Silicone Oil-Assisted Removal of Large Intraocular Foreign Bodies.

PURPOSE: To describe a silicone oil-assisted technique for removal of large intraocular foreign bodies (IOFBs) through an enlarged sclerotomy. METHODS: Description of the surgical technique in two pediatric patients with large posterior-segment IOFBs. RESULTS: Two pediatric patients had large IOFBs, one involving the macular area and the other in the subretinal space. Because of the size of the IOFBs, an air/silicone oil (SO) exchange was performed before extraction. An enlarged sclerotomy was created, and the IOFB was removed with retinal forceps in one case and an external magnet in the ot

Scanning Electron Microscopy-Energy Dispersive Spectroscopy Evaluation of a Retinal Explant Model of Iron Intraocular Foreign Body Injury.

Iron intraocular foreign body (IOFB) can cause retinal damage, which may be related to the release, transfer, and tissue infiltration of iron particles. Sustained retinal impairment may occur after surgical removal of an iron IOFB, while direct in situ evidence of residual retinal iron deposition remains limited. A retinal explant model combined with regional scanning electron microscopy (SEM)-energy-dispersive spectroscopy (EDS) was used to examine retinal morphology and elemental distribution after iron foreign body exposure and removal. SEM analysis showed that, in the Fe group, the retinal

Three Nitrogen Atoms Turn Old Kinase Inhibitors Into New Targetable Remedy.

Pathological neovascularization in the eye is a significant contributor to vision loss in diseases such as age-related macular degeneration and diabetic retinopathy. While anti-VEGF biologics are effective, they require repeated intravitreal injections. Here, we report a novel principle for designing photoactivatable VEGFR2 inhibitors, along with two examples, EYE1090 and EYE1118. Azido-functionalization in these molecules enables light-triggered receptor binding while preserving potent inhibition in the dark. Both compounds exhibit significantly enhanced activity upon exposure to green light

A Peripheral Sulfonium Moiety Engineering Strategy to Enhance AIEgen-Based Antitumor Photodynamic Therapy.

The therapeutic efficacy of photosensitizers (PSs) strongly depends on their capacity to efficiently generate reactive oxygen species (ROS). Herein, we report a peripheral sulfonium moiety engineering strategy and identify a lead aggregation-induced emission (AIE)-based PS, S12. The sulfonium moiety enhances the photosensitization of S12 primarily by increasing spin-orbit coupling (SOC) and reducing ΔEST, thus boosting ROS generation. Moreover, the sulfonium unit enables S12 to simultaneously produce both Type I and Type II ROS, rendering its photosensitization weakly dependent on O2 concentra

Activating transcription factor 6 is involved in the beneficial effects of growth hormone-releasing hormone antagonists in the corneal endothelium.

The corneal endothelium maintains eye transparency through fluid regulation, and its dysfunction has been associated with impaired vision. This study is focused on the possible involvement of activating transcription factor 6 (ATF6) in the beneficial effects of Growth Hormone - Releasing Hormone antagonists (GHRHAnt) in corneal cells. ATF6 is a UPR sensor involved in barrier function, previously shown to exert anti-inflammatory activities. The aim of this study is to evaluate involvement of ATF6 in the effects of GHRHAnt in the corneal endothelium. MTT assay was used to measure the cell viabil

Supercritical Carbon Dioxide-Peracetic Acid Treatment Inactivates Low-Level Bacterial Inocula in Collagen Analogue-Phosphorylcholine Hydrogels while Preserving Functional Properties.

Terminal sterilization of hydrogels is challenging because their high-water-content, chemical structures, and cell-friendly properties are often altered by conventional sterilization methods. Hydrogel implants comprising collagen-like peptide (CLP), polyethylene glycol (PEG), and poly (2-methacryloyloxyethyl phosphorylcholine) (MPC) showed promise in promoting corneal regeneration in preclinical animal studies. However, before these CLP-PEG-MPC implants can be used clinically, they must be sterilized effectively to ensure patient safety. Here, we evaluated supercritical carbon dioxide (scCO2)

Tissue-specific inflammatory responses to systemic LPS in mice with LysM-lineage Hmgb1 deletion.

BACKGROUND: Systemic lipopolysaccharide (LPS) exposure induces peripheral inflammation that can engage neuroimmune pathways. High-mobility group box 1 (HMGB1) may contribute to peripheral-central communication, but the role of lysozyme M (LysM)-expressing cells in circulating HMGB1 and compartment-specific inflammatory responses remains unclear. METHODS: Male 7-8-week-old HMGB1 fl/fl LysM-Cre +  mice and HMGB1 fl/fl LysM-Cre - littermate controls received LPS (5 mg/kg, i.p.) or saline. Plasma HMGB1, midbrain inflammatory gene expression, cervical lymph node (CLN) transcriptional responses, an

Micromotor-enabled platform for early, minimally invasive monitoring of amyloid pathology along a putative plasma-brain-retina axis in a transgenic islet amyloid polypeptide mouse model.

