One-step synthesis of LYH:Tb and LYH:Eu film probes and their application in Bacillus anthracis recognition.

In this study, two new photoluminescent film probes (LYH:Tb and LYH:Eu) were developed through chemical bath deposition. Benefiting from the antenna effect of DPA, these films can enhance the characteristic fluorescence emission of Tb3+ and Eu3+ through energy transfer, enabling stable fluorescence detection of the anthrax spore biomarker, pyridine dicarboxylic acid (DPA). By adding various aromatic organic compounds for selectivity studies, it was found that their fluorescence responses were much lower than that of DPA, indicating that the LYH:Tb and LYH:Eu film probes exhibited excellent sen

Enhancing the in vivo efficacy of anthrax vaccine using trimethylchitosan covalently coated chitosomes in a single-step microfluidic synthesis.

Liposomal vaccines have been developed extensively in recent years, and considerable progress has been made in the prevention of infectious diseases. Quaternary chitosan-coated liposomes were used in the present study. Liposomes coated with TMC (namely chitosomes) were synthesized in a single microfluidic step in which liposomes were formed and coated with trimethylchitosan (TMC) via covalent and/or hydrogen bonds in situ. The endocytosis of chitosomes by JAWS II dendritic cells was detected using flow cytometry. Chitosomes covalently coated with TMC were more efficiently internalized than unc

Persistence in time: the hunt for Bacillus anthracis at a historic tannery site in Austria reveals genetic diversity thought extinct.

UNLABELLED: Identifying and analyzing historic anthrax loci may provide a treasure trove to fill in the gaps of persistence in time and genetic diversity of Bacillus anthracis. In countries where anthrax has become a disease of the past, detailed knowledge of the exact location and stability of spores in soil reservoirs is limited. Reviewing archival records may provide valuable clues to unearthing such forgotten sites. Knowledge of anthrax diversity in Austria is scarce, as the only available isolates-originating from the last outbreak in Austria in 1988-cluster in the B.Br.004 (CNEVA) canoni

Identification and management of a novel Danshen leaf anthracnose caused by Colletotrichum karstii in Salvia miltiorrhiza Bunge in China.

Danshen (Salvia miltiorrhiza Bunge), a member of the genus Salvia within the Lamiaceae family, holds significant economic and medicinal value. Regrettably, the emergence of a novel leaf anthracnose in 2020 has significantly impacted its cultivation, leading to decreased yield and compromised quality. This newly identified pathogen was meticulously isolated from affected leaves, employing meticulous single conidia isolation techniques. Subsequent confirmation of pathogenicity was achieved through strict adherence of Koch's postulates. To ensure precise identification, morphological characterist

A lanthanide metal organic frameworks ratiometric fluorescence sensor for visual detection of anthrax biomarker.

A novel dual-emission fluorescence probe, Eu-TDA-DHTP, was developed based on a ratiometric fluorescence-organic frameworks utilizing Eu3+, 2,5-Thiophenedicarboxylic acid, and 2,5-dihydroxyterephthalic acid for the ratiometric and fluorescent visual sensing of the anthrax biomarker 2,6-pyridine dicarboxylic acid (DPA). The experiments demonstrated that this material can effectively enhance dual emission for DPA detection, enabling clear visualization. The limit of detection for DPA detection using Eu-TDA-DHTP is 1.9 μmol/L in fluorometric mode and 5 μmol/L in colorimetric mode. Additionally, t

Identification of Potential Therapeutic Targets Against Anthrax-Toxin-Induced Liver and Heart Damage.

Anthrax represents a disease resulting from infection by toxin-secreting bacteria, Bacillus anthracis. This research aimed to identify new therapeutic targets to combat anthrax. We performed assays to assess cell viability, apoptosis, glycogen consumption, and compound uptake and release in hepatocytes and cardiomyocytes responding to anthrax toxins. Microarray analysis was carried out to identify the genes potentially involved in toxin-induced toxicity. Knockdown experiments were performed to validate the contributions of the identified genes. Our study showed that anthrax edema toxin (EdTx)

Teixobactin: A Resistance-Evading Antibiotic for Treating Anthrax.

The antimicrobial resistance (AMR) crisis has been associated with millions of deaths. Of particular concern is the threat of bioweapons, exemplified by anthrax. Introduction of novel antibiotics helps mitigate AMR, but does not address the threat of bioweapons with engineered resistance. We reasoned that teixobactin, an antibiotic with no detectable resistance, is uniquely suited to address the challenge of weaponized anthrax. Teixobactin binds to immutable targets, precursors of cell wall polymers. Here we show that teixobactin is highly efficacious in a rabbit model of inhalation anthrax. I

Ratiometric fluorescence sensor based on bimetallic organic frameworks for anthrax biomarker detection.

