Molecular dynamics simulation of a novel multi-epitope vaccine design against Salmonella enterica paratyphi A using a computational approach.

Salmonella enterica serovar Paratyphi A is a significant pathogen responsible for enteric fever, particularly in developing countries, contributing to high morbidity and mortality. Current vaccines predominantly target S. Typhi, leaving a critical gap in protection against Paratyphi A strains. The absence of an effective vaccine for Paratyphi A is a significant public health concern, given its potential for outbreaks and long-term healthcare burdens. Designing a targeted vaccine is essential to reduce the incidence of enteric fever and improve global health outcomes. This study focuses on deve

Genomic characterization of a virulent multidrug-resistant Salmonella enterica serovar Enteritidis strain isolated from meat rabbits (Oryctolagus cuniculus) in China.

BACKGROUND: Salmonella enterica serovar Enteritidis (SE) is a major foodborne pathogen. While poultry is a known source, the threat from other livestock like rabbits (Oryctolagus cuniculus) is not well studied. This work investigates a highly virulent, multidrug-resistant (MDR) SE isolate, SE JL228, from a lethal rabbit farm outbreak in China in 2018. The objective of this study was to elucidate the genetic characteristics, antimicrobial resistance determinants, metal tolerance mechanisms, and virulence potential of a rabbit-derived isolate. This study provides important insights that advance

Zinc molybdenum thiovanadate nanoparticles: synthesis, characterization, and antibacterial activity.

Infection control is a key point for maintaining and developing the healthcare sector. Nanosized metal derivatives have attracted significant interest for preventing infections caused by various bacteria and fungi. This study reports the synthesis of new sulfide nanoparticles containing bioactive Zn, Mo, and V in one structure. Powder X-ray diffraction (PXRD) analysis was used to determine the structure, phase purity, and crystallite size of the ZnMoVS4 nanoparticles (NPs). The results reveal the presence of only one phase corresponding to a cubic close-packed spinel structure in the F4̄3m (21

Preventive-curative functional amino acid-supplemented diet improves health, growth performance, and resilience of growing pigs under an immune challenge.

Immune system stimulation (ISS) alters the metabolic demand for amino acids (AA), and amino acid-based nutritional strategies may mitigate performance reduction in pigs subjected to sanitary challenge conditions. This study aimed to evaluate the effect of extra supplementation of a combination of functional AA (FAA, +20% threonine, methionine, and tryptophan) preventively (pre-ISS), curatively (during-ISS) or both on the performance of growing pigs challenged with Salmonella Typhimurium (ST) and poor housing conditions. Fifty-two entire male pigs (21.7 ± 4.6 kg) were allocated to four dietary

Establishing a chicken embryo model for studying infection control of a novel lipid nanoparticle.

The rise of antibiotic-resistant infections highlights the urgent need for alternative therapeutic strategies. Monoglyceride-based lipid nanoparticles (LNPs) offer both antimicrobial activity and the potential for controlled drug delivery. In this study, LNPs with diameters of 230-350 nm and ζ-potential of approximately -35 mV demonstrated high colloidal stability and strong antibacterial efficacy in vitro, achieving complete growth inhibition of Staphylococcus aureus (S. aureus) at 0.04 mg/mL and Salmonella enterica subsp. enterica serovar Enteritidis (S. Enteritidis) at 0.9 mg/mL. To bridge

Identification of Salmonella Infantis persistence in poultry products in the Netherlands with a role for the pESI plasmid.

As Salmonella is known to be able to persist in processing environments, we investigated whether the detection of Salmonella clusters from retail poultry can be linked to a persistent source. A total of 69 Salmonella Infantis isolates from retail poultry products from four different producers between 2008 and 2020 were sequenced and their sensitivity to antibiotics was determined. The six phylogenetic clusters spanning multiple months identified by sequence analysis indicate persistence of Salmonella. In addition, the pESI megaplasmid, which is known to harbor a diversity of resistance and per

Effects of Ligilactobacillus salivarius on the control of pullorum disease and cecal microbiota in red-feathered native chickens.

Pullorum disease (PD), caused by Salmonella Pullorum (SP), remains a persistent challenge in native chicken production in Asia. Recurrent outbreaks and reliance on antibiotics have raised concerns about antimicrobial resistance. This study established a reproducible clinical PD model in red-feathered native chickens (RFCs) and evaluated Ligilactobacillus salivarius (LS) as a potential alternative to antibiotic. Oral administration of a field SP isolate (SPB6) at 1 × 10⁸ CFU per chick for four consecutive days induced typical PD signs and persistent bacterial colonization, whereas a single-dose

Next-generation live vector vaccine targeting Mycoplasma synoviae and Mycoplasma gallisepticum via recombinant Salmonella.

Avian mycoplasmosis has a considerable financial impact on the global chicken industry with Mycoplasma synoviae (MS) and Mycoplasma gallisepticum (MG) being the dominant causes. Currently, inactivated vaccines provide minimal cross-protection and can't induce high levels of mucosal immunity, indicating the need to develop more effective immunisation platforms. This study aimed at designing and testing rationally engineered live recombinant vaccines using Salmonella enterica serovar Typhimurium expressing MS- and MG-specific antigens, GrpE and CrmA, to produce a robust immune response. Characterization of a Salmonella strain isolated from Tibetan blue sheep.

