Human genetic variation reveals FCRL3 is a lymphocyte receptor for Yersinia pestis.

Yersinia pestis is the bacterium responsible for plague, one of the deadliest diseases in history. To discover human genetic determinants of Y. pestis infection, we utilized nearly 1,000 genetically diverse lymphoblastoid cell lines in a cellular genome-wide association study. A nonsynonymous SNP, rs2282284 (N721S), in Fc receptor-like 3 (FCRL3) was associated with bacterial invasion of host cells (p = 9 × 10-8). Overexpressed FCRL3 facilitated attachment and invasion of Y. pestis and colocalized with Y. pestis at attachment sites. These properties were variably conserved across the FCRL famil

Nanolipoprotein particle (NLP) vaccine confers protection against Yersinia pestis aerosol challenge in a BALB/c mouse model.

INTRODUCTION: Yersinia pestis is the etiological agent of plague, a disease that remains a concern as demonstrated by recent outbreaks in Madagascar. Infection with Y. pestis results in a rapidly progressing illness that can only be successfully treated with antibiotics given shortly after symptom onset. Live attenuated or whole cell inactivated vaccines confer protection against bubonic plague, but pneumonic plague has been more difficult to prevent. Novel effective subunit vaccine formulations may circumvent some of these shortfalls. Here, we compare the immunogenicity generated by an advanc

Ciprofloxacin versus Aminoglycoside-Ciprofloxacin for Bubonic Plague.

BACKGROUND: Plague is a high-consequence infectious disease with epidemic potential. Current treatment guidelines are based on weak evidence. METHODS: We enrolled persons (excluding pregnant persons) in Madagascar who had clinically suspected bubonic plague during 2020-2024. Using an open-label noninferiority design, we compared two treatments included in the national plague guidelines: oral ciprofloxacin for 10 days (ciprofloxacin monotherapy) or injectable aminoglycoside for 3 days followed by oral ciprofloxacin for 7 days (aminoglycoside-ciprofloxacin). The primary end point was treatment

Yersinia pestis BipA is a novel regulator of pesticin and a type 6 secretion system.

Yersinia pestis is a gram-negative bacterium and the causative agent of bubonic, septicemic, and pneumonic plague. Y. pestis is most commonly transmitted to humans by infected fleas that deposit the bacteria into the dermis at the bite site, leading to bubonic plague. The bacteria ultimately access the bloodstream, and after deposition in the lung, can be transmitted person-to-person through infectious respiratory droplets, resulting in primary pneumonic plague, a highly lethal and rapidly progressing pneumonia. Pathogenesis is mediated by a suite of chromosomally encoded and plasmid-borne vir

The long and intimate association between humans and parasites through time.

While the interaction between humans and their parasites is well studied today, taking a long view of infection throughout human evolution helps to place the current picture in context and identify trends in infection over time. After considering how early technologies may have facilitated the transmission of parasites to humans, we examine the association between humans and parasites through time using archaeological and genetic evidence. Techniques such as microscopy, immunoenzymatic assays and DNA analysis have identified a range of protozoa, helminths and ectoparasites in our ancestors. Ev

Performance of diagnostic procedures for bubonic plague in endemic settings in Madagascar: a prospective test accuracy sub-study within the IMASOY trial.

OBJECTIVES: We aimed to assess the diagnostic value of a battery of confirmatory tests included in the WHO guidelines for bubonic plague, combined and individually, including culture, PCR and serology, and the on-site and laboratory performance of the F1 antigen-based lateral flow rapid diagnostic test (F1RDT). METHODS: Bubo aspirates from patients (all ages) with suspected bubonic plague enrolled into the IMASOY trial (NCT04110340) underwent routine laboratory diagnosis complemented with serology to measure IgG F1 antibodies from blood samples taken on days 1, 11, and 21. Yersinia pestis plasminogen activator protease is regulated by the PhoP/PhoQ two-component system.

Yersinia pestis is a gram-negative bacterium and the causative agent of plague. The Y. pestis virulence factor plasminogen activator protease (Pla) is an outer membrane aspartic protease that facilitates the dissemination of bacteria from the site of inoculation to deeper tissue during bubonic plague. During pneumonic plague, Pla acts as an adhesin, which contributes to the suppression of early innate immune responses in the lungs, and as a protease that aids in resisting bacterial killing by neutrophils. Two-component regulatory systems (TCSs) are involved in bacterial adaptation to environme

Imperial Careering: India and the Women's Medical Movement, 1896-1920.

India figured prominently in the women's medical movement of the late nineteenth and early twentieth centuries. It was both a cause-bringing medical aid to Indian women-and a career-offering employment opportunities to qualified British women doctors. Where most studies have focussed on the early years of the movement in India and the creation of the Dufferin Fund in 1885, this article explores the careers, attitudes and experiences of a second generation of white women doctors, from the outbreak of bubonic plague in 1896 to the end of the First World War. As an exercise in imperial careering,

Socio-ecological risk factors associated with human flea infestations of rural household in plague-endemic areas of Madagascar.

Plague is a flea-borne fatal disease caused by the bacterium Yersinia pestis, which persists in rural Madagascar. Although fleas parasitizing rats are considered the primary vectors of Y. pestis, the human flea, Pulex irritans, is abundant in human habitations in Madagascar, and has been found naturally infected by the plague bacterium during outbreaks. While P. irritans may therefore play a role in plague transmission if present in plague endemic areas, the factors associated with infestation and human exposure within such regions are little explored. To determine the socio-ecological risk fa

Legacy of a magic gene-CCR5-∆32: From discovery to clinical benefit in a generation.

