Caspase Cleavage of Kaposi Sarcoma-Associated Herpesvirus Proteins: A role for K5 in Preventing Caspase-Mediated Cell Death during Lytic Replication.

We previously reported that Kaposi sarcoma-associated herpesvirus (KSHV) latency-associated nuclear antigen (LANA) acts as a pseudo-substrate for caspases-1 and 3, thereby interfering with their inflammatory and apoptotic activity, respectively. To determine if other KSHV proteins undergo caspase cleavage, we screened the KSHV proteome for potential caspase cleavage sites. Using SitePrediction (SP), 30 KSHV proteins with potential caspase-cleavage sites were identified. Among those with highest SP score was an early lytic protein, K5. Treatment of BJAB K5-FLAG cells with ⍺Fas, an apoptotic sti

The Psychological Impact of Sarcoma From Pre-Diagnosis to Post-Treatment, a Meta-Synthesis.

OBJECTIVES: Sarcoma is a rare form of cancer affecting soft tissue and bone. Previous research has highlighted psychological challenges associated with this subset of cancer. The purpose of this paper is to draw together existing qualitative research into the psychological experiences of sarcoma patients, and to identify any sarcoma-specific psychological needs. METHODS: A systematic review was conducted with the question: 'What is the psychological impact of sarcoma?'. The protocol for the review was preregistered with PROSPERO (CRD42023439959). Thematic meta-synthesis (meta-ethnography) was

Incidence and risk of cancer emergence among patients post-kidney transplantation in Japan.

BACKGROUND: Cancer is one of the most common complications after kidney transplantation and an important cause of mortality. However, no large, nationally representative study has investigated cancer incidence post-kidney transplantation. This study aimed to determine the standardized incidence ratio (SIR) for cancer after kidney transplantation using the National Database of Health Insurance Claims (NDB). METHODS: We used NDB from April 2013 to March 2022; patients were included if they had been on dialysis for at least one year, were diagnosed with cancer related to post-kidney transplantat

Kaposi sarcoma herpesvirus (KSHV) inflammatory cytokine syndrome (KICS): a case study.

BACKGROUND: Kaposi sarcoma herpesvirus-inflammatory cytokine syndrome (KICS) is a rare, life-threatening condition associated with Kaposi sarcoma and systemic immune dysregulation induced by Human Herpesvirus 8 (HHV-8). With a mortality rate approaching 60%1 (Goncalves, Ziegelbauer, Uldrick, Yarchoan in Curr Opin HIV AIDS 12(1):47-56, 2017), KICS poses significant diagnostic and therapeutic challenges. This report examines a case of KICS in a patient with well-controlled HIV, emphasising the clinical complexities, treatment strategies, and the need for heightened awareness. CASE PRESENTATION:

Vascular lesions of head and neck region: A pictorial review.

Vascular lesions in the head and neck region encompass a wide spectrum of malformations and tumors, ranging from hemangiomas to arteriovenous fistulas. The International Society for the Study of Vascular Anomalies classification system categorizes these anomalies into vascular tumors and malformations. Vascular tumors are further divided into benign, borderline, and malignant types, while vascular malformations are classified based on flow characteristics as high or low flow. Accurate delineation of these lesions is crucial due to their clinical implications, particularly their location and po

The Origin of Kaposi's Sarcoma and Other KSHV-Associated Sarcomas.

Kaposi's sarcoma (KS) is a multicentric, polyclonal malignancy linked to Kaposi's sarcoma-associated herpesvirus (KSHV) infection. While significant advances have been made in understanding KSHV virology and the clinical characteristics of KS, the precise mechanisms of pathogenesis and cellular origins remain elusive. This review integrates clinical observations with the latest cutting-edge research to explore the nature and potential cellular precursors of KS. Multiple cell types have been proposed as potential KS oncogenic progenitors, including endothelial cells, mesenchymal stem cells (MSC

Human immunodeficiency virus negative, immunocompetent primary effusion lymphoma with a complete response on R-miniCHOP.

Primary effusion lymphoma (PEL) is a rare kind of extranodal non-Hodgkin large B-cell lymphoma that develops in the liquid phase in serous membrane-lined body cavities (perineum, pericardium, and pleura) without tumor masses. Kaposi sarcoma-associated human herpesvirus 8 (HHV8) is essential in establishing a diagnosis of PEL. An 84-year-old male with a past medical history of testicular cancer in his 40s presented with a chief complaint of shortness of breath which was attributed to a left pleural effusion. The flow cytometry indicated 32% small T lymphocytes (cluster of differentiation [CD]4:

Incidence rate analysis of cutaneous soft tissue sarcomas in the USA.

