To address the challenges posed by bacterial infection and the persistent oxidative microenvironment in diabetic wounds, a multifunctional intelligent hydrogel dressing (SPC) was developed, comprising sodium alginate (SA) , poly (N-isopropylacrylamide) (PNIPAM) , and an encapsulated photosensitizer. This hydrogel features a dual-crosslinked network and supported by hydrogen bond relay system. The results demonstrate that SPC exhibits unique thermosensitivity, with its lower critical solution temperature (LCST) precisely regulated to 26.8 °C through hydrogen bonding. This thermoresponsive behav

The global spread of multidrug-resistant bacteria poses a serious challenge to effective therapy and public health. The rising resistance to small-molecule antibiotics underscores the limitations of conventional antimicrobial strategies and highlights the urgent need for alternative therapeutic modalities. Lipid nanoparticles (LNPs) have emerged as efficient vehicles for delivering genetic materials, as exemplified by their success in mRNA vaccines. Recent studies suggest that LNPs can also be harnessed to suppress bacterial proliferation and counteract antibiotic resistance through the target

Infected wounds present a formidable challenge to healing due to their complex pathological microenvironment, whereas conventional dressings are often functionally limited and ill-suited to adapt to dynamic wound changes. In this study, based on allantoin-loaded ZIF8 nanoparticles (Alla@ZIF8) and methacrylated quaternary ammonium carboxymethyl chitosan/resveratrol composite hydrogels (RMQCC), we designed and developed an adaptive multifunctional hydrogel system (Alla@ZIF8-Gel). This nanocomposite hydrogel exhibited multiple intelligent features, including pH-responsiveness and morphological ad

Antimicrobial peptides (AMPs) are small, naturally occurring molecules that play a key role in the immune defense of multicellular organisms. They show strong and broad activity against many harmful agents, including bacteria, viruses, fungi, and even cancer cells. Because of their unique ability to target pathogens in multiple ways, AMPs are being studied as promising alternatives to traditional antibiotics-especially important given the growing problem of antibiotic resistance. AMPs can be grouped based on their structure, such as α-helical, β-sheet, loop, or extended forms, and by their sou

BACKGROUND: Therapeutic options for critical infections by carbapenem-resistant Enterobacterales (CRE) are largely limited to polymyxins in India. They have significant toxicity, unpredictable kinetics and prohibitive cost. The objective of the current study was to find the mechanisms of resistance in CRE and to evaluate the in vitro efficacy of potential treatment options.
METHODS: In a prospective study performed during October 2023-March 2024, all Enterobacterales clinical isolates of our laboratory were screened for carbapenem resistance. All CRE isolates were phenotypically and genotypic

Ceftazidime-avibactam (CZA) has revolutionized care for carbapenem-resistant Enterobacterales infections, yet increasing New Delhi metallo-β-lactamase (NDM) prevalence has driven use of CZA plus aztreonam (ATM/CZA). We performed a comprehensive clinical and genomic analysis of ceftazidime-avibactam-resistant Escherichia coli (CZA-R-Ec) collected at a tertiary cancer center (2017-2024) to identify patient- and isolate-level factors associated with reduced susceptibility to ATM/CZA and aztreonam-avibactam (AZA). CZA-R-Ec were isolated from 48 unique patients of whom 28 (58%) had confirmed infect

Piperacillin-tazobactam (TZP) resistance in Klebsiella pneumoniae involves diverse mechanisms with unclear prevalence and phenotypic impact. To elucidate these mechanisms, we analyzed K. pneumoniae clinical isolates resistant to TZP but susceptible to cefotaxime and cefepime. Among 53 isolates, 14 were further studied by MIC testing for TZP, amoxicillin-clavulanic acid (AMC), and ceftazidime (CAZ). Short-read sequencing was performed for all 14 isolates and long-read sequencing for two. Core-genome MLST showed that all were unrelated. Two had a blaOXA-1 gene, one also carrying an ompK35 porin

INTRODUCTION: The global increase in multidrug-resistant (MDR) bacterial infections highlights the urgent need for novel antimicrobial strategies. Antimicrobial peptides (AMPs), such as Dendrocin ZM1 derived from Zataria multiflora Boiss. offer promising therapeutic potential, but face limitations including poor stability and dose-dependent cytotoxicity. We developed a nanoconjugate of Dendrocin ZM1 with carbon quantum dots (CQDs) to enhance antimicrobial efficacy and reduced cytotoxicity.
METHODS: The nanoconjugates were synthesized and characterized using FTIR, XPS, TEM, DLS, and fluorescen

The occurrence of virological failure in a subset of individuals is an inevitable aspect of antiretroviral treatment, and historically has been primarily influenced by suboptimal adherence to oral therapies. The risk of selecting 1- or 2-class human immunodeficiency virus (HIV) drug resistance is influenced by the composition of the regimen, differing significantly depending on the intrinsic barrier to resistance of the regimen. HIV resistance emergence during treatment can be viewed as a regimen-related adverse effect that warrants equal consideration in clinical trials alongside virological

BACKGROUND: Integrase strand transfer inhibitors (INSTIs) are the mainstay of antiretroviral therapy (ART) globally. Virological breakthrough is uncommon but often manifests as low-level viremia, and only 50% of cases have identified drug resistance mutations in the integrase gene. Nonintegrase mutations in the Gag-nucleocapsid protein (NC), envelope glycoprotein (Env), and 3' polypurine tract (3'PPT) have been identified in vitro.
METHODS: Between 2015 and 2021, human immunodeficiency virus type 1 (HIV-1) whole genome sequencing was performed on samples from people with recently acquired HIV

Wound infections significantly impair healing and often result in chronic wounds, burdening healthcare systems substantially. The principal pathogen in such infections is Staphylococcus aureus (S. aureus), which forms biofilms that resist host defences and conventional therapies. To overcome these challenges, recombinant lysostaphin (rLyso) was encapsulated in niosomes using thin-film hydration and evaluated with N-acetylcysteine (NAC) against standard and clinical S. aureus strains. Antibacterial and antibiofilm activities were assessed by determining the minimum inhibitory concentration (MIC
