Counting sheep: Louis Pasteur and the first registered public vaccine trial.

During the COVID-19 pandemic, vaccine development took center stage in the media and minds of the global community. This same daily scrutiny was also applied to the first public registered vaccine trial of anthrax prevention among livestock led by French scientist Louis Pasteur in 1881. At a meeting with the president of the Agricultural Society of Melun, Pasteur outlined what might be considered the first "registered" vaccine trial protocol. The trial proposed a three-arm trial where anthrax-naïve sheep (n = 60) and cows (n = 10) would be assigned either to a treatment group consisting of a 2

Conservation and Specificity in Bacillus Biofilm Dynamics: On Structure and Function of B. cereus Camelysins.

The B. cereus family comprises members highly pathogenic for mammals or insects, with B. anthracis and B. thuringiensis respectively as notable examples. The biofilm operon of these bacteria encodes two TasA-like proteins, the 60% identical Camelysins CalY1 and CalY2. In this study, we observed that at neutral pH CalY2 alone polymerizes readily into filaments, whereas CalY1 forms a polydispersed mixture of oligomers without filament formation. However, at basic or acidic pH CalY1 also modestly polymerizes. CalY2 polymerization into filaments involves ß-sheet remodeling via donor strand complem

Intramolecular DNA Machine Coupled with Catalytic Redox-Recycling Amplification for Highly Efficient Electrochemical Detection of Dipicolinic Acid.

The accurate and timely determination of dipicolinic acid (DPA) is of great concern to prevent an anthrax epidemic. However, conventional methods suffer from poor detection efficiency and limited signal amplification. Herein, we report a highly efficient electrochemical biosensor for DPA analysis, which is based on two core conceptual and mechanistic breakthroughs: (i) a confined intramolecular DNA machine amplification paradigm that replaces the traditional intermolecular diffusion-driven design and (ii) the synergistic coupling of target-triggered intramolecular DNA machine activation with C

Chemical synthesis of the anthrax tetrasaccharide from Bacillus anthracis by an orthogonal one-pot glycosylation strategy.

Tetrasaccharide 1 from Bacillus anthracis represents an important target for the development of carbohydrates-based vaccines and diagnostic tools against the notorious disease anthrax. Here, we report the efficient chemical synthesis of tetrasaccharide 1 through [1 + 1+1 + 1] orthogonal one-pot glycosylation strategy on the basis of glycosyl ortho-(1-phenylvinyl)benzoates, which avoids the potential issues inherent to one-pot glycan assembly with thioglycosides.

Identification and structural characterization of anthrax toxin receptor 2 as the Clostridium perfringens NetF receptor.

Hemolysin β-pore-forming toxins (βPFTs) are key virulence factors of Clostridium perfringens, associated with severe diseases in humans and animals. Yet, the mechanisms by which Clostridium βPFTs recognize and engage specific target cells remain poorly understood. Here, we identify the cellular receptor for C. perfringens necrotizing enteritis toxin F (NetF), a recently discovered toxin implicated in severe enteritis in dogs and foals. We show that NetF binds to the same receptor as anthrax toxin, namely ANTXR2. Using cryo-electron microscopy, we determined the structure of the oligomeric NetF

Layout Types and Efficiency Improvement Strategies for Open Points of Dispensing in Vaccine and Antibiotic Distribution: A Scoping Review.

INTRODUCTION: Regarding pandemics or bioterrorism incidents, prompt and secure distribution of vaccines and prophylactic antibiotics is crucial. Open Points of Dispensing (PODs) are established to serve the public, and their effectiveness depends on the internal spatial layout and operational workflow design. However, studies on systematic classifications of open POD configurations and comprehensive syntheses of strategies for enhancing operational efficiency are lacking. STUDY OBJECTIVE: This scoping review aimed to classify open POD layout types used for vaccine and antibiotic distribution

A novel AAA+ ATPase required for sporulation and stress response in Bacillus anthracis.

UNLABELLED: AAA+ proteins function as molecular machines that utilize ATP to perform diverse cellular functions, including protein homeostasis, stress regulation, and cell cycle/developmental processes. In this study, we identified a novel AAA+ ATPase-related Bacillus anthracis protein BAS0518 (BA PrkA) which has 88% amino acid homology to Bacillus subtilis PrkA. Conserved domain analysis showed that BA PrkA has an N-terminal AAA+ ATPase domain with characteristic Walker A and Walker B motifs and a conserved secondary region of homology (SRH) domain, along with a C-terminal cAMP-dependent prot

A dynamical anthrax toxin nanopore biosensor for high-fidelity single-peptide classification.

