Papillary Tumor of the Pineal Region Presenting With Obstructive Hydrocephalus: A Diagnostic and Management Challenge.

We report a 45-year-old woman with a papillary tumor of the pineal region (PTPR), a neuroepithelial neoplasm of the pineal gland, who presented with a one-year history of headaches, visual disturbance, and gait impairment and was found to have a pineal mass with obstructive hydrocephalus. She underwent endoscopic third ventriculostomy (ETV) with biopsy; the initial radiologic impression favored pineocytoma. Immunohistochemistry was nondiagnostic on the limited sample, and the diagnosis of PTPR was established only after review at two academic neuropathology centers. DNA methylation profiling w

Pineal melatonin modulates responsiveness to exogenous melatonin via the IFN-γ-CXCL9/10-CXCR3 axis in gastric cancer.

BACKGROUND: Melatonin is a pineal-derived neuroendocrine hormone with pleiotropic actions on tumor biology and immune regulation. Although exogenous melatonin has been reported to exert antitumor and immunomodulatory effects, whether these actions depend on an intact endogenous pineal melatonin background remains unclear. In particular, the role of endogenous melatonin in conditioning chemokine-mediated antitumor effects has not been defined. METHODS: A male mouse gastric cancer model was used to investigate the interplay between endogenous and exogenous melatonin in tumor growth, immune infi

Serotonergic Control of Spine Morphology During Growth in Zebrafish.

Scoliosis is a three-dimensional deformity of the vertebral column that commonly emerges during growth, yet the biological mechanisms that initiate and drive curve progression remain poorly understood. Serotonin (5-hydroxytryptamine [5-HT]) has long been linked to scoliosis through its relationship with pineal melatonin signaling and through genetic associations with human disease, but its role in spinal morphogenesis has remained unclear. Here, using zebrafish, we show that serotonergic signaling is required for normal spine morphology. Transient exposure to 5-HT during development induces la

Beyond Brain Parenchyma: A Pictorial Review of Non-Parenchymal and Extracranial Metastases on Cranial MRI.

Brain MRI is the primary imaging technique used to detect, stage, and monitor intracranial metastatic disease. While parenchymal metastases are easily identifiable, metastatic involvement beyond the brain parenchyma is frequently subtle and underreported, despite representing a significant proportion of metastatic findings identified during cranial examinations. Failure to identify these metastases, or identification at a late stage, can lead to incomplete staging, inappropriate treatment selection, and delays in initiating targeted therapies. This pictorial review presents a systematic, compa

Influence of Intravenous Norepinephrine Administration on Melatonin Plasma Concentration in Awake and Anaesthetised Healthy Adult Volunteers.

Melatonin (MEL) is an endogenous hormone that regulates circadian rhythm, immune function and oxidative stress responses. Norepinephrine (NE), an endogenous catecholamine, increases blood pressure through the sympathetic nervous system and is commonly administered intraoperatively or in the clinically ill patient for hemodynamic support. Although endogenous NE is known to stimulate MEL synthesis, the effects of exogenous intravenous NE on plasma MEL concentration, particularly under general anaesthesia (GA), remain poorly characterised. This study examined the influence of intravenous NE on pl

Variable and fixed rate debt: Gambling on the future state of the economy after Covid-19.

Melatonin is a product of the amino acid tryptophan in the pineal gland. Once synthesized, the specific mechanisms governing the release of melatonin from the pineal gland and its functions are largely unknown. Besides its regulatory role in circadian rhythms in mammals, because of its widespread subcellular distribution, melatonin contributes to the reduction of oxidative damage in both the lipid and the aqueous environments of the cell. This postulate is widely supported by the experimental observations showing that melatonin protects lipids in membranes, proteins in the cytosol, and DNA in

Pineal gland organoids illuminate human melatonin and circadian regulation.

Human pineal development and neuroendocrine regulation remain incompletely understood at the molecular and functional levels. Kiral et al.1 establish human pineal gland organoids (hPGOs) that recapitulate pinealocyte maturation, noradrenergic responsiveness, and melatonin synthesis, providing a platform for investigating circadian dysfunction and disease-associated sleep disturbances.

Transcriptomic analysis of pigeon retina and pineal gland illuminated with red light.

1. This study investigated the transcriptomic expression of the pigeon retina and pineal gland when exposed to red light (RL). Light is a crucial environmental factor influencing poultry production, physiology and behaviour. Different wavelengths of light have distinct effects on photoreceptors, including those in the retina and pineal gland, ultimately regulating various production benefits.2. Using transcriptome sequencing, this trial examined the gene expression profiles of the retina and pineal gland of White King pigeons exposed to either red light (RL; 660 nm) or White light (WL; 400-760

Melatonin alleviates gentamicin-induced acute kidney injury through the Keap1/Nrf2/HO-1 signaling pathway.

