Carmofur inhibits ovarian cancer cell proliferation, migration, and invasion via disrupting the YAP-1/TEAD pathway: in vitro and in silico approaches.

The current study aims to present a novel approach to alleviate the aggressiveness of ovarian cancer by pharmacological disruption of the Yes-Associated Protein (YAP) - 1/Transcriptional Enhancer Factor domain (TEAD) heterodimer structure using Carmofur. The Ovcar-3 cell line was incubated with progressively diluted Carmofur solutions. Proliferation inhibition was assessed using the MTT assay. Flow cytometry was employed to determine the pro-apoptotic properties. Cellular morphological alterations were assessed with confocal microscopy. The migration inhibition status was assessed via the scra

Promiscuous binding of CXC chemokines to ORF74 receptors enables immunotoxin approaches in Gammaherpesvirus models.

Kaposi sarcoma-associated herpesvirus (KSHV) encodes the constitutively active chemokine receptor ORF74, a key factor in viral pathogenesis. Murine γ-herpesvirus 68 (MHV68) acts as an in vivo model for KSHV biology, but the ligand-binding and signaling functions of its ORF74 homolog are not fully understood. To address this knowledge gap, we studied interactions between MHV68-ORF74 and human and murine chemokines through binding assays and signaling tests. Additionally, we assessed the cytotoxicity of human chemokine-based fusion toxin proteins (FTPs) targeting ORF74. We found that MHV68-ORF74

Head and Neck Kaposi Sarcoma-An Updated Focus on Clinical and Epidemiological Characteristics: A Comprehensive Review.

BACKGROUND/OBJECTIVES: Interest in Oral Kaposi's sarcoma (OKS) has declined recently, potentially causing diagnostic errors due to physicians' unfamiliarity with its presentation. This review describes clinical and demographic characteristics of OKS patients across epidemiological groups. METHODS: A literature search of studies published from 1957 to December 2024 was conducted using PubMed, Web of Science, Cochrane Library, Scopus, and Google Scholar. Studies with confirmed oral Kaposi sarcoma were included, while those with incomplete data were excluded. Cases were grouped into classic, end

Kaposi's Sarcoma and Acute Limb Ischaemia: A Possible Direct Vascular Link.

Acute limb ischaemia is caused by a sudden interruption in arterial flow. Its occurrence in oncology is uncommon compared with the general population. Generally considered to be part of the paraneoplastic syndrome, its occurrence in a patient with Kaposi's sarcoma (KS) raises the question of a different pathophysiology. We report a case of acute limb ischaemia in a patient with KS. The patient was 51 years old and had hyperpigmented macules on both lower limbs for 6 months, some of which had become nodules. He was recently diagnosed with HIV and had been put on treatment. He presented with pur

Monoclonal neutralizing antibodies elicited by infection with Kaposi sarcoma-associated herpesvirus reveal critical sites of vulnerability on gH/gL.

Kaposi sarcoma-associated herpesvirus (KSHV) is an oncogenic virus that causes Kaposi sarcoma, primary effusion lymphoma and multicentric Castleman disease. A vaccine that prevents KSHV infection or serves in the treatment of KSHV-related diseases represents a critical unmet need, however, the types of immune responses a vaccine should elicit have not been well defined. The gH/gL glycoprotein complex is an important target of KSHV-neutralizing antibodies, but the epitope specificities targeted by these antibodies remain unknown. Here, we isolated 12 gH/gL-specific monoclonal antibodies (mAbs)

COVID vaccination and post-infection cancer signals: Evaluating patterns and potential biological mechanisms.

A growing number of peer-reviewed publications have reported diverse cancer types appearing in temporal association with COVID-19 vaccination or infection. To characterize the nature and scope of these reports, a systematic literature search from January 2020 to October 2025 was conducted based on specified eligibility criteria. A total of 69 publications met inclusion criteria: 66 article-level reports describing 333 patients across 27 countries, 2 retrospective population-level investigations (Italy: ~300,000 cohort, and Korea: ~8.4 million cohort) quantified cancer incidence and mortality t

A rare case of gastroduodenal junction Kaposiform hemangioendothelioma in a cat: pathological findings and follow-up.

Kaposiform hemangioendothelioma (KHE) is a rare vascular neoplasm predominantly affecting pediatric human patients and has rarely been reported in veterinary medicine. We described the clinicopathological features of an uncommon gastroduodenal junction KHE in a 2-year-old male cat presented with a history of intermittent vomiting, acutely worsening over 48 h. Imaging and endoscopic evaluation revealed complete pyloric obstruction caused by an infiltrative mass with enlarged regional lymph nodes, which were surgically excised and examined histologically and immunohistochemically. Specimens were

Kaposi Sarcoma as a Cause of Anemia in Kidney Transplant Recipients: Two Case Reports.

INTRODUCTION: Kaposi Sarcoma (KS), caused by human herpesvirus 8 (HHV-8), may be observed in transplant recipients in the form of immunosuppression-related KS. While it primarily affects the skin, it can also involve internal organs, particularly the gastrointestinal (GI) system. GI involvement is often asymptomatic, but bleeding episodes may occur. Post-transplant anemia (PTA) is a common complication associated with increased mortality and decreased renal function. Early diagnosis and management are crucial. This report discusses 2 cases of KS in kidney transplant recipients presenting with

Malignant vascular neoplasms of the distal female genital tract: a series of 19 cases emphasizing unusual features and diagnostic challenges.

