The salicylic acid receptor NPR1 mediates the polyubiquitination and degradation of the transcription factors ABF3/4 to inhibit drought resistance.

Drought and pathogen infection severely affect plant growth and development. The plant hormones abscisic acid and salicylic acid promote drought and disease resistance, respectively. However, it is largely unknown how plants balance these two stress responses. The salicylic acid receptor NPR1 is required for pathogen resistance and the transcription factors ABF3/4 are required for drought resistance. Here, we show that NPR1 negatively regulates drought resistance by inhibiting ABF3/4 in Arabidopsis. The npr1 mutant is more resistant to drought stress than wild type. Transcriptome analysis reve

Enhancing Antifungal Activity of Nitric Oxide-Releasing Polymers via the Incorporation of Piperine.

As hospital-acquired infections and antibiotic and antifungal resistance increase, there is a need for constant innovation of novel antimicrobial solutions. To this end, the endogenous free-radical gas nitric oxide has been explored, as nitric oxide kills bacteria effectively at low levels without producing significant cytotoxic effects. Biomaterials can be designed to release nitric oxide by incorporating nitric oxide donor molecules such as S-nitrosothiols. Though nitric-oxide-releasing materials show excellent cytocompatibility and antibacterial activity, their efficacy against fungal speci

Vaginal Lactobacillus paragasseri may attenuate fluconazole-resistant phenotypes in the subset of Candida albicans isolates and shows potential as a probiotic adjunct in antifungal therapy.

INTRODUCTION: Candida albicans is the predominant etiological agent of vulvovaginal candidiasis (VVC). Fluconazole (FLU), a first-line azole antifungal agent, has encountered growing resistance among vaginal isolates of C. albicans due to prolonged, high-dose clinical administration. The development of such drug resistance arises from the synergistic interplay of multiple factors, with key mechanisms including enhanced biofilm formation, overexpression of drug efflux pump genes, and structural alterations in target enzymes driven by mutations in the ERG11 gene. Lactobacilli, the dominant comme

Repurposing amlodipine, fluoxetine, and escitalopram as non-antibiotic adjuvants: in vitro enhancement of ciprofloxacin and fluconazole activity against multidrug-resistant bacterial and candida clinical isolates.

INTRODUCTION: Multidrug-resistant (MDR) Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans threaten treatment success, particularly in resource-limited settings. Repurposing licensed, inexpensive non-antibiotic drugs may provide a strategy for enhancing the activity of established antimicrobial agents. METHODS: Clinical isolates from Sana'a, Yemen, and American Type Culture Collection (ATCC) reference strains were tested in vitro with amlodipine, fluoxetine, and escitalopram, individually and in dual or triple combinations with ciprofloxacin or fluconazole. Antimicrobial acti

Novel molecular targets and natural drug candidates for the antifungal therapy of seborrheic dermatitis.

Background: The prevalence of seborrheic dermatitis (SD) involving scalp and non-scalp areas has spread worldwide because of the decreasing susceptibility to azole antifungal drugs and increasing diffusion of the multidrug-resistant fungi of Malassezia species. The antifungal resistance results from structural changes of well-known and new molecular targets and biochemical adaptations of fungi to the standard synthetical treatment. Meanwhile, the research and use of traditional plant products as an alternative therapy strategy for dermatological diseases has increased tremendously over the pas

Antifungal susceptibility and genomic profile associated with biosynthetic gene clusters of bovine mastitis-derived Aspergillus fumigatus DAH1043FM.

The most frequently isolated fungus from mycotic mastitis in cattle is Aspergillus fumigatus, which can cause infection by germinating on the animal's surface to form hyphae that penetrate epithelial and endothelial barriers and initiate mastitis. In this study, the fungal strain DAH1043FM was isolated from the milk of a cow with mastitis and identified as Aspergillus fumigatus by internal transcribed spacer (ITS) region sequencing. Species identity was further confirmed by whole-genome Average Nucleotide Identity (ANI) analysis. A. fumigatus DAH1043FM showed minimum inhibitory concentration (

Susceptibility Profiles Among Sporothrix brasiliensis Genotypes in the Ongoing Zoonotic Sporotrichosis Epidemic in Brazil, Argentina and Paraguay.

Sporothrix brasiliensis is the predominant etiological agent of sporotrichosis in Brazil and has been progressively expanding its geographic distribution to other countries in South America. The human patients infected with S. brasiliensis usually respond favorably to antifungal therapy. However, therapeutic failure may occur, and management may be particularly challenging in cases of severe disease. We recently reported that at least seven S. brasiliensis genotypes circulate across South America. However, their biological and clinical relevance of this genetic diversity remains poorly underst

Genomic and phenotypic characterization of Antarctic Pseudomonas/Aquipseudomonas sp. strains with cold-active antifungal activity against Botrytis cinerea.

Gray mold, caused by the psychrophilic necrotroph Botrytis cinerea, is a leading cause of postharvest losses, and its control is increasingly compromised by the spread of fungicide-resistant field populations and by the failure of conventional biocontrol agents to operate at cold-chain temperatures. Antarctic soils are a rational yet underexploited source of cold-active antagonists. Here we isolated cultivable bacteria from the rhizosphere of Deschampsia antarctica (King George Island) and screened them against B. cinerea at 22 °C and 0 °C. Of 57 bacterial isolates, 44 antagonized the pathogen

Microbial profiles and antimicrobial susceptibility in recurrent and non-recurrent canine otitis externa in Brazil.

