Exacerbated ATP transmission in the carotid body is linked to glomus cell expansion in spontaneously hypertensive rats.

ATP acting on P2X3 receptors contributes to carotid body (CB) hyperexcitability in spontaneously hypertensive rats (SHRs). We investigated whether this reflects altered ATP release and/or impaired ATP metabolism. Using in vitro CB preparations, we quantified ATP release in Wistar rats and SHRs (n = 10 each) and found significantly greater ATP release in SHRs (P = 0.00062). Intracellular ATP depletion in dissociated glomus cells was similar between strains (n = 4 each). Morphometric analysis revealed a disproportionate increase in glomus tissue, measured as tyrosine hydroxylase-positive area, i

Carotid body sensing of systemic inflammation contributes to recruitment of the splanchnic anti-inflammatory pathway.

Systemic inflammation engages autonomic reflexes that modulate immune responses, yet the peripheral sensors and central circuits responsible for recruiting sympathetic anti-inflammatory pathways remain incompletely defined. Here, we identify the carotid bodies as critical afferent sensors required for full engagement of a splanchnic sympathetic reflex during endotoxemia. Using in vivo splanchnic sympathetic nerve activity recordings, cytokine measurements, and brainstem neuronal mapping, we show that intraperitoneal injection of lipopolysaccharide (LPS) activates splanchnic sympathetic outflow

Chronic intermittent hypoxia-induced H2S production promotes carotid body hyperactivity via the upregulation of Sp1 S-sulfhydration and angiotensin II receptor.

Chronic intermittent hypoxia (CIH), a hallmark of obstructive sleep apnea (OSA), induces increased carotid body (CB) activity. Hydrogen sulfide (H2S), a critical oxygen sensor in the CB, promotes protein post-translational modification via S-sulfhydration. This study investigated whether CIH-induced H₂S production drives CB hyperactivity through Sp1 S-sulfhydration-mediated upregulation of angiotensin II receptor type 1 (AT1). Rats were exposed to CIH for 12 weeks. CB activity, H₂S levels, and AT1/Sp1 expression were analyzed. Pharmacological inhibitors [AOAA (H₂S synthase inhibitor) and DTT (

Synchronous Bilateral Shamblin Type III Carotid Body Tumors and Adrenal Pheochromocytoma with SDHD Mutation: A Rare Presentation, with Multimodality Imaging Findings.

Pheochromocytomas and paragangliomas are relatively rare tumors, with an incidence of approximately 0.6 cases per 100,000 person-years. Moreover, the co-occurrence of these tumors is extremely rare and is often associated with pheochromocytomas/paragangliomas-related pathogenic mutations. We present the case of a 33-year-old female diagnosed with bilateral carotid body paragangliomas and a concurrently detected pheochromocytoma, with metastases to the abdominal lymph nodes and lung. DNA analysis revealed a mutation in the succinate dehydrogenase subunit D gene. The tumors displayed high-grade

Respiratory chemoreflex and risk of incident increase in blood pressure.

STUDY OBJECTIVES: Carotid body over-activation increases sympathetic drive and blood pressure but also induces periodic breathing (high loop gain [HLG] sleep apnea). It is plausible that apnea with elevated loop gain has a disproportionate effect on development or worsening of hypertension. METHODS: Subjects from two cohort studies, Sleep Heart Health Study (SHHS) and Osteoporotic Fractures in Men Study (MrOS), were included in this analysis. Respiratory self-similarity (SS, and the related central respiratory event index, CREI) is a method of detecting expressed HLG sleep apnea independent o

Carotid Body Chemoreceptors: from Mountain to Space.

Human acclimatization to both high-altitude and microgravity is crucial for terrestrial and space exploration. Although these extreme environments elicit several similar physiological responses, including autonomic imbalance, metabolic stress, and reduced exercise capacity, the underlying central mechanisms remain unclear. However, these effects appear to be linked to chemoreflex control. This review summarizes current evidence and proposed potential similarities and bioconvergent mechanisms underlying human responses to both terrestrial and extraterrestrial environments.

Amino acid modulation of the carotid body selectively modulates peripheral chemoreceptor respiratory reflex.

Cardiorespiratory homeostasis is maintained by carotid body (CB) peripheral chemoreceptors that monitor blood oxygen and stimulate breathing. While proportionality between peripheral chemoreflex sensitivity and respiratory response is relative to hypoxia severity, the fundamental regulatory mechanisms remain unknown. We hypothesized that intercellular amino acid transmission between glomus cells modulates CB sensitivity. We identified a full complement of glutamate and gamma-aminobutyric acid (GABA) synaptic signalling machinery in the CB. During hypoxia stimulation, glutamate acting via NMDA

A case report of carotid body tumor with rare thoracic vertebral metastasis and literature review.