AIMS: Scarce literature exists regarding the spectrum of vascular neoplasms of the distal female genital tract. Herein, we describe the clinicopathologic features of a cohort of intermediate to malignant endothelial neoplasms of the vulva and vagina. METHODS AND RESULTS: Specimens were identified retrospectively from in-house and consultation files at our institution. Clinicopathologic data was obtained from review of the chart and available histologic material. Nineteen cases were identified in total. Kaposi sarcoma (n = 2) was of the enigmatic 'sporadic' form in elderly non-HIV females. Kap

Primary osseous kaposiform hemangioendothelioma confined to long bones: a retrospective imaging analysis of 12 pediatric cases.

BACKGROUND: Kaposiform hemangioendothelioma (KHE) is a rare, locally aggressive vascular tumor predominantly affecting infants and children. While musculoskeletal involvement is common, cases originating in and confined exclusively to bone are exceptionally rare, and their specific imaging characteristics are not well-defined. This study aims to characterize the clinical and detailed radiological features of primary, intraosseous KHE in a pediatric cohort. METHODS: We conducted a retrospective review of pediatric patients with biopsy-proven KHE admitted to our institution between January 2016

Fatal soft-tissue and bony complications following radiotherapy for HIV-negative Kaposi's sarcoma: a rare case of post-radiation necrotizing fasciitis and osteomyelitis in a resource-limited setting.

BACKGROUND: Kaposi's sarcoma (KS) is a vascular neoplasm strongly associated with HIV infection; however, HIV-negative KS is exceptionally rare, particularly in sub-Saharan Africa. Radiotherapy (RT) remains a standard local treatment for KS, providing excellent symptom relief and local control. Despite its efficacy, severe complications such as necrotizing fasciitis (NF) and osteomyelitis are exceedingly uncommon and scarcely reported in HIV-negative patients. CASE PRESENTATION: We report a 47-year-old HIV-negative man with biopsy-confirmed KS of the right thigh, initially treated with chemot

MRI and MDCT Findings of Kaposiform Hemangioendothelioma in the Oral Cavity of a Neonate: A Case Report and Literature Review.

INTRODUCTION: Kaposiform hemangioendothelioma (KHE) is a rare, locally aggressive vascular tumor of childhood. However, a detailed description of its imaging features, especially for neonatal oral cavity lesions, is lacking in the literature, with no prior reports detailing both prenatal magnetic resonance imaging (MRI) and postnatal multi-detector computed tomography (MDCT) findings. This report presents a case of neonatal KHE, focusing on its distinctive imaging manifestations on both modalities to provide diagnostic insights for this rare condition in an unusual location. Isolated Kaposi Sarcoma of the External Ear in an Immunocompromised Host: A Case Report.

BACKGROUND: A vascular neoplasm linked to HHV-8 infection, Kaposi sarcoma (KS) is common in immunocompromised patients, notably people living with HIV. Isolated external ear involvement is uncommon and may present clinically as keloids, cysts, or infections. CASE PRESENTATION: A 31-year-old man presented with a one-month history of ipsilateral dacryocystitis and a rapidly growing, painful right auricular mass. The auricular lesion was initially presumed to be a sebaceous cyst during his emergency room evaluation, with actively draining purulent material. Labs showed severe pancytopenia with a

Kaposi Sarcoma Associated with Pemphigus Vulgaris in the Setting of Immunosuppressive Therapy: Case Report and Review of Literature.

INTRODUCTION: Pemphigus vulgaris (PV) is a chronic autoimmune blistering disorder characterized by intraepidermal blisters affecting the skin and mucous membranes. The primary treatment for PV is systemic corticosteroid therapy, which is often prolonged due to the disease's chronic and relapsing course. The interplay between autoimmunity and long-term immunosuppressive treatment can predispose patients to secondary complications, including Kaposi sarcoma (KS), an angioproliferative tumor primarily involving the skin and soft tissues. When KS develops in the context of immunosuppressive therapy

Primary Kaposiform hemangioendothelioma of the tibia: report of a case with a previously undescribed GNAQ mutation.

Primary occurrence of Kaposiform hemangioendothelioma (KHE) in bone is extremely rare. We report the radiologic, histopathologic and molecular features of a case of KHE affecting the tibia of an adult male, who presented with pain in the left knee initially associated with previous trauma. Radiographs showed an eccentric well-defined lytic lesion of the tibial epiphysis. On MRI, the lesion showed hypo to isointensity on T1-weighted images and hyperintensity on STIR sequences. A CT-guided biopsy was pathologically inconclusive. MRI performed 11 months later showed lesion progression with cortic

Are Corticosteroids Truly Contraindicated in Pulmonary Kaposi Sarcoma?

Kaposi sarcoma (KS) is an angioproliferative disorder associated with human herpes virus 8 (HHV-8) infection. It has both cutaneous and extracutaneous manifestations. With extracutaneous manifestations, it most commonly involves the oral cavity, gastrointestinal tract, and regional lymph nodes. Visceral organ involvement other than the gastrointestinal tract, such as the liver, lungs, and bones, is extremely rare. Here, we would like to discuss a patient with human immunodeficiency virus (HIV) and acquired immunodeficiency syndrome (AIDS) found to have pulmonary KS manifesting as acute hypoxem

Ultrasound-guided lymph node biopsies: feasible and safe use of pathology services in a resource-limited, high TB/HIV prevalence setting.

BACKGROUND: Enlarged lymph nodes (LN) pose diagnostic challenges for people with HIV (PWH). While tuberculosis (TB) is a common cause in low-income settings, lymphomas and Kaposi's sarcoma must also be considered. Ultrasound and symptoms cannot distinguish between these conditions, and histology is often needed, but limited resources in low-income countries restrict sampling. To minimize the need for excisional biopsies, we introduced an algorithm for ultrasound-guided core-needle biopsies (CNB) after negative fine-needle aspiration (FNA) results by Xpert-Ultra (Cepheid, USA). METHODS: At the
