Emergent insights on the spread of antifungal-resistant Trichophyton indotineae dermatophyte: Clonal expansion, adaptability dynamics and human-animal host adaptation.

Trichophyton indotineae is an emergent, clonally expanding, multidrug-resistant dermatophyte that emerged from the Indian subcontinent and has disseminated internationally, causing extensive, treatment-refractory tinea. Its phenotypic resemblance to Trichophyton mentagrophytes makes classical identification unreliable, so molecular and proteomic tools such as Internal Transcribed Spacer (ITS) with High Mobility Group (HMG) discrimination, updated MALDI-TOF and Squalene Epoxidase gene (SQLE) sequencing are essential. Resistance is driven by SQLE point mutations (notably Leu393Phe, Phe397Leu and

Broad-spectrum antifungal activity of C12/C14 alkyl triphenylphosphonium salts (TPP-C12 and TPP-C14) against clinically relevant pathogens.

INTRODUCTION: Human fungal infections affect billions of people and result in more than 2 million deaths every year, however, they have historically been neglected as a cause of infectious disease-related deaths worldwide. Fungal drug resistance has become an increasingly serious problem with the wide use of antifungal drugs and the adaptive evolution of fungi. Resistance to all commonly used antifungal drugs has been reported, and the development of non-traditional antifungal drugs is urgently needed. METHODS: Minimal inhibitory concentrations (MICs) of clinical pathogenic fungi were assesse

Antifungal and anti-biofilm activity of patchouli essential oil (Pogostemon cablin) against oral isolates of Candida albicans.

The Candida albicans species is part of the human microbiota and is associated with different types of infections. It is the most commonly isolated yeast in samples of oral cavity infections. The number of infections attributed to the Candida genus has grown significantly on a global scale and due to the rigorous and improper use of current antimicrobials, antimicrobial resistance has also increased. Pogostemon cablin Benth., popularly known as patchouli, is a medicinal herb of great relevance, with several pharmacological properties reported in the literature. The aim of this study was to eva

Reduced-Resistances Model for Enhanced Drug Permeation via a Solubilizing Receiver Medium: A Mechanistic Study with Hollow Fiber Membranes.

A solubilizing receiver medium has been documented to increase drug flux in vitro, but the mechanisms underlying this effect remain poorly understood. This study investigated these mechanisms and established a mathematical model to describe the increase in apparent permeability. Flow rate experiments were performed to quantify the individual boundary layer and membrane resistances associated with diffusion. The impact of nine solubilizing receiver additives, including surfactants, cyclodextrins, and bovine serum albumin, on the flux of griseofulvin was investigated. The increase in apparent pe

Gim3 buffers and potentiates de novo mutations that affect fluconazole susceptibility in yeast.

Gim3 is an evolutionarily conserved component of the prefoldin chaperone complex, involved in protein folding. We previously found that GIM3 genetically interacts with many de novo mutations in Saccharomyces cerevisiae. Removing GIM3 from mutagenized S. cerevisiae cells significantly affected the fitness effect of mutations. This indicates that Gim3 might change the evolutionary impact of de novo mutations by either buffering (hiding) or potentiating (increasing) their phenotypic effects, depending on the environmental or genetic context. Here, we investigated Gim3's role in shaping the evolut

Galleria mellonella as a versatile model for investigating Candida glabrata virulence and antifungal resistance.

Candida glabrata is an opportunistic fungal pathogen and a leading cause of invasive candidiasis, particularly in immunocompromised patients, where treatment is increasingly compromised by intrinsic and acquired antifungal resistance. Despite lacking morphological plasticity, C. glabrata employs distinct virulence strategies, including adhesin-mediated host colonization, intracellular survival within phagocytes, stress tolerance, iron acquisition and biofilm formation. This review synthesizes current knowledge of C. glabrata virulence and antifungal resistance mechanisms, with a particular foc

A metabolism-specific drug-repurposing screen reveals itraconazole as a potent OXPHOS inhibitor in acute myeloid leukemia.

Targeting mitochondrial oxidative phosphorylation (OXPHOS) enhances the effects of standard chemotherapy and overcomes treatment resistance in preclinical models of acute myeloid leukemia (AML). So far, the few clinically available OXPHOS inhibitors have shown adverse effects or limited potency in clinical trials; therefore, the identification of safe and effective drugs that target mitochondrial metabolism in AML is critical. Here, we performed a high-throughput drug-repurposing screen designed to identify clinically applicable OXPHOS-specific inhibitors through nutrient sensing. We uncover i

Natural product based approaches to overcome Candida glabrata and emerging AMR threats.

The rise of C. glabrata as a serious, multidrug-resistant organism poses a significant and global challenge to the human health. The reasons C. glabrata has developed resistance to standard antifungal drugs, include the activation of efflux pumps, the production of biofilms, and changes in ergosterol biosynthesis. In light of the threat posed by C. glabrata, the potential of phytochemicals as therapeutic alternatives should be considered due to their diverse structures and ability to exhibit more than one type of antifungal activity. This review summarizes advances in the use of plant-based na

G521 is the gatekeeper and a key transmembrane domain contact residue of Candida albicans Cdr1.

UNLABELLED: Candida albicans Cdr1 plays an important role in antifungal drug resistance through the extrusion of structurally diverse substrates. However, the molecular basis of its substrate preference remains unclear. A G521R mutation at the center of transmembrane segment 1 (TMS1) was previously found to impair the efflux of large substrates and to confer resistance to large efflux pump inhibitors, suggesting a role for G521 in regulating substrate and inhibitor access. To explore the structural and functional basis of this phenotype, we analyzed a series of Saccharomyces cerevisiae strains

Comprehensive Insights into Sugar Transporters of Candidozyma auris and Their Roles in Antifungal Resistance.

In Candida species, including Candidozyma auris (formerly Candida auris), overexpression of efflux pumps is a well-established mechanism of antifungal resistance. However, accumulating evidence indicates that impaired drug import may also significantly contribute to reduced antifungal susceptibility. Sugar importers, historically viewed solely as hexose transporters (HGTs), are now emerging as potential indirect modulators of antifungal uptake. Here, we performed a comprehensive inventory and functional analysis of the HGT family in C. auris to assess its contribution to antifungal import. Phy

Aneuploidy, polyploidy and loss of heterozygosity distinguish serial bloodstream isolates of Candida albicans.

The opportunistic pathogen Candida albicans is the leading cause of invasive Candida infections worldwide. Genomic variation between C. albicans clinical isolates complicates efforts to pinpoint novel genetic variants underlying antifungal drug resistance and tolerance. Studies of serial isolates from individual patients are uncommon, limiting our understanding of genomic and phenotypic diversity during invasive Candida infections. We performed comparative genomic analyses of 101 C. albicans bloodstream isolates from 55 patients in the Twin Cities region of Minnesota, including serial isolates

Cell Wall Proteome of Candida albicans Reveals Proteins Associated with Tolerance to Antibiofilm Activity of a Lippia graveolens Kunth Stem Extract.

Candida albicans is an opportunistic fungus capable of forming biofilms that are resistant to conventional antifungal treatments. This study evaluated the antibiofilm activity of an ethanolic extract from Mexican oregano (Lippia graveolens Kunth) stem and its impact on the protein composition of the C. albicans cell wall (CW). The proteomic analysis was restricted to the adherent cells that persisted after treatment, representing the more tolerant subpopulation. First, the biofilm-forming capacity of 18 clinical isolates was characterized. Subsequently, the effect of the Lippia graveolens Kunt
