Neural Organization of Episodic Memory and Navigation in Children and Adults.

Episodic memory and spatial navigation are often viewed as interlinked, in part due to their shared reliance on medial-temporal lobe (MTL) structures. At the same time, there are important differences, for example, natural spatial navigation requires moving one's body through space while episodic memory does not. We investigated whether naturalistically encoded memories that were either episodic or additionally embedded in a navigational experience relied on overlapping or distinct neural networks. Children ages 8-13 years old and young adults viewed pictures of objects that they had previousl

"When the word ends, the story begins": word offsets improve encoding of natural speech in a MEG study.

The segmentation of continuous speech into words was investigated using the temporal response function (TRF) approach for source-space magnetoencephalography (MEG) signals from 54 participants. Although the onset of words has been extensively studied, the contribution of their offset remains unclear. Acoustic and semantic predictors were used to test the relative contribution of word onsets and offsets to the processing of natural speech in the auditory temporal cortex. Circular shifting of word onsets and offsets significantly reduced predictive power (p < 0.001 for both). The corrupted word

Integrated Mesh-Based Analysis of Neuronal Morphology, Synaptic Subcompartments, and Laminar Inputs in the H01 Human Temporal Cortex.

Quantitative nanoscale descriptions of human cortical neurons remain limited, particularly when neuronal morphology, synaptic geometry, and laminar input organization are examined within a unified connectomic framework. Using publicly available H01 human temporal cortex data and open-source tools, we developed a reproducible pipeline to decode sharded data, reconstruct neuronal and synaptic mesh models, and integrate mesh-based morphometry with annotation-derived laminar input analysis. The associated custom scripts will be released in a public repository upon publication. Morphometric analysi

Motifs of brain cortical folding from birth to adulthood.

Cortical folding of the brain is widely regarded as an interplay between genetic programming and biomechanical forces, closely linked to cytoarchitectonic regionalisation. Abnormal folding patterns are frequently observed in neurodevelopmental conditions and psychiatric disorders. However, significant inter-individual variability of secondary and tertiary folds obscures the detection of shape biomarkers and confounds the investigation of folding-functional relationships. Here, we investigate cortical folding heterogeneity at a fine scale, using Multimodal Surface Matching with Hierarchical Tem

Lack of tau expression modifies epileptogenesis associated neuroplasticity in the ventral dentate gyrus in a mouse model of temporal lobe epilepsy.

Deletion of the microtubule-associated protein, tau reduces seizure burden in rodent models of genetic epilepsies and suppresses evoked seizures. In the intrahippocampal kainate (IHK) model of acquired temporal lobe epilepsy (TLE), mice lacking tau expression develop spontaneous seizures, but at a lower rate than controls. Mechanisms by which tau deletion modifies the development of acquired TLE are not well understood, but a modification of the synaptic reorganization that accompanies epileptogenesis may contribute to modified seizure expression. Increased inhibitory synaptic input to dorsal

Robust Induction of Spontaneous Recurrent Seizures and Hippocampal Sclerosis with Synthetic Kainic Acid in the Intrahippocampal Kainate Model.

Intrahippocampal kainate (IHK) is a well-established mouse model of temporal lobe epilepsy, in which focal hippocampal injection of kainic acid induces excitotoxic injury followed by the development of chronic spontaneous seizures. Commercial sources of kainic acid (KA) have increasingly transitioned from naturally derived to synthetic KA, which has introduced a potential source of variability to the implementation of the IHK model. Here, we evaluated injection parameters for synthetic KA, with the goal of identifying conditions that reliably produce spontaneous seizures while minimizing morta

The development of slow and fast theta oscillations in the human brain.

Theta oscillations are pivotal for attention and memory, with developmental shifts in theta activity and connectivity tracking memory ability across the lifespan. Building on initial evidence that the theta band separates into slow and fast sub-bands across development within the medial temporal lobe (MTL) and the prefrontal cortex (PFC), this preregistered study ( https://osf.io/gsru7 ) characterized the frequencies of slow (∼1.5-4.5 Hz) and fast (∼4.5-9.0 Hz) theta oscillations across the brain in a large developmental cohort. We analyzed intracranial EEG (iEEG) recordings from widespread br

Decoding brain anatomy from neuronal neighborhoods with MYCEL.

Extracellular recordings carry information about neuronal location, and pooling per-unit predictions across neighboring electrodes improves its readout, but whether this local pooling should be learned is untested. We present MYCEL (Message-passing Yields Cellular Embeddings of Location), a graph neural network whose nodes are spike-sorted units carrying waveform and spike-timing features selected on cell-type ground truth, and whose edges encode recording geometry. On the Allen Brain Observatory Visual Coding Neuropixels dataset, learned message passing outperforms fixed aggregation, with the

Transient Prosopagnosia after Right Chronic Subdural Hematoma Surgery: Top-Down Squash on Fusiform Gyrus or a Sign of Preexisting Dementia?

Face blindness, also called prosopagnosia, is the inability to recognize familiar faces and is often secondary to dysfunction of the fusiform gyrus. This symptom can be part of dementia or be related to a lesion in this anatomical structure. We present the case of a 78-year-old female patient, with a diagnosis of dementia without prosopagnosia, who developed an episode of transient loss of consciousness. A brain computed tomography scan revealed an extensive right hemispheric chronic subdural hematoma. Patient underwent urgent hematoma drainage after deterioration of the neurological condition

Chorioretinal Scars in Congenital Cytomegalovirus Infection Following Prenatal Valacyclovir Therapy.

