Polysaccharide-mediated modulation of gut microbiota and lysine lactylation: implications for tumor microenvironment remodeling and cancer immunotherapy.

Despite the revolutionary clinical benefits of immune checkpoint inhibitors (ICIs), their therapeutic efficacy is largely hampered by the immunosuppressive tumor microenvironment (TME) as well as primary and acquired drug resistance. Natural polysaccharides serve as promising immunomodulators to boost anti-tumor immunity, yet their systematic regulatory network remains poorly characterized. Accumulating evidence demonstrates that gut microbiota and its fermentative metabolites, including short-chain fatty acids and lactate, govern systemic immune surveillance and determine patient responsivene

Targeting SLC1A5-mediated glutamine metabolism overcomes PARP inhibitor resistance in ovarian cancer.

Poly(ADP-ribose) polymerase inhibitors (PARPi), such as olaparib, have significantly improved outcomes in ovarian cancer (OC); however, therapy resistance remains a major clinical challenge. Here, we showed that glutamine metabolism was upregulated in PARPi-resistant OC cells and identified the glutamine transporter SLC1A5 as a key mediator of this metabolic adaptation. Inhibition of SLC1A5 increased the sensitivity of OC cells to olaparib both in vitro and in vivo. Mechanistically, we demonstrated that the deubiquitinating enzyme USP14 directly interacted with and stabilized SLC1A5 by removin

A Graphene Oxide-Doped CuMOF Nanoplatform Enhances Chemodynamic Therapy and Suppresses CYP2J2-Mediated Linoleic Acid Metabolism in Castration-Resistant Prostate Cancer.

PURPOSE: Castration-resistant prostate cancer (CRPC) responds poorly to current therapies. A tumor microenvironment-responsive nanoplatform was developed to couple chemodynamic therapy with modulation of oncogenic lipid metabolism. METHODS: A graphene oxide-doped copper-based metal-organic framework (GO-CuMOF) was prepared by ultrasonicating solvothermally synthesized CuMOF with graphene oxide, and was characterized physicochemically. Its generation of hydroxyl radicals (•OH) was confirmed by a methylene blue degradation assay. Antitumor activity was evaluated in DU145 and PC3 cells and in a

1,2,3-Triazole-Indole Derived Hybrids: Recent Progress and Therapeutic Perspectives for Cancer Treatment.

1,2,3-Triazole and indole-derived scaffolds are privileged nitrogen-containing heterocyclic pharmacophores that form abundant non-covalent interactions with tumor-related biomacromolecules. Molecular hybridization that integrates these two scaffolds into a single molecular scaffold yields 1,2,3-triazole-indole-derived hybrids, which can mitigate intrinsic drawbacks of single-heterocycle anticancer small-molecule agents, such as insufficient target selectivity, unsatisfactory pharmacokinetic performance, and acquired multidrug resistance. This review summarizes research advances on 1,2,3-triazo

Protection of Bevacizumab From Silicone Oil-Induced Inactivation by Liposomal Encapsulation.

PURPOSE: Silicone oil endotamponades in retinal surgery induce aggregation and thus deactivate antibody drug formulations. Liposomes represent an approach to protect and deliver these drugs when administered intravitreally; however, their interactions with silicone oil endotamponades and their influence on encapsulated antibody agents have never been investigated. METHODS: Bevacizumab was encapsulated in liposomes using dual centrifugation. Size, polydispersity index, zeta potential, and ultrastructure were characterized using dynamic light scattering, laser Doppler electrophoresis, and elect

Gut microbiome-shaped chemotherapy responses across cancer types: mechanisms of resistance, biomarkers, and targeted interventions.

Cytotoxic chemotherapy remains an important component of multimodal treatment for many solid tumors, but resistance and dose-limiting toxicities continue to constrain clinical benefit. The gut microbiome can influence chemotherapy response through microbial drug biotransformation, metabolite signaling, intestinal-barrier integrity, systemic immune regulation, and tumor-cell stress and death pathways. Across solid tumors, the evidence is strongest in colorectal cancer, where clinical associations are supported by increasingly detailed mechanistic and interventional studies; evidence in many ext

Nano-/Nanobiotechnology-Augmented Pyroptosis in the Treatment of Triple-Negative Breast Cancer: An Updated Review.

Triple-negative breast cancer (TNBC), a breast cancer subset defined by the absence of estrogen receptor, progesterone receptor, and human epidermal growth factor receptor 2 expression, faces persistent clinical challenges including narrow first-line treatment choices and inferior long-term survival rates. Immune checkpoint blockade has transformed clinical management across multiple malignancies, yet unselected TNBC patients exhibit only modest and heterogeneous clinical responses. This therapeutic resistance is primarily attributed to an immune-excluded tumor microenvironment (TME) with supp

Pentahydroxyflavone and Metformin Mitigate Diabetes-Induced Hepatic Dysfunction in Gamma-Irradiated Rats Through the AMPK-SREBP-1c-SCD1 Pathway.

