Non-Relapse Mortality With Bispecific Antibodies in Hematologic Malignancies: A Dual Systematic Review and Pharmacovigilance Analysis.

Bispecific antibodies (BsAbs) redirect T-cells to tumor antigens in an MHC-independent manner. These universally available, off-the-shelf products have demonstrated unprecedented efficacy in heavily pretreated patients with relapsed/refractory (R/R) hematological malignancies (HMs). The indefinite treatment duration associated with most BsAb products, coupled with neutropenia, B-cell aplasia, and hypogammaglobulinemia result in significant immune-related toxicities and infections. Hence, non-relapse mortality (NRM) which represents the cumulative burden of treatment-related toxicities, defined

Reversible atrioventricular block associated with transdermal fentanyl use: a paediatric case with non-Hodgkin lymphoma.

Pain is a common and significant clinical problem in childhood cancers, arising both from the disease itself and from administered treatments. Although pain control is primarily achieved with routine analgesics, this may be insufficient in chronic or progressive stages of the disease. In such cases, treatment with potent analgesics, such as opioid derivatives, comes into play. Transdermal fentanyl is widely preferred in paediatric oncology practice due to its ease of administration and effective analgesic properties. The most common adverse effects of opioids include tolerance, nausea, vomitin

Adenopathy and Extensive Skin Patch Overlying a Plasmacytoma (AESOP) Syndrome Associated With a Solitary Bone Plasmacytoma of the Left Scapula: A Rare Paraneoplastic Presentation Managed With High-Precision Radiotherapy.

Adenopathy and extensive skin patch overlying a plasmacytoma (AESOP) syndrome is a rare paraneoplastic manifestation characterized by three cardinal components: the underlying pathology (plasma cell dyscrasia), the cutaneous lesion (vascular cutaneous lesion), and the skeletal/nodal manifestation (solitary plasmacytoma/regional lymphadenopathy). Scapular involvement is exceptionally uncommon and may represent an early indicator of progression within the polyneuropathy, organomegaly, endocrinopathy, monoclonal plasma cell disorder, and skin changes (POEMS) spectrum. The Bird's Nest Sign as an Initial Presentation of Primary Pulmonary B-Cell Non-Hodgkin Lymphoma With Plasmacytic Differentiation.

The bird's nest sign (BNS) has been described as a sign on chest computed tomography (CT) in various pulmonary diseases. We report a rare case of primary pulmonary B-cell non-Hodgkin lymphoma with plasmacytic differentiation presenting with BNS, mimicking an invasive fungal disease at first. A 61-year-old Vietnamese man with a history of idiopathic thrombocytopenia and splenectomy 3 years prior to admission presented with prolonged dry cough, mild dyspnoea on exertion and chest discomfort for 3 weeks. His laboratory tests were unremarkable, but his chest CT showed bilateral consolidation with

Therapeutic potential of oncolytic viruses in hematopoietic malignancies: from preclinical insights to clinical realities.

While certain forms of lymphoma-such as classical Hodgkin lymphoma-are classified as curable, with over half of all patients achieving remission following first-line therapy, a significant proportion of patients still suffer from relapsed or refractory disease. Through autologous stem cell transplantation (ASCT) and chimeric antigen receptor (CAR) T-cell therapy can significantly improve survival rates, their widespread clinical utility is constrained by logistical bottlenecks, manufacturing complexities, and high financial burdens. Similarly, antibody-drug conjugates and bispecific antibodies

Open-label extension study of leniolisib in activated PI3Kδ syndrome: Long-term efficacy and safety.

Activated phosphoinositide 3-kinase delta syndrome (APDS) is an inborn error of immunity characterized by immune deficiency and dysregulation. Leniolisib is a selective phosphoinositide 3-kinase delta inhibitor approved to treat APDS in patients ≥12 years old. We report final results from a single-arm, open-label extension study in 37 patients with APDS (NCT02859727). Median leniolisib exposure was 4.3 years (range, 1.2-7.0 years). Adverse events (AE) were reported in 34 patients; 87% were grade 1. Serious AEs, reported in 10 patients, were deemed unrelated to leniolisib. Two patients disconti

Pulmonary malignant lymphoma requiring surgical lung biopsy for diagnosis following coronavirus disease 2019 infection: a case report.

