Bacillus cereus biovar anthracis causes inhalational anthrax-like disease in rabbits that is treatable with medical countermeasures.

Bacillus anthracis is a zoonotic organism that causes the disease anthrax due to the activity of virulence factors harbored on plasmids pXO1 and pXO2. Inhalation of B. anthracis spores results in pneumonic disease that progresses quickly, and often results in lethality in the absence of medical countermeasure (MCM) intervention. Recently, reports have identified Bacillus cereus isolates that possess pXO1 and pXO2-like plasmids and cause an anthrax-like disease. These isolates have been named B. cereus biovar anthracis, or Bcbva. To evaluate disease course of Bcbva, the inhalational median leth

Decontamination of Soil Contaminated at the Surface with Bacillus anthracis (Anthrax) Surrogate Spores Using Steam Vapor.

In the event of a wide-area release of Bacillus anthracis spores, soils and other outdoor materials will likely become contaminated with the biological agent. Soils may also become contaminated with B. anthracis when livestock or wildlife succumb to anthrax disease. This study was conducted to assess the in situ remediation of soil using steam vapor to inactivate a B. anthracis spore surrogate (Bacillus atrophaeus) inoculated into soil samples. Tests were conducted using small columns (~0.04 m3 of soil) filled with either loam, clay, or a sandy soil. Following steam treatment, the B. atrophaeu

Distinct off-on fluorescence signal from dual-response lanthanide metal-organic frameworks for ratiometric sensing of anthrax biomarker.

Bacillus anthrax is a highly dangerous zoonotic pathogen that represents a significant threat to public health. 2,6-Dipicolinic acid (DPA) is typically designated as the primary biomarker of Bacillus anthrax, which is a distinctive major component of anthrax spores. The development of sensitive, efficient, and rapid detection of this marker has significant implications for the elimination of potential threats and the safeguarding of public health and food safety. In this study, we have designed a dual lanthanide ion (Ce3+, Eu3+) functionalized metal-organic framework fluorescent probe, Ce@Eu@M

The Anthrax Toxin Lethal Factor in Solution Does Not Have the Protein's Crystallized Structure.

The bacterium Bacillus anthracis secretes three protein exotoxins: Protective Antigen 83 (PA83), Lethal Factor (LF), and Edema Factor (EF). A cleaved form of PA83 (PA63) aids LF and EF entry into the cytoplasm, which leads to anthrax-induced cell death. The Protein Data Bank (PDB) has more than 25 structures of LF: the monomer alone, bound with inhibitors, or bound to PA63. The structures are all-with only minor shifts of a few Ångströms-nearly congruent. We have measured the structure of LF at equilibrium in D2O solution by small-angle neutron scattering (SANS). The shape is modeled well by a

Pleiotropic functions of CpdB in Bacillus anthracis.

Bacillus anthracis is the causative agent of anthrax disease. Our prior research indicated that cyclic (c)-di-AMP accumulation attenuated bacterial virulence in mice. However, the function of c-di-AMP secretion and its regulatory mechanism in anthrax have not been revealed. We characterized the role of the ecto-nucleotidase CpdB in B. anthracis. CpdB exhibits phosphodiesterase activity towards c-di-AMP and 2'3'-cGMP-AMP, and nucleotidase activity towards several mononucleotides, including the preferred substrates c-di-AMP and pApA. Our results demonstrated that inactivation of cpdB altered pur

Exploring community knowledge, perceptions, and the impacts of anthrax among farming communities living in game management areas in Zambia: A qualitative study using a hybrid approach.

Anthrax remains a neglected zoonotic disease of critical public and animal health significance in Zambia, particularly in regions with active human-wildlife-livestock interfaces such as the Western, Southern and Eastern provinces of Zambia. This study explores the socio-ecological drivers of anthrax transmission and examines the role of legal and illegal wildlife trade value chains in sustaining outbreaks. Secondly, the study explores the methodology used to investigate community knowledge, perceptions, and the impacts of anthrax through focus group discussions (FGDs) and a hybrid approach com

Serological correlates of protection and long-term efficacy of a novel recombinant protective antigen-based anthrax vaccine in a rabbit model.

The novel anthrax vaccine candidate GC1109, composed of recombinant protective antigen, has shown robust antibody responses and safety in preclinical and clinical studies. However, the assessment of GC1109 vaccine efficacy was limited in a rodent model and could not be applied in clinical studies due to ethical issues. In this study, we aim to provide predictive insights for human applications of GC1109 by examining the correlation between anthrax toxin-neutralizing antibodies (TNAs) and protection against Bacillus anthracis infection in rabbit. The threshold level of TNAs necessary for protec

Development of a high-throughput method for processing sponge-stick samples to detect viable Bacillus anthracis spores.

