Identification of physiological races of Puccinia striiformis f. sp. tritici and molecular docking of some biological treatments as prospective fungal inhibitor candidates in wheat.

Stripe rust caused by Puccinia striiformis f. sp. tritici (Pst), represents one of the most significant challenges to the world's wheat production. This study aimed to investigate five aggressive physiological races of Pst impacting significant wheat cultivars in Egypt's northern Delta, which are essential for breeding programs. Moreover, the study evaluated the antifungal activity and mechanisms of action of chitosan nanoparticles, Trichoderma harzianum, and Sargassum latifolium against Pst using molecular docking analysis. During the 2024 growing season, an increase in the virulence of newly

Genetic confirmation of terbinafine resistance in Trichophyton rubrum mediated by the squalene epoxidase Leu393Phe mutation via targeted gene replacement.

Terbinafine (TBF) is a first-line allylamine antifungal used to treat dermatophytosis caused by Trichophyton species, targeting squalene epoxidase (SQLE). The recent emergence of TBF-resistant Trichophyton rubrum (T. rubrum) strains represents a significant global clinical challenge. Although the SQLE Leu393Phe substitution is a commonly reported resistance mechanism in Japan, direct genetic evidence demonstrating its effect when introduced into a T. rubrum recipient strain has been lacking. In this study, we established the causal relationship between the SQLE Leu393Phe substitution and TBF r

Emerging antifungal resistance: an updated review on current scenario, pathogens involved and tackling strategies.

Antifungal resistance (AFR), an emerging and significant threat to humanity, receives far less attention than antibacterial resistance. AFR management is plagued by rising incidence in both immunocompromised and immunocompetent populations, zoonotic infections, limited fungal diagnostics, limited antifungal armamentarium with even fewer in pipeline, cross-resistance between environmental fungicides and clinically used antifungals, lack of strong antifungal stewardship programs, limited applications of novel strategies for human AFR, to name a few. In this review, we compile available informati

Evaluation of the Antifungal Activity of Z24 Against Botrytis cinerea.

The gray mold disease of plants caused by Botrytis cinerea leads to severe economic losses. Our previous studies showed that the neocryptolepine derivative Z24 exhibits excellent antifungal activity, particularly against B. cinerea. To assess the application value of Z24, this study further evaluated its antifungal effects. The results of this study show that N-methyl-2-pyrrolidone, dimethyl sulfoxide, and cinnamon oil can all serve as solvents for Z24. The EC50 value of Z24 against B. cinerea was 0.8466 μg/ml, which was significantly lower than that of neocryptolepine, and there was no cross-

Deciphering subcellular localization-dependent functions of Hog1 MAPK in Cryptococcus neoformans.

The Hog1 mitogen-activated protein kinase regulates stress adaptation, sexual differentiation, and virulence traits by dynamically shuttling between the cytoplasm and nucleus in the fungal pathogen Cryptococcus neoformans, a leading cause of fatal fungal meningoencephalitis worldwide. However, how spatial partitioning governs Hog1 pathway specificity remains poorly understood. Here, we generated genetically engineered strains expressing localization-restricted Hog1 variants including membrane-tethered and constitutively nuclear, and we compared their phenotypic traits with those of a fully fun

A Chickpea NCR Peptide Suppresses Postharvest Alternaria alternata Infection by Disrupting Fungal Membranes, Mitochondria and Respiration.

Postharvest fungal diseases cause substantial losses in horticultural crops and are commonly managed using chemical fungicides, which raise concerns regarding resistance development and environmental impact. Plant-derived antifungal peptides represent a promising alternative for sustainable disease control. Here, we evaluated the antifungal activity and mode of action of three nodule-specific cysteine-rich (NCR) peptides-NCR044, NCR192 and NCR13_PFV2-against major postharvest fungal pathogens. Among the peptides tested, NCR13_PFV2 exhibited the strongest inhibitory activity against Alternaria

Biosynthesized selenium nanoparticles increase soybean resistance to root rot by recruiting beneficial microbes and reprogramming host metabolism.

BACKGROUND: Soybean root rot, primarily caused by Fusarium species, is a devastating soil-borne disease that severely threatens global soybean production. Conventional chemical controls are problematic due to their environmental impact and the risk of fostering pathogen resistance. There is therefore an urgent need to develop sustainable and eco-friendly alternative strategies. RESULTS: This study demonstrated that selenium nanoparticles (SeNPs) biosynthesized by Bacillus subtilis ZY56 significantly increased soybean resistance to F. oxysporum. Characterization confirmed that the obtained SeN

Role of C17 fengycin B from Bacillus subtilis DB9011 in enterotoxigenic Escherichia coli inhibition.

