Helminth infections in grazing ruminants are of major concern for animal welfare and cause substantial economic losses, prompting the widespread use of ivermectin (IVM). The emergence of IVM resistance, driven by complex and poorly understood mechanisms, increasingly compromises treatment efficacy. Drug efflux transporters, particularly P-glycoproteins (PGPs), are suspected to contribute to resistance. Yet, the study of their individual and functional role is hindered by their diversity in nematodes. This study aimed to dissect the role of specific PGPs in mediating IVM resistance. Thus, the C

In recent years, following the COVID-19 pandemic, the emergence of antibiotic-resistant microorganisms has increased. This surge has prompted global research into new biologically active molecules, particularly those focused on combination therapies involving antifungal agents. Synthetic antimicrobial peptides have emerged as promising candidates to address this demand. Therefore, the objective of this study was to assess the synergistic therapeutic effects of the synthetic peptide γ31-45PvD1++ combined with amphotericin B (AmB) on Candida tropicalis and Saccharomyces cerevisiae. This study ai

The adoption of targeted selective treatment (TST) is an alternative for the sustainable management of gastrointestinal nematodes in sheep. TST mitigates the development of anthelmintic resistance by reducing the number of animals drenched while preserving the parasite refugia population. This study analysed the reduction of anthelmintic (AH) usage and assessed the AH cost reduction derived from a TST program compared to the previous whole-flock treatment regimen on a commercial smallholder sheep farm. Data acquisition spanned a 4.75-year period on a smallholder meat sheep farm in Colombo, Par

Insecticides can greatly limit the impacts of cockroaches on indoor environment and human health. Insecticide resistance, however, reduces the efficacy of insecticides and increases insecticide use by both residents and pest control professionals. This study examined the relationship between insecticide cross-resistance and associated gene expression across nine different German cockroach populations from the eastern U.S. Using vial bioassays, we identified significantly correlated resistance between three insecticides from different classes (indoxacarb, lambda-cyhalothrin, fipronil), with pot

BACKGROUND: Blastocystis sp. is a highly prevalent anaerobic intestinal protozoan increasingly associated with gastrointestinal disorders. Its survival within the gut microenvironment depends on metabolic pathways such as glycolysis, where glyceraldehyde-3-phosphate dehydrogenase (GAPDH) plays a central role, alongside interactions with nitric oxide (NO) derived from host defenses and surrounding microbiota. The emergence of metronidazole resistance necessitates exploration of alternative therapeutic strategies.
OBJECTIVE: To evaluate the in vitro effects of metronidazole, metformin, and Alli

BACKGROUND: Resistance to Artemisinin-based combination therapies (ACTs) threatens the backbone of Ghana's success in Malaria control. In addition, malaria remains prevalent in the Central Region, raising concerns about the potential emergence of drug resistance. Mutations in the PfKelch13 gene associated with reduced susceptibility have been reported across Africa and in some parts of Ghana, highlighting the need for molecular surveillance in these endemic settings.
METHOD: Dried blood spots were prepared from whole blood, and genomic DNA was amplified using established Plasmodium-specific p

BACKGROUND: Substandard and falsified (S&F) antimalarial medicines undermine malaria control efforts across sub-Saharan Africa, with an estimated 19% of antimalarials failing to meet quality standards and contributing to 40,000-160,000 preventable deaths annually. ACTs, particularly artemether-lumefantrine (AL), are the cornerstone of malaria treatment, yet their effectiveness is threatened by poor pharmaceutical quality and emerging drug resistance. This study evaluated the pharmaceutical quality of AL circulating in Rwanda's public health supply chain to assess compliance with international

Blue mold caused by Penicillium expansum leads to significant fruit loss to pome fruit packers and raises food safety concerns for fruit processors owing to patulin (PAT) contamination in processed products. The disease is primarily managed through postharvest applications of thiabendazole (TBZ), pyrimethanil (PYR), and fludioxonil (FDL). However, P. expansum has developed resistance to these commonly used fungicides. In 2021 and 2022, high levels of PAT were detected in several processed apple lots in Washington State, raising concerns about a potential link between fungicide resistance and i

