Endometrial fibrosis leads to subfertility when periglandular fibrosis impairs endometrial gland function. Identifying distinct protein expressions in mares suffering from this condition is essential for understanding the pathogenesis and developing treatments to delay fibrotic progression. To gain a better understanding of protein dynamics underlying this condition, Data-Independent Acquisition Mass Spectrometry was utilized to comprehensively characterize and compare the equine endometrial and plasma proteome from mares with different severities of endometrial fibrosis. Mares in diestrus wer

The endometrium is a hormonally responsive tissue that undergoes cyclic remodeling. Although endometrial organoids have been established in several species, detailed characterization remains limited. Here, we assessed the structural and molecular fidelity of equine endometrial organoids across reproductive cycle stages and during extended culture. Organoids were generated from biopsies collected during estrus and diestrus and analyzed using histology, immunohistochemistry, electron microscopy, and bulk- and single-cell RNA sequencing. Organoids formed polarized cystic structures composed of co

Intrauterine adhesion (IUA) is an adhesion of the uterine cavity or cervical canal resulting from damage to the basal layer of the endometrium, usually accompanied by fibrosis of the endometrium. This study analyzed endometrial samples from three IUA patients and three healthy control participants and screened for differentially expressed genes (DEGs) using RNA-seq and bioinformatics techniques. A total of 2402 DEGs were identified in endometrial tissues compared to normal endometrial tissues, among which microfibrillar-associated protein 5 (MFAP5) was upregulated in IUA tissues (log2FoldChang

We developed a hormonally responsive organoid model of the bovine endometrium. Bovine glandular epithelial cells isolated from reproductive tracts were cultured in an extracellular matrix hydrogel (Cultrex 2) at 38.5°C, 5% CO2 (n = 3). RNA was extracted and qPCR confirmed the presence of gland markers: leukemia inhibitory factor, mucin-1, insulin-like growth factor binding protein-1, kruppel-like factor-5, and forkhead box protein-A2. Organoids were imaged at specific time-points to monitor growth. Morphologically, organoids were spherical and fast-growing at passages 0 and 1, but this decline

Recent advances in the development of stem-cell-based embryo models and endometrial assembloids have fuelled understanding of their respective biology. However, a faithful combined approach is required to truly advance our understanding of implantation processes. This mini-review considers the most recent developments in producing reliable in vitro models of the human endometrium and human embryo, and the next steps required to combine their respective potential. While the fundamental biology of implantation is the primary driver of in vitro model development, the combined effort of embryo and

STUDY QUESTION: Can patient-derived organoid models be reliably established from diverse surgical phenotypes of endometriosis, and how do clinical factors such as hormonal treatment affect their growth success and morphology?
SUMMARY ANSWER: Endometriosis organoids can be established across all major surgical phenotypes with variable efficiency, and hormonal treatment at the time of biospecimen collection significantly reduces organoid establishment success.
WHAT IS KNOWN ALREADY: Organoid cultures have been developed from eutopic endometrium and select endometriosis tissue biospecimens prev

Pregnancy establishment in mammals requires a complex sequence of events, including bi-lateral embryo-maternal communication, leading up to implantation. This is the time when most pregnancy loss occurs in mammals (including humans and food production species) and dysregulation in embryo-maternal communication contributes to pregnancy loss. Embryo-derived factors modify the function of the endometrium for pregnancy success. We hypothesise that these previously unexplored conceptus-derived proteins may be involved in altering the function of the endometrium to facilitate early pregnancy events

Endometrial epithelial cell proliferation is essential for establishing endometrial receptivity during embryo implantation. In ruminants, embryonic interferon-tau (IFNT) mediates receptivity establishment by activating endometrial interferon-stimulated genes. This study investigates how IFNT-induced interferon alpha-inducible protein 6 (IFI6) regulates bovine endometrial epithelial cells (bEECs) proliferation. We identified the JAK2-STAT3 (rather than STAT1) as the primary signaling axis driving IFNT-mediated IFI6 expression. Functional experiments revealed that IFI6 knockdown markedly impaire

Non-receptive endometrium is one major cause of female fertility defects. While transcriptomic analyses have illuminated gene expression changes, the role of dynamic phosphoproteomic modifications remains under-investigated. We profiled the proteomic and phosphoproteomic landscape of the human endometrium during the establishment of endometrial receptivity at pre-receptive LH + 5, receptive LH + 7 and LH + 9 in which thousands of proteins and phosphor sites displayed dynamic changes. Among them, GSK3β protein levels were constant but Thr390 phosphorylation in human endometrium were increased f

