Investigation of cerebellar structure and static and dynamic functional connectivity in cervical spondylotic myelopathy patients.

PURPOSE: To investigate cerebellar structural and functional alterations in cervical spondylotic myelopathy (CSM) using multimodal MRI. METHODS: 127 CSM patients and 91 healthy controls (HCs) underwent resting-state fMRI. Gray matter volume (GMV), static functional connectivity (sFC), and dynamic functional connectivity (dFC) were analyzed across 34 cerebellar subregions using the spatially unbiased infratentorial template (SUIT) atlas. Associations between altered imaging metrics and clinical measures were assessed. RESULTS: (1) CSM patients showed reduced GMV in left lobule V and vermal Cr

Reduced inhibition of hippocampal adult-born granule cells by parvalbumin interneurons after TBI.

Traumatic brain injury (TBI) is one of the leading causes of acquired temporal lobe epilepsy. TBI drives hippocampal circuit rearrangements that may contribute to increased seizure risk, such as altered inhibitory circuit function and aberrant post-traumatic neurogenesis. In the hippocampal dentate gyrus, adult-born dentate granule cells (DGCs) acquire inhibitory synaptic inputs from parvalbumin-expressing (PV) interneurons early in their maturation. PV inputs are important for circuit integration and feedforward inhibition of adult-born DGCs. We used genetically modified mice, retroviral vect

Lead-Free Halide Double Perovskites: A Review of Recent Advances in Synthesis, Optical Properties and Applications.

Analyzing multimodal brain networks offers new insights into the mechanisms of brain disorders. However, most studies focus on unimodal networks or use global/local graph metrics, often missing critical information about disrupted pathways. In this study, we propose a novel framework for estimating static and dynamic multimodal brain graphs by integrating time-resolved functional network connectivity (FNC) and gray matter features using a Gaussian graphical model. We then apply a path-based graph theory analysis to identify connectors and disconnectors, edges that respectively introduce or eli

Loneliness relates to smaller hippocampal subfield volumes in individuals with elevated plasma pTau217: results from IGNITE.

Loneliness is associated with cognitive decline and dementia risk, but the role of Alzheimer's disease (AD) pathology in its associations with medial temporal lobe (MTL) morphology remains unclear. We examined whether loneliness was related to total hippocampal volume, hippocampal subfields, and entorhinal cortex (ERC) volumes, and whether plasma phosphorylated tau 217 (pTau217) moderated these relationships. We analyzed baseline data from cognitively unimpaired older adults aged 65-80 years enrolled in IGNITE. Loneliness was assessed using the revised UCLA Loneliness Scale. Structural 3 T MRI

Patterns and predictive factors of hearing loss following microvascular decompression for hemifacial spasm: a systematic review.

Microvascular is a safe and effective therapeutic intervention for hemifacial spasm; however, its potential association with hearing impairment remains a concern. This study aimed to determine the prevalence, patterns, and risk factors of hearing loss following microvascular decompression in patients with hemifacial spasm. In accordance with PRISMA guidelines, a systematic literature search was conducted across PubMed, Cochrane Library, ScienceDirect, and Web of Science from inception to April 2025. Eligible studies included adult patients undergoing microvascular decompression for hemifacial

Reconfiguration of the Cognitive Control Network Induced by Cerebral and Cerebro-Cerebellar Transcranial Direct Current Stimulation.

Cognitive control can be modulated by noninvasive brain stimulation, but it remains unclear how different stimulation protocols are associated with task-related network organization. To characterize behavioral and brain-network profiles, we recorded brain activity and behavioral performance while participants completed a cognitive control task during functional magnetic resonance imaging. Profiles were compared across a pre-stimulation session and two sessions following high-definition transcranial direct current stimulation (HD-tDCS): single-site stimulation of the presupplementary motor area

What is a standard temporal lobectomy? A survey of epilepsy neurosurgeons.

OBJECTIVE: Anterior temporal lobectomy (ATL) is the most common surgical procedure used to treat drug-resistant epilepsy. ATL fundamentally involves removal of the anterior temporal neocortex and mesial structures; however, there may be substantial variability in resection extent across surgeons. The objective of this study was to evaluate the practice patterns and ATL surgical techniques of epilepsy surgeons. METHODS: An anonymous REDCap-based survey of epilepsy neurosurgeons was conducted to collect demographic and resection data. Resection extent was assessed by asking participants to draw

Enhancing Ion Yields in MALDI-2-MSI Using Dopant-Assisted Post Ionization for High-Resolution MS-Imaging of Lipids and Metabolites.

Matrix-assisted laser desorption ionization mass spectrometry imaging (MALDI-MSI) is the most widely used label-free method for mapping the molecular distribution of lipids and metabolites from tissue sections and cell cultures. To mitigate the problem of partially low MALDI ion yields for several of these analyte classes, different post ionization techniques have recently been developed─for example, MALDI-2, where a second UV-laser pulse intersects the initial particle plume to generate additional charges. Here, we show that the MALDI-2 signal intensities of important analyte classes can be f

Prematurity insults remodel cerebellar development and behaviour.

