Psycho-Neuro-EndocrinE-Immunology Therapy of Cancer, Autoimmunity, Geriatric Disorders, Covid-19, and Hypertension.

Despite the great number of experimental investigations in the area of psycho-neuro-endocrine-immunology showing that endocrine, nervous, and immune systems cannot be in vivo physiologically separated, the diagnosis and therapies of the pathologies of these three functional biological systems continue to be separately performed from a clinical practice point of view. The separation between experimental and clinical medicine became dramatic after the discovery of more than 10 human molecules provided by anti-inflammatory and antitumor activity, completely devoid of any toxicity, which may be su

Papillary Tumor of the Pineal Gland With Discordant FDG and FAPi Uptake.

In recent years, 68Ga-FAP inhibitor has emerged as a possible substitute radiotracer to FDG for imaging of most of the tumors. Here, we report a 7-year-old girl diagnosed with recurrent papillary tumor of the pineal gland, presenting with complaints of headache and vomiting. 18F-FDG PET/CT revealed an FDG-avid enhancing lesion in the posterior part of the third ventricle. Negligible FAPI uptake was noted on the corresponding 68Ga-FAPI-46 PET/CT images, highlighting the scope of 18F-FDG PET/CT in this realm.

Multimodality Imaging in a Patient with Erdheim Chester Disease with Cardiovascular Involvement.

We hereby report a 40-year-old woman presenting with chest pain and recurrent pericardial effusion. Cardiac MRI revealed diffuse sheath-like mass involving predominantly the right heart chambers. She also had bony and perirenal involvement. Additionally, she had a pineal gland tumor, a biopsy from which revealed xanthogranulomatous infiltration with foamy histiocytes which were CD 68 positive and CD1a negative, diagnostic of Erdheim Chester disease. Erdheim Chester disease is a rare form of non-Langerhans cell histiocytosis. We highlight the importance of multimodality imaging for comprehensiv

Ultrasound modulation of melatonin release from pineal glands in vitro.

OBJECTIVES: In this study, we aim to investigate whether therapeutic ultrasound can modulate the release of melatonin from the pineal gland-either increasing or decreasing its levels-and to assess the safety of this technique. This research could address a significant clinical need by providing a noninvasive method to potentially regulate sleep and circadian rhythms through the targeted modulation of melatonin. METHODS: Rat pineal glands were placed in a well with a Krebs Ringer Buffer solution. Ultrasound was applied to the glands using unfocused transducers set at an average intensity of 1

Chronic Dexamethasone Disturbs the Circadian Rhythm of Melatonin and Clock Genes in Goats.

Dex is a drug commonly used as an immunosuppressive and anti-inflammatory agent in humans and animals. GCs have a profound impact on melatonin expression and biological rhythm. However, the effect of chronic exposure to Dex on melatonin secretion and biological clock gene expression in ruminants is still unclear. Ten goats were randomly divided into two groups: the control group was injected with saline, and the Dex-treated group was intramuscularly injected daily for 21 d with 0.2 mg/kg Dex. The rhythm of melatonin secretion in the plasma was disturbed in the Dex group, and the plasma and col

Data Mining Approach to Melatonin Treatment in Alzheimer's Disease: New Gene Targets MMP2 and NR3C1.

Melatonin is a hormone released by the pineal gland that regulates the sleep-wake cycle. It has been widely studied for its therapeutic effects on Alzheimer's disease (AD), particularly through the amyloidosis, oxidative stress, and neuroinflammation pathways. Nevertheless, the mechanisms through which it exerts its neuroprotective effects in AD are still largely unknown. Data mining was used to identify potential gene targets that link melatonin's effects to AD pathways, yielding a comprehensive view of the underlying molecular mechanisms. We identified 3397 genes related to AD from DisGeNet

Visualizing the spatial distribution of Alprazolam in the mouse brain following nasal administration using desorption electrospray ionization mass spectrometry imaging.

Alprazolam (Alp), a triazolobenzodiazepine, is widely prescribed for the treatment of sleep disorders, anxiety, and panic disorder. While oral administration remains the standard route, its slow onset of action has prompted interest in intranasal delivery as an alternative, offers the potential for direct drug delivery to the brain. This study aims to develop a fast-acting intranasal formulation of Alp (Alp-nd). Safety evaluations were conducted using mucociliary toxicity tests in bullfrogs and ciliary toxicity tests in rats, demonstrating that Alp-nd was safe and did not cause damage to the n

Dysregulation of the molecular clock by blood-borne factors in Alzheimer's disease patients.

