Central monoamine dysfunction in Prader-Willi syndrome: a mechanistic framework for core clinical features.

Prader-Willi syndrome (PWS) is a rare genetic neurodevelopmental and neurobehavioral disorder characterized by hyperphagia and behavioral, endocrine, arousal, and cognitive dysregulation. Although caused by loss of paternally expressed, maternally imprinted genes in 15q11-q13, the mechanisms linking this lesion to the broad clinical phenotype remain incompletely understood. This review synthesizes evidence that central monoaminergic dysfunction, encompassing serotonin, dopamine, and norepinephrine, provides an integrative, though not exclusive, mechanistic framework for core features of PWS. Neuronal surface autoantibodies at cerebellar and brainstem synapses: mechanisms, phenotypes, and translational gaps.

Pathogenic neuronal-surface autoantibodies can disrupt extracellular receptors, ion-channel complexes, and synaptic proteins, yet their regional neurological phenotypes differ substantially. This variability is particularly informative in the cerebellum and brainstem, where precisely timed excitatory and inhibitory signaling supports motor coordination, ocular-motor control, autonomic regulation, arousal, and respiration. In this Mini Review, we examine how antibody effector mechanisms, regional antigen distribution, and local synaptic physiology interact to shape cerebellar and brainstem mani

The cerebellum in migraine: a narrative review of anatomical, imaging, and clinical evidence.

Migraine is usually framed through trigeminovascular pathways and interactions among the brainstem, hypothalamus, and cortex. The cerebellum has received less attention, although human studies place it within circuits that process trigeminal nociception and vestibular signals. This narrative review examines anatomical and experimental work together with imaging, genetic, and clinical studies, giving greatest weight to direct human migraine evidence. Repeated-measures imaging suggests that cerebellar connectivity changes across the migraine cycle, but the cohorts are small. Structural findings

Very Low Frequency of SCA27B Among Undiagnosed Cerebellar Ataxia Patients in Peru.

Spinocerebellar ataxia type 27B (SCA27B) is a recently described autosomal dominant ataxia caused by a pathogenic GAA repeat expansion in the FGF14 gene. The frequency and the distribution of FGF14 repeat alleles remain largely unexplored in underrepresented populations, particularly in Latin America. To investigate the frequency of SCA27B and characterize the allelic distribution of the GAA-FGF14 microsatellite locus in a Peruvian cohort of individuals with ataxia of unknown molecular etiology. We analysed a cohort of affected individuals evaluated at a referral center for neurogenetic disord

Hydrocephalus and hearing loss associated with chronic Brucella ceti infection in stranded bottlenose dolphins Tursiops truncatus.

Brucella ceti has a global distribution and is associated with neurologic disease, systemic infection, and reproductive losses in cetaceans. B. ceti has been associated with perinate mortality and sporadic systemic infections in bottlenose dolphins (BNDs) Tursiops truncatus from strandings both during and outside of unusual mortality events on the southern and eastern coasts of the USA. In 2014-2015, 2 BND from the waters of Texas and Florida stranded on separate occasions, were admitted to rehabilitation facilities, and were later placed under permanent professional care. Both dolphins had su

Obesity-induced oxidative stress and cGMP depletion suppress cerebellar α2,3-Na+,K+-ATPase activity in female mice.

Obesity is a major metabolic disorder linked to neuroinflammation and cognitive decline. Beyond hypothalamic alterations, emerging evidence indicates that extra-hypothalamic regions, including the cerebellum, are vulnerable to obesity-induced neurotoxicity. This study investigated whether high-fat diet (HFD)-induced obesity causes cerebellar oxidative stress, impairs Na+,K+-ATPase (NKA) activity, and triggers molecular features resembling brain aging in female Balb/C mice. Chronic 10-week HFD intake established a marked obese phenotype, characterized by increased adiposity, fasting hyperglycem

Chemically Modified ω-Agatoxin IVA Enables Exploration of P/Q-Type Voltage-Dependent Calcium Channels.

P/Q-type voltage-dependent calcium channels (VDCCs) are involved in the regulation of neurotransmitter release and in the integration of dendritic inputs in the central nervous system (CNS). However, their precise localization and protein networks in the CNS remain elusive due to the lack of molecular tools applicable to the delicate and complex three-dimensional environment of the living brain. In this study, we synthesized peptide-based probes using ω-agatoxin IVA (AgTX), a specific channel blocker for P/Q-type VDCCs derived from the funnel-web spider Agelenopsis aperta. The far-red fluoroph

Machine learning of connectome-wide functional connectivity in spinal muscular atrophy.

While traditional view posed SMA as a pure motor neuron disorder, accumulating evidence pointed to the broader involvement of central nervous system. This study aimed to combined connectome-wide resting-state functional connectivity with machine learning algorithms to investigate the underlying brain mechanisms in SMA and capture the potential neuroimaging biomarker. Resting-state functional magnetic resonance imaging were acquired from 43 SMA (26 SMA type 2 and 17 type 3) and 45 healthy controls (HCs). Connectome-wide functional connectivity, defined based on the Automated Anatomical Labeling

Hierarchical reorganization of cortical and cerebellar functional gradients in Parkinson's disease.

INTRODUCTION: Parkinson's disease (PD) is a common movement disorder with heterogeneous clinical manifestations, and its tremor-predominant presentations may overlap with essential tremor (ET), complicating clinical differentiation in some patients. Functional gradient analysis provides a continuous framework for characterizing macroscale brain hierarchical organization and may offer candidate imaging features for disease characterization. METHODS: We first investigated surface-based resting-state functional measures and cerebral perfusion in 50 patients with idiopathic PD and 33 healthy cont

Cytoplasmic TDP-43 Accumulation in Autosomal Recessive Spastic Ataxia of Charlevoix-Saguenay.