Growing evidence indicates that metabolic dysfunction contributes to neurodegeneration, leading to the "type 3 diabetes" hypothesis. In this context, human islet amyloid polypeptide (hIAPP), a highly amyloidogenic peptide, has emerged as a potential molecular link between peripheral metabolic dysfunction and neurodegeneration. Here, we developed a hybrid ex vivo platform integrating micromotor (MMGO-AuNPs)-based electrochemical aptassay for the detection of circulating amyloid-β42 oligomers (AβO42) in human IAPP-overexpressing transgenic (Tg) and wild-type (Wt) mice, under high-fat diet (HFD).

Employing the Role of SLC40A1-Mediated Microglia Inflammatory Response in Diabetic Retinopathy Based on Single-Cell Transcriptomics Technology.

Diabetic retinopathy (DR) is one of the most severe complications of diabetes. Existing treatments are effective but still have many limitations. Thus, it is particularly crucial to uncover novel therapeutic targets and develop more efficacious treatment strategies. In this study, single-cell datasets of retinal and normal retinal tissues of DR patients were obtained from the Gene Expression Omnibus (GEO) database. Using cell annotation, Gene Ontology (GO) pathway enrichment analysis, and cell trajectory analysis, key cell subgroups and their enriched pathways affecting DR progression were sel

Supervised Reduction Protocol for Management of Herpetic Stromal Keratitis.

PURPOSE: Herpetic stromal keratitis (SK) caused by herpes simplex virus (HSV) or varicella zoster virus (VZV) can cause sight-threatening corneal scarring. Given high recurrence rates of herpetic SK with steroid reduction and limited consensus on treatment duration, we examine outcomes of an approach we term the supervised reduction protocol (SRP). METHODS: Among 162 patients with ocular HSV or VZV evaluated between September 2016 and June 2025, 53 had SK managed with SRP. Under the SRP, patients underwent frequent slit-lamp evaluation allowing for early detection and management of stromal in

The evolving landscape of fungal keratitis: from emerging pathogens to precision diagnostics and novel therapeutics.

Fungal keratitis (FK) remains a major cause of vision-threatening corneal infection, especially in tropical and subtropical regions, with an expanding epidemiological spectrum driven by environmental, climatic, occupational and host-related factors. Fusarium and Aspergillus were the most common pathogens reported. Greater etiological diversity is reflected by increasing recognition of dematiaceous fungi, uncommon molds, and cryptic species. The literature search was performed by searching bibliographic databases (PubMed and Google Scholar), covering publications from January 2000 to August 202

Identification of key cell subpopulations and associated biomarkers for mouse dry eye disease based on the Scissor-CIBERSORTx algorithm integration strategy.

Dry eye disease (DED) is characterized by tear film instability and chronic inflammation, with the cornea serving as a key target site vulnerable to inflammatory damage. This study integrated single-cell and bulk transcriptomics combined with Scissor and CIBERSORTx algorithms to screen corneal key cell populations and biomarkers in murine DED. Corneal wing cells were identified as core disease-associated cells, among which interferon-response wing cells (IFN-response-wing) were the pivotal pathogenic subtype. Three IFN-response-wing signature biomarkers Upk3b, Dusp1 and Ly6e were screened, enr

Liraglutide reduces lipid accumulation in human orbital fibroblasts in Graves' orbitopathy.

Graves' orbitopathy (GO) is characterized by proliferation and differentiation of orbital fibroblasts. Glucagon-like peptide-1 receptor agonists (GLP-1RAs) are widely used for metabolic disorders and have been reported to modulate cell proliferation and differentiation. The present study aimed to examine the effects of GLP-1RAs on key fibroblast-mediated pathogenic processes relevant to GO using human orbital fibroblasts. Orbital fibroblasts were isolated from patients with GO and non-GO controls. Cells were treated with the GLP-1RAs liraglutide (100 nM) or semaglutide (200 nM). Fibroblast pro

[Bardet-Biedl syndrome-diagnosis, care and treatment approaches].

The Bardet-Biedl syndrome (BBS) is a genetically determined multiorgan disorder characterized by congenital postaxial polydactyly, obesity, hyperphagia and retinal dystrophy that develops in childhood. The condition is heterogeneous both in terms of molecular genetics and clinically: it is caused by mutations in at least 26 genes, and the variable extraocular manifestations include developmental delay, diabetes mellitus and fatty liver. The retinal dystrophy is also heterogeneous and presents with a broad phenotypic spectrum. While there is no specific treatment option for the retinal dystroph

Tolosa-Hunt Syndrome: A Diagnostic Challenge and the Complexities of Long-Term Corticosteroid Management.

Tolosa-Hunt syndrome (THS) is an uncommon cause of painful ophthalmoplegia resulting from idiopathic granulomatous inflammation involving the cavernous sinus, superior orbital fissure, or orbit. Its diagnosis remains challenging because early manifestations may be nonspecific and its clinical and radiological features overlap with several vascular, infectious, inflammatory, and neoplastic conditions. We report the case of a 59-year-old woman with type 2 diabetes mellitus, arterial hypertension, dyslipidemia, and post-thyroidectomy hypothyroidism who initially presented to her family physician
