Immunological effects of β-glucan as a trained immunity-based adjuvant in vaccinated goats against anthrax.

Trained immunity-based adjuvants (TIbAs) have emerged as a promising strategy to enhance vaccine-induced protection through functional reprogramming of innate immune cells. The present study evaluated the adjuvant potential of β-glucan derived from Debaryomyces hansenii CBS8339 in a vaccine against anthrax in goats. Peripheral blood monocyte-like cells from vaccinated goats were assessed for effector and metabolic responses following ex vivo lipopolysaccharide stimulation. Co-administration of β-glucan and anthrax vaccine significantly enhanced crucial innate immune effector mechanisms, includ

Hypoxia-induced regulations of cell adhesion molecules and their implications for pathogenesis, diagnosis, and treatment of endometriosis.

Endometriosis is a chronic inflammatory disease characterized by the presence of endometrial-like tissue outside the uterine cavity, affecting up to 10% of women of reproductive age. Despite extensive research, its pathogenesis remains incompletely understood, and clinically useful non-invasive diagnostic tools are still lacking. Increasing evidence identifies hypoxia as a key microenvironmental factor promoting lesion establishment and persistence. Cellular responses to hypoxia are mediated by hypoxia-inducible factor 1 alpha (HIF-1α), which coordinates transcriptional programs involved in an

Rational Engineering of the Anthrax Toxin Nanopore Interface for Orthogonal Peptide Classification.

The development of high-performance functional biointerfaces requires precise control over molecular recognition mechanisms at the nanoscale. A fundamental limitation in current biological nanopore materials is that wild-type (WT) channels often lack the specific physicochemical "grip" required to resolve chemically similar analytes. Here, we demonstrate that rationally engineering a key active site, called the φ-clamp, in the anthrax toxin protective antigen (PA) nanopore creates a tunable biointerface with orthogonal selectivity. By mutating the φ-clamp constriction (F427A), we generated a v

A One Health investigation of a Crimean-Congo Hemorrhagic fever outbreak reveals high seropositivity in livestock in Lyantonde District, Uganda, 2024.

In January 2024, an outbreak of Crimean-Congo hemorrhagic fever (CCHF) was confirmed in Buyanja village, Lyantonde District, Uganda. Initially suspected to be anthrax, laboratory tests at the Uganda Virus Research Institute (UVRI) confirmed CCHF, prompting a One Health response. Five (5) human cases were confirmed through both Reverse Transcription Polymerase Chain Reaction(RT-PCR) and IgM Enzyme-Linked Immunosorbent Assay (ELISA) testing, all of which were associated with direct contact with livestock, particularly goats. Among the twenty-six goats sampled, 88% (23/26) tested positive for CCH

Subclinical Anthrax Exposure in Railroad Workers Following Soil Disruption in an Endemic Region: A Seroprevalence Study in Kars, Türkiye.

During construction of the Baku-Tbilisi-Kars railroad between Kars City in Türkiye and Tbilisi in Georgia, blasting operations in an anthrax-endemic region disrupted a burial pit containing carcasses of cattle that had died of anthrax. Railroad workers expressed concerns that release of this material could result in them developing anthrax infection. We therefore undertook a seroprevalence study six months later to seek evidence of exposure to Bacillus anthracis spores. We used an optimised Enzyme-Linked Immunosorbent Assay (ELISA) to screen serum for antigen-specific IgG antibodies to the ant

Dynamic S-acylation controls CMG2 maturation extracellular matrix regulation and anthrax toxin susceptibility in vivo.

CMG2/ANTXR2 functions as a Collagen VI receptor required for extracellular matrix homeostasis and as the primary portal for anthrax toxin entry. Mutations in CMG2 cause Hyaline Fibromatosis Syndrome (HFS), a rare and often fatal genetic disorder characterized by excessive extracellular matrix accumulation, yet the molecular mechanisms regulating CMG2 function remain poorly understood. We show that CMG2 is controlled by ordered cycles of S-acylation and deacylation that regulate its folding, trafficking, and signalling competence. S-acylation by ZDHHC7 on two juxtamembrane cysteines protects CM

A Eu/Tb-Functionalized Hydrogen-Bonded Organic Framework for Ratiometric Sensing of Anthrax Biomarker and Integrated Intelligent Bactericidal Efficacy Evaluation.

Luminescent materials with stimulus-responsive and discriminable optical outputs are highly desirable for advanced on-site sensing and biosecurity monitoring technologies. Dipicolinic acid (DPA) is a specific Bacillus anthracis spore biomarker for anthrax diagnosis, while 1-bromo-3-chloro-5,5-dimethylhydantoin (BCDMH), an efficient anthrax decontaminant, poses health and environmental risks when overused. Therefore, the simultaneous detection of DPA and BCDMH is critical for anthrax diagnosis and disinfection monitoring, yet it remains challenging for existing sensing platforms. Herein, we syn

In vitro activity and in vivo efficacy of sulopenem against Bacillus anthracis in inhalational anthrax mouse and rabbit models.

Bacillus anthracis is a gram-positive, spore-forming bacterium and the etiological agent of anthrax. As a Tier 1 Select Agent, the pathogen can be isolated from the natural environment, and the endospore is a significant biological threat that can be produced in large quantities, stored, and disseminated by aerosolization. The intentional release of B. anthracis spores has the potential for mass casualties and is a serious threat to public and military health. Fluoroquinolones and tetracyclines are common antibiotics used for post-exposure prophylaxis and treatment in the United States (U.S.),
