Tularemia is a zoonotic disease, occurring throughout the Northern Hemisphere. The causative agent, the bacterium Francisella tularensis, is represented by two main types. Type A is found in North America, whereas type B is mainly found in Asia and Europe and to a minor extent in North America. No routine technique for rapid diagnosis of tularemia has been generally applied. We have partially sequenced 16S rRNAs of two F. tularensis strains, as well as the closely related Francisella novicida. Of 550 nucleotides analyzed, only one difference in 16S rRNA primary sequence was found. This 16S rRN

A microagglutination test with safranin-stained Francisella tularensis antigen was compared with a conventional tube agglutination test for the serodiagnosis of tularemia. The microagglutination test was performed in round-bottom microtiter plates by using 0.025 ml of the antisera and of the antigen. The antibody titers obtained by using the microagglutination test were 8 to 64 times higher than those seen with the tube agglutination. By the microagglutination test, the serum agglutinins were detected 3 days earlier in rabbits and 9 days earlier in humans than by the tube agglutination test. T

A comparative study of the antibody titer against Francisella tularensis biovar palaearctica in serum and lung extract was made using different immunoassays. Samples were taken from experimentally-infected goshawks (Accipiter gentilis) and European beavers (Castor fiber), and in a field survey of wild beavers. Good accordance between the antibody titer in serum and in lung extract was found in the experimental studies. However, the antibody titer was generally one- to three-fold lower in lung extract than was the titer of serum. Results in the field survey were less reliable. Estimating antibo

1. Blood-seeking diel activity cycles of mosquitoes are described from 24 h landing catches on human bait in a field and a forest habitat during June-August in central Sweden. 2. Daytime activity in the field was low or absent, whilst in the forest activity was relatively even throughout the diel. 3. In both habitats, activity peaks generally occurred about sunset and sunrise and were more distinct in the field than in the forest. 4. Data from this and other studies show that, in general, Aedes cinereus Meigen feeds at night in field-type habitats and Ae.communis (DeGeer) during the diurnal-cr

The occurrence of tularemia was studied in 1,500 hares submitted to the National Veterinary Institute, Uppsala, Sweden for postmortem examination during 1973 through 1985. A total of 109 tularemia cases was recorded based on the fluorescent antibody (FA) test for Francisella tularensis and on the gross and microscopic pathology. Tularemia was diagnosed only in the varying hare (Lepus timidus) and not in the European brown hare (Lepus europaeus). The geographical distribution of the 109 cases indicates that tularemia has not spread in Sweden during the last 45 yr, with the exception of an endem

The host response to experimental murine tularemia was examined in different inbred mouse strains. The kinetics of growth of Francisella tularensis live vaccine strain (LVS) in the livers and spleens of A and C57BL/6 mice were monitored, and it was observed that mice of the A strain were more susceptible to the proliferation of LVS than were C57BL/6 mice. The difference was most marked 5 days following infection, when the number of bacteria isolated from the spleens of A mice was found to exceed that of C57BL/6 mice by 100-fold. In addition, the C57BL/6 strain exhibited a more pronounced splen

A mathematical model of antibiotic and immunostimulator (IMS) combined effect on various elements of the immune system and general state of patients with infectious diseases is described. The model was constructed as a system including 6 usual differential equations of the 1st order. With the use of this model and a computer many diverse variants of infection development under conditions of treatment with IMS at the background of antibiotic therapy were modeled. Ii was shown that IMS-antibiotic complexes markedly improved the indices of antibiotic therapy as compared to the use of the antibiot

Patients with atypical pneumonias, whether caused by bacterial, fungi, or viruses are associated with pleural effusions. The effusions generally are small and ipsilateral to the parenchymal infiltrate. Usually, the pleural fluid is a serous exudate with a predominance of mononuclear cells. The pleural fluid glucose and, presumably, the pleural fluid pH are not low. The etiologic organism has been isolated from pleural fluid but usually is not necessary to establish the diagnosis. Pleural biopsy is not helpful in diagnosis with the exception of acute coccidioidal pneumonia and effusion. Pleural

Between June 1 and July 15, 1984, twenty persons with glandular tularemia were identified on the Lower Brule and Crow Creek Indian reservations in South Dakota. The median age of the patients was 6 years (range, 2 to 20 years). The clinical illness was mild, consisting of fever, headache, and lymphadenopathy. All lymphadenopathy was in the head and neck area. Dermacentor variabilis ticks were identified as the vector. Although the mild clinical illness suggested Francisella tularensis, type B, was the agent, both type A and type B strains of F tularensis were isolated from ticks collected from

