How does a tadpole know when to metamorphose? Integrating evolutionary, ecological, and developmental biology with the neuroendocrinology of amphibian metamorphosis.

Amphibian metamorphosis is an excellent model for studying how selective pressures, environmental cues, endocrine signals, and molecular mechanisms influence developmental timing. In this minireview, I discuss key questions about the timing of metamorphosis, including: when and how do larvae acquire the ability to initiate metamorphosis; what historical and current ecological factors influence the optimal timing of transformation; and what are the physiological mechanisms that translate environmental information into a developmental response? From an ecological perspective, the timing of amphi

Redox Perturbation in Cadmium-Mediated Hypothalamic, Pituitary, and Testicular Impairment and Protective Effects of Zinc in Rats Is Accompanied by Modulation of Tryptophan Catabolism Enzyme/Protein.

Cadmium (Cd) is a toxic heavy metal found in the environment from natural (volcanic eruptions) and industrial sources, including mining, smelting, batteries, and fuel combustion. Studies have shown that oxidative stress, inflammation, and abnormal immune response are associated with Cd-induced hypothalamic-pituitary-testicular (HPT) toxicity. The dietary trace element zinc (Zn) prevents oxidative and inflammatory effects in diverse experimental models. The literature has reported that Zn inhibits Cd-mediated HPT toxicity. Our objective is to investigate the mechanisms by which Zn inhibits the

A feasibility study of functional preservation in craniospinal irradiation with photon for pediatric medulloblastoma.

BACKGROUND: Craniospinal irradiation (CSI) is essential for treating pediatric medulloblastoma (MB) but causes significant long-term toxicities. Existing dose-reduction or partial-sparing strategies improve neurocognitive outcomes but may compromise survival or fail to address other late effects. METHODS: A new functional preservation CSI (FP-CSI) technique was developed to spare the hippocampus, hypothalamic-pituitary axis (HPA), cochlea, and scalp while ensuring homogeneous vertebral coverage. Eight pediatric patients with average-risk MB were retrospectively planned with volumetric modulat

Ultrasound-guided percutaneous versus trans-nasal pterygopalatine fossa block in endoscopic trans-sphenoidal pituitary gland surgery: a randomized controlled trial.

BACKGROUND: Various techniques are being used to create a relatively bloodless surgical field during endoscopic trans-sphenoidal pituitary gland surgery (ETS). Pterygopalatine fossa (PPF) block helps to improve the field of surgery and control haemodynamic fluctuations. METHODS: Eligible patients with the American Society of Anaesthesiologists class I or II were randomly classified by closed envelope method. Group TN received bilateral transnasal PPFB with 4 mL of 0.25% bupivacaine after induction of general anaesthesia. Group PC received bilateral ultrasound-guided infrazygomatic percutaneous

PPP1R12A Mutation Presenting With Congenital Jejunal Atresia and Short Stature: A Pediatric Endocrinology Case Report.

We report an 11-year-old Hispanic male with a PPP1R12A gene de novo heterozygous likely pathogenic mutation, p. (Gln13Arg) (CAG>CGG), c.38 A > G in Exon 1 (NM_002480.2), detected on whole-exome trio sequencing during his short-stature evaluation. His medical history is remarkable for congenital jejunal atresia diagnosed prenatally and repaired surgically shortly after birth. Notably, he lacks genitourinary anomalies, which are frequently described in individuals with PPP1R12A-related urogenital and brain malformation syndrome (UBMS). An endocrine evaluation revealed growth hormone deficiency w

Dynamic Transcriptomic Profiling of the HPG Axis Reveals Stage-Specific Regulatory Networks Underlying Natural Sex Change in Orange-Spotted Grouper (Epinephelus coioides).

Protogynous sex change in teleosts involves dramatic gonadal restructuring, governed by the hypothalamic-pituitary-gonadal (HPG) axis. Here, we established the dynamic regulatory atlas of the HPG axis during natural sex change in protogynous orange-spotted grouper (Epinephelus coioides) through multi-tissue transcriptomics. Profiling hypothalamus, pituitary, and gonad tissues across ovarian, intersex, and testicular stages revealed minimal transcriptomic changes in the hypothalamus during ovarian-to-intersex transition but significant differential expression during intersex-to-testis progressi

Primordial germ cell-like cells residing in the pituitary may serve as the origin of intracranial germ cell tumors.

Intracranial germ cell tumors (iGCTs) are rare, histologically classified as extragonadal germ cell tumors, and predominantly affect children and adolescents. These tumors are commonly found in the midline of the brain, with a notable male predominance and varying geographic incidences. The origins of iGCTs have been debated, particularly whether they arise from abnormal migration of primordial germ cells (PGCs) or transformed embryonic stem cells (ESCs). In this study, we hypothesize the potential presence of PGC-like cells in the pituitary gland, and these cells may differentiate into iGCTs

Concurrent Pituitary and Thyroid Immune-Related Adverse Events after Immune Checkpoint Inhibitors Associated with HLA-DR15-Related Haplotypes.

Some patients undergoing immune checkpoint blockade treatment develop immune-related adverse events (irAE) affecting multiple organs, but whether pituitary and thyroid dysfunction is prone to co-occur is unclear. A total of 1,014 patients treated at Nagoya University Hospital were prospectively assessed for pituitary and thyroid function, and human leukocyte antigens (HLA) were analyzed in patients with pituitary and/or thyroid irAEs. Pituitary irAE and thyroid irAE developed in 68 and 128 patients, respectively. The incidence of thyroid irAE was significantly higher in patients who developed

Multiplex in situ hybridization chain reaction reveals the spatial organization of six pituitary hormone mRNAs in the Japanese eel.