Root-knot nematodes (Meloidogyne spp.) cause catastrophic yield losses in global agriculture. This study identified itaconic acid (IA), through comparative metabolomic analysis (the study of small molecules in biological systems), as a key virulence-related metabolite produced by the fungus Trichoderma citrinoviride Snef1910. In vitro assays demonstrated potent nematicidal activity of IA, with an LC50 value of 243.4 mg/liter against J2s of M. incognita and 78.4% egg-hatching inhibition at 800 mg/liter, performance that is comparable with the structural analog trans-aconitic acid. To date, 21 distinct Colletotrichum species have been identified as causal agents of bitter rot on apple fruit. However, whereas bitter rot occurs in nearly all apple-growing regions, the Colletotrichum species responsible for this disease are not globally prevalent and have distinct regional distributions. Because Colletotrichum species can differ in pathogenicity, virulence, and fungicide sensitivities, characterizing the species present in a growing region is essential for effective disease management. In this study, we obtained 130 Colletotrichum isolates from symptomatic fruit collected i

Global temperature increases require the reevaluation of pesticide efficacy under thermal stress conditions; however, most studies have focused on acute high-temperature exposure, neglecting the effects of natural diurnal temperature fluctuations and cumulative acclimation. This study investigated the effect of thermal acclimation during developmental on the sensitivity of Mythimna separata (Walker) (Lepidoptera: Noctuidae) to emamectin benzoate (EB). Larvae were reared under cyclic thermal regimes (25→34/38→25 °C), experiencing daily high-temperature exposures of 2, 4, or 6 hours from the egg

Background: Melanoma management faces the dual challenge of preventing tumor recurrence while ensuring optimal post-surgical wound healing, particularly problematic given melanoma's high recurrence rates and therapeutic resistance. Artesunate (ARS) emerges as a promising multimodal agent with concurrent anticancer, anti-inflammatory, and tissue-regenerative properties. However, its anti-melanoma mechanisms remain incompletely characterized, and clinical translation has been limited by suboptimal pharmacokinetics. Methods: We employed transcriptomic profiling (RNA-seq) to identify novel ARS-reg

Botrytis fruit rot is a major disease affecting strawberry production worldwide, involving several species. In this study, 1,222 isolates were collected from California and Northeastern U.S. strawberry fields to analyze their resistance phenotypes and genotypes. High frequencies of resistance were detected to thiophanate-methyl and pyraclostrobin, whereas lower but increasing frequencies were found to newer fungicides such as succinate dehydrogenase inhibitors (SDHIs). Botrytis isolates from California strawberry fields showed higher resistance frequencies than those from Northeastern states,

Ovarian cancer remains a leading cause of gynecologic cancer mortality, in part due to the persistence of ovarian cancer stem cells (OCSCs) that drive tumor recurrence, metastasis, and drug resistance. Anisomycin, a natural antibiotic derived from Streptomyces coelicolor, has previously been shown to exert antitumor effects, but the mechanisms by which it targets OCSCs remain unclear. In this study, primary human OCSCs were isolated and treated with anisomycin to investigate its biological and molecular effects. Cell proliferation, apoptosis, migration, and colony formation were assessed in vi

BACKGROUND: The resistant rice stem borer (Chilo suppressalis) poses a serious threat to rice yield and food security. For two decades, fipronil was a key insecticide for its control, but its use has been restricted owing to the toxicity of its degradation products to aquatic life and bees. To develop superior alternatives, we structurally modified fipronil-5-amino through acylation, benzylation and carbamation, and evaluated the insecticidal activity of the resulting derivatives against resistant C. suppressalis, as well as their toxicity to zebrafish and honeybees.
RESULTS: Twenty-seven fip
