The effects of interaural time and level differences on the binaural interaction component of the auditory brainstem response in Cope's gray treefrogs.

Processing binaural cues is crucial for many acoustic communication tasks, and understanding the neural mechanisms of binaural sound processing gives important insights into how receivers localize signalers, which in anurans is a key component of mate selection. We characterized the binaural interaction component (BIC) of the auditory brainstem response (ABR) by collecting monaural and binaural ABRs using click stimuli with varying interaural time difference (ITD) and interaural level difference (ILD) in male and female Cope's gray treefrogs (Hyla chrysoscelis). We hypothesized that the latenc

A tale of two ears: development of binaural auditory processing in the big brown bat.

Big brown bat (Eptesicus fuscus) pups are born naked, blind, and with closed external ear canals. We examined the ears of newborn bat pups (n = 82; 42 females) and found that in a majority of pups (80%), the ear canals open asymmetrically. What remains unknown is whether this developmental asymmetry influences hearing. We recorded auditory brainstem response (ABR) waveforms in a subset of pups with both ears open by placing subdermal needle electrodes near the base of the auditory bullae and repeatedly presenting broadband acoustic clicks and averaging the evoked neural response. The resultant

Slow-wave sleep engages brainstem circuitry to prevent stress-induced anxiety.

The beneficial effects of sleep on anxiety are established, but the mechanisms remain unclear. We identify a GABAergic circuit from the parafacial zone (PZ) to the lateral parabrachial nucleus (LPB) neurons that project to the oval bed nucleus of the stria terminalis (ovBNST) as a node for slow-wave sleep (SWS)-mediated anxiolysis. Optogenetic activation of PZ GABAergic neurons following social defeat stress induces time-locked SWS and prevents anxiety. Multi-region Ca2+ recording reveals suppressed activity in LPB and ovBNST during natural and PZ-initiated SWS. The LPB-ovBNST pathway is requi

Use of trans-tympanic electrically evoked auditory brainstem responses (TT-EABR) in decision making of cochlear implants in hypoplastic auditory nerve cases.

INTRODUCTION: Cochlear Nerve Deficiency, particularly Hypoplastic Auditory Nerve (HAN), creates uncertainty regarding Cochlear Implant (CI) outcomes. This study evaluated the utility of Transtympanic Electrically Evoked Auditory Brainstem Response (TT-EABR) in managing HAN cases. METHODS: Twenty-two children with MRI-confirmed HAN were assessed using TT-EABR to guide ear selection for CI vs Auditory Brainstem Implantation (ABI) candidacy. Consequently, 13 children received CIs (HAN -CI group). Their auditory and speech outcomes -measured via CAP, SIR, and MAIS/IT-MAIS at baseline and 12 month

Asc-1/SLC7A10 regulates NMDA receptor co-agonist availability at hypoglossal motoneurons.

The Asc-1/SLC7A10 transporter regulates extracellular levels of the NMDA receptor co-agonists glycine and D-serine, yet its role at excitatory synapses in the brainstem remains poorly understood. We investigated how pharmacological Asc-1 blockade modulates NMDA receptor-mediated synaptic transmission at hypoglossal motoneurons and whether glycine or D-serine is the functionally dominant co-agonist at these synapses. Whole-cell patch-clamp recordings were performed in Mg2+-free ACSF from hypoglossal motoneurons in brainstem slices of neonatal mice. Bath application of the selective Asc-1 inhibi

Cut through the noise: vocal signals and auditory processing vary along an urbanization gradient in Pacific treefrogs.

From dense assemblages of vocalizing organisms in neotropical rainforests, to fast-flowing streams and waterfalls in the Old World, Peter Narins' work has been instrumental to our understanding of how animals, particularly frogs, communicate in noisy environments. With increasing levels of human-generated noise, there has been an explosion of studies on how vocal signals and signaling behaviors change in urban soundscapes. Surprisingly, we know little about changes in sensory-processing mechanisms of receivers. We asked whether vocal signals, hearing sensitivity, and the ability to extract sig

Vestibular function in degenerative parkinsonism.

