Danshen Injection for 5-Fluorouracil-Resistant Gastric Cancer: Mitochondrial Apoptosis and Enhanced Drug Activity.

Gastric cancer ranks among the leading causes of cancer-related death worldwide, and the progressive emergence of 5-fluorouracil (5-FU) resistance represents a critical barrier to the efficacy of conventional chemotherapy, particularly 5-FU-based regimens. Danshen injection (DI), a clinically approved intravenous preparation derived from Salvia miltiorrhiza, has demonstrated preliminary antineoplastic activity; however, the mechanistic basis underlying its reversal of 5-FU resistance in gastric cancer has not been systematically defined. This study aimed to evaluate the antitumor efficacy of D

Ferroptosis in cancer: molecular mechanisms, tumour microenvironment, therapeutic resistance, and clinical translation.

Ferroptosis is a distinct form of regulated cell death driven by iron-dependent lipid peroxidation and oxidative membrane damage, and it differs fundamentally from apoptosis, necroptosis, and pyroptosis in its molecular mechanisms and execution pathways. Increasing evidence indicates that ferroptosis plays a pivotal role in cancer biology by regulating tumour initiation, progression, metastasis, therapeutic resistance, and interactions within the tumour microenvironment (TME). Dysregulation of iron metabolism, lipid peroxidation, amino acid metabolism, and antioxidant defence systems, includin

Targeting Cancer-Associated Fibroblasts with Small-Molecule Inhibitors: Molecular Mechanisms and Therapeutic Potential.

The multifaceted role of cancer-associated fibroblasts (CAFs) in orchestrating extracellular matrix remodeling, immune evasion, and therapeutic resistance positions these stromal cells as critical yet underexploited nodes in the tumor microenvironment (TME). While small-molecule agents offer practical advantages over biologics for stromal modulation, a systematic understanding of their mechanistic diversity and translational barriers remains incomplete. To address this gap, this review consolidates current knowledge on the principal signaling axes-TGF-β/STAT3, NF-κB, Notch, Hedgehog, PI3K/AKT,

Auranofin Potentiates Cisplatin Response in the Setting of Context-Dependent NOTCH-Associated Signaling States in Endometrial Cancer.

UNLABELLED: Therapeutic resistance remains a major challenge in advanced and recurrent endometrial cancer. Aberrant NOTCH signaling has been associated with aggressive tumor behavior and therapeutic resistance across multiple malignancies, yet its therapeutic significance in endometrial cancer remains incompletely defined. We investigated whether auranofin (AuR), a pleiotropic gold-containing compound that inhibits thioredoxin reductase and can disrupt RBPJ chromatin occupancy, alters platinum responsiveness in endometrial cancer models. Upper-quartile NOTCH3 copy number was associated with po

Multi-omics profiling reveals coordinated metabolic and immuno-inflammatory remodeling in a high-fat diet and single-dose streptozotocin-induced mouse model of diabetic cardiomyopathy.

BACKGROUND: Diabetic cardiomyopathy (DCM) is a major myocardial complication of type 2 diabetes and a contributor to heart failure. However, coordinated remodeling across the cardiac transcriptome, proteome and metabolome in DCM remains incompletely defined. METHODS: Type 2 diabetes was induced in mice by a high-fat diet combined with single-dose streptozotocin. DCM was characterized by echocardiography, histology, transmission electron microscopy, and systemic metabolic and inflammatory assessments. Left ventricular myocardium underwent RNA sequencing, quantitative proteomics and untargeted

PD-L1 in Merkel Cell Carcinoma: From Tumor Microenvironment to Pathological Interpretation.

Merkel cell carcinoma (MCC) is a rare, aggressive cutaneous neuroendocrine carcinoma in which Merkel cell polyomavirus (MCPyV) oncoproteins and ultraviolet-induced mutations provide distinct sources of tumor immunogenicity. This narrative review examines the biological determinants, immunohistochemical assessment, and prognostic and therapeutic relevance of programmed death ligand 1 (PD-L1) in MCC, linking tumor immunobiology to pathological interpretation. PD-L1 expression on tumor and immune cells is often concentrated at tumor-immune interfaces, consistent with interferon-γ-driven adaptive

Anti-Glioma Agents and Hyperbaric Oxygen for Prospective Therapeutic Approaches.

