Treatment of bacterial biothreat agents with a novel purified bioactive lactoferrin affects both growth and biofilm formation.

Lactoferrin is known to exhibit broad spectrum activity against a multitude of bacteria, fungi, and viruses due to its multi-functional mode of action. Recently, Lactea Therapeutics and its affiliates have developed a novel, patent-pending technology to purify naturally derived bovine lactoferrin (Lactea Lf) for use as a medical countermeasure that was not previously available. To assess the efficacy of Lactea Lf against biothreat pathogens, we performed biofilm inhibition assays and generated dose-response curves against Burkholderia pseudomallei, Burkholderia mallei, and Francisella tularens

New insights into the evolutionary origin of Francisella tularensis subsp. holarctica.

BACKGROUND: As a global expansion of Francisella tularensis subsp. holarctica in the northern hemisphere, its origin has been of great interest. Based on the phylogenetic analysis of the whole genome sequences (WGS) available in public databases, estimating temporal signal in accordance with random permutations of sampling dates, and searching the powerful SNPs within the B.16 clade, we intend to explore the historical origin of this subspecies and routes of dispersal across its regional distribution. METHODS: We constructed a phylogenetic tree for the four main clades based on 395 WGSs, incl

Unusual increase in tularemia incidence in Alsace, Northeastern France in 2024.

Tularemia is a bacterial zoonosis endemic to the Northern Hemisphere, transmitted through various modes and presenting with diverse clinical manifestations. In recent years, the number of notifications (tularemia is a reportable disease in France) have increased and the spectrum of clinical forms have changed. We conducted a retrospective study of tularemia incidence diagnosed in 2024 at two major hospitals in Alsace, a small region of Northeastern France. This study included 10 cases (7 men and 3 women; mean age = 52 years) reported in 2024. Clinical forms comprised six ulceroglandular or gla

An Outbreak of Pulmonary Tularemia in Slovenia in Summer 2024.

Background: Tularemia is a rarely identified disease in Slovenia. In summer 2024, we detected a tularemia outbreak in the Kranjsko-Sorško polje, located in North-Western part of Slovenia. Aim: To describe the epidemiological investigations and preventive measures to contain the outbreak. Methods: The patients with confirmed tularemia were interviewed. Serology and PCR was used for microbiological confirmation of tularemia and in some patients by isolation from blood or by RT-PCR. Results: The majority of confirmed tularemia cases in 2024 were infected in the geographically limited area in Nort

The stealth Francisella: The first fatality caused by Francisella novicida bacteraemia in Malaysia.

Francisella novicida is a rare opportunistic pathogen that is closely related to Francisella tularensis subspecies tularensis and subspecies holartica. However, it does not cause tularaemia and is not transmitted to humans through arthropod bites. This case is the first time this bacterium has been isolated from a patient in Malaysia. The patient was a 69-year-old woman with hypertension and dyslipidaemia, who unfortunately passed away due to the infection. She presented with infective gastroenteritis and deteriorated into full blown Gram-negative bacteraemia. Despite treatment with meropenem,

Guidelines for tick-borne diseases (excluding Lyme borreliosis): diagnosis and management.

Other tick-borne microorganisms can cause symptoms that differ from those of Lyme borreliosis, often with a more acute course. Fever is the primary clinical warning sign, which may be accompanied by various other symptoms-cutaneous, neuromeningeal, and lymphatic-that help guide diagnosis. A clinical approach makes it relatively straightforward to distinguish Lyme borreliosis from other tick-borne diseases, with the common factor being the tick as the vector. Biological abnormalities (anaemia, thrombocytopenia, cytolysis, etc.) are often present in tick-borne diseases, unlike in Lyme borreliosi

Activation of B1 B cells by F. tularensis atypical LPS depends on classical complement and C3a.

