Antimicrobial peptides (AMPs) are small, naturally occurring molecules that play a key role in the immune defense of multicellular organisms. They show strong and broad activity against many harmful agents, including bacteria, viruses, fungi, and even cancer cells. Because of their unique ability to target pathogens in multiple ways, AMPs are being studied as promising alternatives to traditional antibiotics-especially important given the growing problem of antibiotic resistance. AMPs can be grouped based on their structure, such as α-helical, β-sheet, loop, or extended forms, and by their sou

BACKGROUND: Therapeutic options for critical infections by carbapenem-resistant Enterobacterales (CRE) are largely limited to polymyxins in India. They have significant toxicity, unpredictable kinetics and prohibitive cost. The objective of the current study was to find the mechanisms of resistance in CRE and to evaluate the in vitro efficacy of potential treatment options.
METHODS: In a prospective study performed during October 2023-March 2024, all Enterobacterales clinical isolates of our laboratory were screened for carbapenem resistance. All CRE isolates were phenotypically and genotypic

Ceftazidime-avibactam (CZA) has revolutionized care for carbapenem-resistant Enterobacterales infections, yet increasing New Delhi metallo-β-lactamase (NDM) prevalence has driven use of CZA plus aztreonam (ATM/CZA). We performed a comprehensive clinical and genomic analysis of ceftazidime-avibactam-resistant Escherichia coli (CZA-R-Ec) collected at a tertiary cancer center (2017-2024) to identify patient- and isolate-level factors associated with reduced susceptibility to ATM/CZA and aztreonam-avibactam (AZA). CZA-R-Ec were isolated from 48 unique patients of whom 28 (58%) had confirmed infect

Piperacillin-tazobactam (TZP) resistance in Klebsiella pneumoniae involves diverse mechanisms with unclear prevalence and phenotypic impact. To elucidate these mechanisms, we analyzed K. pneumoniae clinical isolates resistant to TZP but susceptible to cefotaxime and cefepime. Among 53 isolates, 14 were further studied by MIC testing for TZP, amoxicillin-clavulanic acid (AMC), and ceftazidime (CAZ). Short-read sequencing was performed for all 14 isolates and long-read sequencing for two. Core-genome MLST showed that all were unrelated. Two had a blaOXA-1 gene, one also carrying an ompK35 porin

INTRODUCTION: The global increase in multidrug-resistant (MDR) bacterial infections highlights the urgent need for novel antimicrobial strategies. Antimicrobial peptides (AMPs), such as Dendrocin ZM1 derived from Zataria multiflora Boiss. offer promising therapeutic potential, but face limitations including poor stability and dose-dependent cytotoxicity. We developed a nanoconjugate of Dendrocin ZM1 with carbon quantum dots (CQDs) to enhance antimicrobial efficacy and reduced cytotoxicity.
METHODS: The nanoconjugates were synthesized and characterized using FTIR, XPS, TEM, DLS, and fluorescen

Pseudomonas aeruginosa is a major cause of health care-associated infections, particularly in intensive care units (ICUs). To the best of our knowledge, this is one of the rare studies reporting the prevalence and antimicrobial resistance patterns of P. aeruginosa in a Tunisian ICU. This retrospective study included all P. aeruginosa strains isolated from ICU patients in Sfax, Tunisia, between 2017 and 2024. A total of 1,126 nonredundant P. aeruginosa strains were obtained from 767 patients. P. aeruginosa strains accounted for 21.1% of all clinical isolates in the ICU. The majority of strains

Klebsiella pneumoniae has a long history of causing infections as an opportunistic pathogen. There is an upsurge in infections caused by K. pneumoniae, mainly due to its two pathotypes, classical K. pneumoniae (cKP) and hypervirulent K. pneumoniae (hvKP). Various infections associated with these pathotypes include pneumonia, urinary tract infections, bloodstream infections, and pyogenic liver abscesses. This review provides an update on the pathogenicity, virulence factors, mechanisms of virulence, emergence and dissemination of multidrug resistance, as well as the convergence of virulence and

Despite decades of research, current understanding of the spectrum of targets bound by kinase inhibitors remains incomplete. This complicates mechanism of action studies, drug repurposing, and development of new therapies. Here, we describe kinome-wide profiling of an optimal kinase library (OKL) comprising 192 small molecules selected based on stage of clinical development, chemical diversity, and target coverage. Our results show that polypharmacology is widespread and independent of regulatory approval. The generally understood ("assigned") targets of approved molecules are not necessarily

Proteus mirabilis is an opportunistic pathogen frequently associated with catheter-associated urinary tract infections (CAUTIs), where robust crystalline biofilms contribute to chronicity, antimicrobial tolerance, and recurrence. The rising prevalence of multidrug-resistant (MDR) P. mirabilis has prompted interest in bacteriophage therapy as an alternative to conventional antibiotics. In this study, we isolated a lytic bacteriophage, vB_PmiM_ZX7, from sewage using a highly virulent clinical strain, P. mirabilis YV2, as the host. vB_PmiM_ZX7 displayed a broad host range among MDR P. mirabilis i

Candida albicans is ubiquitous on human mucosal surfaces and easy to cause infectious diseases. The widespread use of antifungal drugs has led to the increasing drug resistance of C. albicans, posing a significant challenge to clinical treatment. Berberine hydrochloride (BBH), a quaternary ammonium alkaloid isolated from Coptidis Rhizoma, exhibits broad-spectrum antifungal activity. This study explored the antifungal effect and mechanism of BBH combined with fluconazole (FLC) against drug-resistant C. albicans. Results showed that BBH could enhance the antifungal effect of FLC against the drug

The SARS-CoV-2 public health challenges have highlighted the urgent need for coronavirus-targeting life-saving therapeutics. Given the emergence of drug-resistant strains, the development of antivirals against viral proteins beyond the commonly targeted main protease or RNA-dependent RNA polymerase is critical. The SARS-CoV-2 nonstructural protein 13 (nsp13) is a highly conserved RNA helicase and an essential component of the viral replication-transcription complex (RTC). It unwinds double-stranded RNA to facilitate viral transcription and replication, making it a strong target for drug develo
