Synaptic Facilitation Enhances the Reliability and Precision of High-Frequency Neurotransmission.

The small and tortuous volume of synaptic clefts limits the diffusion of Ca2+ ions during high-frequency spiking. Extracellular Ca2+ levels ([Ca2+]o) of 0.8 mM or lower have been measured or calculated for different synapses. Here, we recorded evoked postsynaptic potentials (EPSPs) and action potentials (APs) from young-adult male and female mouse auditory brainstem principal neurons to investigate the relationship between neurotransmission reliability, stimulation frequency, and [Ca2+]o In 0.8 mM [Ca2+]o, we observed AP failures during stimulation at 100 Hz. Surprisingly, AP failures, EPSP-AP

Cortical and Sensory Integration in Brainstem Circuits for Mastication.

Mastication is a vital function that relies on precise synchronization among multiple brainstem regions, known as being part of a central pattern generator (CPG). Movements can be triggered either by stimulating a sensory-motor region called the cortical masticatory area (CMA), well-documented in various species but not yet formally identified in mice, or by stimulating the orofacial sensory primary afferents, the fibers of which form the trigeminal tract (Vtr). However, it is unclear whether these different inputs activate distinct components of the CPG or converge on the same. This study aim

The NERD model: reflex circuit dysfunction as a systems-level driver of persistent post-concussion symptoms.

Persistent post-concussive symptoms are often attributed to diffuse cortical dysfunction, yet this perspective may overlook key systems-level mechanisms. We propose a conceptual framework in which dysfunction arises from the dynamic interplay among five functional nodes: the sensory interface, reflex-brain stem hub, cerebellar module, basal ganglia-thalamic modulator, and cerebral cortex. Grounded in clinical observation and systems-level dynamic modeling, this framework treats the brain as a time-evolving control network that processes inputs, integrates them across hierarchical nodes, and ge

Associations of standard and extended high-frequency thresholds with suprathreshold auditory brainstem response morphology in normal-hearing adults.

The Auditory Brainstem Response (ABR) is essential for assessing neural integrity, yet the influence of subclinical extended high-frequency (EHF) deficits on waveform morphology remains insufficiently characterized. This cross-sectional study evaluated the distinct predictive utility of Global Standard Averages (0.125-8 kHz) versus EHF thresholds (9-20 kHz) for ABR latencies and amplitudes in 89 otologically healthy adults. While all but one participants exhibited clinically normal hearing thresholds (≤20 dB HL) across standard frequencies, multiple linear regression analyses were employed to

A dual-delivery nanotherapeutic strategy using chitosan-epigallocatechin-3-gallate nanoparticles encapsulated in exosomes: A novel approach to combat brain aging.

Brain aging is a multifactorial process associated with oxidative stress, chronic neuroinflammation, and synaptic dysfunction, ultimately leading to cognitive decline and increased susceptibility to neurodegenerative disorders. Epigallocatechin gallate (EGCG) is a potent antioxidant and anti-inflammatory agent, but its therapeutic potential is limited by poor stability and bioavailability. In this study, a dual nano delivery system was developed by loading chitosan-EGCG nanoparticles into mesenchymal stem cell-derived exosomes (Ex-Chit-EGCG NPs) and evaluated for neuroprotective efficacy in a

Pre-Neural Source of the Envelope-Following Response Revealed in Cases of Auditory Neuropathy.

OBJECTIVES: Complex stimuli such as two-tone stimuli (f 1 and f 2 ) elicit frequency-following responses that reflect phase-locking to the stimulus envelope (f 2 - f 1 ) and temporal fine structure at multiple stages of the auditory pathway, from the cochlea to the cortex. The relative contribution of these structures to the scalp-recorded envelope-following response (EFR) remains a matter of debate. Although subcortical sources have been proposed as the main contributors, near-field recordings close to the cochlea contain pre-neural components and have recently been reported in auditory stead

Targeting Pediatric Glioblastomas by Combining OLIG2 Inhibitor CT-179 with Fractionated Radiation in a Panel of Patient-Derived Orthotopic Xenograft Mouse Models.

The poor clinical outcomes of pediatric high-grade glioma (pHGG) highlight the urgent need for new therapies. Oligodendrocyte lineage transcription factor 2 (OLIG2) is a pro-mitotic transcription factor highly expressed in glioma stem cells and may represent a novel therapeutic target. To evaluate the therapeutic efficacy of an OLIG2 inhibitor CT-179 in pHGG, we determined the OLIG2 mRNA expression in 10 patient-derived orthotopic xenograft (PDOX) models. In vitro activities of CT-179 were analyzed in monolayer and neurosphere cells (0-10 µM) with and without radiation (XRT) (0-8 Gy), brain pe

XKR8 Deletion Protects Against Noise-Induced Hearing Loss by Attenuating Apoptosis and Preserving Mitochondrial Bioenergetics in the Cochlea.

This study elucidated the role of the XKR8 gene in noise-induced hearing loss (NIHL) through animal experiments, PCR experiments, inner ear morphological experiments, ATP and mitochondrial complex I and II assays, and Western blotting experiments. The results showed that pre-exposure ABR showed no significant difference in baseline hearing thresholds between XKR8-/-and WT mice (P > 0.05). On day 1 post-exposure, ABR results indicated hearing thresholds around 85 dB in both groups. On days 3, 7, and 14, XKR8-110 dB mice had lower hearing thresholds than WT-110 dB mice (P < 0.05). The succinate prodrug NV354 prevents brain lesions and late-stage motor dysfunction in mitochondrial complex I deficiency.

