Longitudinal MRI studies of children with attention-deficit/hyperactivity disorder: A systematic review.

We reviewed longitudinal magnetic resonance imaging (MRI) studies exploring the brain development of children with attention-deficit/hyperactivity disorder (ADHD). Specifically, we conducted a systematic review of papers published until 9 September 2025 in English on pediatric patients with ADHD. After screening 3385 studies, 32 were selected for review. The age range at baseline for the participants in the review ranged from 4 to 18 years old. Children with ADHD exhibited delays in brain volume and cortical thickness compared to typically developing (TD) children, particularly at baseline, wi

Fractal dynamics in spontaneous motor activity associate with effective antidepressant chronotherapeutic response, cerebellar volumes, and white matter microstructure in bipolar disorder.

AIM: Bipolar disorder (BD) is characterized by circadian dysregulation, altered sleep-wake behaviors, and heterogeneous antidepressant response. Chronotherapeutic interventions such as total sleep deprivation combined with light therapy (TSD + LT) produce rapid antidepressant effects, but predictors of outcome are lacking. We investigated whether fractal dynamics of motor activity, indexed by the short-term scaling exponent α₁, are associated with antidepressant response, sleep-wake regulation, and structural brain measures and may serve as a biomarker of treatment responsiveness. METHODS: Si

Intact motor sequence learning but altered error-related brain activation in writer's cramp.

Writer's cramp (WC) is a task-specific arm dystonia with involuntary contractions during writing. While motor circuit dysfunction is well-characterized in WC, the roles of motor learning and error processing remain unclear. We investigated implicit motor sequence learning in WC patients using fMRI during Serial Reaction Time Task (SRTT), focusing on cerebellar activity and error-related processing. 25 WC patients and 17 healthy controls were compared. Both groups showed a comparable sequence-specific advantage with faster reaction times and lower error rates in sequence versus random blocks. W

Brain Dynamics Underlying Reaching Movements Under Uncertainty.

Motor responses are influenced by inferred predictability, but it remains unclear how uncertainty, or its inverse, precision, is encoded within motor control networks. In this study, we combined computational modeling of reaching movement response speed with functional magnetic resonance imaging (fMRI). We used a cueing paradigm with changing cue predictability (~50%, ~70%, and ~90%) to test how predictive inference about left/right reaching responses affected cortical activity and network connectivity in 28 healthy young adults. Behavioral results showed that the response speed differences be

High-SNR Whole-Brain Mesoscale Diffusion MRI Using Rotating-View Acquisitions (ROVER-dMRI).

PURPOSE: We aim to acquire and reconstruct high signal-to-noise ratio (SNR), whole-brain, mesoscale (isotropic submillimeter) diffusion MRI (dMRI) in vivo in a clinically feasible scan time. METHODS: We present ROVER-dMRI, a unified rotating-view acquisition and super-resolution reconstruction framework. Thick slices (up to 10 mm) acquired across many rotated orientations improve the base SNR, and a shared multi-resolution hash-encoded implicit neural representation (INR) with per-direction MLP head and a physics-informed, Bloch-simulated slice-profile forward model reconstructs these views i

Frequency-dependent cerebellar circuits independently gate social vocalizations and movement.

Communication depends on precise coordination between motor execution and cognition. Here we reveal that the cerebellum exerts real-time control over social vocalizations in adult mice. Optogenetic activation of excitatory cerebellar output in the superior cerebellar peduncle suppressed ultrasonic vocalizations with frequency-dependent potency while inducing distinct motor phenotypes. Systematic functional mapping across cerebellar regions and cell types revealed that vocal suppression can occur in the absence of overt motor impairment, suggesting selective control of vocal output beyond gross

Effects of Cerebellar Transcranial Direct Current Stimulation on Motor Learning in Healthy Individuals: A Systematic Review and Meta-Analysis.

Cerebellar transcranial direct current stimulation (tDCS) may enhance motor learning, but findings vary across tasks and stimulation protocols. This systematic review and meta-analysis quantified its effects on motor adaptation and motor skill learning in healthy adults and examined potential moderators. PubMed, the Cochrane Library, MEDLINE, Scopus, Web of Science and Embase were searched from inception to 17 August 2026. The protocol was registered with PROSPERO (CRD42024596088). Randomized controlled and crossover studies comparing cerebellar tDCS with sham stimulation and reporting practic

From Contrasts to Representations: Rethinking fMRI Approaches for Cortico-Cerebellar Networks.

The relationship between the cerebellum and cerebrum remains poorly understood in the neuroimaging literature, despite decades of functional magnetic resonance (fMRI) research. In this opinion article, we argue that this gap reflects fundamental methodological constraints rather than merely a lack of scientific interest. We first discuss challenges in acquiring fMRI data and analysis, with an emphasis on the cerebellum. We then critically evaluate the most common analysis approaches including functional connectivity, univariate contrasts, multivariate pattern analysis, and encoding models, exa

Perinatal insults, genetic susceptibility, and the pathogenesis of neurodevelopmental disorders: from epigenetic remodeling to neural circuit dysfunction-a narrative review.

