Molecular identification and phylogenetic analysis of marmots in the Xinjiang's Altai Mountains, a newly discovered plague focus of China.

The Altai Mountains in Xinjiang have been recognized as a new plague focus in China since 2023. Although previous morphological studies suggested that marmots in this region belong to Marmota baibacina, this classification remained unconfirmed at the molecular level. In this study, 37 marmot samples were collected: including 10 M. baibacina from the northern Tianshan Mountains, 9 M. baibacina from the southern Tianshan Mountains, and 18 unverified individuals from the Altai Mountains. The COI, Cytb, and D-loop gene fragments were sequenced and subjected to phylogenetic analysis. Homology compa

ERAD inhibitor Eeyarestatin I (EerI) suppresses duck plague virus by restoring STING-mediated antiviral immunity.

Duck plague (DP), caused by the duck plague virus (DPV), is a highly contagious disease characterized by high morbidity and mortality, causing significant economic losses and biosecurity risks to the duck farming industry. Since current vaccines do not provide complete protection, the development of antiviral drugs is urgently needed. In this study, we evaluated the anti-DPV activity of the ER-associated degradation (ERAD) pathway inhibitor Eeyarestatin I (EerI) in vitro. Our results demonstrated that EerI suppressed DPV proliferation in duck embryo fibroblast (DEF) cells in a dose-dependent m

Development and evaluation of an assay for the detection of tick-borne encephalitis virus RNA via real-time PCR with reverse transcription.

BACKGROUND: Tick-borne encephalitis virus (TBEV, Orthoflavivirus encephalitidis) is an arbovirus of the family Flaviviridae. It is the etiological agent of tick-borne encephalitis (TBE), a severe disease affecting the central nervous system. Among arboviral infections, TBE represents the greatest burden in northern Eurasia, both in terms of emerging infection risk and mortality. Globalization and climate change increase the risk of TBEV introduction into nonendemic countries. They may also lead to the emergence of new viral variants featuring increased virulence for humans or altered antigenic

Functional characterization of UL50 gene reveals its essential role in duck enteritis virus replication and pathogenesis.

Duck plague, caused by a highly contagious α-herpesvirus, poses a major threat to waterfowl farming. Although UL50 homologs have been studied in mammalian α-herpesviruses such as HSV-1 and PRV, their role in avian herpesviruses remains unknown. Here, we investigated the function of the DEV UL50 gene, which encodes a conserved viral dUTPase, using bioinformatics, molecular biology, and virological approaches. Sequence analysis confirmed that DEV UL50 retains conserved catalytic motifs and structural features characteristic of α-herpesvirus homologs. A UL50-deleted mutant (ΔUL50) was constructed

Spatiotemporal analysis of plague risk in Tibet: Multi-source data-driven ensemble model development and validation.

BACKGROUND: Plague, caused by the highly lethal bacterium Yersinia pestis, remains a critical public health threat. Tibet's unique alpine environment fosters sensitive ecosystems, yet plague dynamics there are understudied. This study reveals environmental drivers and distribution evolution of plague foci in Tibet. METHODS: Using Tibet plague surveillance data (2000-2021), we integrated eight environmental variables (maximum temperature, precipitation, normalized difference vegetation index, land use and land cover, soil moisture, slope, aspect, and population spatial distribution). Ten algor

Edible baits for systemic flea control, plague mitigation, and wildlife conservation: evaluation of four active ingredients with three rodent species in western North America.

The flea-borne agent of plague, Yersinia pestis, is lethal to endangered black-footed ferrets (Mustela nigripes, BFFs) and the prairie dogs (Cynomys spp., PDs) on which BFFs depend for habitat and prey. We developed bait pellets containing insecticides for flea control with PDs. Individual baits contained 0.46, 0.91, 1.21, or 1.52 mg fipronil, 5.40 mg afoxolaner, 50.62 mg fluralaner, or 85.20 mg spinosad. From 2023 to 2025, we tested the baits with black-tailed PDs (C. ludovicianus, BTPDs), Gunnison's PDs (C. gunnisoni, GPDs), and Richardson's ground squirrels (Urocitellus richardsonii, RGSs).

Markers of tuberculous infection in 17th-century Lucca (central Italy): An investigation through granular impressions on the endocranial surface.

OBJECTIVE: Granular impressions (GIs) on the endocranial surface have been regarded as evidence of tuberculous meningitis. However, recent studies suggest that these subtle lesions more likely reflect disseminated tuberculosis. This study examines the frequency of GIs in an epidemic skeletal assemblage from Lucca (Italy) dating to the 17th century CE. MATERIALS: Excavations at the convent of St Francesco uncovered a cemetery archaeologically and historically linked to an epidemic outbreak, likely the plague of 1630-31. The assemblage comprises 98 individuals of poor preservation. METHODS: En

Tularemia: Historical Perspectives and Current Challenges of a Re-Emerging Zoonosis.

Tularemia is a plague-like, potentially fatal zoonosis caused by the coccobacillus Francisella tularensis. It was discovered at the beginning of the last century in the United States and was soon recognized in Japan and in the former Soviet Union as the cause of clinical conditions that had been known for one and two centuries, respectively. More than 250 animal species are susceptible to infection, with rodents and lagomorphs serving as key reservoirs, and several vectors may transmit the disease, mainly ticks and mosquitoes. Humans are incidental hosts and are infected primarily by two F. tu

Bacterial Strains from Soybean Nodules in the Lower Volga Region Belong to a New Subspecies Bradyrhizobium japonicum subsp. saratovii subsp. nov.