Leigh syndrome is a fatal pediatric neurodegenerative disease caused by mitochondrial dysfunction, which can be modeled in the Ndufs4 KO mouse with mitochondrial respiratory chain complex I (CI) deficiency. This study explores NV354, a prodrug of succinate with enhanced oral bioavailability and brain uptake, as a potential therapy to counteract this devastating condition. NV354 modulated whole-body respiration and metabolic flexibility, prevented late-stage motor dysfunction, delayed clinical ataxia scores, and improved body weight development, but had otherwise minimal effect on neurobehavior

Nitric oxide synthase activation in nTS is essential to NMDA receptor dependent encoding of the cough reflex.

Polymodal vagal afferent nerves terminating in the mucosa of the large conducting airways play essential roles in regulating cough. These cough receptors project bilaterally from the nodose ganglia, are activated by protons and by mechanical stimulation, and terminate centrally in the medial (SolM) solitary tract nuclei (nTS). Although many vagal afferent nerves utilize non-NMDA type glutamate receptors exclusively for transmission centrally, cough receptor signaling requires NMDA receptor activation. We hypothesized that nTS nitric oxide synthase (NOS) activation and a cGMP transduction casca

Modeling GABAergic hyperexcitability in sleep bruxism patient-derived brainstem neurons using a multielectrode array platform.

OBJECTIVES: Sleep bruxism (SB) involves involuntary jaw movements during sleep and has been linked to impaired inhibitory regulation of brainstem circuits (particularly those involving GABAergic neurons). While previous studies using patch-clamp electrophysiology have demonstrated intrinsic hyperexcitability in neurons differentiated from SB patient-derived human induced pluripotent stem cells (hiPSCs), the low-throughput nature of this technique limits large-scale phenotypic screening. We aimed to establish a robust, high-throughput, multielectrode array (MEA)-based platform capable of quanti

An open-source deep learning-based toolbox for automated auditory brainstem response analyses (ABRA).

Hearing loss is a pervasive global health challenge with profound impacts on communication, cognitive function, and quality of life. Recent studies have established age-related hearing loss as a significant risk factor for dementia, highlighting the importance of hearing loss research. Auditory brainstem responses (ABRs), which are electrophysiological recordings of acoustically evoked synchronized neural activity from the auditory nerve and brainstem, serve as in vivo correlates for sensory hair cell and synaptic function, hearing sensitivity, and other critical readouts of auditory pathway p

The sympathoregulatory region of the mouse rostral brainstem relies on both GABA and glycine to generate inhibitory currents.

Presympathetic neurons in the ventrolateral and ventromedial medulla (VLM/VMM) control sympathetic tone, and GABA and glycine were identified as inhibitory neurotransmitters. Although glycinergic inhibition of presympathetic neurons in the VLM/VMM has been shown, the sources of glycinergic inputs and the mechanism of glycine release are ill-defined. Here, we tested whether presympathetic neurons in the VLM/VMM receive glycinergic inputs. Glycine is recycled by glycine transporter 2 (GlyT2), which is a reliable marker of glycinergic neurons. GlyT2 Cre mice were cross bred with channelrhodopsin

Differences in auditory brainstem responses between laboratory-reared and wild-caught prairie voles (Microtus ochrogaster).

Prairie voles (Microtus ochrogaster) are a semi-fossorial rodent that are an emerging model in social neuroscience. Comparing laboratory-reared and wild-caught individuals is essential for understanding how environmental history shapes neural and sensory traits and for assessing the ecological validity of laboratory findings. Despite this, relatively few studies have taken this approach. We used auditory brainstem responses (ABRs) to compare ABR thresholds and ABR wave characteristics between laboratory-reared and wild-caught prairie voles. ABR recordings show that, similar to other semi-fosso

Therapeutic potential of Cuscutae Semen and Rehmanniae Radix Preparata in age-related central hearing loss associated with Gad1 regulation.

ETHNOPHARMACOLOGICAL RELEVANCE: Cuscutae Semen and Rehmanniae Radix Preparata have traditionally been prescribed in East Asian medicine for auditory disorders associated with aging-related kidney deficiency, as documented in classical texts including Mingyi Bielu, Bencao Gangmu, Donguibogam, and Bangyakhappyeon. However, their relevance to age-related hearing loss (ARHL) and underlying mechanisms remains unclear. AIM OF THE STUDY: This study aimed to evaluate the protective effects of a combined extract of Cuscutae Semen and Rehmanniae Radix Preparata (TS) against ARHL, focusing on its effect

Lateral hypothalamus directs stress-induced modulation of acute and psoriatic itch.

Stress modulates itch, with acute stress suppressing and chronic stress exacerbating pruritus, yet the underlying neural mechanisms remain unclear. In this study, we investigate the role of lateral hypothalamic area (LHA) neurons in stress-induced itch modulation. Using neural-activity-dependent genetic labeling and chemogenetic tools, we selectively engaged a stress-sensitive population of LHA neurons (LHAstress-TRAP neurons). Transient activation of these neurons elicited anxiety-like behavior and place aversion while suppressing both acute (chloroquine-induced) and chronic (psoriatic) itch;

Morpho-functional correlates of gait impairment in early Parkinson's disease patients: insights from a multimodal path modeling framework.

OBJECTIVE: To deepen the underlying pathophysiology of gait impairment in early-stage Parkinson's disease (PD) through a multimodal assessment, including MR-based brainstem morphometry and EEG functional connectivity (FC). METHODS: 73 PD patients and 60 healthy controls (HC) were included. The MR Parkinsonism Index (MRPI) was calculated for each subject. FC was measured using HD-EEG in θ-α-β-low-γ-high-γ bands. Partial Least Squares Path Modeling (PLS-PM) was used to explore the causal relationships between MRPI, FC, and clinical data. RESULTS: MRPI was higher in PD than HC (p = 0.001) and c