Forty-two subjects with acute tularemia were studied for the occurrence of C-reactive protein (CRP), and 73 subjects with acute tularemia or experience of the disease within the last 11 years were studied for immunoglobulin M (IgM), IgA, and IgG class-specific antibodies, agglutinating antibodies, and complement-fixing antibodies to Francisella tularensis by using an enzyme-linked immunosorbent assay (ELISA), the tube agglutination test, and a complement-fixing ELISA. The incubation time between infection and the outbreak of symptoms varied from 1 to 10 days, averaging 6.5 days. Elevated CRP c

Mice with generalized influenza or tularemia of similar lethality were studied in an effort to compare biochemical responses of the myocardium during infections of viral and bacterial etiology. A progressive loss of body weight characterized the course of both infections. Accompanying this, the myocardial content of protein and the activities of lactate dehydrogenase, citrate synthase, and cytochrome c oxidase all decreased. However, myocardial protein degradation appeared earlier and was more pronounced in influenza, and the protein changes were accompanied by a rapid decline of myocardial RN

Tularemia occurs in Sweden as an epizootic among the mountain hare. Little is known about the occurrence of the disease in other animals in this country. For this reason serum samples from 28 cattle, 83 moose, 110 beavers and 97 mountain hares were investigated for the presence of antibodies against Francisella tularensis. The antibody levels against F. tularensis found in moose and cattle were generally low and also in a low incidence. This indicates that these species are not susceptible to tularemia and not involved in the epizootiology of this disease. Beaver titres were found to vary from

A 37-year-old woman experienced local symptoms on returning from a camping trip. Within three weeks she developed generalized symptoms and was hospitalized. Tularemia, and often overlooked cause of bubo formation, is endemic in Nevada. The diagnosis should be considered when patients who have visited such an area present with enlarged, painful lymph nodes, skin pustules, ulcers, headache, myalgia, malaise and nausea. Cultures tend to be negative, because the causative organism, Francisella tularensis, needs cysteine. While there are five clinical forms of tularemia, man tends to get the ulcero

To investigate the effects of strenuous forced exercise on the course and complications of a bacterial infection and on myocardial responses and performance capacity, rats with tularemia (characterized by pyogranulomatous hepatic and splenic lesions) were exercised by swimming on days 0-6 of infection. Levels of glutamic oxaloacetic and pyruvic transaminases in plasma, densities of pyogranulomatous lesions, and bacterial counts in blood, liver, and spleen were similar in exercising and resting rats. Although a few exercising rats showed an unusual dissemination of infection, the antibody respo

Live attenuated Francisella tularensis vaccine induced long-lasting humoral and cell-mediated immune responses in all 13 subjects studied. Lymphocyte blast transformation reactivity to F. tularensis appeared 2 weeks after vaccination in most subjects and remained unchanged for up to 1.5 years. Similarly, in most recipients, antibodies against F. tularensis were detectable by both the enzyme-linked immunosorbent assay (ELISA) and the agglutination method from 2 weeks after vaccination, although diagnostically significant agglutination titers (greater than or equal to 80) were not detectable unt

Although rare among humans in Canada, tularemia is often endemic in wildlife. The inhabitants of rural areas are especially likely to be exposed to the causative bacterium, Francisella tularensis, through trapping or through the bites of arthropods. Muskrats have replaced rabbits as the principal source of infection, as illustrated by a familial outbreak of oropharyngeal tularemia in Saskatchewan. In humans the disease has six distinct forms and can be asymptomatic, but it generally comes to medical attention as fever, persistent ulcers and enlarged lymph nodes. Serologic tests will confirm th

The enzyme-linked immunosorbent assay (ELISA) was applied for immunological diagnosis of human tularemia, using lipopolysaccharide from Francisella tularensis as antigen. Sera collected from patients, healthy individuals, and vaccinated volunteers were investigated for antibodies against F. tularensis by ELISA and tube agglutination. In ELISA all sera were titrated with a polyspecific anti-immunoglobulin enzyme conjugate. A limited number of consecutive sera from individual patients were also investigated for immunoglobulin G (IgG) and IgM antibodies by means of immunoglobulin class-specific c

There was revealed a regular reduction of plague, tularemia and pseudotuberculosis bacteria count in the lungs of guinea pigs the first 12 hours after aerosol infection. Generalization of the infectious process and associated septicemia occurred in pulmonary plague and pulmonary tularemia on the 1st-2nd day, and in pulmonary pseudotuberculosis - on the 4th-5th day. Limits of accumulation of the causative agents in the organs and the blood at various stages of the infectious process were established.