Since the national validation of the sponge-stick based method for detection of Bacillus anthracis spores in environmental samples, there have not been focused efforts to address the low throughput nature of the method, which processes only one sample at one time. Sample processing remains a serious bottleneck for rapidly analyzing large numbers of samples expected from a biological warfare attack. Therefore, we developed a high-throughput method to simultaneously process multiple sponge-stick samples to be better prepared for rapid response and recovery after wide area anthrax incidents. In t

A novel one-tube RPA/CRISPR melting curve detection sensing system based on unique 3'-toehold nucleic acid aptamer for Bacillus anthracis detection.

Developing effective detection methods for Bacillus anthracis is essential for our public health system to accurately detect hidden anthrax outbreaks. Herein, we introduce a unique 3'-toehold nucleic acid aptamer (probes) into Cas12a biosensor, combined with RPA, to establish a rapid (1h), specific, and sensitive (1copy/μL) detection method for B. anthracis. The design behind this approach is that the target sequence is amplified via RPA, and the amplification product triggers the crRNA/Cas12a complex to degrade the 3' toehold probes, which are analyzed using melting curve analysis on a specif

FapR regulates HssRS-mediated heme homeostasis in Bacillus anthracis.

UNLABELLED: Bacillus anthracis, a Gram-positive facultative anaerobe and the causative agent of anthrax, multiplies to extraordinarily high numbers in vertebrate blood, resulting in considerable heme exposure. Heme is an essential nutrient and the preferred iron source for bacteria during vertebrate colonization, but its high redox potential makes it toxic in excess. To regulate heme homeostasis, many Gram-positive bacteria, including B. anthracis, rely on the two-component signaling system HssRS. HssRS comprises the heme sensing histidine kinase HssS, which modulates the activity of the HssR

A Nasal Vaccine Displaying Anthrax Antigen on the Surface of Lactiplantibacillus plantarum Induces Protective Mucosal Immunity against Anthrax Toxin.

Anthrax, particularly inhalational anthrax, poses a severe threat due to its high fatality rate and potential misuse as a biological weapon. Current anthrax vaccines require multiple injections, which limits their practicality and underscores the need for improved, user-friendly vaccination strategies. In this study, we developed a novel nasal booster vaccine platform aimed at preventing inhalational anthrax by inducing mucosal immunity at the pathogen's entry site, the respiratory tract. We successfully engineered a biologically safe Lactiplantibacillus plantarum (L. plantarum) strain to disp

Redox-Active Europium-Based MOFs Showing Ratiometric Fluorescence Sensing for the Anthrax Biomarker and Oxytetracycline, and Proton Conduction.

Redox-active metal-organic frameworks have received increasing attention for potential applications in conductivity, sensing, catalysis, and magnetism. In this work, a novel redox-active europium-based metal-organic framework, {[(CH3)2NH2][Eu(BTA-2OH)(H2O)2]·2H2O}n (Eu-MOF, H4BTA-2OH = 2,3,5,6-tetrakis(p-carboxyphenyl)hydroquinone), is prepared and structurally characterized. The Eu-MOF is characterized by a well-defined structure featuring a two-dimensional channel along the c-axis. Eu-MOF can be converted into Eu-MOF-Ox through the oxidation of hydroquinone to benzoquinone, which can maintai

Pore formation by the CDTb component of the Clostridioides difficile binary toxin is Ca2+-dependent.

Clostridioides difficile infection (CDI) is one of the five most urgent bacterial threats in the United States. Furthermore, hypervirulent CDI strains express a third toxin termed the C. difficile binary toxin (CDT), and its molecular mechanism for entering host cells is not fully elucidated. Like other AB-type binary toxins, CDT enters host cells via endosomes. Here we show via surface plasmon resonance and electrochemical impedance spectroscopy that the cell-binding component of CDT, termed CDTb, binds and form pores in lipid bilayers in the absence of its enzymatic component, CDTa. This occ

Study on population genomics of Bacillus anthracis based on multiple types of genetic variations.

Bacillus anthracis, the causative agent of the deadly infectious disease anthrax, is also a typical biological warfare agent. It primarily infects livestock such as cattle and sheep, as well as humans, causing significant economic losses to the livestock industry and posing a threat to human society. A more profound insight into the genetic diversity and evolutionary drivers of this species is essential for studying its virulence mechanisms and conducting anthrax surveillance and control. However, current research in this area is insufficient, particularly lacking population genomic studies ba

The virgin lands campaign and the occurrence of foot-and-mouth disease and anthrax in the Republic of Kazakhstan (1955-1970).

The campaign for the development of virgin lands in Kazakhstan (1955-1970) was one of the most ambitious programs implemented by the Soviet government, which, arguably, resulted in both positive and negative consequences for the country. The campaign brought, at the same time, development, environmental degradation, and a dramatic cultural change to Kazakhstan. A barely explored aspect of the virgin lands campaign is related to its impact on the epidemiology of animal diseases. This paper describes, for the first time, the changes experienced by Kazakhstan during the implementation of the virg
