Carmofur inhibits ovarian cancer cell proliferation, migration, and invasion via disrupting the YAP-1/TEAD pathway: in vitro and in silico approaches.

The current study aims to present a novel approach to alleviate the aggressiveness of ovarian cancer by pharmacological disruption of the Yes-Associated Protein (YAP) - 1/Transcriptional Enhancer Factor domain (TEAD) heterodimer structure using Carmofur. The Ovcar-3 cell line was incubated with progressively diluted Carmofur solutions. Proliferation inhibition was assessed using the MTT assay. Flow cytometry was employed to determine the pro-apoptotic properties. Cellular morphological alterations were assessed with confocal microscopy. The migration inhibition status was assessed via the scra

Malignant vascular neoplasms of the distal female genital tract: a series of 19 cases emphasizing unusual features and diagnostic challenges.

AIMS: Scarce literature exists regarding the spectrum of vascular neoplasms of the distal female genital tract. Herein, we describe the clinicopathologic features of a cohort of intermediate to malignant endothelial neoplasms of the vulva and vagina. METHODS AND RESULTS: Specimens were identified retrospectively from in-house and consultation files at our institution. Clinicopathologic data was obtained from review of the chart and available histologic material. Nineteen cases were identified in total. Kaposi sarcoma (n = 2) was of the enigmatic 'sporadic' form in elderly non-HIV females. Kap

Primary osseous kaposiform hemangioendothelioma confined to long bones: a retrospective imaging analysis of 12 pediatric cases.

BACKGROUND: Kaposiform hemangioendothelioma (KHE) is a rare, locally aggressive vascular tumor predominantly affecting infants and children. While musculoskeletal involvement is common, cases originating in and confined exclusively to bone are exceptionally rare, and their specific imaging characteristics are not well-defined. This study aims to characterize the clinical and detailed radiological features of primary, intraosseous KHE in a pediatric cohort. METHODS: We conducted a retrospective review of pediatric patients with biopsy-proven KHE admitted to our institution between January 2016

Fatal soft-tissue and bony complications following radiotherapy for HIV-negative Kaposi's sarcoma: a rare case of post-radiation necrotizing fasciitis and osteomyelitis in a resource-limited setting.

BACKGROUND: Kaposi's sarcoma (KS) is a vascular neoplasm strongly associated with HIV infection; however, HIV-negative KS is exceptionally rare, particularly in sub-Saharan Africa. Radiotherapy (RT) remains a standard local treatment for KS, providing excellent symptom relief and local control. Despite its efficacy, severe complications such as necrotizing fasciitis (NF) and osteomyelitis are exceedingly uncommon and scarcely reported in HIV-negative patients. CASE PRESENTATION: We report a 47-year-old HIV-negative man with biopsy-confirmed KS of the right thigh, initially treated with chemot

MRI and MDCT Findings of Kaposiform Hemangioendothelioma in the Oral Cavity of a Neonate: A Case Report and Literature Review.

INTRODUCTION: Kaposiform hemangioendothelioma (KHE) is a rare, locally aggressive vascular tumor of childhood. However, a detailed description of its imaging features, especially for neonatal oral cavity lesions, is lacking in the literature, with no prior reports detailing both prenatal magnetic resonance imaging (MRI) and postnatal multi-detector computed tomography (MDCT) findings. This report presents a case of neonatal KHE, focusing on its distinctive imaging manifestations on both modalities to provide diagnostic insights for this rare condition in an unusual location. Isolated Kaposi Sarcoma of the External Ear in an Immunocompromised Host: A Case Report.

BACKGROUND: A vascular neoplasm linked to HHV-8 infection, Kaposi sarcoma (KS) is common in immunocompromised patients, notably people living with HIV. Isolated external ear involvement is uncommon and may present clinically as keloids, cysts, or infections. CASE PRESENTATION: A 31-year-old man presented with a one-month history of ipsilateral dacryocystitis and a rapidly growing, painful right auricular mass. The auricular lesion was initially presumed to be a sebaceous cyst during his emergency room evaluation, with actively draining purulent material. Labs showed severe pancytopenia with a

Kaposi Sarcoma Associated with Pemphigus Vulgaris in the Setting of Immunosuppressive Therapy: Case Report and Review of Literature.

INTRODUCTION: Pemphigus vulgaris (PV) is a chronic autoimmune blistering disorder characterized by intraepidermal blisters affecting the skin and mucous membranes. The primary treatment for PV is systemic corticosteroid therapy, which is often prolonged due to the disease's chronic and relapsing course. The interplay between autoimmunity and long-term immunosuppressive treatment can predispose patients to secondary complications, including Kaposi sarcoma (KS), an angioproliferative tumor primarily involving the skin and soft tissues. When KS develops in the context of immunosuppressive therapy

Primary Kaposiform hemangioendothelioma of the tibia: report of a case with a previously undescribed GNAQ mutation.

