Preparation of Postmortem Human Formalin-Fixed Paraffin-Embedded Frontal Cortex Tissue for Profiling Pyramidal Neurons Via Digital Spatial Profiling.

Digital spatial profiling (DSP) facilitates RNA sequencing (RNA-seq) of small populations of cells, specifically ~20 pyramidal neurons in postmortem formalin-fixed paraffin-embedded (FFPE) human brain tissue. This approach enables precise expression profiling via RNA-seq data linked to spatially characterized samples utilizing tissue-bound probes. We present a comprehensive protocol for manual slide preparation using the GeoMx DSP system as preliminary steps to spatial characterization of lamina-specific pyramidal neurons in human postmortem Brodmann area 9 (BA9) frontal cortex brain tissue.

Neuroprotective and memory-enhancing effects of Aloysia gratissima extract and rutin in an MPTP-induced model of Parkinson's disease.

Parkinson's disease (PD) is the second most prevalent neurodegenerative disorder worldwide and currently has no cure. Available treatments mainly alleviate symptoms but show limited long-term efficacy, underscoring the need for new therapeutic strategies. This work aimed to investigate the neuroprotective potential of Aloysia gratissima leaf extract and the flavonoid rutin on memory deficits, inflammatory markers, and neurotransmitter bioavailability in an MPTP-induced model of PD in rats. At 12-13 weeks of age Wistar rats were intranasally administered 1 mg of MPTP to induce PD-like symptoms.

Transformer Mechanisms Mimic Frontostriatal Gating Operations When Trained on Human Working Memory Tasks.

Working memory (WM) is thought to rely on sophisticated frontostriatal mechanisms for selective gating, supporting updating and readout of information to and from distinct memory "addresses" (neural populations). According to computational models, capacity limitations in WM arise due to challenges in "role addressability": learning to correctly bind items in memory to their respective roles and assigning credit to the corresponding gating operations. However, these conclusions are based on particular assumptions about biological neural networks, and it is unclear whether the principles general

Resting-state functional connectivity in default mode and frontoparietal networks predicts directed forgetting.

Directed forgetting (DF) refers to the goal-directed modulation of memory, typically manifested as superior memory for information cued to be remembered (TBR) compared to information cued to be forgotten (TBF). Although prior studies have emphasized task-evoked neural mechanisms of DF, it remains unclear whether intrinsic brain network organization contributes to individual differences in memory control. The present study examined whether resting-state functional connectivity within and between the default mode network (DMN) and frontoparietal network (FPN) predicts individual differences in D

The Cost of Scrutiny: Subclinical Social Anxiety and Performance Context Reshape Frontal-Midline Theta Network Reconfigurations during Probabilistic Learning.

Social anxiety is reliably characterized by biases toward avoidance and aversive learning. Frontal-midline theta (FM-theta) oscillations have been implicated as a key neural mechanism underlying these learning biases. Here, we combined a dynamic network neuroscience approach with computational modeling to investigate how FM-theta oscillatory activity coordinates large-scale functional network interactions on a trial-to-trial basis during feedback-guided learning. Additionally, we examined how these neural dynamics are associated with social anxiety across different social contexts. A sample of

Clinical and imaging correlates of lateralized rhythmic delta activity: A multicenter cohort study.

OBJECTIVE: Determine the clinical and imaging correlates and outcomes of lateralized rhythmic delta activity (LRDA). METHODS: Multicenter retrospective study of hospitalized patients (age ≥ 18) undergoing continuous EEG. Rhythmic and periodic patterns (RPPs) including LRDA, lateralized and generalized periodic discharges (LPDs, GPDs) were extracted from EEG reports. RESULTS: Among 3163 adults, LRDA occurred in 199 (6.3%); 137 (68.8%) had isolated LRDA and 62 (31.2%) had LRDA with other RPPs. Patients with LRDA were more likely to have LPDs compared to patients without LRDA (27.6% vs 6.8%; p

Temporal Interference Modulates Medial Temporal and Frontoparietal Activity During Mnemonic Discrimination: A High-Resolution Whole-Brain fMRI Investigation.

Mnemonic discrimination-the ability to distinguish between existing memories and similar new inputs-supports accurate memory in the face of interference (e.g., in a card game, recognizing that the card you are currently seeing, the King of Hearts, is distinct from the King of Diamonds you encountered earlier in the same hand). Although hippocampal pattern separation has been emphasized as a key mechanism for this process, recent theoretical frameworks and empirical findings suggest that surrounding medial temporal lobe and cortical regions also contribute by resolving interference. An importan

Coexistence of Ebstein Anomaly and Cerebral Cavernous Malformation: A Case Report.

In most patients who have Ebstein anomaly (EA), neurological symptoms are either associated with thromboembolic episodes or due to arrhythmic complications. Cerebral cavernous malformation (CCM) is a vascular anomaly made up of sinusoidally dilated blood vessels, often presenting with seizure activity or intracranial bleeding. There has never been any documented report regarding coexisting EA and CCM. Herein, we have reported a 32-year-old female with an established history of Ebstein anomaly diagnosed in 2013 who had a history of generalized tonic-clonic fits since 2020. Neuroimaging studies

Neuroimaging signatures of facial onset sensory and motor neuronopathy (FOSMN).

