Tularemia following a squirrel bite in Southern Germany: a case report.

BACKGROUND: Tularemia is a zoonotic infection caused by Francisella tularensis. Human cases in Germany are emerging and are usually associated with hare or tick exposure. Transmission via squirrel bites has only been reported once before in Germany. CASE PRESENTATION: We describe a 23-year-old woman who developed tularemia 11 days after being bitten by an injured red squirrel (Sciurus vulgaris) in southern Germany. Initial symptoms included headache, chills and fatigue, followed by painful right axillary lymphadenopathy without fever. Empirical ciprofloxacin (500 mg twice daily) was initiated

Rickettsia parkeri subverts nicotinamide adenine dinucleotide (NAD+) metabolic pathway for its survival in endothelial cells.

Rickettsia parkeri rickettsiosis is an emerging tick-borne disease in humans. The bacterium R. parkeri is primarily transmitted to humans by the Gulf Coast tick, Amblyomma maculatum. Upon transmission to the host, R. parkeri infects and multiplies in host endothelial cells. In this study, we provide evidence that R. parkeri modulates the host nicotinamide adenine dinucleotide (NAD+) pathway for its survival in endothelial cells. Analysis performed with arrays containing antibodies against proteins involved in different metabolic pathways revealed increased levels of BST1 (an NAD+-dependent mol

The iucC gene is a major contributor to the virulence of hypervirulent Klebsiella pneumoniae.

BACKGROUND: Hypervirulent Klebsiella pneumoniae (hvKp) is a critical pathogen that causes highly lethal invasive infections with its virulence linked to that of aerobactin, the core siderophore system of hvKp. METHODS: Here, 20 porcine-derived K. pneumoniae strains were subjected to capsular genotyping and the virulence of the strains was evaluated through mouse lethality assays. Whole-genome sequencing and analysis was performed on two K2 serotype strains (including highly and weakly virulent strains), and the differential virulence genes were analyzed. The virulence-related gene iucC, ident

Two Cases of Q Fever in Pregnancy, including Management of the Newborn, Australia.

Optimal management of the birthing parent with Q fever in pregnancy and of the infant has not been established. Coxiella burnetii expresses a tropism for the placenta; resulting infection can potentially lead to spontaneous abortion and fetal demise. Although evidence around preventing transmission and infection in the peripartum and postpartum period is lacking, reports of healthy babies born to mothers with acute or chronic Q fever in pregnancy are increasing. Historically, many clinicians have recommended against breastfeeding in this setting because of a theoretical risk for bacterial tran

Predictive multi-omic biomarkers for urban zoonotic spillover detection: an integrative review.

Urban wildlife is an overlooked yet critical component of zoonotic disease surveillance, especially in biodiversity hotspots where human-animal interfaces accelerate spillover risk. This review synthesizes five complementary omics layers: Host microRNAs, host-pathogen genetic markers, bacterial microbiome profiling, viromics, and host transcriptomics into a single predictive framework for early spillover detection. Across taxa and pathogen classes, we highlight convergent molecular signatures of infection, from receptor polymorphisms and shifts in MHC diversity to pathogen-responsive miRNAs, h

Unlocking the full potential of nanopore sequencing: tips, tricks, and advanced data analysis techniques.

Nucleic acid sequencing is the process of identifying the sequence of DNA or RNA, with DNA used for genomes and RNA for transcriptomes. Deciphering this information has the potential to greatly advance our understanding of genomic features and cellular functions. In comparison to other available sequencing methods, nanopore sequencing stands out due to its unique advantages of processing long nucleic acid strands in real time, within a small portable device, enabling the rapid analysis of samples in diverse settings. Evolving over the past decade, nanopore sequencing remains in a state of ongo

Tracking spread of carbapenemase-producing Enterobacterales between humans and companion animals: successes and challenges.

Carbapenemase-producing Enterobacterales (CPE) pose a critical public health threat. Recent reports highlight that CPE emergence in companion animals mirrors that found in humans, underscoring the need for a One Health approach to investigating transmission routes. This Perspective article outlines an interdisciplinary model developed as part of the Centers for Disease Control and Prevention-funded Pathogen Genomics Center of Excellence at the Minnesota Department of Health (MDH) to investigate CPE transmission across human and animal populations. This represents one of the first operational O

The impact of STING signaling on myeloid cells highlights macrophages as the major player in controlling Brucella abortus.