Adjuvant treatments for IDH-wild-type adult high-grade gliomas (HGGs) are being explored to improve therapeutic efficacy. Tumor hypoxia poses a major therapeutic challenge. In this narrative review, we discuss the potential of hyperbaric oxygen to counteract hypoxia and therapeutic resistance in HGG, with particular emphasis on glioblastoma. We delineate the pros and cons of combining treatment with hyperbaric oxygen (HBO) with established and experimental treatments for glioblastoma. HBO has been investigated as an adjuvant treatment alongside radiotherapy and chemotherapy, including new inve

Biomarkers of subclinical cholestasis predict cancer immunotherapy outcomes.

In a recent paper published in Cancer Cell, we identified subclinical cholestasis as an accessible imprint of gut dysbiosis predicting resistance to immune checkpoint blockade. Our work connects γ-glutamyl transferase, bile-acid remodeling, and MAdCAM-1 loss to compromised antitumor immunity, opening avenues for novel biomarker-guided microbiota interventions.

The multifaceted role of 3-methyladenine in cancer therapy.

3-Methyladenine (3-MA), an early autophagy inhibitor, blocks autophagosome formation mainly by inhibiting class III phosphatidylinositol 3-kinase (PI3K) vacuolar protein sorting 34, while its concurrent inhibition of class I PI3K can produce opposing effects on autophagic flux depending on nutrient status. This Review examines the context-dependent roles of 3-MA across cancer types. When autophagy serves a pro-survival function, 3-MA-mediated inhibition tends to attenuate this protective response, thereby potentiating the cytotoxicity of chemotherapeutics, targeted drugs, and natural compounds

MAP2-mediated integrin dysregulation reprograms cancer cell plasticity in hepatocellular carcinoma.

Targeting tumor cell plasticity is central to overcome drug resistance and metastasis. Through lineage-tracing and lineage-ablation studies, our team has previously shown hepatocellular carcinoma (HCC) cells marked by CD133 to represent a functional subset displaying dedifferentiated status with stemness traits, yet there are limited therapies effective for eradicating such resilient cell populations. Here, we explore the therapeutic potential of repurposing an existing drug estramustine phosphate (EMP) for targeting HCC plasticity through inhibiting MAP2, a gene uniquely enriched in CD133+ 'H

The RNA-binding protein La/SSB is associated with HNSCC progression and TFAP2C/FSCN1-linked transcriptional regulation.

Head and neck squamous cell carcinoma (HNSCC) is characterized by aggressive progression, frequent therapeutic resistance, and poor clinical outcomes. Although the RNA-binding protein La/SSB is aberrantly expressed in multiple malignancies, its functional and mechanistic role in HNSCC remains incompletely understood. Here, by integrating transcriptomic and chromatin accessibility profiling with comprehensive in vitro and in vivo analyses, we identify La/SSB as a putative regulator associated with HNSCC progression and cisplatin (CDDP) resistance. La/SSB was markedly upregulated in HNSCC tissue

Comparative anthelmintic activity of artemisinin, dihydroartemisinin, and artesunate against sheep gastrointestinal nematodes: in vitro evaluation and exploratory β-tubulin molecular modeling.

BACKGROUND: Gastrointestinal nematode infections are an important constraint on small-ruminant production, and increasing anthelmintic resistance has intensified the search for alternative therapeutic compounds. Artemisinin and its derivatives have demonstrated activity against several helminth species, although comparative information regarding their effects on gastrointestinal nematodes remains limited. This study compared the in vitro anthelmintic activity of artemisinin (ART), dihydroartemisinin (DHA), and artesunate (ARTE) and explored β-tubulin as a candidate molecular target using compl

Effective bio-activities of the metal nanoparticles mycosynthesized by the plants' fungal endophytes.

Fungal endophytes act as bio-nano factories and are favored over bacteria and plant extracts, as they secrete large quantities of proteins and enzymes, facilitating stable, efficient, and economic nanoparticle synthesis. This review highlights the advantages of plant-associated fungal endophytes as bio-factories for metal nanoparticles (MNPs) synthesis and critically examines the biological activities and potential applications of these MNPs in human health, food technology, and agriculture. Compared to chemical synthesis, biogenic MNPs are more resistant to aggregation due to the involvement

Distinct Mitochondrial Recovery Dynamics Following Taxol Removal in Triple-Negative Breast Cancer Cells.