Considering the ecological importance of blue sheep and the lack of information regarding their infection status, this study aimed to determine the causes of diarrhea and death in blue sheep in the Shigatse region. A key strain that caused diarrhea and even death in blue sheep was isolated from fecal samples and analyzed using morphological observation, biochemical tests, 16S rRNA genetic analysis, mouse pathogenicity tests, and drug susceptibility tests. The strains were grouped into pairs, short chains, or Gram-negative bacilli. The biochemical results were consistent with those obtained for

Invasive non-typhoidal Salmonella infections in China (1961-2024): a retrospective systematic analysis of multicentre case reports.

BACKGROUND: Salmonella can be classified as either typhoidal or non-typhoidal. The former primarily causes invasive infections, while the latter typically results in self-limiting diarrhoea. Infections caused by invasive non-typhoidal Salmonella (iNTS) are becoming an emerging global health burden, particularly in low- and middle-income regions. While they account for thousands of deaths each year, we still lack systematic analysis on their burden and epidemiology in Asia, particularly in China. METHODS: We searched Web of Science, PubMed, China National Knowledge Infrastructure, Wanfang Data

An ecological cascade links climatic variability to avian irruptions and zoonotic salmonellosis outbreaks.

Salmonellosis is a common bacterial infection that can lead to severe illness and death in humans as well as domesticated and wild animals. In bird populations, salmonellosis outbreaks are temporally irregular but occur simultaneously across large spatial extents, frequently leading to widespread avian mortality with occasional spillover to humans. Here, we test the hypothesis that avian salmonellosis epidemics result from a cascade of ecological processes triggered by climate variability in coniferous forests. Building on well-evidenced ecological mechanisms, we find strong evidence for multi

Longitudinal Trends in Pediatric Non-Salmonella Gram-Negative Enterobacterales Infections at a Tertiary Care Center in West Africa, 2005-2023.

BACKGROUND: Despite data demonstrating the high prevalence of third-generation cephalosporin resistant Gram-negative Enterobacterales (GNE) among children in sub-Saharan Africa, longitudinal descriptions of resistance among these pathogens from Francophone West Africa remain rare. METHODS: We conducted a retrospective analysis of pathogen-positive children 0-15 years old included in an invasive bacterial infection study at l'Hôpital Gabriel Touré (in Bamako, Mali) from 2005 to 2023. We aimed to describe changes in pathogen burden and non-Salmonella GNE resistance over time and compare in-hosp

Phage treatment significantly reduces Salmonella shedding in layer hens and Salmonella abundance on the surface of the eggs they produce.

Non-typhoidal Salmonella species cause 85% of human foodborne cases and infections caused by multidrug-resistant Salmonella strains are increasing. Therefore, alternative antimicrobials are needed, and phage therapy offers a promising tool to reduce the spread of Salmonella in the human food chain. The aim of this study was to determine if a phage cocktail delivered in water could reduce Salmonella shedding in infected egg laying hens, and Salmonella abundance on the surface of eggshells in eggs produced by infected hens. 240 56-week-old layer hens, which were environmentally infected with Sal

Salmonella virulence factor sptP activates the NLRP3/Caspase-1 pathway to induce pyroptosis and exacerbate intestinal injury in chicks.

Salmonella enterica serovar Enteritidis (SE) is a major foodborne pathogen causing severe intestinal diseases in humans and animals, posing significant global public health and economic burdens. The Salmonella protein tyrosine phosphatase (sptP), a type III secretion system effector and critical SE virulence factor, contributes to pathogenesis, but its precise role in SE-induced intestinal injury remains unclear. Specifically, whether sptP modulates the NLRP3/Caspase-1 inflammasome (a central inflammatory regulator) is undefined. To address this, we constructed an sptP gene deficient SE strain

Acute Abdominal Pain attributable to Salmonella Typhi-infected endometriotic cyst: A rare case report.

Endometriotic cyst infection is rare and typically associated with invasive pelvic procedures or pelvic inflammatory disease. We present a case of a 34-year-old nulligravida female with no recent surgical or gynaecological interventions, who developed acute abdominal pain due to a ruptured endometriotic cyst infected with Salmonella enterica serovar Typhi. This case underscores the potential for hematogenous spread of infection and the importance of considering infected endometriomas in the differential diagnosis of acute abdomen.

Diagnostic and pathological characterization of senecavirus A-associated epidemic transient neonatal losses in swine.

Senecavirus A (SVA) has emerged as a significant viral pathogen of swine, traditionally associated with idiopathic vesicular disease but more recently linked to epidemic transient neonatal losses (ETNL), a syndrome characterized by increased mortality in piglets without consistent vesicular lesions. The mechanisms underlying SVA-associated neonatal mortality remain unclear due to the lack of specific clinical signs, frequent co-infections, and limited tissue characterization. This retrospective study analyzed 186 neonatal submissions (<24 days old) that were tested for SVA by RT-qPCR at the Io