INTRODUCTION: Brucellosis, caused by Brucella abortus (Ba), is one of the most common zoonoses worldwide. Some components of the bacteria can be recognized by the innate immune system and understanding these mechanisms is necessary for directing future vaccines and therapies against brucellosis. Our group has demonstrated that STING molecule contributes to host protection against Ba in vivo. Considering that Ba has ability to subvert the immune response, the study of the mechanisms of host cell activation becomes fundamental for the development of measures to control bacterial replication. Enzymatic Synthesis of 2-Amino-adenosine Triphosphate as a Noncanonical Nucleotide Precursor of Z-Modified RNA.

2-amino-deoxyadenosine (dZ) occurs naturally in certain bacteriophage genomes, where it replaces deoxyadenosine, forming three hydrogen bonds with thymidine. This noncanonical deoxyribonucleoside underlies the unique biophysical properties of dZ-DNA. Its corresponding ribonucleoside has been introduced to RNA to form Z-modified RNA, with promising applications in vaccine production and biomedicine. Unlike dZ-DNA, Z-modified RNA has only been synthesized in vitro, which required the addition of chemically synthesized 2-amino-adenosine triphosphate (ZTP) as a precursor. Here, we describe enzyme

Seasonal dynamics, antimicrobial resistance, and genetic lineages of thermotolerant Campylobacter isolates from South American camelids.

BACKGROUND: South American camelids (SACs) are increasingly kept as companion animals, but their role in zoonotic transmission is poorly understood. Thermotolerant Campylobacter spp. are a leading cause of bacterial zoonoses globally. This study investigated the occurrence and zoonotic potential of thermotolerant Campylobacter spp. in SACs on German farms. METHODS: Fecal swabs from up to 20 animals on 10 farms were collected across four seasons. Campylobacter spp. were isolated, and genomes were typed to assess multilocus sequence types (STs), virulence, and antimicrobial resistance. RESULTS

Vaccination against H5 HP avian influenza virus leads to persistent immune response in wild king penguins.

Since 2021, the panzootic nature of high pathogenicity avian influenza (HPAI) represents an increasing threat to wild vertebrate populations. In this context, recent vaccines developed for poultry could provide tools for the conservation of wild endangered birds populations. The king penguin (Aptenodytes patagonicus), a long-lived seabird breeding in dense colonies with an extended chick-rearing period, has been identified as a possible surrogate species for a vaccination trial in a sub-Antarctic natural setting. Here we investigate the immune response of king penguin chicks to a self-amplifyi

Exploring antibiotic stewardship interventions within a One Health context: a scoping review.

Antibiotic resistance (ABR) presents a global threat to human health, animal health, and the environment. While ABR is a natural phenomenon, the overuse and misuse of antibiotics in human health, animal health, and food production is a major driver of ABR. As interactions between humans, animals, and the environment are central to the emergence and spread of ABR, adopting a One Health approach is essential to effectively address the issue. In this context, a large range of antibiotic stewardship interventions has been developed to optimize the use of antibiotics. However, we lack a comprehensi

AddaVax-Adjuvanted Inactivated Vaccine Confers Protection Against Listeria monocytogenes Challenge in Mice.

Listeria monocytogenes (Lm) is a facultative intracellular pathogen responsible for life-threatening listeriosis, characterized by high mortality rates. Currently, there is no vaccine available for the prevention or treatment of listeriosis. Here, we developed two adjuvanted inactivated vaccines and evaluated their safety, immunogenicity, and protective efficacy. The hypervirulent Lm strain XYSN was inactivated with β-propiolactone (BPL) and formulated with AddaVax or Alum to produce inactivated vaccines (ADIV and ALIV) respectively. Local reactogenicity and histopathological observations reve

Autophagy in emerging and highly concerned severe zoonotic infectious diseases.

Autophagy is a conserved cellular process that mediates degradation of damaged organelles, misfolded proteins, and invading pathogens, playing critical roles in intracellular homeostasis and immune regulation. Given that over 70% of infectious diseases and 60% of emerging infectious diseases are zoonotic, posing a major threat to global health, this review aims to summarize the cellular and molecular mechanisms underlying the crosstalk between autophagy and key zoonotic pathogens. We comprehensively retrieved relevant research literature from the PubMed, Web of Science, and Scopus databases (w