Francisella tularensis (Ft), a Gram-negative bacterium that causes tularemia, possesses a non-inflammatory atypical LPS (LPSFt) that is highly immunogenic through unknown mechanism. We previously showed that immunization with LPSFt, a type 2 T-independent (TI) antigen, elicits protective LPSFt-specific IgM (IgMFt) and IgG3Ft by B1 cells in a mechanism dependent on the IL-5 produced by innate lymphoid cells type 2 (ILC2). Here, we examined the role of complement in the B1 cells' response against LPSFt. C3-/-, C1q-/- and C4-/- mice immunized with LPSFt failed to produce IgMFt and IgG3Ft. In cont

Animal modelling with the Francisella tularensis subspecies holarctica strain OR96-0246.

Tularemia is a zoonotic disease caused by Francisella tularensis. Most human cases are caused by F. tularensis ssp. tularensis (type A) or F. tularensis ssp. holarctica (type B), with the former considered more virulent. For this reason, type A isolates are often the benchmark for the testing of new vaccines or antibiotics. However, both subspecies cause considerable disease and can differ in their responsiveness to medical countermeasures. Accordingly, there is a need to identify and characterize representative type B isolates that are available to qualified research institutions to ensure th

A clinical pitfall in caseating necrotizing granulomatous lymphadenitis: Tularemia.

Tularemia, caused by Francisella tularensis, is a zoonotic infection that typically presents with lymphadenopathy, acute tonsillitis, or ulcerative skin lesions. Various infectious agents and malignancies may underlie lymphadenopathy. Although necrotizing granulomatous reactions are common in suppurative lymphadenitis, caseating granulomas are rare and often raise suspicion for tuberculosis. We report two tularemia cases presenting with lymph node enlargement and caseating granulomas on histopathology. Case 1 involved a 27-year-old woman with axillary lymphadenopathy and no systemic symptoms.

In-silico core proteome analysis for chimeric vaccine development against tick-borne tularemia.

Tularemia is an extremely contagious zoonotic illness resulting from infection with the intracellular bacterium Francisella tularensis. It is transmitted primarily via vector bites particularly from ticks, flies, and mosquitoes and is a severe public health threat. Because of its high virulence, low infective dose, aerosol transmissibility, and potential for mass casualties, F. tularensis is also considered a potential biological warfare agent. Despite its severity, there is presently no licensed vaccine against this pathogen. In the present work, a subtractive proteomics pipeline was implemen

A Comprehensive Review of Nanoadjuvants in Cancer Vaccines and Their Immunomodulatory Role and Clinical Applications.

INTRODUCTION: Hepatitis C virus (HCV) is a viral infection affecting 71 million people worldwide. A high prevalence of co-infection has been observed with parasitic infections, such as Schistosoma mansoni, Fasciola sp., and Toxoplasma gondii, all of which can contribute to the progression of liver disease. This study aimed to investigate the prevalence of co-parasitic infections with HCV-positive individuals within Egyptian populations and the resulting biochemical changes in liver and kidney biomarkers. MATERIALS AND METHODS: A total of three hundred and thirty-seven blood samples were scree

Tularemia With Necrotizing Mediastinal Lymph Nodes.

Tularemia, also known as rabbit fever, is a rare zoonotic infectious disease transmitted by air, arthropods, food, and water caused by bacteria of the genus Francisella. Because of its multiple transmission routes and low infectious doses, Francisella tularensis is listed as a biological weapon. In rare cases, tularemia can lead to pneumonia and necrotizing mediastinal and hilar lymph nodes. Herein we present the case of a 43-year-old female patient with an isolated mediastinal mass, initially thought to be a solid tumor. Further investigations led to the diagnosis of tularemia. The patient's

Tularemia for clinicians: An up-to-date review on epidemiology, diagnosis, prevention and treatment.

