacteria. We report sequence-selective, molecularly imprinted nanoparticles (MINPs) targeting MRP1 in human cancer cells. These nanoparticles mask different segments of the long, flexible linker connecting NBD1 (nucl

Within-host evolution drives the emergence of ceftazidime-avibactam resistance mediated by IncN plasmid-encoded blaNDM-1 and blaKPC-33 in ST11-KL64 hypervirulent Klebsiella pneumoniae.

UNLABELLED: Ceftazidime-avibactam (CZA) is considered one of the most effective antimicrobial agents for treating infections caused by Klebsiella pneumoniae carbapenemase-producing Klebsiella pneumoniae. However, rising CZA resistance poses a critical clinical challenge, primarily driven by blaKPC mutations. From a single CZA-treated patient, we isolated two CZA-susceptible and three CZA-resistant K. pneumoniae strains. Whole-genome sequencing of CZA-resistant strains CYH4028 and CYH1535 revealed two key resistance pathways: (i) acquisition of a conjugative IncN plasmid harboring blaNDM-1 and

The emerging pathogen Candida auris: host interactions and disease drivers.

Candida auris has emerged over the past fifteen years as a multidrug-resistant human fungal pathogen responsible for hospital-acquired infections associated with high mortality rates. Clinical isolates frequently exhibit resistance to one or more first-line antifungal drugs, and management is further complicated by persistent colonization, biofilm formation, and historical misidentification in diagnostic laboratories. Rapid species-level identification and accurate antifungal susceptibility testing are critical for effective patient care and infection control. In this article, we summarize the

CaO2 nanoparticle-loaded injectable hydrogel with sustained oxygen release and ROS-scavenging functions for accelerating frostbite wound healing at high altitude.

Frostbite is common in high-altitude regions, where low oxygen levels and the cold climate aggravate wound hypoxia and reactive oxygen species (ROS) accumulation, making frostbite lesions prone to becoming hard-to-heal wounds. Traditional hydrogel dressings remain unsatisfactory under such extreme conditions, as they lack freezing resistance, low-temperature adhesion, sustained oxygen release, and ROS-scavenging activity. Herein, we report a multifunctional hydrogel for the management of high-altitude frostbite wounds, formed from polyvinylpyrrolidone-coated CaO2 nanoparticles (CPO NPs), polyv

Concentration- and Time-Dependent Virucidal Responses of Major Transboundary Animal Disease Viruses to Disinfectants.

Transboundary animal diseases (TADs) pose persistent threats to global livestock production, and chemical disinfection remains a critical component of biosecurity. However, virucidal efficacy is commonly assessed using single-condition endpoints, limiting comparative interpretation across biologically heterogeneous viruses. In this study, an experimental framework explicitly structured across virus species, disinfectant concentration, and contact time was applied to systematically compare virucidal response patterns across four major TAD viruses-avian influenza virus, African swine fever virus

KPC-33 and ompK37 mutations: Unraveling the mechanism of ceftazidime/avibactam resistance in ST11 carbapenem-resistant Klebsiella pneumoniae.

BACKGROUND: Global surveillance indicates rising ceftazidime-avibactam resistance among carbapenem-resistant Klebsiella pneumoniae (CRKP), with sequence type 11 predominating in China. The contribution of blaKPC variants and porin alterations to high-level resistance remains significant. METHODS: We employed whole-genome sequencing and functional analyses to characterise a CZA-resistant ST11 CRKP isolate recovered from a patient without prior exposure to ceftazidime-avibactam or carbapenems. RESULTS: The isolate harboured blaKPC-33 on a non-conjugative plasmid and multiple non-synonymous mut

Repurposing sulfamethoxazole via gold nanocluster modification for antibacterial and anti-biofilm activity against multidrug-resistant Gram-negative bacteria.

OBJECTIVES: The global spread of antimicrobial resistance represents a severe threat to public health, with infections caused by multidrug-resistant Gram-negative bacteria (MDR-GNB) emerging as a major clinical challenge. The clinical utility of traditional antimicrobial agents, such as sulfamethoxazole (SMZ), has become increasingly restricted by the emergence of bacterial resistance. Recent advances in nanomedicine have opened opportunities for 'old drugs, new uses' strategies to improve the antibacterial performance of existing agents. METHODS: We synthesized SMZ-modified gold nanoclusters