Drug resistance in TNBC is associated with metabolic plasticity, yet little is known about how mitochondrial and cellular states evolve after chemotherapeutic withdrawal. We used optical redox imaging (ORI) microscopy and complementary functional measurements to characterize post-Taxol recovery in two TNBC cell lines with different Taxol sensitivities. Whole-dish nuclear counting following 48 h Taxol treatment confirmed that HCC1806 cells responded at lower Taxol concentrations than MDA-MB-436 cells. Recovery was subsequently monitored following extracellular Taxol withdrawal using mitochondri

Redox Storm in the Tumor Microenvironment (ΤΜΕ): Multifaceted Redox Injury and Immunometabolic Reprogramming Induced by Immune Checkpoint Blockade in B16F10 Melanoma: An Ultrastructural Study of the Peritumoral Interface.

Immune checkpoint blockade (ICB) has revolutionized melanoma treatment, yet the in vivo ultrastructural consequences within the tumor microenvironment (TME) remain poorly characterized. This study investigates the morphological alterations induced by ICB at the peritumoral interface through a "redox storm" paradigm. B16F10 melanoma-bearing mice were treated with anti-CTLA-4, anti-PD-1, or their combination, and evaluated by transmission electron microscopy (TEM) at Days 20, 30, and 40; quantitative morphometric analyses were performed for comparative purposes. Acquired Chemoresistance in Colorectal Cancer: Therapy-Induced Signalling Adaptation, Cellular Plasticity and Mechanism-Guided Therapeutic Strategies.

The effectiveness of chemotherapy in colorectal cancer (CRC) is significantly reduced by acquired chemoresistance to 5-fluorouracil (5-FU), oxaliplatin and irinotecan. This structured narrative review synthesises mechanistic, preclinical, translational and clinical evidence underlying resistance to these agents. Distinct drug-specific molecular mechanisms can underlie acquired chemoresistance, whereas shared adaptive survival processes support treatment persistence and may, in some settings, contribute to the emergence of stable acquired resistance. 5-FU resistance is associated with factors s

Cell Death Induction by MO2455, a Novel Potential Anti-Cancer Agent in Monocytic and T-Cell Leukemia Cells.

Despite advances in targeted therapies, treatment resistance and disease recurrence remain major obstacles in leukemia management. Structural modification of clinically successful kinase inhibitors represents a promising approach for discovering novel anti-leukemic agents with improved biological activities. In the present study, we investigated the anti-leukemic activity and mechanism of action of MO2455, a newly synthesized imatinib-derived compound, in monocytic leukemia U937, THP-1, and T-cell leukemia CCRF-CEM cells. MO2455 showed substantial cytotoxic activity against all three cell line

Antiviral Activity of an Ethanolic Extract of Cinnamomum cassia and Cinnamaldehyde Against the Herpesviruses HSV-1 and VZV.

Herpes simplex virus type 1 (HSV-1) and varicella-zoster virus (VZV) are clinically important alphaherpesviruses that establish lifelong latent infections and remain significant causes of morbidity despite the availability of antiviral drugs. The emergence of drug resistance and the inability of current therapies to eliminate latent virus reservoirs underscore the need for novel antiviral agents with alternative mechanisms of action. In the present study, we evaluated the antiviral activity of an ethanolic extract prepared from commercial cinnamon bark marketed as Cinnamomum cassia and cinnama

A Combination of Ursolic Acid and Oleanolic Acid Disrupts Bioenergetic Homeostasis in Prostate Cancer and Inhibits Cancer Growth by Cytotoxic Autophagy.

Oleanolic acid (OA) and ursolic acid (UA) are bioactive pentacyclic triterpenoids (PTTs) and potential therapeutics for cancers. If tested individually, their therapeutic efficacy is limited. The antitumor potential of OA, UA, and their combination was tested against castration-resistant prostate cancer cells (CRPCs) in vitro and in vivo. UA, OA, and UA + OA (1:1) inhibited the cell proliferation and clonogenic growth of CRPC cells. Compared to the potency of individual OA and UA on these cells, their potency was synergistically enhanced when tested as an OA + UA mix. Neither of these compound
