Transcriptional activation of EPHB1 by SOX2 promotes epithelial-mesenchymal transition and metastasis in esophageal squamous cell carcinoma.

Esophageal squamous cell carcinoma (ESCC) is a highly aggressive malignancy with an unfavorable prognosis, and the molecular mechanisms underlying its metastatic progression remain incompletely defined. Here, we investigated the functional role of ephrin type-B receptor 1 (EPHB1), a member of the ephrin receptor family, in ESCC metastasis. Integrated analyses of RNA-sequencing data and publicly available databases demonstrated that EPHB1 expression is significantly upregulated in ESCC tissues and is positively associated with advanced tumor grade and poor patient survival. Gain- and loss-of-fu

Predictive modeling of lung cancer subtype with integration of clinical and thoracic imaging features to guide early brain metastasis management.

PURPOSE: Optimal treatment for synchronous brain metastases at lung cancer diagnosis depends on subtype, but histologic/molecular profiling is often delayed. We developed machine-learning models using clinical and imaging data to classify lung cancer into three subtypes with distinct brain metastasis treatment implications: small cell lung cancer (SCLC), EGFR-mutated non-small cell lung cancer (EGFR+), and EGFR-wild-type non-small cell lung cancer (EGFR-). METHODS: We retrospectively identified patients with metastatic lung cancer diagnosed from 2016 to 2025 at a single institution. Patients

Efficacy and Safety of First-Line Treatment Options for Oncogene Wild-Type Lung Adenocarcinoma With Liver Metastases.

OBJECTIVE: Liver metastasis represents one of the frequent drivers for cancer-related mortality in lung adenocarcinoma. For advanced lung adenocarcinoma harboring wild-type oncogene drivers, chemotherapies in combination with immune checkpoint inhibitors (ICIs) and/or bevacizumab (named IC, BC, and IBC, respectively) are alternative first-line schemes as recommended by canonical treatment guidelines. However, due to the immunosuppressive tumor microenvironment in the liver, patients with liver metastases are less effective to ICIs. For this subpopulation, the optimal regimen is still undetermi

High-Throughput DNA Barcoding Identifies Lipid Nanoparticles for Lung Delivery of TRAIL-mRNA to Induce Tumor Cell Death in Experimental Metastasis Model.

INTRODUCTION: Lipid nanoparticles (LNPs) have enabled the clinical translation of nucleic acid therapeutics, and their composition plays a critical role in organ targeting and route-specific delivery. Intranasal (IN) administration offers direct access to the lungs, while reducing systemic exposure. However, designing LNPs capable of overcoming respiratory tract barriers remains a challenge. METHODS: We used DNA barcoding (b-DNA) for high-throughput in vivo screening of an LNP library following IN administration to investigate the impact of helper lipid identity and changes in the relative he

Redefining leptomeningeal metastasis in non-small cell lung cancer: From terminal complication to molecularly managed disease.

Leptomeningeal disease (LMD) is one of the most devastating complications of advanced non-small cell lung cancer (NSCLC), historically associated with profound neurologic morbidity and poor survival. Advances in cerebrospinal fluid (CSF)-based diagnostics, improved molecular characterization, and the development of central nervous system (CNS)-penetrant targeted therapies have begun to reshape both disease assessment and treatment expectations, particularly in oncogene-driven NSCLC. Available data suggest that outcomes in LMD are highly heterogeneous and are influenced by molecular subtype, pe

Advance Learning in Oncology - A CT Imaging Dataset of Lung Nodule Metastases from Bone and Soft Tissue Tumors.

Bone and soft tissue tumors (BSTTs) are prone to metastasize to the lungs to form pulmonary nodules, which are quite different from other metastatic nodules or primary nodules, such as those from lung, breast, or colorectal cancers. Early detection of these specific lung nodules is crucial for timely intervention and treatment of BSTTs. To promote the development of BSTTs detection algorithm based on deep learning, this paper releases a Computed Tomography (CT) dataset containing 59 patients and 779 BSTTs metastatic pulmonary nodules with pixel-level annotations. We further publish benchmark l

UBE2L6 promotes invasion and metastasis of triple-negative breast cancer by enhancing autophagy through STK38 ISGylation.

Although aberrant activation of autophagy is well known in triple-negative breast cancer (TNBC), its functional roles and underlying mechanisms remain largely unknown. In the present study, we found that high UBE2L6 expression was strongly associated with aggressive clinical features in TNBC. We demonstrated that UBE2L6 promoted migration, invasion, and lung metastasis of TNBC by enhancing STK38-mediated autophagy. Mechanistically, UBE2L6 stabilized STK38 by promoting its ISGylation and inhibiting its ubiquitin-proteasomal degradation. Therefore, targeting UBE2L6 and modulating the STK38 ISGyl

Targeting Cystine Addiction Suppresses Breast Cancer Lung Metastasis.

