Papillary thyroid cancer (PTC) typically has a favorable prognosis. However, in some cases, the presence of angioinvasion can result in local invasion, recurrence, and distant metastasis, leading to poorer outcomes. Identifying disrupted pathways associated with angioinvasion could lead to the discovery of novel angioinvasive markers in PTC, which, in turn, may suggest more aggressive clinical management strategies, as radioiodine therapy (RAI). This study aims to identify potential markers associated with angioinvasive PTC, with a particular focus on nitrosative stress and endothelial damage.

BACKGROUND: Per- and polyfluoroalkyl substances (PFAS) comprise a class of persistent environmental pollutants with potential carcinogenic effects, but their impact on childhood cancer remains underexplored. A child's exposure to PFAS can occur through various pathways postnatally, including contaminated food, water, and consumer products; and in utero, as PFAS can cross the placenta.
METHODS: To investigate the association between early-life PFAS exposure and the risk of childhood acute lymphoblastic leukemia (ALL), we analyzed archived blood samples from children diagnosed with ALL and matc

The extreme lethality and limited treatment options for pancreatic ductal adenocarcinoma (PDAC) underscore the urgent need for innovative therapeutic strategies. This study presents the first preclinical investigation of a cell-free gene-directed enzyme prodrug therapy (GDEPT) based on conditioned medium (CM) from placenta-derived mesenchymal stem cells (PlacMSCs) engineered to express the yeast cytosine deaminase::uracil phosphoribosyltransferase (yCD::UPRT) fusion enzyme. The CM was concentrated tenfold (cCM) and characterized by proteomics, transmission electron microscopy, and Western blot

BACKGROUND: Choriocarcinoma (CC), a highly aggressive and malignant subtype of gestational trophoblastic disease (GTD), arises from the dysregulated proliferation of trophoblastic cells, which normally mediate placental development during pregnancy. This study aimed to characterize the expression and functional significance of metastasis-associated lung adenocarcinoma transcript 1 (MALAT1), a nuclear-enriched long non-coding RNA, in CC pathogenesis.
METHODS: We analyzed MALAT1 expression levels in 30 normal placental villi, 46 hydatidiform moles (repressive subtype), and 52 CC specimens. To i

Polycystic ovary syndrome (PCOS) is a common endocrine disorder with a complex pathogenesis that includes disordered follicle development, hypothalamic-pituitary-ovarian (HPO) axis dysfunction, hyperandrogenemia, and insulin resistance. The risk of complications during pregnancy, such as gestational diabetes mellitus (GDM) and preeclampsia (PE), among PCOS patients is higher than that in the general population. Kisspeptin (KP) is a peptide hormone widely expressed in the hypothalamus, limbic system, gonads, pancreas, and liver; it is highly expressed in the placenta and is considered to play a

Estrogen receptor positive breast cancer (ERBC) is among the most common type of breast cancers. Currently aromatase inhibition is the mainstay therapy of ERBC. However, the emergence of drug resistance, and relapse of tumor progression has created the need of identification of new targets for drug development against ERBC. Hence, steroid sulfatase (STS) has emerged as a new drug target for the treatment of ERBC, as it is involved in the biosynthesis of hormones, including estrogens and androgens. The current study focuses on the purification of STS from human placentae, followed by identifica

BACKGROUND: Placental mesenchymal dysplasia (PMD) is a placental abnormality resembling partial hydatidiform moles without trophoblastic proliferation. Although many PMD cases involve androgenetic/biparental mosaicism or chimerism, we recently reported that approximately 30% of cases retain biparental genomes (BiPMD) and exhibit aberrant methylation at multiple imprinted differentially methylated regions (DMRs). This resembles multilocus imprinting disturbances (MLIDs) and biparental hydatidiform moles (BiHMs), which are associated with pathogenic variants in subcortical maternal complex (SCMC

Stromal regulation of cancer dissemination is well recognized, however causal genes remain unidentified. We previously demonstrated that epitheliochorial species have acquired stromal resistance to placental invasion, correlating with reduced rate of cancer malignancies, identifying stromal genes correlating with depth of placental invasion called ELI (Evolved Levels of Invasibility) genes. Similarly, decidualization of human endometrial fibroblasts confers resistance to placental invasion. We hypothesized that both trajectories may convergently use similar pathways, providing an opportunity t