The anterior pituitary gland regulates growth, development, metabolism, reproduction, and stress responses. In this study, we established a two-round multiplex in situ hybridization chain reaction (isHCR) procedure combined with a fluorescence-quenching device to visualize the spatial distribution of six pituitary hormone mRNAs (gh, lhb, fshb, tshb, prl, and pomc) in cultured immature Japanese eels. In pituitary sections, prl and tshb were localized to the rostral pars distalis (RPD), whereas lhb, fshb, and gh were distributed in the proximal pars distalis. Pomc was detected in both the RPD an

Molecular biology of pituitary neuroendocrine tumors.

BACKGROUND: Pituitary neuroendocrine tumors (PitNETs) represent a heterogeneous group of intracranial neoplasms arising from the anterior pituitary gland. While most tumors are benign, certain subsets can display aggressive behavior marked by invasiveness, treatment resistance, and familial clustering. The current World Health Organization (WHO) classification emphasizes the role of lineage-specific transcription factors in better identifying cell types. However, this methodology is not sufficient to ensure fully accurate prediction of tumor behavior; therefore, new, more in-depth methods are

Dense sampling for mapping pituitary growth dynamics before, during, and after pregnancy.

Pregnancy represents a period of profound endocrine activity and neural reorganization. While recent evidence highlights pituitary volume as a biomarker of endocrine dynamics during pregnancy, its precise trajectory (timing and relative magnitude of effect) across human pregnancy remains undescribed. Three healthy women (59 total observations) underwent T1-weighted MRI before conception (5 baseline observations), during pregnancy (38 total observations, spanning gestational weeks 1-36), and up to 1 year postpartum (16 total observations). Anterior and posterior pituitary lobes were manually de

Identification of a novel THRB mutation causing thyroid hormone resistance syndrome.

Resistance to thyroid hormone syndrome (RTHS) is a rare disorder caused by mutations in the thyroid hormone receptor beta (THRB) gene, resulting in impaired action of thyroid hormones on target tissues and organs. We report a case of a 57-year-old Chinese male who presented with palpitations and hand tremors. Laboratory tests revealed elevated serum thyroid hormone levels, while serum thyroid-stimulating hormone (TSH) levels remained within the normal range. Enhanced magnetic resonance imaging of the pituitary gland showed no abnormalities. Through genetic testing, we identified a rare heteroz

Pituitary lesions in captive chimpanzees (Pan troglodytes): clinical presentation and histologic characterization.

Pituitary lesions have been sporadically reported in captive chimpanzee (Pan troglodytes) populations. These lesions can be functional (hormone secreting) or non-functional. Functional lesions produce different clinical syndromes depending on the hormone produced. Previous reports of pituitary adenomas in chimpanzees have not characterized the tumors through clinical syndromes or by immunohistochemical features of the lesions. A retrospective review of necropsy and medical records from the Chimpanzee Care Center (CCC) at The University of Texas MD Anderson Cancer Center were queried to identif

DRD2 Deficiency Underlies Pituitary Adenoma Dependent on Escherichia coli Translocation from the Gut.

Pituitary adenoma (PA) are common intracranial tumor types that have harmful effects on human health. However, the pathogenesis of PA remains unclear yet. The intratumoral microbiome has been reported playing an important impact on the occurrence, metastasis, immune monitoring, and drug resistance of various tumors. While normal dopamine receptor D2 (DRD2) expression is enriched in the apical junction of pituitary epithelium and colonic enterocytes, various factors-induced drd2 loss dampened its expression at both sites. DRD2 deficiencies are characterized by chronic hyperprolactinemia, pituit

Quantification of pituitary 5-HT1B receptors with positron emission tomography: Negligible specific binding despite conspicuous uptake.

While Positron Emission Tomography (PET) images with the 5-HT1B receptor specific radioligand [11C]AZ10419369 show pronounced uptake in the pituitary region, and experimental studies support 5-HT1B receptor involvement in pituitary hormonal release, this uptake remains unquantified. In the present study we applied invasive and non-invasive models to evaluate pituitary 5-HT1B receptor binding. Ten subjects underwent PET with [11C]AZ10419369, of which six participated in three additional PET-examinations after pretreatment with increasing doses of the 5-HT1B receptor antagonist AZD3783. While [1

Periovulatory neurohormone dynamics reveal an association between secretoneurin and GnRH across the mouse estrous cycle.

INTRODUCTION: Surge release of luteinizing hormone (LH) from the pituitary is essential for fertility, as it triggers ovulation. Secretoneurin (SN), a conserved peptide derived from secretogranin-2, stimulates LH release, but its relationship to periovulatory changes in classical reproductive hormones remains unclear. METHODS: We measured fluctuations of two reproductive steroids and three peptides in the hypothalamus, pituitary, and ovary of female mice during the periovulatory period using a novel nano LC-MS/MS protocol. Immunohistochemistry for SN and GnRH was performed on brain samples. G

A Medically Managed Case of Acromegaly: A Case Report.

Endocrine glands are specialized cells responsible for producing and secreting hormones that serve many functions in various body tissues. The pituitary gland is an endocrine gland located in the brain, composed of anterior and posterior parts, both of which are responsible for the secretion of many hormones. The anterior pituitary gland secretes six hormones. Hypersecretion and undersecretion result in abnormalities in the body. Autonomous growth hormone production due to a pituitary adenoma leads to various changes in tissues and organs. Excess growth hormone results in gigantism during chil