Facial onset sensory and motor neuronopathy (FOSMN) is an exceptionally rare condition characterized by progressive facial paresthesia and the subsequent appearance of bulbar symptoms with rostro-caudal motor and sensory deficits sometimes extending to the hand muscles. The pathophysiological understanding of this disorder is very limited. In order to identify patterns of neuroanatomical involvement within the brain, we applied advanced magnetic resonance imaging-based techniques to a group of six patients with the diagnosis of FOSMN, using unbiased analysis techniques with atlas-based volumet

Frontal aperiodic activity and sensorimotor theta clustering are differentially modulated by tDCS during proprioceptive training in Parkinson's disease: An EEG study.

Proprioceptive deficits are a core feature of Parkinson's disease (PD), but the neural mechanisms by which transcranial direct current stimulation (tDCS) may augment proprioceptive learning remain unknown. This study investigated whether six sessions of anodal tDCS combined with proprioceptive training modulates EEG‑derived aperiodic activity, Lempel‑Ziv complexity (LZC), and theta clustering in individuals with PD. Twenty‑four participants with mild‑to‑moderate PD were randomly assigned to active (2 mA, 20 min) or sham tDCS over the supplementary motor area, combined with a proprioceptive exe

Greater temporo-occipital gray matter volume and frontoparietal cortical thickness in post-menopausal women: a UK Biobank study.

Menopause is a critical transitional stage in women's lives, yet evidence on its impact on brain structure remains inconsistent, and large-sample neuroimaging studies are limited. Here, we applied whole-brain morphometry techniques to evaluate differences between pre- and post-menopausal women in terms of gray matter volume, cortical thickness and cortical gyrification. We processed and analyzed T1-weighted brain scans of 4098 women (1033 pre-menopausal; 3065 post-menopausal) from the UK Biobank. Post-menopausal women exhibited greater gray matter volume than pre-menopausal women primarily in

Long-term survival with immune checkpoint inhibitors combined with bevacizumab in advanced clear cell renal cell carcinoma: A case report.

RATIONALE: Renal cell carcinoma is a highly heterogeneous urological malignancy, with clear cell renal cell carcinoma (ccRCC) as the most common pathological subtype. A considerable proportion of patients are diagnosed at advanced stages or experience postoperative recurrence and distant metastasis, leading to poor long-term prognosis. Although targeted therapy and immune checkpoint inhibitors (ICIs) have greatly improved the treatment landscape of advanced ccRCC, optimal sequential and combination treatment strategies for heavily pretreated metastatic patients remain to be further explored.

Psilocybin's effect on human brain synaptic plasticity.

Psychedelics such as psilocybin have been linked to enhanced neuroplasticity and symptom relief in affective disorders, but the neurobiological mechanisms and impact of environmental context remain unclear. Here, we tested whether a single dose of psilocybin (0.3 mg/kg) alters synaptic density in healthy individuals and whether setting-dependent subjective experience shapes this effect. Fifteen healthy participants had a psilocybin-induced psychedelic experience either inside an MRI scanner or in a therapeutic-like room. We assessed synaptic density changes by measuring the Synaptic Vesicle gl

Cognitive outcomes following neurosurgery for frontal intracerebral tumors: A systematic review and meta-analysis.

OBJECTIVE: Neurosurgical resection of frontal lobe tumors poses specific challenges to the patients. Even when lesions involve the so-called "non-eloquent" areas, such as the prefrontal cortex, neurosurgery can still affect critical functions, because the prefrontal regions are essential for executive functions and other complex cognitive processes. The aim of this study is to systematically review the literature and quantitatively assess the impact of frontal lobe tumor resection on patients' cognitive functioning. METHODS: In the present systematic review and meta-analysis, we synthesized d

What shapes health-related quality of life in drug-resistant epilepsy? Clinical, emotional, and cognitive predictors.

PURPOSE: Clinical and psychological factors associated with health-related quality of life (HRQoL) in drug-resistant epilepsy (DRE) are well established; however, the contribution of alexithymia and specific neurocognitive domains remains less well characterized. This study aimed to examine clinical, emotional, and neuropsychological correlates of HRQoL in patients with focal DRE, with particular emphasis on alexithymia and cognitive functioning. METHODS: Thirty-two patients with frontal lobe epilepsy (FLE) and 38 with mesial temporal lobe epilepsy (mTLE) underwent comprehensive clinical, cog

Dynamic functional connectivity in left frontal and temporal tumors.

This study examined dynamic resting-state functional connectivity and graph-theoretic measures in 71 patients with left intra-axial tumors (40 left frontal lobe tumors and 31 left temporal lobe tumors) and 53 age- and sex-matched healthy controls. Dynamic functional connectivity was estimated across 8 80-s sliding windows, region of interest-to-region of interest Fisher-z connectivity values from 132 atlas regions were harmonized using ComBat, and graph metrics were calculated at a 20% proportional density. The primary within-study comparison was restricted to left frontal lobe tumors and left

Clinical characteristics and semiological pitfalls in patients with focal epilepsy and functional/dissociative seizures: A comparative analysis of correct and incorrect initial diagnoses.

OBJECTIVE: Both epilepsy and functional/dissociative seizures (FDS) can present with a wide variety of event manifestations which can appear similar in nature and contribute to misdiagnosis. Whereas many studies have addressed the misdiagnosis of FDS as epilepsy, research into the inverse, where epilepsy is mistaken for a functional disorder, remains limited. Accurate differentiation is vital, because management differs greatly, ranging from antiseizure medications and surgery to specialized mental health interventions. METHODS: A retrospective review was conducted of patients undergoing vide