UNLABELLED: Metastatic disease remains a major cause of cancer-related mortality. Recent studies suggest that dissemination to other organs comes with metabolic changes that allow the metastasizing cancer cells to adapt to new microenvironments. A deeper knowledge of these specific metabolic features and associated vulnerabilities could lead to the development of more effective therapies against metastasis. We used in vivo and ex vivo models of MYC-driven breast tumorigenesis to explore the key metabolic pathways that change when mammary gland tumor (MGT) cells metastasize to the lung. Stable

Single-cell and spatial transcriptomics map lung-adaptive metastasis programs through Exoc4, Cd36-vasculogenic mimicry, and Cxcr2 signaling in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is an aggressive subtype associated with high metastatic potential and poor outcomes, with lung metastasis representing one of the most frequent and life-threatening events. However, the mechanism how cellular plasticity and reprogramming that enable TNBC cells to colonize in the lung remain incompletely understood. Here, we integrated single-cell RNA sequencing and spatial transcriptomics on paired MMTV-PyMT primary mammary tumors and lung metastatic tumors, complemented by functional assays in 4T1 mouse models, to delineate the cellular heterogeneity and

Targetable S100-RAGE signaling mediates intrinsic trastuzumab deruxtecan resistance in metastatic breast cancer.

PURPOSE: Trastuzumab deruxtecan (T-DXd) has improved outcomes in metastatic breast cancer; however, a substantial subset of patients experience early lack of clinical benefit that is not reliably predicted by routine clinicopathologic variables. In an institutional cohort of 109 T-DXd-treated metastatic lesions, HER2 expression level, ER/PR status, Ki-67 index, and metastatic site were not significantly associated with response, highlighting the need to define mechanistic determinants of intrinsic resistance. EXPERIMENTAL DESIGN: We performed spatial transcriptomic and proteomic profiling usi

Predicting local control of brain metastases after Gamma Knife radiosurgery: a multimodal deep learning-radiomics approach.

BACKGROUND: Accurate prediction of local control (LC) following Gamma Knife radiosurgery (GKRS) remains clinically challenging for patients with brain metastases (BMs) secondary to breast or lung cancer. The scarcity of validated pre-therapeutic biomarkers currently hinders the development of individualized treatment decision-making strategies. METHODS: This retrospective analysis included 324 BMs from 195 patients. To develop and benchmark various predictive frameworks, several models were constructed: clinical-only, radiomics-only, 2D deep learning (DL)-only, fused radiomics-DL (DLR), and a

A Review on EGFR-Tyrosine Kinase Inhibitors and their Resistance Mechanisms.

BACKGROUND: An essential component of cell development, proliferation, and survival is the transmembrane receptor known as the epidermal growth factor receptor (EGFR). Dysregulated EGFR signalling is an appealing pathway that has been linked to the genesis and progression of several cancer types. EGFR tyrosine kinase inhibitors (TKIs) are targeted drugs that show promise in the fight against cancer. EGFR tyrosine kinase inhibitors obstruct cancer growth and survival signalling pathways by blocking the receptor's tyrosine kinase domain. Patients with non-small cell lung cancer (NSCLC) that have

The multidimensional role of cancer epidemiology in cancer prevention: discovery science and beyond.

The pivotal role of epidemiology in the identification of the causes of cancer is well recognized. However, after this identification, the translation of those findings into cancer prevention typically requires further epidemiological research. The role of cancer epidemiology in these next steps and in other aspects of cancer prevention is perhaps less well appreciated. Here we describe a framework for the multidimensional role of cancer epidemiology in cancer prevention including (1) hazard identification, (2) risk assessment, (3) understanding natural history, and (4) evaluating biological t

Stress Promotes Lung Metastasis in Breast Cancer by Altering Neutrophil Differentiation.

UNLABELLED: Mental stress is widely recognized as a significant risk factor for breast cancer, exerting detrimental effects on both progression and prognosis. Herein, we investigated the role of stress in regulating breast cancer metastasis. In genetically engineered and transplantation breast cancer mouse models, chronic stress stimulation increased tumor growth and lung metastasis. Single-cell RNA sequencing analysis of the premetastatic lung microenvironment revealed induction of a previously unrecognized subtype of cancer stress-primed (CSP) neutrophils, characterized by the overexpression