BACKGROUND: Autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS) is a rare, early-onset neurodegenerative disorder characterized by progressive cerebellar ataxia, spasticity and peripheral neuropathy. It is caused by biallelic mutations in the SACS gene, which encodes sacsin, a large protein involved in cytoskeletal organization and mitochondrial homeostasis, and protein quality control. Despite advances in understanding sacsin biology, the molecular mechanisms underlying neurodegeneration in ARSACS remain incompletely understood. Here, we report the first neuropathological evide

A transcriptomic resilience gap between the substantia nigra and cerebellum is associated with regional vulnerability in Parkinson's disease.

The mechanisms underlying the selective vulnerability of the substantia nigra (SN) in Parkinson's disease (PD) are incompletely understood. We computed the tissue-specificity index τ for 19,355 protein-coding genes across 68 GTEx v11 tissues, defining a gene as tissue-specific when τ ≥ 0.8, median TPM > 1, and the tissue of interest was the absolute maximum. Under this definition, the SN harboured only 12 SN-specific genes whereas the cerebellum (CB) contained 367 CB-specific genes. The direct pairwise contrast was not statistically significant (exact p = 0.090). The primary evidence is a nega

Effect of Different Cortical Targets of Transcranial Magnetic Stimulation on Freezing of Gait in Parkinsonism: A Systematic Review.

BACKGROUND AND PURPOSE: Freezing of gait (FOG) is a disabling and episodic gait disturbance in patients with Parkinson's, with varying responses to noninvasive brain stimulation. Transcranial magnetic stimulation (TMS) has been explored as a therapeutic option; however, the optimal cortical target for reducing FOG remains unclear. This systematic review synthesizes and critically evaluates the available evidence regarding the effects of TMS applied to different cortical targets on FOG-related outcomes, with emphasis on the indirect nature of comparisons between targets. METHODS: A systematic

Cellular scaling laws in the mammalian brain.

Recent whole-brain cell atlases have uncovered a consistent cytoarchitectural feature: the greatest diversity of discrete neuronal cell types resides not in the regions with the most neurons (e.g., the cortex and cerebellum) but rather in deep subcortical structures like the hypothalamus and brain stem. We propose that this discrepancy reflects a fundamental algorithmic division in the vertebrate brain between a "learning subsystem" and a "steering subsystem." The learning subsystem (cortex, striatum, and cerebellum) scales via the replication of repetitive modules to maximize computational ca

Basal Forebrain Volume is Associated With Cortical Amyloid Burden in Cognitively Unimpaired Older Adults at Varying Genetic Risk for Alzheimer's Disease.

BACKGROUND: In mild cognitive impairment and dementia due to Alzheimer's disease (AD), postmortem and in vivo neuroimaging studies have demonstrated significant neuronal loss in the basal forebrain cholinergic system (BFCS), which provides the primary cholinergic input to the cerebral cortex. Within the BFCS, atrophy is most prominent in the nucleus basalis of Meynert (nbM), a posteriorly located cluster of magnocellular neurons. However, less is known surrounding the relationship between amyloid deposition, BFCS atrophy, and medial temporal lobe (MTL) volume loss in preclinical AD. This study

Thymoquinone-Loaded Hexosomal Nanoparticles Attenuate Neuroinflammation and Improve Cognitive Function in an Alzheimer's Disease Mouse Model.

INTRODUCTION: Alzheimer's disease (AD) involves neuroinflammation that worsens cognitive decline. Thymoquinone (TQ) has potent anti-inflammatory activity, but its therapeutic application is limited by poor solubility, stability, and systemic bioavailability, which may restrict effective brain exposure. This study developed TQ-loaded hexosomal nanoparticles (TQ-HNP) to enhance TQ delivery and therapeutic efficacy in a dizocilpine (MK-801)- induced mouse model of AD. METHODS: TQ-HNP were prepared using glyceryl monooleate, oleic acid, and poloxamer 407, optimized to ~104 nm size, -31 mV zeta po

WDGTA: a wavelet-denoised graph attention framework for spatial transcriptomics deconvolution.

Spatial transcriptomics (ST) facilitates the spatial resolution of gene expression profiling; however, many widely used spot-based sequencing platforms capture transcripts from multiple neighboring cells at each location, producing mixed profiles that require accurate cell-type deconvolution. The wavelet-denoised graph attention framework for spatial transcriptomics deconvolution (WDGTA), a graph attention-based framework for ST cell-type deconvolution, was developed and validated on five datasets from two species and four organs. Through the integration of real with pseudo-ST spots, WDGTA int

Carbon-12 and Gamma-Rays Combined Irradiation Ameliorates Behavioral and Biochemical Alterations in Tau P301S Homozygous Transgenic Mice.

No effective treatments currently exist for tauopathies. Given emerging evidence that moderate-dose ionizing radiation (IR) may exert neuroreparative effects in neurodegeneration, we investigated the impact of combined IR exposure on disease progression in the Tau P301S mouse model. Tau P301S mice received combined gamma-ray (0.24 Gy) and carbon-12 nuclei (0.18 Gy, 400 MeV/n) irradiation at the presymptomatic stage. Following longitudinal behavioral testing until the terminal stage, post-mortem brains were analyzed via RT-qPCR, ELISA, and HPLC to assess gene expression, protein levels, and mon