Tularemia, also known as rabbit fever, caused by Francisella tularensis, is re-emerging in the Northern Hemisphere and in particular in Europe in recent years. It is a bacterial zoonotic, arthropod-borne, air-borne, food- and water-borne disease and the multiple routes of infection result in six typical clinical pictures (ulceroglandular, glandular, oculoglandular, oropharyngeal, typhoidal, and pneumonic) along with several other uncommon presentations, potentially affecting any body site. In addition, due to the ease of airborne transmission, F. tularensis is a category A agent for bioterrori

Tularemia: A Unique Presentation for a Rare Disease.

This report highlights an uncommon presentation of tularemia in a high-risk pediatric patient to increase awareness and broaden the differential diagnosis among clinicians. A previously healthy eight-year-old female presented to the emergency department multiple times within two weeks with nonspecific flu-like symptoms, worsening purulent conjunctival injection, fever, and left-sided facial swelling. She was initially diagnosed with several conditions, including corneal abrasion, preseptal cellulitis, and dacryocystitis, and was treated with antibiotics without symptom resolution. Upon admissi

A case report of ulceroglandular tularemia.

Tularemia, caused by the bacterium Francisella tularensis, is a zoonotic disease that has been reported in many countries across the globe. It may manifest in many forms, like pneumonic, oropharyngeal, glandular, ulceroglandular, or oculoglandular. In Poland, one of the most common types of tularemia is ulceroglandular. Typical symptoms of ulceroglandular tularemia include painful ulcers at the site of the insect or animal bite, along with systemic flu-like symptoms. Tularemia is generally effectively treated with appropriate antibiotics, however, frequent misdiagnosis and subsequent improper

[Tularaemia acquired in Denmark].

Tularaemia is a rare infection caused by Francisella tularensis. We present three cases of ulceroglandular tularaemia. The patients were diagnosed within the same month and had possibly been exposed to the bacteria in Djursland, Denmark. All patients were treated with antibiotics for weeks. Two patients needed surgical revision of suppurative lymphadenitis before complete remission of their symptoms. Tularaemia should be considered if a patient presents with fever, ulcer and regional lymphadenopathy, especially if there is a history of tick or mosquito bites or direct contact with animals.

Organic electrolyte gated field effect transistor for detecting alpha synuclein longitudinally in Parkinsonism mouse model.

BACKGROUND: Enthesopathy is an evolving area for applied clinical research. MRI is the gold standard in the diagnosis of elbow joint pathology, but recent reports indicate that ultrasound imaging is more sensitive and accurate than MRI in detecting enthesopathy of the heels and knees. Too many patients are under-diagnosed and/or misdiagnosed because the early pathological changes of enthesitis in the different types of seronegative arthropathies are not detected. OBJECTIVES: This study was undertaken to describe the ultrasound features of elbow enthesitis in patients with seronegative arthrop

Characterization of the ciliary beating efficiency in primary diffuse chronic rhinosinusitis.

The Pacific coast tick, Dermacentor occidentalis Marx, is found throughout California and can harbor agents that cause human diseases such as anaplasmosis, ehrlichiosis, tularemia, Rocky Mountain spotted fever and rickettsiosis 364D. Previous studies have demonstrated that nonpathogenic endosymbiotic bacteria can interfere with Rickettsia co-infections in other tick species. We hypothesized that within D. occidentalis ticks, interference may exist between different nonpathogenic endosymbiotic or nonendosymbiotic bacteria and Spotted Fever group Rickettsia (SFGR). Neuroinvasive Francisella tularensis Infection: Report of 2 Cases and Review of the Literature.

BACKGROUND: Neuroinvasive infection with Francisella tularensis, the causative agent of tularemia, is rare. Establishing clinical suspicion is challenging if risk factors or clinical features classically associated with tularemia are absent. Tularemia is treatable with antibiotics; however, there are limited data to inform management of potentially fatal neuroinvasive infection. METHODS: We collected epidemiologic and clinical data on 2 recent US cases of neuroinvasive F. tularensis infection, and performed a literature review of cases of neuroinvasive F. tularensis infection published after