BACKGROUND AND AIM: Canine otitis externa is a common inflammatory disorder in which bacterial and yeast proliferation may complicate treatment and contribute to recurrence. In Brazil, limited information is available comparing recurrent and non-recurrent cases while simultaneously characterizing microbial isolates and their antimicrobial susceptibility. This study compared clinical, otoscopic, and cytological findings between recurrent and non-recurrent otitis externa, identified bacterial and yeast isolates, evaluated their in vitro susceptibility to selected fluoroquinolones and antifungal

When the fungus is susceptible, but the patient is not: reading antifungal susceptibility from bench to bedside.

Antifungal susceptibility testing is increasingly available and is central to the management of invasive, refractory, recurrent, and breakthrough fungal infections. However, minimum inhibitory concentrations (MICs) are not self-interpreting results. They measure in vitro antifungal activity under defined laboratory conditions, but clinical outcome is shaped by species identification, method, breakpoint or epidemiological cutoff values, availability, pharmacokinetic and pharmacodynamic exposure, infection site, fungal burden, source control, biofilm formation, and host immunity. Non-classic fun

Harnessing probiotics against candidiasis: a systematic review of experimental and mechanistic evidence.

BACKGROUND: Candida species are among the most important opportunistic fungal pathogens responsible for superficial mucosal infections and life-threatening invasive candidiasis, particularly in immunocompromised individuals. The increasing prevalence of antifungal resistance, biofilm-associated tolerance, and the emergence of multidrug-resistant species such as Candida auris have highlighted the need for alternative or adjunctive therapeutic strategies. Probiotics have emerged as promising microbiome-based interventions owing to their ability to inhibit Candida growth, suppress virulence trait

Disseminated Aspergillus flavus infection presenting as spondylodiscitis: a fatal tuberculosis mimicker in an immunocompetent host - case report and literature review.

INTRODUCTION: Disseminated invasive aspergillosis is a rare but life-threatening fungal infection that predominantly affects immunocompromised individuals. Vertebral involvement is an uncommon manifestation, and diagnosis in immunocompetent patients is particularly challenging because of its nonspecific clinical and radiological features, often resulting in delayed diagnosis and inappropriate therapy. We report a fatal case of disseminated Aspergillus flavus aspergillosis presenting as spondylodiscitis in an immunocompetent host, highlighting the diagnostic challenges and therapeutic implicati

Comparative transcriptome and proteome analysis of resistant and susceptible Talaromyces marneffei clinical isolates reveals a systemic multitiered defense network against azoles.

Talaromyces marneffei is a thermally dimorphic fungus causing life-threatening systemic mycosis in immunocompromised patients. The emergence of azole-resistant clinical isolates, especially those with reduced susceptibility to voriconazole (VOC) and fluconazole (FLC), presents a critical therapeutic challenge. Although target-gene alterations and efflux-related mechanisms have been reported in pathogenic fungi, the broader molecular basis distinguishing high-minimum-inhibitory concentration (MIC) and low-MIC T. marneffei clinical isolates remains incompletely defined. Here, we performed integr

A receptor-like kinase pair integrates extracellular ATP and salicylic acid signaling to activate plant immunity.

Extracellular Adenosine 5'-triphosphate (eATP) is a damage-associated molecular pattern in both plants and animals, playing an important role as a danger signal in response to various environmental stresses. eATP works both independently and in coordination with other signaling pathways. Here, we identify a previously uncharacterized proline-rich extensin-like receptor kinase 15 (PERK15), which interacts with the plant eATP receptor P2K1 and integrates eATP and salicylic acid (SA)-mediated pathogen defense responses. perk15 mutant plants display reduced eATP responses and compromised pathogen

The Antagonistic Effect of 2-Phenylethanol Produced by Serratia liquefaciens Strain JS-239 Against Botrytis cinerea.

Botrytis cinerea is a necrotrophic fungal pathogen responsible for gray mold diseases in economically significant crops. Although conventional fungicides are commonly used to control these diseases, they pose environmental hazards, such as soil and water contamination, harm to nontarget organisms, and ecosystem disruption. Additionally, overuse promotes the development of fungicide-resistant strains, reducing their efficacy. Consequently, finding sustainable alternatives is essential for agriculture. In this study, strain JS-239 was isolated from the Northern Unit of Kettle Moraine State Fores

Multi-Omics Profiling Unveils Antifungal Secondary Metabolites Mediating Epicoccum layuense LQ's Biocontrol Efficacy Against Colletotrichum fructicola.

Colletotrichum fructicola is a significant phytopathogen in both pre- and postharvest stages of fruit development and storage. The development of environmentally friendly biological control agents has attracted increasing research interest. In this study, we characterized a fungal strain (Epicoccum layuense LQ) that strongly inhibits C. fructicola. A potato dextrose broth culture filtrate of strain LQ inhibited the vegetative growth of C. fructicola by approximately 80% at a 1:10 (vol/vol) dilution. Cytological observations revealed that the filtrate disrupted mitosis and cellular polarity dur