BACKGROUND AND OBJECTIVE: Neurodevelopmental disorders (NDDs)-including autism spectrum disorder (ASD), attention-deficit/hyperactivity disorder (ADHD), intellectual disability (ID), epilepsy, and schizophrenia-affect approximately 10-15% of children worldwide. Emerging evidence suggests that NDDs arise from complex interactions between genetic susceptibility and environmental insults, particularly during perinatal periods. This narrative review aims to synthesize current evidence on the molecular and circuit-level mechanisms linking perinatal insults and genetic vulnerabilities to NDD pathoge

Brain imaging-derived phenotypes influence risk of bipolar disorder: comprehensive exposome-wide association and wide-angled genetic analyses.

BACKGROUND: Despite many reported imaging markers for bipolar disorder (BD), systematic assessment across diverse brain imaging-derived phenotypes (IDPs) is lacking. We used epidemiological and genetic analyses to progress from association to causality and identify candidate biomarkers and potential pathways of BD. METHODS: We conducted an Exposome-Wide Association Study (ExWAS) in 35,680 UK Biobank participants to assess associations of 686 brain IDPs with BD. Genetic correlations were estimated via linkage disequilibrium score regression (LDSC). Bidirectional two-sample Mendelian randomizat

Dynamic functional abnormalities of cerebellar networks in adolescents with obsessive-compulsive disorder.

BACKGROUND: Obsessive-compulsive disorder (OCD) commonly begins during adolescence, yet its neurobiological basis remains unclear. Beyond the classical cortico-striatal framework, the cerebellum has been implicated in cognitive, affective, and motor regulation, relevant to OCD. However, it is unknown whether adolescent OCD involves abnormal dynamic coordination of large-scale cerebellar networks. METHODS: Thirty-eight adolescents with OCD and 29 matched controls underwent dynamic functional network connectivity (dFNC) analysis of cerebellar networks using a sliding-window approach and k-means

The Computational Functions of the Inferior Olive: Clues from Evolution.

Precerebellar structures, mainly studied in weakly electric fish, act to remove the effects of the fish's own movements on the electrical signals they receive, but do not directly control movement, nor do they have a clear error signal. The cerebellum does directly control movement - arguably the most prominent difference in function between the two structures. The most pronounced anatomical difference between precerebellar structures and the cerebellum itself is that the latter possesses an inferior olive. This conjunction suggests that the inferior olive carries out a function that is necess

Analysis of brain iron deposition in an in vivo model of pediatric autosomal dominant polycystic kidney disease.

Polycystic kidney disease (PKD) represents one form of chronic kidney disease (CKD) affecting the pediatric population. CKD in early life has been associated with neurocognitive deficits and disturbances in iron metabolism. Excess iron accumulation may exacerbate inflammatory and oxidative stress pathways linked to neurocognitive decline in CKD. No animal studies have yet investigated brain iron metabolism in early-life CKD, including PKD, or confirmed these findings through histological analysis. Utilizing a conditional Pkd2 knockout mouse, we investigated the effect of kidney disease on earl

Altered resting-state brain function in diabetic retinopathy: a meta-analysis of combined transcriptome-neuroimaging study.

INTRODUCTION: Patients with diabetes may develop cognitive dysfunction, particularly at advanced stages of the disease, manifested by impairments in learning, memory, language, and judgment. Previous studies have reported abnormal brain functional alterations in patients with diabetic retinopathy (DR), which may be associated with cognitive impairment in diabetes; however, the molecular correlates of these brain alterations remain poorly understood. METHODS: In this study, we performed a coordinate-based neuroimaging meta-analysis to identify convergent resting-state brain functional alterati

Whole-brain precision functional mapping of a proximal upper-extremity motor task.

Upper-extremity motor control has been widely studied with hand-task functional magnetic resonance imaging (fMRI) in healthy and disease states. However, proximal-arm motor tasks, such as at the shoulder, are uncommon during fMRI due to practical limitations, though they are critical to a complete understanding of upper-extremity motor control. Previous fMRI studies of proximal upper-extremity control have generally used unconstrained movements or a single low force or torque condition, leaving unknown how distributed supraspinal brain activity scales with increasing proximal motor output. Haploinsufficiency, de novo and biallelic missense variants in CSMD2 are associated with neurodevelopmental disorders.

CUB and Sushi domains-containing CSMD proteins are involved in neuron development. Variants in the CSMD1-3 paralogs were GWAS-associated with cognition, attention-deficit hyperactivity disorder (ADHD), schizophrenia and brain structure, suggesting that these genes could be linked to neurodevelopmental disorders (NDD). Indeed, rare biallelic variants in all three CSMD paralogs were recently associated with epilepsy, while rare biallelic variants in CSMD1 were also linked with intellectual disability and cortical malformations. We identified nine individuals from eight families carrying CSMD2 bi

Cerebellar Network Organization in Parkinson's Disease: Functional Connectivity across Motor and Cognitive Circuits.

BACKGROUND: The cerebellum is increasingly recognized as a key contributor to cognitive reserve and network organization in Parkinson's disease (PD). However, the relationship of cerebello-cortical and cerebello-basal ganglia connectivity with cognitive status and nigrostriatal integrity remains incompletely understood. METHODS: Resting-state functional magnetic resonance imaging (fMRI) data from 172 participants with PD in the Parkinson's Precision Medicine Initiative were analyzed. Cerebello-basal ganglia and cerebello-cortical connectivity were investigated using region of interest (ROI)-t