BACKGROUND: Pulmonary involvement by peripheral T-cell lymphoma, not otherwise specified (PTCL-NOS), is an exceedingly rare extranodal manifestation with nonspecific radiological and clinical features. Its computed tomography (CT) findings-including ground-glass and consolidative opacities and, in some areas, a crazy-paving appearance-overlap with those of resolving infectious pneumonia and drug-induced lung injury, so that pulmonary lymphoma is readily mistaken for these more common conditions and the diagnosis is often delayed. CASE DESCRIPTION: A 62-year-old man with a smoking history and

Primary Hepatic Mucosa-Associated Lymphoid Tissue Lymphoma and Primary Biliary Cholangitis: A Case Report of a Rare Association.

INTRODUCTION: Primary hepatic mucosa-associated lymphoid tissue (MALT) lymphoma is an exceedingly rare subtype of extranodal marginal zone B-cell lymphoma within the spectrum of non-Hodgkin lymphomas. Chronic inflammatory and autoimmune conditions are recognized risk factors for lymphomagenesis; however, the association between primary hepatic MALT lymphoma and primary biliary cholangitis (PBC) has been rarely reported. CASE PRESENTATION: We report the case of a 50-year-old woman with a 9-year history of non-cirrhotic PBC, treated with ursodeoxycholic acid and with only partial biochemical re

Impact of EBV status and histology on patient outcomes with nivolumab-AVD vs BV-AVD in advanced classic Hodgkin lymphoma (S1826).

Classic Hodgkin lymphoma (cHL) associated with Epstein-Barr virus (EBV) positive status as well as nonnodular sclerosis (non-NS) histologic subtypes has demonstrated poorer outcomes compared with EBV- and NS tumors. We report a prespecified subset analysis of patients enrolled in the phase 3 SWOG S1826 trial to evaluate outcomes based on EBV status and histologic subtype in patients treated with N-AVD (nivolumab-AVD) or BV-AVD (brentuximab vedotin-AVD). In the phase 3 SWOG S1826 trial, patients with stage III to IV cHL were randomized to N-AVD or BV-AVD treatment. Of 970 eligible patients, 522

CUE domain-containing 2 promotes diffuse large B-cell lymphoma progression via PP2A-B55β degradation and shapes macrophage-mediated immune responses.

CUE domain-containing 2 (CUEDC2) regulates ubiquitin-mediated protein turnover, cell-cycle progression, and inflammatory signaling, but its role in diffuse large B-cell lymphoma (DLBCL) remains unclear. DLBCL is the most common aggressive non-Hodgkin lymphoma, and a substantial proportion of patients develop relapsed or refractory disease. Here, by integrating clinical bioinformatics, immunohistochemistry, and functional studies in DLBCL cell lines, we demonstrate that CUEDC2 is overexpressed in DLBCL and is associated with advanced disease stage and poor prognosis. Mechanistically, proteomic

Hodgkin Lymphoma With Sarcoid-Like Reaction: Importance of Clinicopathological Correlation in Granulomatous Lymphadenopathy.

BACKGROUND Sarcoid-like reaction, characterized by noncaseating epithelioid granulomas, occurs in 13.8% of Hodgkin lymphoma cases. When the granulomatous component is prominent and neoplastic cells are scarce, the initial biopsy may lead to a misdiagnosis of sarcoidosis. Reliance on pathological findings alone, without integration of the clinical context, can result in considerable diagnostic delay. CASE REPORT A 63-year-old man with systemic lymphadenopathy underwent cervical lymph node biopsy, which showed epithelioid granulomas without identifiable neoplastic cells. Sarcoidosis was initiall

Impact of Prophylactic Steroids on CAR T-cell Related Toxicity for Relapsed and Refractory Non-Hodgkin Lymphoma.