Enterotoxigenic Escherichia coli (ETEC) is a leading cause of post-weaning diarrhea in piglets, and the rise of antimicrobial resistance underscores the urgent need for antibiotic alternatives in swine production. Bacillus species are promising candidates owing to their ability to produce antimicrobial secondary metabolites, particularly cyclic lipopeptides such as surfactins, iturins, and fengycins. Although Bacillus lipopeptides are known to exhibit antibacterial activity against E. coli, the specific compounds responsible for the inhibition of ETEC in probiotic-relevant Bacillus strains rem

Single Amino Acid Change in the FvHIK1 Protein Associated with Fludioxonil Resistance in Fusarium verticillioides.

Fusarium stalk rot, caused by Fusarium verticillioides, is a major disease affecting maize production. Currently, chemical control serves as the primary management strategy, and our preliminary studies have demonstrated that fludioxonil exhibits excellent antifungal activity against F. verticillioides. However, the resistance risk and potential resistance mechanisms have not been fully established. In this study, we investigated four resistant laboratory isolates of F. verticillioides that had fludioxonil EC50 values ranging from 43.3 to 53.7 μg/ml. Evaluation of their biological characteristi

Engineered pectin-coated copper oxide nanocarriers for controlled fungicide release: a mechanistic exploration of fungal pectinase-mediated delivery and its role in advancing sustainable agriculture.

Stimuli-responsive agrochemical formulations hold significant promise for advancing the sustainability of modern agriculture. In this study, we developed pectinase-responsive controlled-release fungicide formulations by covalently grafting pectin onto tricyclazole-loaded mesoporous copper oxide nanoparticles. While copper oxide nanoparticles (CuONPs) alone exhibited inherent antifungal activity and performed effectively against Aspergillus niger, a major phytopathogenic fungus, the TCP formulation proved substantially more potent due to the combined action of CuONPs and tricyclazole. Although

Physiologically Based Pharmacokinetic Model to Predict Drug-Drug Interactions With the Antibody-Drug Conjugate Trastuzumab Deruxtecan.

The topoisomerase I inhibitor payload (DXd) released from the human epidermal growth factor receptor 2-targeted antibody-drug conjugate (ADC) trastuzumab deruxtecan (T-DXd) is eliminated by hepatic uptake via OATP1B1/3, metabolism by CYP3A, biliary excretion via P-glycoprotein and breast cancer resistance protein, and urinary excretion. In a clinical drug-drug interaction (DDI) study between T-DXd and ritonavir (a strong cytochrome P450 [CYP] 3A/organic anion transporting polypeptide 1B inhibitor) or itraconazole (a strong CYP3A inhibitor), a 1.2-fold increase in DXd exposure was observed. The Role of the Mediator Complex in Modulating CjXDR1-Induced Multidrug Resistance and Virulence in Clarireedia jacksonii.

The Mediator complex is a central regulator of gene expression in pathogenic fungi; however, its composition and functions remain uncharacterized in Clarireedia jacksonii, a major fungal pathogen in turfgrass. In this study, we systematically analyzed the Mediator complex in C. jacksonii and identified its key role in transcriptional activation. Functional characterization of 10 knockout mutants revealed distinct roles for Mediator subunits in cell growth, fungicide resistance, pathogenicity, and stress responses. Notably, Med1, Med31, Med15, and Cdk8 were essential for pathogenicity, and subu

Evaluation of in vitro and in vivo antifungal activities of a deuterated tetrazole-based CYP51 inhibitor.

Invasive fungal infections represent a significant global health threat, and increasing prevalence of azole-resistant fungal pathogens has severely undermined the efficacy of current antifungal therapies. Tetrazole-based CYP51 inhibitors have recently emerged as promising next-generation antifungal agents, largely due to their reduced potential for drug-drug interactions. In our previous study, tetrazole compound 15, containing a pyrazole side chain, exhibited potent in vitro antifungal activity but suffered from poor metabolic stability, potentially limiting its in vivo efficacy. Assessing antifungal stewardship competency among pharmacy students: key findings and implications for clinical practice.

BACKGROUND: Antifungal resistance (AFR) is a significant public health concern. It restricts the available treatments for fungal infections, making them more difficult to possibility of treatment and increasing the risk of serious consequences. Pharmacists could significantly promote the rational use of antifungals, lessen their overuse, and support public health initiatives to stop the spread of resistant fungal infections by participating in antifungal stewardship (AFS) programs. Hence, the current study aimed to evaluate undergraduate pharmacy students' knowledge, attitudes and practices to