Endometriosis (EMs) is a common gynecological disorder. According to the most widely recognized theory of retrograde menstruation, endometrial cells require completion of three key steps during ectopic implantation: adhesion, invasion, and angiogenesis. Although kisspeptin exerts anti-invasive and anti-angiogenic effects in multiple tumors, its potential inhibitory effects mediated through the KISS1 receptor (KISS1R) on EMs-related invasion and angiogenesis remain uncharacterized. This study aimed to identify regulatory genes in EMs pathogenesis via RNA sequencing and elucidate underlying mole

Embryonic implantation is a crucial developmental phase characterized by intricate molecular crosstalk between the embryo and endometrium, with emerging evidence implicating circular RNAs (circRNAs) as important regulators. This study elucidated the role of hsa_circ_0001550 in impairing embryo adhesion and evaluated recombinant human granulocyte colony-stimulating factor (rhG-CSF) as a therapeutic strategy. Overexpression of hsa_circ_0001550 in an in vitro adhesion model using Ishikawa cells reduced the adhesion capacity of BeWo or JAR spheroids and diminished the murine blastocyst implantatio

INTRODUCTION: Rural and remote communities across Canada face persistent physician shortages. Internationally trained physicians (ITPs) have become a critical component of the rural physician workforce, yet little is known about how specific workload characteristics influence physicians' perceptions of workload sustainability and long-term rural retention. Objectives of this study were to identify workload-related factors associated with perceived workload sustainability among ITPs practicing in rural Saskatchewan.
METHODS: We conducted a secondary cross-sectional analysis of prospectively co

Despite the proven efficacy of tamoxifen in breast cancer prevention, its uptake remains limited due to concerns over side effects. A phase 3 trial showed that low-dose tamoxifen (5 mg/d for 3 years) substantially reduces recurrence with minimal toxicity. After 10 years of follow-up, we observed no clinically significant differences in benign gynecological or breast events between the tamoxifen and placebo arms. The uterus was the most frequently affected site (30 cases per arm), followed by the breast (18 cases) and ovary (5 cases vs 3 cases for tamoxifen and placebo, respectively). Endometri

Endometrial cancer (EC) is the most common gynecological malignancy, yet reliable screening and diagnostic approaches remain limited. We developed a weakly supervised deep multi-instance learning model (DSMIL) to classify hematoxylin and eosin-stained whole-slide images (WSIs) of endometrial tissue. A total of 885 WSIs from 442 patients, including EC, atypical endometrial hyperplasia (AEH), endometrial hyperplasia without atypia (EH), and normal endometrium (NE), were analyzed. DSMIL achieved an average AUROC of 0.9776 for four-class classification, with inter-class AUROCs of 0.9876 for EC, 0.

The endometrium is the uterine lining that supports implantation and pregnancy. Existing in vitro systems only partly capture epithelial structure and function. We built a microfluidic model of the human endometrial epithelium using patient-derived organoids and defined a parameterized device and ECM conditions that yield a stable, polarized monolayer on chip. We specify the geometry, surface treatments, and collagen-based hydrogel or coating conditions, and we link these parameters to epithelial morphology and barrier integrity readouts. The epithelial layer maintains histologic features and

Obesity represents a major contributor to reproductive dysfunction during early mammalian pregnancy, yet the underlying mechanisms by which it impairs endometrial receptivity remain largely unknown. Through integrative metabolomic and transcriptomic profiling of uterine tissues from obese and control sows on gestational day 13, glutathione peroxidase 3 (GPX3) was identified as a key molecular determinant of endometrial receptivity. GPX3 expression was markedly reduced in the uterine tissue of obese sows and was closely associated with impaired receptivity. Functionally, GPX3 knockdown induced

The aim of this study was to develop an advanced in vitro model of human endometrium using single-cell-derived endometrial epithelial organoids, enabling the study of embryo secretome-endometrial crosstalk at the maternal-fetal interface. Single-cell-derived organoids generated from endometrial tissue of a parous 39-year-old woman recapitulated hormone-responsive decidualization, as demonstrated by expression of SPP1 and acetyl-α-tubulin. When cultured in embryo culture media, organoid viability was maintained with no cytotoxicity, but proliferation was suppressed, likely due to the lower conc