Yin Yang 1 (YY1) is a highly conserved multifunctional transcription factor with multiple biological functions including cell proliferation, differentiation and transcriptional regulation. In recent years, the discovery of the functional activity of YY1 in the reproductive tissues of animals, including humans has attracted more and more attention. YY1 plays important roles in the development of follicular, embryo and placenta, as well as spermatogenesis. In addition, the role of YY1 in reproductive diseases including infertility, polycystic ovarian syndrome, recurrent spontaneous abortion (RSA

Cholangiocarcinoma (CCA) is an aggressive bile duct malignancy with poor survival rates due to late detection, rapid metastasis, and treatment resistance. Therefore, new therapeutic strategies are needed to improve treatment outcomes. Previous studies have reported that mesenchymal stem cells (MSCs) secrete several soluble factors that modulate intracellular signaling pathways that are critical for the regulation of cancer cell function. The present study aimed to investigate the effects and molecular mechanisms of conditioned media from chorion-derived human MSCs (CH-CM) and placenta-derived

Placental mesenchymal dysplasia (PMD) and hepatic mesenchymal hamartomas are rare diseases associated with Beckwith-Wiedemann Syndrome (BWS). We present the case of a 22-year-old diagnosed with PMD at 24 weeks gestation who required emergent delivery secondary to fetal distress and preeclampsia at 30 weeks' gestation. The neonate was diagnosed with a hepatic mesenchymal hamartoma following delivery. While genetic testing returned negative, a clinical diagnosis was made for BWS due to persistent hypoglycemic episodes, left lower extremity hyperplasia, an umbilical hernia, nephromegaly, bilatera

BACKGROUND: Complete hydatidiform mole with a coexisting fetus (CMCF) is a rare form of twin pregnancy associated with high maternal and perinatal risks, posing complex diagnostic and therapeutic challenges. The standard approach ranges from termination of pregnancy to conservative management to fetal viability. Recent evidence suggests that spontaneous regression of molar tissue may occur, supporting conservative treatment in selected cases. We present a new case and discuss the clinical consequences of this phenomenon.
CASE: We report a 29-year-old woman with a history of a previous molar p

INTRODUCTION: Whilst about 18 % of all cancers in females are gynaecological cancers, more than 50 % of these can be classified as rare tumours (defined as an annual incidence of <6 per 100,000). Such cancers represent an important challenge for small countries like Malta where the small caseload may limit the expertise of clinicians in the diagnosis and treatment of such cancers.
METHODOLOGY: The study uses data from the Maltese population-based cancer registry to examine trends in incidence rate of the rare gynaecological cancers for the 12-year period between 2010 and 2021. It employs the

Cancers of female genital tract (FGTC) account in Europe for 14 % of all female malignancies, and many of them are rare. FGTC epidemiological indicators are usually reported by anatomical site, but they are a heterogeneous group of diseases in terms of morphological characteristics and clinical management. We show incidence and survival population-based estimates and time trends in the European population for FGTC, defined combining topography and morphology codes. We analyzed data of females aged 15 years or more diagnosed with 25 FGTC of which 23 rare. Data were provided by 100 European popu

Deciduosis is a benign condition characterised by the transformation of subepithelial connective tissue into decidual-like cells, typically occurring in response to hormonal changes during pregnancy. Although benign, cervical deciduosis may macroscopically resemble malignant lesions, leading to diagnostic uncertainty. This report presents two cases of cervical deciduosis diagnosed in early pregnancy. Both patients presented with first-trimester vaginal bleeding, and speculum examination revealed extensive wart-like lesions covering the cervix. Histopathological examination of cervical biopsies

INTRODUCTION: Gestational Trophoblastic Disease (GTD) encompasses a spectrum of placenta-related disorders with significant health risks. Despite the varying reported prevalence rates in East Africa, no comprehensive meta-analysis has been conducted to synthesize the overall burden and predictors of GTD in this region.
METHODS: Following the PRISMA guidelines, we conducted a systematic review and meta-analysis of studies from PubMed, Cochrane Library, and Google Scholar (search period: 2000-2024). We assessed heterogeneity (I2 and Q tests), pooled estimates using a random-effects model, and e