Nanopore sensing holds the potential to revolutionize proteomics, yet current methods often rely on ensemble aggregation, where thousands of events must be statistically aggregated or averaged to identify a protein or peptide signature. While effective for pure samples, this aggregation strategy fails in complex, heterogeneous mixtures where the identity of individual molecules must be determined in real-time. Here, we demonstrate high-fidelity classification of peptides from single, individual translocation events, eliminating the need for ensemble averaging. This sensitivity is achieved usin

Identification of the biological properties of three novel lytic bacteriophages capable of infecting Bacillus anthracis: Novel members of the class Caudoviricetes.

Bacillus anthracis spores can remain viable for many years in regions where anthrax cases are reported or where deceased animals are buried. Anthrax occurrences may be more common in areas where livestock farming is a primary source of livelihood. In this study, 43 Bacillus anthracis isolates were used, which were isolated from samples collected in different provinces of Türkiye. Three lytic bacteriophages(BaTR8, BaTR13, and BaTR16) were isolated from environmental soil samples. These three lytic bacteriophages' biological properties (host range, burst size, and phage morphology) were defined,

Effects of chemical and biological warfare agent decontaminants on trace survival: Impact on fingermarks deposited on paper.

Chemical and biological events present a challenging environment for forensic scientists to perform their work. This research is the follow-up of the previous article that investigated the impact of decontaminants on fingermarks deposited on glass. In the present study, the focus shifted to paper, a porous and sensitive substrate where fingermark residues are absorbed and cannot be optically seen, making their detection more difficult. Paper is also highly relevant in forensic casework, as demonstrated by the anthrax letter events. Here the impact of 16 decontaminants on fingermarks deposited

Decoding Virulence Mechanisms of Bacillus anthracis Using a Galleria mellonella Infection Model: Differential Host Response Profiles Elicited by AtxA and PlcR.

A thorough understanding of the functions of virulence regulators in Bacillus anthracis evolution and host adaptation, particularly the systematic host responses they trigger, requires an efficient infection model capable of resolving subtle mechanisms. This study constructed a high-resolution host immune response decoder based on Galleria mellonella to analyze the specific response profiles elicited by different virulence regulators in a capsule-deficient background. By integrating transcriptomic, histopathological, and bacterial colonization analyses, the research delineated distinct host im

The Dual Role of RsiP in Regulating Virulence and Host Adaptation in Bacillus anthracis.

Bacillus anthracis displays susceptibility to penicillin despite harboring a β-lactamase gene, a phenotype governed by the anti-sigma factor RsiP. While RsiP represses σP-dependent β-lactamase expression, its broader roles in physiology and virulence remain unclear. This study aimed to define the global regulatory functions of RsiP beyond antibiotic resistance. Deletion of rsiP significantly upregulated the nprR gene, which is an important quorum-sensing (QS) system regulator and enhanced protease secretion. The ΔrsiP mutant caused higher mortality in cellular and Galleria mellonella models an

Anthrax-causing bacteria form biofilms on a soil-inspired porous glass bead model.

Bacillus anthracis and the emerging pathogen Bacillus cereus biovar anthracis (Bcbva) are causative agents of the lethal disease anthrax. Their ability to form highly resistant endospores enables them to persist in the environment, posing significant threats to wild herbivores, livestock, and also humans globally, especially considering their potential use as bioweapons. Despite the importance of these pathogens, the exact mechanisms underlying host infection remain poorly understood. Notably, both require relatively high infectious doses to cause disease. Proposed transmission routes include

Stigmatizing bioterrorism as a public health concern: The case of anthrax in US media.

This work approaches the role of media in stigmatizing anthrax as a bioweapon in the United States, transitioning the disease from a relatively obscure zoonotic pathogen to a symbol of bioterrorism. By analyzing over 20,000 articles published between 1979 and 2023, we trace how the 2001 anthrax attacks in the US significantly shifted media coverage to focus anthrax as a national security, terrorism, and public health concern. This framing reinforced the stigmatization of anthrax as a militarized threat, even during periods of low assessed risk. The study uses a novel data science approach inte