BACKGROUND: Congenital cytomegalovirus infection is the most common congenital infection. Ocular abnormalities affect approximately 14% of symptomatic children, but fewer than 1% of asymptomatic children. Prenatal valacyclovir reduces vertical transmission, but ophthalmic outcomes in exposed infants are poorly described. METHODS: We describe 2 neonates with congenital cytomegalovirus confirmed by amniotic fluid and urine polymerase chain reaction following first-trimester maternal primary infection. Both mothers received high-dose valacyclovir (8 g daily) from 13 weeks' gestation. Infants und

Inhibition of Cerebellar Dentate Nucleus-Zona Incerta Glutamatergic Circuit Improves Sleep Disturbances in Chronic Temporal Lobe Epilepsy Model.

BACKGROUND: Patients with chronic temporal lobe epilepsy (TLE) frequently experience various sleep disorders, characterized by reduced sleep duration, diminished sleep stages, and disrupted sleep patterns. Recent research has identified a population of GABAergic neurons in the ventral zona incerta (ZI) that is instrumental in promoting sleep, along with a neural pathway that connects the cerebellar dentate nucleus (CDN) to the ZI. However, the precise role of this circuit in regulating sleep patterns remains inadequately understood. METHODS: This study employed anatomical, chemogenetic, optog

Endoscopic transorbital approach for resection of Meckel's cave aspergillosis: illustrative case.

BACKGROUND: Aspergillosis involving Meckel's cave (MC) is rare. Only 5 previous case reports exist in the literature. The primary treatment for this disease is surgical debridement combined with antifungal therapy. The authors present a minimally invasive technique using an endoscopic transorbital approach (ETOA) to manage this condition. OBSERVATIONS: A 26-year-old male corn farmer presented with progressive right-sided facial pain and numbness for 1 month. MRI demonstrated a markedly hypointense T2 signal lesion in the right MC with gadolinium enhancement. CT revealed sphenoid sinus opacifi

Awake resection of temporal encephalocele associated with drug-resistant epilepsy: illustrative case.

BACKGROUND: The association between temporal encephaloceles (TEs) and drug-resistant epilepsy (DRE) is well recognized, yet the literature regarding therapeutic management remains scarce and largely retrospective. Reported strategies vary from conservative approaches to extensive resection of encephaloceles including the adjacent epileptogenic zone (EZ). Surprisingly, although the EZ may be localized within or adjacent to eloquent areas, surgeries under awake conditions have not been described. One plausible explanation is the potential risk of intraoperative pain during surgical manipulation

Neurosurgical Outcomes of Epilepsy Surgery Between 2017 and 2025: A Retrospective ESSQ-19 Study From a Tertiary Care Center.

Introduction Epilepsy is a common neurological disorder. Despite being on anti-epileptic drug therapy, a subset of patients continue to experience seizures. In such patients with drug-resistant epilepsy, surgery remains a viable treatment option. Patient-reported satisfaction provides an important perspective on post-epilepsy surgery outcomes. This study aimed to descriptively assess postoperative patient satisfaction using the validated Epilepsy Surgery Satisfaction Questionnaire-19 (ESSQ-19). Materials and methods This was a retrospective observational study conducted at a tertiary care cent

Temporal Interference Modulates Medial Temporal and Frontoparietal Activity During Mnemonic Discrimination: A High-Resolution Whole-Brain fMRI Investigation.

Mnemonic discrimination-the ability to distinguish between existing memories and similar new inputs-supports accurate memory in the face of interference (e.g., in a card game, recognizing that the card you are currently seeing, the King of Hearts, is distinct from the King of Diamonds you encountered earlier in the same hand). Although hippocampal pattern separation has been emphasized as a key mechanism for this process, recent theoretical frameworks and empirical findings suggest that surrounding medial temporal lobe and cortical regions also contribute by resolving interference. An importan

Lateralized amyloid PET uptake and beta-band connectivity in relation to cognition in temporal lobe epilepsy.

We examined focus-relative amyloid PET asymmetry and its associations with resting-state EEG connectivity and processing speed in mesial temporal lobe epilepsy (MTLE). Thirty adults with clinically assigned MTLE (19 right-focus, 11 left-focus) underwent 18F-florapronol PET and resting-state EEG. Regional standardized uptake value ratios (SUVRs) and sensor-level weighted phase lag index (wPLI) were evaluated using paired comparisons, rank correlations and exploratory bootstrap indirect-association models. Cognitive data were available for 26 participants, including Digit Symbol Coding (DSC) for

Comprehensive transcriptomic and proteomic profiling of mTOR-related epilepsy.

OBJECTIVE: Malformations of cortical development (MCDs) are a major cause of drug-resistant epilepsy (DRE) in children. A subset of MCDs, including focal cortical dysplasia type II, tuberous sclerosis complex, and hemimegalencephaly, shares characteristic histopathological features and somatic mutations in genes within the Phosphoinositide 3-kinase (PI3K)-AKT-mechanistic target of rapamycin (mTOR) signaling cascade, and is collectively termed mTORopathies. However, mechanisms driving epileptogenesis and drug-resistant seizures remain poorly understood. METHODS: We performed integrated genomic