INTRODUCTION: The degenerative pathology in Parkinson's disease (PD), multisystem atrophy (MSA) and progressive supranuclear palsy (PSP) have different spatial distribution in the brainstem. Our aim was find out in what way this pathology affects brainstem vestibular reflex pathways in each of these diseases. METHODS: We studied 30 patients: 15 with PD, 9 with PSP (Richardson syndrome) and 6 with MSA (cerebellar) and compared their results with results from 37 age-matched healthy controls (HC). Motor functions of the patient groups were evaluated with the Unified Parkinson's Disease Rating Sc

Transcriptomic Analysis Identifies the PVT1-miR-34a Axis as a Novel Non-Coding RNA Therapeutic Target in Diffuse Intrinsic Pontine Glioma.

OBJECTIVES: Diffuse intrinsic pontine glioma (DIPG) is a lethal paediatric brainstem tumour with a median survival under one year and no effective therapy. Although non-coding RNAs (ncRNAs) regulate tumour progression, their regulatory networks in DIPG remain poorly characterised. This study aimed to identify dysregulated lncRNA- miRNA-mRNA axes in DIPG through integrated transcriptomic analysis, computationally validate these networks, and investigate their downstream impact on cancer-related pathways to reveal novel therapeutic targets for RNAbased precision medicine. METHOD: This study was

Prevalence and risk factors for failed newborn hearing screening among preterm infants: a prospective cohort study in Nairobi, Kenya.

BACKGROUND: Despite the high burden of global childhood hearing loss, compliance with universal newborn hearing screening (UNHS) guidelines is low in many low- and middle-income countries, including Kenya. Preterm infants are especially prone to an increased risk of hearing loss due to the associated perinatal and neonatal morbidities. The objectives of this study were to investigate the prevalence and risk factors of failed hearing screening in preterm newborns. METHODS: We conducted a hospital-based prospective cohort study of preterm infants at the Kenyatta National Hospital (KNH) and the

Brainstem volume and arterial stiffness interact to support memory in midlife.

Historically regarded as a hub for autonomic control, the brainstem is now increasingly recognized for its role in cognition. However, little information is available regarding its specific contribution to cognitive function, particularly in midlife, when modifiable cardiovascular risks begin to shape brain health trajectories. Because the brainstem houses key nuclei that regulate cardiovascular dynamics and cerebral perfusion, we focused on carotid-femoral pulse wave velocity (cfPWV), a well-established index of central arterial stiffness that directly reflects cerebral hemodynamic load. This

Minimal invasive surgery for primary brainstem intracerebral hemorrhage: a critical mini review.

CONTEXT: Intracerebral hemorrhage (ICH) accounts for half of stroke attributable deaths and two thirds of DALYs loss. Past well designed trials (2005 STICH, 2013 STICH 2, 2019 MISTIE 3, 2024 ENRICH, 2024 SWITCH) did not demonstrate benefits of surgery, aside from ENRICH which suggested improved functional outcome thanks to minimal invasive surgery for lobar ICH, but not for deep seated thalamic ICH. Since 2020, retrospective studies suggest great functional benefit of minimal invasive surgery for brainstem ICH. MATERIALS AND METHODS: Review of weel designed trials and guidelines regarding sur

Long-term integrative personalized medicine care in massive brainstem hemorrhage: A CARE-compliant case report.

RATIONALE: Brainstem hemorrhage is a rare but devastating subtype of intracerebral hemorrhage that frequently presents with impaired consciousness, quadriplegia, and recurrent infections. Integrative personalized medicine care (IPMC), which combines essential conventional treatments with personalized traditional Korean medicine interventions, has been suggested to improve functional outcomes and support immune regulation. By closely monitoring a patient with massive brainstem hemorrhage treated with IPMC, we observed neurological improvement, effective control of recurrent infections and infla

Progressive motor impairment from intracerebral critical demyelinating lesions.