BACKGROUND: Anti-CD19 chimeric antigen receptor T-cell therapy (CAR-T) has transformed the treatment of relapsed/refractory (R/R) non-Hodgkin lymphoma (NHL). However, toxicities including cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS) remain significant. Prophylactic corticosteroid use increased after cohort 6 of ZUMA-1, which suggested reduced severe CRS and ICANS, although the lack of a comparator arm limits interpretation. OBJECTIVE: To evaluate the impact of steroid prophylaxis on outcomes and toxicities in a real-world setting. STUDY D

A Human Genome-wide CRISPR-Cas9 Screen Reveals Host Factors That Support Epstein-Barr Virus B cell infection.

UNLABELLED: Epstein-Barr virus (EBV) infects 90% of the population worldwide and is associated with multiple malignancies, including Burkitt, Hodgkin and post-transplant lymphomas, nasopharyngeal and gastric carcinoma. EBV also triggers autoimmune diseases, including multiple sclerosis and systemic lupus erythematosus. While host factors important for EBV B-cell entry have been defined, including CD21/CR2, MHC class II and R9AP, much remains to be learned about host factors that support post-entry steps of EBV infection. To gain insight, we conducted a human genome-wide CRISPR-Cas9 screen. In

Fibrous dysplasia of the jaw and diffuse large B-cell lymphoma coexistence, diagnosis and treatment experience and analysis: a case report.

BACKGROUND: Fibrous dysplasia (FD) is a rare benign fibro-osseous bone lesion, while diffuse large B-cell lymphoma (DLBCL) is the most common aggressive non-Hodgkin lymphoma. Their clinical and imaging manifestations overlap substantially, which complicates diagnosis, particularly when DLBCL manifests as extranodal bone lesions without peripheral lymphadenopathy. The coexistence of jaw FD and DLBCL is extremely rare, and relevant clinical experience is insufficient. CASE DESCRIPTION: This report presents a 46-year-old female with a 40-year history of jaw FD who presented with progressive righ

Delayed diagnosis of selected thoracic and mediastinal conditions in young adults: a focused narrative review of clinical pitfalls, cognitive biases, and system barriers.

BACKGROUND: Thoracic and mediastinal conditions affecting the esophagus, diaphragm, mediastinum, and airway carry considerable morbidity when diagnosis is delayed. Young adults are often perceived as low risk for serious thoracic disease, and low clinical suspicion, atypical symptoms, and cognitive and healthcare-system barriers can delay recognition. METHODS: This focused narrative review searched PubMed and Scopus, supplemented by Google Scholar, for studies published between January 2000 and May 2026, with selected earlier landmark studies retained where essential. Advances in CAR-T and CAR-NK cell therapy in follicular lymphoma: a review.

Follicular lymphoma (FL) is the most common indolent B-cell non-Hodgkin lymphoma and is characterized by prolonged survival but recurrent relapses. The unmet need is greatest among patients with high-risk disease, including progression of disease within 24 months of first-line immunochemotherapy (POD24), double-refractory disease, histologic transformation, and exposure to multiple previous lines of therapy. In pivotal single-arm studies enrolling heavily pretreated patients, CD19-directed chimeric antigen receptor T-cell (CAR-T) therapies produced overall response rates of 86-97% and complete

[Whole blood exchange for a critically ill patient with Hodgkin lymphoma complicated by severe autoimmune hemolytic anemia: a case report and literature review].

This article reports the management of a patient with Hodgkin lymphoma (HL) complicated by refractory critical autoimmune hemolytic anemia (AIHA) and explores the value of whole blood exchange (WBE) after failure of conventional therapy and plasma exchange. The patient was a 52-year-old woman with a history of classical HL treated with anti-PD-1 therapy. She was admitted due to progressive fatigue and altered mental status, with a nadir hemoglobin level of 24 g/L and a positive direct antiglobulin test for both anti-immunoglobulin G and anti-C3d (1:256). She also presented with impaired consci