Drug-related factors represent a primary cause of acute kidney injury. Gentamicin (GM), while being one of the most effective and commonly used clinical agents against Gram-negative bacteria, frequently induces nephrotoxicity and triggers acute kidney injury during treatment. Melatonin, a natural antioxidant produced by the pineal gland, has been shown in recent studies to mitigate drug-induced nephrotoxicity. This study aimed to delineate the dose-dependent effects and underlying mechanisms of GM-induced acute kidney injury, along with the protective role of melatonin. Results demonstrated th

Physiological relevance of autocrine melatonin signaling in pineal and extrapineal sites: a systematic review.

Beyond the pineal gland, melatonin is produced locally in many extrapineal organs, where it mediates local tissue homeostasis. However, little attention is paid to the physiological role of autocrine melatonin signaling across body organs. This study synthesizes original data to address this gap, as gaining insight into this topic could lead to new therapeutic approaches for diseases associated with melatonin. This systematic review used a narrative synthesis, following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) 2020 guideline standards to synthesize origin

Serum melatonin predicts hepatic encephalopathy in patients with liver cirrhosis.

AIM: Hepatic encephalopathy (HE) is a serious complication in patients with liver cirrhosis and is strongly associated with morbidity and mortality. Melatonin, a neuroprotective hormone secreted by the pineal gland and enterochromaffin cells of the gastrointestinal tract is involved in the sleep cycle in normal people. This may be disrupted in cirrhotic patients. We aimed at evaluating the role of serum melatonin in cirrhotic patients with HE. METHODS: A prospective observational study was conducted in hospitalized patients with liver cirrhosis after meeting the inclusion and exclusion criter

The pineal gland in ageing and alzheimer's disease: age-related molecular changes.

The pineal gland is an organ that undergoes significant degeneration with age, and these degenerative changes lead to numerous physiological alterations. This study investigated age-related molecular changes and Alzheimer's disease (AD)-associated pathology in the human pineal gland using histopathological analyses. This study collected a total of 54 human pineal gland specimens. Of these, 47 were categorised into five age groups: 0-20, 21-40, 41-60, 61-80, and 81-100 years. A further 7 cases with confirmed AD-related neuropathological changes were assigned to the AD group, while matched to 7

Novel Insights into the Distribution and Effects of Perfluorooctanesulfonic acid (PFOS) in the Nervous System of a Frog Tadpole Model by Mass Spectrometry Imaging.

Determining how environmental contaminants localize within tissues is essential for understanding the consequences of sublethal exposures. Therefore, we optimized a mass spectrometry imaging (MSI) workflow to study the distribution and biological effects of contaminants in an environmentally relevant frog tadpole model. We applied the method to the entire tadpole head postexperimental exposure to the legacy contaminant, perfluorooctanesulfonic acid (PFOS; 0.1-100 μg/L, 48 h). At 10 μg/L, PFOS unexpectedly localized mainly to the olfactory epithelium. At 100 μg/L, PFOS was distributed throughou

Four camera-type eyes in the earliest vertebrates from the Cambrian Period.

Vertebrate vision is mainly accommodated by a pair of lateral image-forming camera-type eyes and is supplemented in non-mammalian vertebrates by a dorsal pineal complex (pineal and parapineal organs) functioning as photoreceptive and/or endocrine organs1. The pineal complex shares a common genetic and embryological basis with the lateral eyes, both derived from evaginations during the development of diencephalon2. Despite being widely heralded as the 'third eye' in crown vertebrates3, the nature of the pineal complex and its presumed visual capability in early vertebrates2 remain unknown. Here

Pinopsin Regulates Melatonin Production and Daily Locomotor Activity: Functional Insights From Gene-Edited Xenopus Tadpoles.

Circadian rhythm alignment depends on environmental light detection via opsins. Pinopsin, originally identified in the pineal organ of birds and later in amphibian pineal complex and eyes, may play a role in this process, though its function has not been genetically tested. Evolutionary analysis suggests pinopsin was independently lost in several vertebrate lineages, including mammals (Synapsida), some reptiles (e.g. snakes and crocodiles), and teleost fish, but retained in birds, turtles, lizards, and non-teleost Actinopterygii. We conducted a detailed genomic search of the pinopsin gene acro