The isolation of locally adapted rhizobial strains with high symbiotic activity represents an effective strategy for increasing soybean yield under extreme environmental conditions. In this study, seven novel strains were isolated from nodules of soybeans grown in a greenhouse using field soil from the Lower Volga region. Five genomes were assembled into complete circular chromosomes, whereas two strains yielded near-complete chromosomes containing single repeat-mediated junctions. All strains had putative plasmids that were independently validated as circular by long-read mapping and confirme

Epidemic times: epidemic ontologies and temporalities in medieval islamic medicine.

Recent scholarship on the Black Death has uncovered crucial new insights into the disease and its spread, and has charted ways for historians to collaborate with the biological sciences. However, this approach has emphasized the primacy of a modern Western biomedical ontology that understands the disease and its spread through Yersinia pestis. In Islamic history, this meant a focus on tracing how medieval authors distinguished modern biological plague symptoms from those of other conditions, when and where the disease first emerged. Building on postcolonial methodological approaches, this arti

Host traits shape flea infestation patterns in small mammals: a case study of Spermophilus undulatus and associated flea species in northern Xinjiang, China.

OBJECTIVE: Fleas are obligate ectoparasites of mammals and play an important role in the transmission of zoonotic pathogens, including Yersinia pestis. Describing empirical patterns of flea infestation across host communities is essential for understanding host-parasite associations in plague-endemic regions. The aim of this study is to investigate the relationship between fleas and their host animals by examining how host traits influence flea parasitism, with the goal of offering novel insights for future plague prevention strategies and related research. METHODS: From 2022 to 2025, small m

Assessing the effects of fluids and antibiotics in an acute murine model of sepsis: study protocol for the National Preclinical Sepsis Platform-01 (NPSP-01) Study.

BACKGROUND: Sepsis remains a leading cause of mortality in critical care. Despite extensive preclinical research on sepsis pathophysiology, the development of effective therapies has been largely unsuccessful. Key obstacles include limited construct validity of animal models, insufficient methodological rigor and the lack of collaborative frameworks akin to clinical trials. These issues plague not only sepsis research, but preclinical research in general. The National Preclinical Sepsis Platform (NPSP), an interdisciplinary network under Sepsis Canada, addresses these challenges in sepsis rese

Ecologic Investigative Strategies to Determine Human Plague Exposure Sites, United States, 1991-2018.

Plague is a rare but potentially life-threatening fleaborne zoonotic disease caused by Yersinia pestis. Public health agencies in the United States use multiple concurrent epidemiologic and ecologic strategies to determine Y. pestis exposure sites. We reviewed 196 plague case files from 1991-2018 to describe effort and yield of implemented strategies. All files included an epidemiologic component, and 71% were followed up with environmental investigations. Environmental samples were collected for laboratory testing in 88% of investigations. The percentages of investigations yielding laboratory

The plague of 1720 and migration in Martigues (France) in the 17th and 18th centuries.

One-off events such as wars or epidemics can change the structure of populations, by encouraging the mobility or causing the death of certain categories of people. They can also lead to a demographic slump, made up for by the arrival of new migrants. The town of Martigues (Bouches-du-Rhône, France) provides an example of this kind of local population renewal. Between the seventeenth and eighteenth centuries, its population was partially renewed after the plague of 1720. To measure the effect of the plague as a disruptive event, data on the surnames of people born in Martigues before and after

An evolutionary perspective on the nucleocytoplasmic shuttling mechanism of duck plague virus UL54 protein.

Duck plague virus (DPV), a member of the alpha-herpesvirus, causes duck plague (DP), thereby posing a serious threat to the waterfowl industry. The DPV UL54-encoded protein shuttles between the nucleus and cytoplasm to modulate viral replication. However, the role of the UL54 protein in the evolutionary process remains unknown. This study is conducted from an evolutionary perspective to explore the nuclear-cytoplasmic shuttling characteristic of UL54 protein. First, we analyzed the co-evolution on DPV whole genome and its UL54 gene using molecular evolutionary methods. Next, we constructed phy

Temporal shifts in polygenic traits track major epidemics in Western Eurasia.

Infectious diseases are recognized as one of the strongest selective forces, exerting a profound influence on the genetic makeup of human populations over time. Recently, large-scale genome-wide association studies (GWAS) on immunological traits have underscored the notion that genetic predisposition to infectious diseases likely stems from the contribution of several thousand loci across the human genome. To model the polygenic inheritance of these traits, multiple variants can be combined into polygenic risk scores (PRS), which estimate an individual's genetic predisposition for a trait. By

Changing drivers of the Great Atlantic Sargassum Belt from physical forcing to ecological control.

Inundations of pelagic Sargassum plague the tropical Atlantic, with size and impacts steadily increasing to surpass 30 million tons in 2025. Understanding the drivers of Sargassum growth in the so-called Great Atlantic Sargassum Belt is fundamental to developing effective mitigation strategies for affected nations. We present a nonlinear regression model that both explains the seasonal and interannual variability observed between 2011 and 2022 and predicts Sargassum concentrations in 2023 and 2024. The growth of Sargassum, initiated by a prolonged negative phase of the North Atlantic Oscillati