Soft tissue sarcomas (STS) are rare and heterogeneous, with cutaneous STS (CSTS) lacking recent epidemiological data. This study analyses CSTS incidence from 2000 to 2019 using SEER data, identifying 18 181 cases. CSTS was more common in men and non-Hispanic Whites, with Kaposi sarcoma (KS) and dermatofibrosarcoma protuberans (DFSP) being predominant. The overall CSTS incidence was 11.2 per 1 000 000 person-years, with KS having the highest rate. The study reveals a 1.4% annual decline in CSTS rates and highlights ethnic disparities, especially in KS incidence among Non-Hispanic Black patients

Incidence of rare cancers during the COVID-19 pandemic: a US population-based study.

INTRODUCTION: Recent media reporting has suggested an increase in the incidence of rare cancers in 2021 as a result of the COVID-19 pandemic, raising concern for possible carcinogenic effects of severe acute respiratory syndrome coronavirus 2 infection. We aimed to identify cancers that increased in incidence in the United States during 2021 compared with 2018-2019. METHODS: We analyzed data from Surveillance, Epidemiology, and End Results (SEER-22) using a systematic classification schema including rare cancers. We estimated age-standardized incidence rate ratios (aIRRs) comparing the year 2

Patient-Reported Outcome Measurement and Reporting for Patients with Soft Tissue Tumors: A Scoping Literature Review.

(1) Background: Quality of life (QoL) assessment is a crucial aspect for patients diagnosed with cancer. Over the years, different tools have been developed to measure QoL, both generic and pathology specific, but the inclusion of quality of life among other indicators of efficacy in randomized controlled trials (RCTs) remains a controversial issue. In this review, we aim to review the frequency and modality of QoL assessment in RCTs, enrolling patients diagnosed with mesenchymal tumors. (2) Methods: An electronic literature search of bone and soft tissue sarcoma and GIST-related RCTs publishe

Cutaneous Squamous Cell Carcinoma Mimicking Kaposi Sarcoma in an HIV-Positive Patient: Diagnostic Challenges and Therapeutic Response to Liposomal Doxorubicin.

Cutaneous malignancies are among the most common cancers worldwide, with cutaneous squamous cell carcinoma (cSCC) being the second most frequent non-melanoma skin cancer. In people living with HIV (PLHIV), cSCC may present atypically and overlap clinically with opportunistic tumors such as Kaposi sarcoma (KS), complicating diagnosis and management. Immunosuppression increases the risk of aggressive and multifocal cSCC, often delaying diagnosis. Liposomal doxorubicin, an anthracycline-based chemotherapy standardly used for KS, has shown limited off-label activity against other cutaneous maligna

Transmission of human herpesvirus-8 through liver transplantation and development of post-transplantation Kaposi sarcoma: A case report.

INTRODUCTION: Kaposi sarcoma is a vascular tumour associated with human herpesvirus-8 (HHV-8) infection. Due to immunosuppressive treatment, solid organ transplant recipients infected with HHV-8 have increased risk of Kaposi sarcoma. The risk of being infected with HHV-8 is associated with higher age, male sex, non-White ethnicity, being a man who has sex with men (MSM) and use of recreational drugs. Post-transplant Kaposi sarcoma is rare in HHV-8 low-prevalence areas. We report the first case of a liver transplant recipient developing Kaposi sarcoma following donor-transmitted HHV-8 infection

Intranodal Hemangioma as a Differential Diagnosis of Melanoma Metastasis: A Case Report of a Rare Entity.

Dear editor, Haemangiomas are common benign vascular tumours usually detected on skin, mucosal surfaces, and soft tissues. In some extremely rare cases, haemangiomas can occur within a lymph node and as such are classified as primary benign vascular tumours of the lymph nodes, along with lymphangiomas, haemangioendotheliomas, and angiomyomatous hamartomas (1-2). A study reported that the overall prevalence of benign primary vascular intranodal tumours is only 0.3% (3). Even though intranodal haemangiomas are benign and usually asymptomatic, in clinical practice it is often challenging to diffe

Kaposi sarcoma in an HIV-infected patient with high CD4 count: a case report and literature review.

Kaposi sarcoma is a tumor commonly associated with human herpesvirus 8 (HHV-8) infection and is more prevalent in individuals with immunosuppression, such as those with HIV/AIDS, organ transplant recipients, or other conditions causing immune dysfunction. It typically presents as purple patches or nodules commonly affecting the skin or mucous membrane of the gastrointestinal tract, and can spread to the lymph nodes and lungs. This case report describes an 18-year-old male with vertical HIV infection, managed on a regimen of dolutegravir/lamivudine/tenofovir (DTG/3TC/TDF), and with a recent CD4

REST Is Restless in Neuronal and Non-Neuronal Virus Infections: An In Silico Analysis-Based Perspective.

Repressor element-1 silencing transcription factor or neuron-restrictive silencer factor (REST/NRSF) is an extensively studied neuronal gene regulator both in neuronal cells and non-neuronal cells. Even though the role of REST in host cellular gene regulation is well established, its role in the establishment of viral infections and its capability to stabilize and destabilize such viral infections are scarcely studied. Co-repressor and DNA modifiers are involved in REST-mediated repressive action of its target genes. The role of REST and co-repressors together or individually in the regulation