Diabetes mellitus (DM) is a common complication among cancer patients, potentially worsening the adverse effects of radiation therapy. This study aimed to evaluate whether pentahydroxy flavone (PHF) and Metformin (MET) could mitigate liver damage associated with streptozotocin (STZ)-induced diabetes in rats exposed to gamma irradiation (IR). Diabetes was induced in rats by a single dose of STZ (35 mg/kg) and exposure to a cumulative IR dose (4 Gy). Over one month, each treatment group received either PHF (50 mg/kg) or MET (200 mg/kg). Combined treatment with PHF and MET improved lipid profiles

Exploring Non-Covalent Pin1-PROTACs Insights Into Degradation Efficiency in Multiple Myeloma Cell Line.

Pin1 is a peptidyl-prolyl isomerase involved in key signalling pathways and upregulated in different cancers, including Multiple Myeloma (MM), where it is linked to poor prognosis/drug resistance. Despite its therapeutic relevance, Pin1 remains challenging to target with conventional inhibitors thereby motivating interest in Targeted Protein Degradation (TPD) strategies. All reported Pin1-PROTACs rely on covalent Pin1 ligands, leaving non-covalent degraders largely unexplored. Since PROTACs (PROteolysis TArgeting Chimeras) act catalytically/reversibly, non-covalent designs are attractive. Here

RCCD1-JMJD5 coordinate histone demethylation to promote platinum resistance in non-small cell lung cancer via FN1-mediated epithelial-mesenchymal transition.

Chemoresistance remains a major obstacle to effective treatment of non-small cell lung cancer (NSCLC), yet the epigenetic mechanisms underlying platinum resistance remain incompletely understood. Here, we show that Jumonji C domain-containing protein 5 (JMJD5) promotes platinum resistance in NSCLC cells and xenograft tumors while inducing a slow-proliferative, mesenchymal-like phenotype characterized by enhanced migration, invasion, spheroid formation, and epithelial-mesenchymal transition (EMT). Mechanistically, JMJD5 interacted with RCC1 domain-containing protein 1 (RCCD1), and cisplatin tre

A Dual-Action CXCR4-Targeted Peptide-Drug Conjugate Engineered to Enhance Antitumor Efficacy and Mitigate Myelosuppression in Pancreatic Cancer.

Gemcitabine (GEM) is a first-line therapeutic option for pancreatic cancer; however, it has low efficacy due to rapidly developed drug resistance and severe dose-limiting myelosuppression. To enhance its therapeutic effect, this study developed a novel peptide-drug conjugate using a CXCR4 antagonistic peptide as the targeting head and gemcitabine as the drug payload, based on the characteristics of pancreatic tumor cells highly expressing CXCR4 that mediates immunosuppression and tumor progression through interaction with its specific ligand CXCL12. The therapeutic effect of the conjugate (P12

Glucocorticoid Receptor-Mediated Engineered Cancer Stem Cell-Led Therapy and Inhibition of Recurrence in Colorectal Cancer.

Dysregulated wnt/β-catenin signaling cascades drive self-renewal, proliferation, differentiation, and therapeutic resistance in cancer stem cells (CSCs), thus contributing to their tumor aggression and poor clinical outcomes. Earlier, we demonstrated that a glucocorticoid receptor (GR)-targeted liposomal formulation carrying dexamethasone (Dex) and the wnt/β-catenin inhibitor FH535 (D1XFH) selectively sensitizes colorectal cancer (CRC) cells and suppresses tumor growth. Herein, we investigate the immuno-therapeutic potential of D1XFH, especially within colorectal CSCs and against tumor recurre

A V-type starch-lycopene complex with enhanced cis-lycopene content alleviates hyperglycemia and glycogen fragility in type 2 diabetic db/db mice via gut microbiota.

Type 2 diabetes mellitus (T2DM) is a major global health challenge. Resistant starch type 5 (RS5), formed by complexing starch with guest components into V-type crystalline structures, represents a promising approach for glycemia control. Native lycopene has anti-diabetic potential but limited bioactivity. Here we developed a V-type corn starch-lycopene RS5 complex (CS-LYC) and evaluated its effects in T2DM db/db mice, focusing on glycemic control and gut microbiota-liver glycogen connection. CS-LYC contained 26.07% resistant starch and 11.8 mg/g lycopene, with complexation promoting isomeriza

Protein arginine methyltransferase crosstalk: decoding the synergistic-antagonistic axis in cancer therapy.

BACKGROUND: Protein arginine methyltransferases (PRMTs) are abnormally expressed in various tumors and participate in multiple tumorigenic processes, including proliferation, metastasis, drug resistance, and metabolic reprogramming. However, in recent years, with the in-depth exploration of the functions of PRMTs and their associated tumorigenic mechanisms, the complex crosstalk relationships among different PRMTs have been increasingly highlighted. Different PRMTs may not only exhibit synergistic tumor-promoting effects but also demonstrate antagonistic effects in certain tumor biological fun