BACKGROUND: Focal critical demyelinating lesions anatomically associated with progressive motor impairment are described in multiple sclerosis (MS) and restricted demyelinating diseases, typically within the spinal cord lateral column and ventral cervicomedullary junction (CMJ). This study describes such lesions within the intracerebral corticospinal tracts. METHODS: We retrospectively reviewed people seen at Mayo MS Clinic (2002-2025) with progressive central nervous system demyelinating disease and progressive (≥ 1 year) upper motor neuron weakness anatomically associated with a presumed de

Castanopsis echinocarpa extract attenuates diabetes-associated peripheral-to-central auditory electrophysiological dysfunction in streptozotocin-induced diabetic mice.

Diabetes-associated auditory dysfunction may extend beyond cochlear threshold elevation and involve broader metabolic and auditory neural abnormalities. This study investigated whether Castanopsis echinocarpa extract (CAE) attenuates peripheral-to-central auditory electrophysiological dysfunction in streptozotocin (STZ)-induced diabetic mice and modulates related auditory and pancreatic stress responses. HEI-OC1 auditory cells were exposed to neomycin or inflammatory cytokines, and Tnf, Il1b, Trpv1, and Gjb2 mRNA expression was analyzed. Post-exposure otic hair cell recovery was assessed in ne

Age-related changes in acoustic cue use for speech-in-speech perception.

Acoustic cues such as pitch and spatial location allow listeners to attend to a target speaker and ignore competing talkers, aiding speech recognition in background noise. Diminished ability to utilize acoustic cues for speech stream segregation may thus contribute to older adults' challenges hearing in noise. Adults aged 18-74 completed a speech-in-speech identification task with three conditions containing 1) only pitch cues (fundamental frequency), 2) only spatial cues (interaural time differences; ITDs), or 3) both pitch and spatial cues for segregating a target talker from competing talke

Longitudinal changes in DTI-ALPS index are associated with clinical progression and brainstem atrophy in spinocerebellar ataxia type 3.

BACKGROUND: The perivascular and neurofluid-related tissue environment is implicated in neurodegeneration, but its clinical relevance in spinocerebellar ataxia type 3 (SCA3) remains unclear. We investigated alterations in diffusion tensor image analysis along the perivascular space (DTI-ALPS) across disease stages, and its longitudinal associations with clinical progression and brain atrophy. METHODS: We included 129 SCA3 (44 preclinical, 85 ataxic) at baseline, 51 with longitudinal follow-up, and 78 controls. Redefining PHOX2B-Related Neurodevelopmental Disorders as Circuitopathies: A New Paradigm in Autonomic Network Medicine.

The paired-like homeobox transcription factor 2B (PHOX2B) is a master regulator whose mutations cause a spectrum of severe neurological and neurodevelopmental disorders, including Congenital central hypoventilation syndrome (CCHS), Hirschsprung disease, and neuroblastoma. For decades, a central paradox has challenged the field: how can PHOX2B's seemingly restricted expression in the brainstem account for the broad, multi-system clinical phenotypes observed in patients? Here, we elucidate how this paradox has been resolved by recent methodological advances. We synthesize evidence from multiple

Hearing Aid Benefit in Auditory Neuropathy Due to Friedreich Ataxia: A Case Report.

PURPOSE: Friedreich ataxia is one of the most common recessively inherited ataxias, with hearing loss due to auditory neuropathy being a recognized complication. However, there are limited data on the best interventions for hearing loss in these patients. We report the significant behavioral and subjective benefits of binaural amplification in a patient with auditory neuropathy due to Friedreich ataxia. METHOD: Auditory function was assessed in a 44-year-old woman with genetically confirmed Friedreich ataxia using pure-tone audiometry, speech audiometry, and immittance testing, along with aud