BACKGROUND: Malignant carotid body tumor (CBT), a type of carotid body paraganglioma (PGL), is defined by the presence of distant metastasis; histology alone cannot reliably distinguish benign from malignant CBT. We present a 70-year-old female patient who had been suspected of having CBT for more than 10 years without specific treatment, and complained of abdominal pain for 6 months. Contrast-enhanced MRI and CT revealed a T11 vertebral lesion, followed by percutaneous biopsy of the lesion, which showed atypical cell nests. Immunohistochemical staining was positive for CD56, CgA, Syn and S-10

Opioid-induced respiratory depression: an underappreciated role of the peripheral nervous system.

Opioid overdose often leads to death, largely due to opioid-induced respiratory depression (OIRD). Extensive investigations have focused on the effects of opioids on brainstem breathing circuits. Peripheral sensory pathways also play an important role in shaping breathing behavior. This review highlights the effects of selected opioids and enkephalins on peripheral sensory inputs controlling breathing. Morphine and enkephalins activate vagal pulmonary C-fiber afferents, causing apnea in rodent models, along with increased laryngeal nerve activity and airway constriction; these effects can be r

Glomus cell heterogeneity underpins distinct carotid body chemoreflex pathways: implications for hypertension.

AIMS: The carotid body (CB) is a multimodal chemosensory organ, yet how it encodes differential external stimuli to drive distinct peripheral chemoreflex responses remains unclear. This study aimed to define differences in intrinsic cellular mechanisms underlying CB signalling and to investigate how these processes are altered in pre-hypertension. METHODS AND RESULTS: We used potassium cyanide (KCN) as a hypoxia mimetic to activate the CB.K+ channels implicated in hypoxia sensing were pharmacologically inhibited, with TWIK-related acid-sensitive K+ (TASK) channels targeted using Ba2+ or ML365

Combination of the BK-Channel Antagonist ENA-001 and Naloxone Is More Effective Than Either Agent Alone in Reversing Fentanyl-Induced Respiratory Suppression in Rats.

Background Fentanyl overdose in humans produces two life-threatening effects: central μ-opioid receptor (MOR)-mediated hypoventilation and generalized muscle rigidity that further impairs ventilation. Naloxone, the current standard of care, reverses both effects via competitive antagonism at MOR; however, its clinical utility is limited, particularly in the setting of polydrug overdose, where nonopioid mechanism(s) may also contribute to respiratory suppression. ENA-001 restores ventilation as a peripherally directed (carotid body) BK-channel antagonist with a mechanism independent of MOR, and

Evolution and plasticity of the control of breathing in high-altitude natives: Insights from deer mice.

Breathing and the hypoxic ventilatory response are vital determinants of gas exchange and metabolism in hypoxia. High-altitude natives are valuable models for elucidating how plasticity and evolutionary adaptation can shape the control of breathing in chronic hypoxia. Here, we review studies on this issue in deer mice, a species found from sea level to over 4300 m elevation. Distinct populations from high and low altitudes have been bred in captivity and compared in common conditions of normoxia or hypoxia. Low-altitude populations exhibit ventilatory acclimatization to hypoxia (VAH), in which

Microvascular remodeling in the carotid body of spontaneously hypertensive rats: A stereological study.

The carotid body (CB) serves as the primary peripheral arterial chemoreceptor in mammals and plays a key role in the regulation of blood pressure by sensing changes in arterial blood gas levels and influencing sympathetic nerve activity. It is one of the most highly vascularized organs, with blood vessels occupying approximately 25% of its volume. Although hypertensive conditions are known to induce CB structural plasticity, detailed morphometric analysis of its vasculature remains limited. We performed a quantitative stereological study to assess microvascular remodeling in the CB of spontane

The influence of metaboreflex activation on pulmonary pressure with combined chemoreflex activation in acute and chronic hypoxia.

Sympathetic activation arising from both the peripheral chemoreflex in hypoxia and muscle metaboreflex during exercise mediate an increase in pulmonary artery systolic pressure (PASP). We tested the hypothesis that coactivation of the chemoreflex and metaboreflex would augment the PASP response, as has been shown previously for heart rate and ventilation. We conducted two experimental studies: (i) a laboratory study (n = 15) in normoxia and acute isocapnic hypoxia (45 mmHg P ET O 2 ${{P}_{{\mathrm{ET}}{{{\mathrm{O}}}_2}}}$ ) and (ii) a field study (n = 14) at sea level (344 m) and after 4-