It is of great significance to construct ratiometric fluorescence sensors with simple operation and desirable anti-interference ability. In this study, a bimetallic organic framework was prepared for the first time by a one-pot solvothermal method, using 4,4'-biphenyl dicarboxylic acid as ligand, lanthanide metal terbium ions (Tb3+) and transition metal zirconium ions (Zr4+) as central metal ions. This preparation method was easy to carry out. On this basis, a novel ratiometric fluorescence sensor Tb-Zr-MOF was constructed successfully for the detection of anthrax biomarker (2,6-pyridinedicarb

Regulating the aromatic structure in carbon dots to enhance the internal filter effect for sensitive on-site monitoring of anthrax biomarkers.

Anthrax, a fatal infectious disease caused by Bacillus anthracis (B. anthracis), poses a significant threat to public health. Herein, a novel type of carbon dots rich in π → π∗ transitions (R-CDs) was developed to fabricate an R-CDs/Eu3+ hybrid for the rapid on-site detection of the B. anthracis biomarker dipicolinic acid (DPA). The R-CDs/Eu3+ hybrid exhibits two distinct fluorescence responses to DPA: (1) quenching of the R-CDs fluorescence due to the internal filter effect, and (2) enhancement of Eu3+ fluorescence resulting from the antenna effect and the protective role of R-CDs, enabling r

Important Diseases of Small Ruminants in Sub-Saharan Africa: A Review with a Focus on Current Strategies for Treatment and Control in Smallholder Systems.

Sheep and goats are an important source of livelihood for smallholder farmers in sub-Saharan Africa (SSA). These livestock are almost entirely managed by resource-poor, smallholder farmers and pastoralists. Despite the large number of sheep and goats in SSA, their productivity is low, mainly due to diseases, poor feed, and inferior breeds. This review aims to summarize the most important diseases in small ruminants in SSA, with a focus on current treatment and control strategies. The following diseases were identified as the most significant constraints for small ruminant farmers: helminthoses

ERK pathway reactivation prevents anthrax toxin lethality in mice.

Lethal toxin (LT), the major virulence factor of Bacillus anthracis, proteolytically inactivates MEKs and disables downstream ERK, p38 and JNK pathway signalling leading to tissue damage and mortality. Therapies for LT-induced damage after host cell internalization of the toxin are lacking. Here we constructed MEK variants in which the LT proteolytic site was modified: MEK2(P10V/A11D), MEK3(I27D) and MEK6(I15D). These variants were resistant to proteolysis by LT. Expression in cells enabled sustained activation of ERK and p38 pathways and promoted cell survival upon LT treatment. Survival of L

Antiviral siRNA delivered using attenuated, anthrax toxin protects cells from the cytopathic effects of Zika virus.

Curative drugs are needed for the treatment of viral infections. Small interfering (si)RNA offer such a prospect but require the development of safe, effective and non-hepatotropic subcellular delivery systems. Here, 5 candidate siRNA molecules targeting defined sequences within the Zika Virus (ZIKV) genome were assayed for their ability to reduce ZIKV induced cytopathic effects in vitro. The protection of Huh-7 cells from ZIKV cytopathic effects was recorded after electroporation and the siRNA Feron-Zv2, resulting in 122.7 ± 5.3% cell viability (n = 3 ± standard error of the mean (SEM), 100 n

A machine learning computational approach for the mathematical anthrax disease system in animals.

OBJECTIVES: The current research investigations present the numerical solutions of the anthrax disease system in animals by designing a machine learning stochastic procedure. The mathematical anthrax disease system in animals is classified into susceptible, infected, recovered and vaccinated. METHOD: A Runge-Kutta solver is applied to collect the dataset, which decreases the mean square error by dividing into training as 78%, testing 12% and verification 10%. The proposed stochastic computing technique is performed through the logistic sigmoid activation function, and a single hidden layer co

Association of the ADRB2 rs1042714 variant with retinopathy of prematurity highlights the importance of the renin-angiotensin-aldosterone system.

Retinopathy of prematurity (ROP) remains a leading cause of childhood blindness globally. The clinical progression of ROP exhibits notable similarities to infantile hemangioma (IH), suggesting shared risk factors and underlying mechanisms. This study aimed to investigate the influence of variants in genes postulated for IH-specifically, anthrax toxin receptor 1 (ANTXR1), beta-2-adrenergic receptor (ADRB2), Fms-related tyrosine kinase 4 receptor (FLT4), kinase insert domain receptor (KDR), and insulin-like growth factor 1 receptor (IGF1R)-on the development and severity of ROP. Genetic Insights Into Hyaline Fibromatosis Syndrome: A Case Report of an ANTXR2 Mutation Featuring a Rare Variant c.697+1G>A.

Hyaline fibromatosis syndrome (HFS) is a rare genetic disorder encompassing juvenile hyaline fibromatosis (JHF) and infantile systemic hyalinosis (ISH), caused by mutations in the anthrax toxin receptor 2 gene (ANTXR2). This condition leads to the accumulation of hyaline plaques in the skin and organs, resulting in symptoms such as skin lesions, joint contractures, and digestive issues, often culminating in early mortality due to infections or diarrhea. By 2005, 20 mutations in ANTXR2 linked to ISH and JHF had been documented, impairing cellular adhesion to the laminin matrix. In this study, w