Preterm survivors often develop motor and socio-cognitive impairments that implicate altered cerebellar development, yet the underlying mechanisms remain poorly understood. A key challenge is that prematurity involves overlapping perinatal insults that converge on the developing brain, making their individual effects difficult to disentangle. Here, we model two major prematurity-associated insults, maternal immune activation (MIA) and neonatal hypoxia (Hx), in mice. By controlling timing and sequence, we define how these insults shape cerebellar assembly during rapid maturation. Comparative an

Paratonia is inversely correlated to premotor areas perfusion.

OBJECTIVE: Paratonia is defined as an inability to relax the muscles during tone assessment, resulting in participants either opposing or facilitating the examiner's movement involuntarily. This phenomenon is frequently found in patients with cognitive impairment and has been attributed to frontal lobe dysfunction, although a direct correlation has not been established. This study aimed to explore the correlation between frontal lobe function, as assessed by functional brain imaging, and paratonia, as assessed by electromyography. METHODS: Memory clinic outpatients with suspected Alzheimer's

prickle-mediated epilepsy in flies is a brain-derived neurodevelopmental defect.

Damaging DNA variants in PRICKLE genes cause epilepsy from flies to humans. While classic work, primarily in flies, has demonstrated that PRICKLE proteins work with other planar cell polarity proteins to polarize epithelial sheets, the function of PRICKLE proteins in the nervous system is still unclear and may play distinct roles in the nervous system of different organisms. We thus sought to compare and contrast Drosophila prickle with PRICKLE genes in vertebrate systems in order to identify unifying roles across species. First, we find that similar to expression of the mouse Prickles, fly pr

Nanoplasmonic Aptasensing Enables Real-Time Optical Monitoring of Neurotransmitters in Living Brainstem Organoids.

Disruption of dopamine signaling is central to Parkinson's disease (PD) pathology and a primary target of symptomatic therapy. However, direct measurement of dopamine dynamics in human neural tissue with high sensitivity, temporal resolution, and compatibility with three-dimensional models remains technically challenging. Here, an organoid-on-a-chip platform that integrates a cavity-coupled nanoplasmonic aptasensor with human brainstem organoids (hBSOs) and microfluidic control is employed for label-free monitoring of the extracellular dopamine response. The plasmonic aptasensor achieves a lim

Formal Thought Disorders.

Formal thought disorder (FTD), changes in the form (as opposed to the content) of thinking, has a largely unknown pathophysiological basis or bases, although language, frontal/executive, and semantic memory mechanisms have been implicated. FTD is a major symptom of schizophrenia and is also common in mania, with there being a marked overlap of clinical features between the 2 disorders. Changes in thought form are also seen in major depression, autistic spectrum disorder, and the frontal lobe syndrome; the presentations in these latter disorders differ from those seen in schizophrenia and mania

The entorhinal cortex atrophy score for diagnostic distinction between phases of the Alzheimer's disease continuum.

BACKGROUND: Alzheimer's disease (AD) is the most common type of dementia, causing functional impairment in millions of individuals and accounting for a large amount of healthcare costs. The entorhinal cortex appears to be one of the first brain structures affected by AD in its early stages. The Entorhinal Cortex Atrophy (ERICA) score is a visual atrophy score used by neuroradiologists in clinical practice for the diagnosis of AD. OBJECTIVE: To analyze the effectiveness of the ERICA score for distinguishing between patients with subjective cognitive decline (SCD), mild cognitive impairment (MC

Central monoamine dysfunction in Prader-Willi syndrome: a mechanistic framework for core clinical features.

Prader-Willi syndrome (PWS) is a rare genetic neurodevelopmental and neurobehavioral disorder characterized by hyperphagia and behavioral, endocrine, arousal, and cognitive dysregulation. Although caused by loss of paternally expressed, maternally imprinted genes in 15q11-q13, the mechanisms linking this lesion to the broad clinical phenotype remain incompletely understood. This review synthesizes evidence that central monoaminergic dysfunction, encompassing serotonin, dopamine, and norepinephrine, provides an integrative, though not exclusive, mechanistic framework for core features of PWS. Neuronal surface autoantibodies at cerebellar and brainstem synapses: mechanisms, phenotypes, and translational gaps.

Pathogenic neuronal-surface autoantibodies can disrupt extracellular receptors, ion-channel complexes, and synaptic proteins, yet their regional neurological phenotypes differ substantially. This variability is particularly informative in the cerebellum and brainstem, where precisely timed excitatory and inhibitory signaling supports motor coordination, ocular-motor control, autonomic regulation, arousal, and respiration. In this Mini Review, we examine how antibody effector mechanisms, regional antigen distribution, and local synaptic physiology interact to shape cerebellar and brainstem mani