BACKGROUND: Circadian disruptions are increasingly recognized in Alzheimer's disease (AD) patients and may influence disease onset and progression. This study examines how AD pathology affects blood-borne factors that regulate circadian rhythms. METHODS: Eighty-five participants from the Sino Longitudinal Study on Cognitive Decline were enrolled: 35 amyloid-beta negative normal controls (Aβ- NCs), 23 amyloid-beta positive normal controls (Aβ+ NCs), 15 patients with amnestic mild cognitive impairment (aMCI), and 12 with Alzheimer's disease dementia (ADD). Patients with aMCI and ADD were groupe

Transcriptome and DNA methylation analysis of the goat pineal gland during puberty.

Previous studies have confirmed that methylation regulates gene transcription in the hypothalamus-pituitary-gonadal axis during puberty initiation, but little is known about the regulation of DNA methylation on gene expression in the pineal gland. To screen pineal gland candidate genes related to the onset of goat puberty and regulated by genome methylation, we collected pineal glands from prepubertal and pubertal female goats, then, determined the DNA methylation profile by whole genome bisulfite sequencing and the transcriptome by RNA sequencing on Illumina HiSeqTM2500. We analyzed different

Interplay of Light, Melatonin, and Circadian Genes in Skin Pigmentation Regulation.

Circadian regulation of skin pigmentation is essential for thermoregulation, ultraviolet (UV) protection, and synchronization of skin cell renewal. This regulation involves both cell-autonomous photic responses and non-cell-autonomous hormonal control, particularly through melatonin produced in a light-sensitive manner. Photosensitive opsins, cryptochromes, and melatonin regulate circadian rhythms in skin pigment cells. We studied light/dark cycles and melatonin coordination in melanin synthesis and cell proliferation of Xenopus laevis melanophores. In vivo, tadpole pigmentation shows robust c

MT1/cAMP/PKA Pathway in Melatonin-Regulated Sperm Capacitation.

Melatonin is mainly synthesized and secreted by pineal gland, and plays multiple functions, including its regulating effects on reproductive processes. Sperm capacitation is necessary for fertilization, but the effect of melatonin on mouse sperm capacitation remains to be elucidated. We thus investigated the roles of melatonin on capacitation by culturing the sperms from mouse cauda epididymis in the medium with different doses of melatonin. The results showed that 10-7 mol/L melatonin significantly enhanced the sperm capacitation by increasing the sperm tyrosine phosphorylation level, percent

Effects of light on biological functions and human sleep.

The nonvisual effects of light in humans are mainly conveyed by a subset of retinal ganglion cells that contain the pigment melanopsin which renders them intrinsically photosensitive (= intrinsically photosensitive retinal ganglion cells, ipRGCs). They have direct connections to the main circadian clock in the suprachiasmatic nuclei (SCN) of the hypothalamus and modulate a variety of physiological processes, pineal melatonin secretion, autonomic functions, cognitive processes such as attention, and behavior, including sleep and wakefulness. This is because efferent projections from the SCN rea

Melatonin influence on miRNA expression in sperm, hypothalamus, pre-frontal cortex and cerebellum of Wistar rats.

Melatonin is a pineal hormone synthesized exclusively at night, in several organisms. Its action on sperm is of particular interest, since they transfer genetic and epigenetic information to the offspring, including microRNAs, configuring a mechanism of paternal epigenetic inheritance. MicroRNAs are known to participate in a wide variety of mechanisms in basically all cells and tissues, including the brain and the sperm cells, which are known, respectively, to present 70% of all identified microRNAs and to transfer these molecules to the embryo. MicroRNAs from sperm have been associated with m

Growth Hormone Increase Induced by Oral Administration of Melatonin in a Young Woman With Sleep Disturbances.

Growth hormone (GH) secretion by the pituitary is regulated by stimulatory and inhibitory pathways such as growth hormone releasing hormone (GHRH) and somatostatin, respectively, being also modulated by different neurotransmitters acting at the hypothalamic/pituitary level. The pineal gland hormone melatonin regulates GH secretion in many mammals, including humans, although its role in modulating GH secretion has been debated. We describe the case of a young woman chronically taking melatonin for sleep disturbances, referring to her general practitioner for flushing that appeared just after st

The microsurgical infratentorial supracerebellar approach for lesions of the pineal gland: feasibility, morbidity, and functional outcomes from a single-center experience.

Pineal gland lesions pose a significant surgical challenge due to the deep-seated nature of the pineal gland, as well as the limited field of view, and the complex vascular anatomy. The mainstay of surgical treatment, when necessary, is always histopathological clarity and gross total resection (GTR). We evaluate the surgical outcomes for pineal gland lesions, shedding light on functional outcomes, histological findings, and surgical complications. We analyzed patients with pineal gland lesions who underwent surgical treatment in our institution between September 2013 and May 2022. Patient dem