Primary occurrence of Kaposiform hemangioendothelioma (KHE) in bone is extremely rare. We report the radiologic, histopathologic and molecular features of a case of KHE affecting the tibia of an adult male, who presented with pain in the left knee initially associated with previous trauma. Radiographs showed an eccentric well-defined lytic lesion of the tibial epiphysis. On MRI, the lesion showed hypo to isointensity on T1-weighted images and hyperintensity on STIR sequences. A CT-guided biopsy was pathologically inconclusive. MRI performed 11 months later showed lesion progression with cortic

Association between KSHV genotype and immune response to Kaposi sarcoma.

Kaposi sarcoma (KS) is a locally aggressive endothelial tumor that can be classified into four clinical types: classic, AIDS-related, iatrogenic, and endemic. KS usually affects the skin and soft tissues, but it can develop into mucosal, lymph node, and multiple-organ lesions. KS is caused by Kaposi sarcoma-associated herpes virus (KSHV), which has six genotypes (A, B, C, D, E, and F). However, it remains unclear whether the KSHV genotype affects clinical presentation. In this study, we investigated clinical stage, cellular proliferation, NF-κB activity, and lymphocyte infiltration around the

Extracavitary Primary Effusion Lymphoma: Diagnostic Challenges and Differential Diagnosis in a Resource-Constrained South African Setting.

AimsPrimary effusion lymphoma (PEL) is an aggressive, human herpesvirus 8 (HHV8)-associated large B-cell lymphoma characterised by malignant effusions in serous cavities (classic PEL). However, a subset of tumours may present exclusively as solid masses, referred to as extracavitary primary effusion lymphoma (EC-PEL). Despite the high regional prevalence of human immunodeficiency virus (HIV) and Kaposi sarcoma-associated virus /HHV8, PEL is likely under-reported in Sub-Saharan Africa. This study reports on the clinicopathologic features and differential diagnoses of EC-PEL.MethodsA retrospecti

Secondary changes in the peripheral liver due to kaposiform haemangioendothelioma in the hepatic hilum: a report of two transplant cases.

AIMS: Kaposiform haemangioendothelioma (KHE) is a rare vascular neoplasm of infancy and childhood that typically affects the soft tissues of the extremities; however, a variety of visceral sites have been reported. When KHE is located within the liver hilum, it may involve hilar structures and lead to secondary changes in the hepatic parenchyma. We report the histological changes in the explant livers of two patients transplanted for liver failure secondary to clinically unsuspected KHE at the hepatic hilum. METHODS AND RESULTS: Patient A initially presented with the signs and symptoms of liv

Harnessing Artificial Intelligence to Restore Standards in Medical and Dental Education in Pakistan.

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l

Relative Tumor Density of Soft-Tissue Sarcoma in Korean Population: An Institutional Review.

BACKGROUND: Comprehensive studies on the tumor burden of soft-tissue sarcoma (STS) by anatomical site are lacking in Asian populations. OBJECTIVE: To investigate the anatomical distribution of STS via relative tumor density (RTD) in a Korean cohort. METHODS: The RTDs of patients with STS at a single-institution from 2007-2022 were retrospectively analyzed. To describe the STS locations, the body was divided into 4 anatomical sites, and the RTD of each was calculated to the compare topographic tumor burden. RESULTS: Fifty-nine cases in 58 individuals, 35 male (60.3%) and 23 female (39.7%), w

Electrochemotherapy for cutaneous Kaposi Sarcoma: A European register-based cohort study from the International Network for Sharing Practices of electrochemotherapy (InspECT).

INTRODUCTION: Kaposi sarcoma (KS) is a cancer of endothelial cells involving the skin and visceral organs. Electrochemotherapy (ECT) is an alternative treatment option for patients with cutaneous malignancies of skin and non-skin origin not suitable for conventional treatments. ECT exerts anti-tumour activity through enhanced drug delivery to tumour cells and cytotoxicity, inducing immunogenic tumour cell death and through a vascular-disrupting effect. MATERIALS AND METHODS: In this multi-institutional prospective, observational study, we aimed to evaluate the outcomes of using ECT as a treat

Vascular lesions of head and neck region: A pictorial review.

Vascular lesions in the head and neck region encompass a wide spectrum of malformations and tumors, ranging from hemangiomas to arteriovenous fistulas. The International Society for the Study of Vascular Anomalies classification system categorizes these anomalies into vascular tumors and malformations. Vascular tumors are further divided into benign, borderline, and malignant types, while vascular malformations are classified based on flow characteristics as high or low flow. Accurate delineation of these lesions is crucial due to their clinical implications, particularly their location and po