The discovery of the 32-bp deletion allele of the chemokine receptor gene CCR5 showed that homozygous carriers display near-complete resistance to HIV infection, irrespective of exposure. Algorithms of molecular evolutionary theory suggested that the CCR5-∆32 mutation occurred but once in the last millennium and rose by strong selective pressure relatively recently to a ~10% allele frequency in Europeans. Several lines of evidence support the hypothesis that CCR5-∆32 was selected due to its protective influence to resist Yersinia pestis, the agent of the Black Death/bubonic plague of the 14th

Decrypting the multi-genome data for chimeric vaccine designing against the antibiotic resistant Yersinia pestis.

Yersinia pestis, the causative agent of plague, is a gram-negative bacterium that can be fatal if not treated properly. Three types of plague are currently known: bubonic, septicemic, and pneumonic plague, among which the fatality rate of septicemic and pneumonic plague is very high. Bubonic plague can be treated, but only if antibiotics are used at the initial stage of the infection. But unfortunately, Y. pestis has also shown resistance to certain antibiotics such as kanamycin, minocycline, tetracycline, streptomycin, sulfonamides, spectinomycin, and chloramphenicol. Despite tremendous progr

Past anthropogenic impacts revealed by trace elements in cave guano.

Natural and human-induced toxic elements can accumulate in the environment, posing significant risks to human health and ecosystems. This study explores cave bat guano, an unconventional and relatively under-researched environmental repository, to reveal historical pollution trends and sources. Through trace elements analysis of a 1.5-m-thick guano deposit from Zidită Cave (Romania), we track changes in mining and metallurgy from 1000 CE-2012. We identified substantial pollution primarily from porphyry copper and Au-Ag-Te mines, but also impacts from usage of leaded gasoline and agricultural p

The Need for Historical Fluency in Pandemic Law and Policy.

The primary claim of this essay is that historical fluency is required for effective work in crafting legal and policy interventions as a part of public health emergency preparedness and response (PHEPR). At a broad level, public health law is explicitly recognized as a key systems-level component of PHEPR practice.1 This essay therefore focuses on the extent to which historical fluency is necessary or at least useful to all aspects of PHEPR that draw on or deploy legal and policy mechanisms (e.g., design, planning, implementation, dissemination, monitoring and evaluation, etc.). The essay col

Knowledge and practices related to plague persistence in plague-endemic foci, Mbulu District, Tanzania.

INTRODUCTION: Plague continues to be a major public health concern in African countries. Several social practices and environmental conditions have been associated with the reoccurrence of bubonic plague, especially in places where the disease is prevalent. Therefore, it remains important to understand people knowledge, behavior and practices related to disease risks in order to identify factors that may hinder prevention and control strategies in the foci. METHODS AND RESULTS: A study survey of 100 households was conducted in Mbulu district to assess plague knowledge, factors that influence

Sex differences in immune protection in mice conferred by heterologous vaccines for pneumonic plague.

BACKGROUND: Yersinia pestis is the etiological agent of plague, which can manifest as bubonic, septicemic, and/or pneumonic disease. Plague is a severe and rapidly progressing illness that can only be successfully treated with antibiotics initiated early after infection. There are no FDA-approved vaccines for plague, and some vaccine candidates may be less effective against pneumonic plague than bubonic plague. Y. pestis is not known to impact males and females differently in mechanisms of pathogenesis or severity of infection. However, one previous study reported sex-biased vaccine effectiven

Differentiation of Synanthropic Fleas from Andalusia (Spain) through Geometric Morphometrics Analysis.

Fleas (Siphonaptera) are ectoparasitic hematophagous insects responsible for causing bites and itchy skin conditions in both humans and animals. Furthermore, they can act as vectors of different pathogens of a wide variety of diseases worldwide, including bartonellosis, rickettsiosis, and bubonic plague. Accurate identification of fleas is necessary for the study of their epidemiology, prevention, and control. In addition to traditional morphological classification approaches and molecular biology techniques, geometric morphometrics is increasingly proving to be a useful complementary tool for

Public health threat of novel zoonotic diseases: literature review.

Zoonoses, diseases transmitted from animals to humans, continue to challenge public health despite advancements in controlling infectious diseases. The intricate link between human, animal, and environmental health is emphasised by the fact that zoonoses contribute to 60% of emerging human infections. Wet markets, wildlife hunting, intensive wildlife farming, and interactions between domestic animals and humans are key transmission sources. Historical examples like the bubonic plague and English Sweats illustrate the longstanding impact of zoonotic diseases. With new transmission patterns emer

Yersinia pestis and pneumonic plague: Insight into how a lethal pathogen interfaces with innate immune populations in the lung to cause severe disease.

Yersinia pestis is the causative agent of bubonic, septicemic and pneumonic plague. The historical importance and potential of plague to re-emerge as a threat worldwide are indisputable. The most severe manifestion of plague is pneumonic plague, which results in disease that is 100% lethal without treatment. Y. pestis suppresses host immune responses early in the lung to establish infection. The later stages of infection see the rapid onset of hyperinflammatory responses that prove lethal. The study of Y. pestis host/pathogen interactions have largely been investigated during bubonic plague an

Differential pathogenicity and lethality of bubonic plague (1720-1945) by sex, age and place.

COVID-19 brought back to the attention of the scientific community that males are more susceptible to infectious diseases. What is clear for other infections-that sex and gender differences influence both risk of infection and mortality-is not yet fully elucidated for plague, particularly bubonic plague, although this knowledge can help find specific defences against a disease for which a vaccine is not yet available. To address this question, we analysed data on plague from hospitals in different parts of the world since the early eighteenth century, which provide demographic information on i
