Differential Effects of Ischemia and Inflammation on Plasma-Derived Extracellular Vesicle Characteristics and Function in a Mouse Model.

Extracellular vesicles (EVs) have long been understood to be important mediators of cell-to-cell communication and may lead to the molecular aftermath and exacerbation of brain injuries such as stroke. This study explored how the source of the EVs influenced their characteristics and the effect these differences had on naïve brain tissue. EVs were isolated from mice post-stroke in the acute or chronic stages of recovery in animals with and without reperfusion (transient and permanent middle cerebral artery occlusion) and from a model of systemic inflammation (i.p. lipopolysaccharide). The data

Comparing Organs at Risk Sparing Between Intensity-Modulated Radiotherapy and Pencil-Beam Scanning Plans Based on Disease Location for Hodgkin and Non-Hodgkin Lymphoma.

PURPOSE: Radiotherapy (RT) is often used as a combined modality therapy for patients with Hodgkin (HL) and aggressive Non-Hodgkin Lymphoma (aNHL). RT options include photon intensity-modulated radiotherapy (IMRT) or proton therapy (PT). As PT is a limited and costly resource, it is important to identify cases in which PT may best benefit in order to offer providers and patients the best possible information for decision-making. Accordingly, the purpose of this study was to assess the difference in OAR sparing between photon IMRT and pencil-beam scanning (PBS) based on disease location using th

Effects of physical activity and exercise interventions in health-related variables in Hodgkin's and non-Hodgkin's lymphoma patients during clinical treatment: a systematic review and single-arm meta-analysis.

INTRODUCTION: Cancer-related fatigue and reduced health-related quality of life (HRQOL) are common among lymphoma patients undergoing treatment. Exercise may be safe and feasible for individuals with hematological malignancies, with potential benefits in mitigating treatment-related toxicity and enhancing treatment tolerance. This systematic review and meta-analysis aimed to evaluate the effects of exercise interventions on health-related outcomes such as HRQOL, muscle strength, cardiorespiratory fitness (CRF), fatigue, and body composition in this population. EVIDENCE ACQUISITION: Electronic

Automated RECOMIA AI-based total metabolic tumor volume in lymphoma - a retrospective study.

BACKGROUND:  Increasing evidence suggests that total metabolic tumor volume (tMTV) measured before treatment in lymphoma patients undergoing [18F]fluorodeoxyglucose (FDG) PET/CT scans can predict prognosis. However, there is a lack of fast, reliable, and easy-to-perform multilesional segmentation tools with an urgent need to improve tMTV segmentation workflow in clinical practice. Here, we develop an artificial intelligence (AI)-based tool that automatically calculates tMTV in untreated lymphoma patients undergoing FDG PET/CT. The RECOMIA AI-based tool is a 3D U-Net convolutional neural networ

Efficacy of Standard-Dose Versus Low-Dose Nivolumab in Relapsed/Refractory Hodgkin Lymphoma-A Retrospective Analysis.

PURPOSE: As there is a paucity of data comparing outcomes with different dosing regimens of PD-1 inhibitors in relapsed/refractory Hodgkin lymphoma (R/R HL), this retrospective analysis was undertaken to compare the efficacy of low-dose nivolumab (fixed at 40 mg) with the standard dose of 3 mg/kg. METHODS: Single-center retrospective analysis of patients with R/R HL who failed at least one salvage regimen and were treated with nivolumab between 2015 and 2023, at either 3 mg/kg (standard-dose nivolumab [SD-NIV]) or a flat dose of 40 mg (NIV40), both administered once in 2 weeks. Primary Diffuse Large B-Cell Lymphoma Mimicking a Dental Abscess: A Case Report.

Diffuse large B-cell lymphoma (DLBCL) is the most common subtype of non-Hodgkin lymphoma, yet its presentation in the maxillofacial region can pose diagnostic challenges, particularly in patients with confounding medical histories. We report the case of a 40-year-old Iranian male with a history of ulcerative colitis on mesalazine therapy, who presented with progressive left facial swelling. Symptoms persisted for over a year, during which initial fine-needle aspiration (FNA) of a cervical lymph node showed only chronic lymphadenitis. This result prompted repeated empiric treatments with antibi

Understanding hairy cell leukemia in the context of mature B-cell neoplasms: tumor microenvironment and extracellular vesicle contribution to disease pathogenesis.

Mature B-cell neoplasms constitute a biologically and clinically heterogeneous group of hematologic malignancies, defined by the clonal proliferation and accumulation of monotypic mature B lymphocytes, which may involve the peripheral blood, bone marrow (BM), lymphoid tissues, or present primarily in extranodal sites. While the pathogenesis of common subtypes, such as chronic lymphocytic leukemia, multiple myeloma, Hodgkin Lymphoma and Diffuse Large B-cell Lymphoma has been extensively studied, those of rare entities like Hairy Cell Leukemia (HCL) remains poorly understood. HCL is a distinct B

Evolution of outcomes in autologous stem cell transplantation for hodgkin lymphoma over 2 decades at King Hussein cancer center.

Autologous hematopoietic stem cell transplantation (auto-HCT) remains a cornerstone in relapsed Hodgkin lymphoma (HL), especially where access to novel agents is limited. We evaluated trends in outcomes after auto-HCT at King Hussein Cancer Center (KHCC) over two decades. We retrospectively analyzed adult HL patients who underwent auto-HCT between 2003 and 2020, divided into two eras: group A (2003-2015) and group B (2016-2020). Survival was estimated by Kaplan-Meier and compared by log-rank; prognostic factors were assessed with Cox regression. We identified 265 patients (group A: N = 149, 56

CD30 CAR-T cells in combination with anti-PD-1 camrelizumab in relapsed/refractory CD30+ lymphomas.

INTRODUCTION: Although CD30-directed chimeric antigen receptor (CAR)-T cell therapy has demonstrated antitumor activity in CD30+ lymphomas, its therapeutic efficacy remains suboptimal. We therefore conducted a prospective, phase II, single-arm, multicenter clinical trial (ChiCTR-2100046763) to evaluate the efficacy and safety of CD30 CAR-T cell therapy in combination with camrelizumab, an immune checkpoint inhibitor, in patients with relapsed/refractory (r/r) CD30+ lymphomas. METHODS: All participants received a lymphodepleting regimen followed by infusion of CD30 CAR-T cells at a dose of 1 ×

Put the CAR-T before the HRS: Advances in Anti-CD30 Immunotherapy Targeting Hodgkin/Reed-Sternberg Cells in Classical Hodgkin Lymphoma.

Classical Hodgkin lymphoma (cHL) is characterized by rare Hodgkin/Reed-Sternberg (HRS) tumor cells that uniformly express cluster of differentiation (CD)30 molecules and orchestrate an immunosuppressive tumor microenvironment, making CD30 an attractive and selective therapeutic target. We summarize the biological rationale for CD30 as a therapeutic target and the preclinical and clinical evidence across major platforms: antibody-drug conjugates (brentuximab vedotin), monoclonal antibodies (including acimtamig and its combinations with Natural Killer cells), second- and third-generation chimeri

Bibliometric analysis of global hodgkin lymphoma research from 2002 to 2022.

BACKGROUND: Hodgkin lymphoma is a rare neoplasm of the lymphatic system, representing one of the most common cancers in young adults. It is characterised by a low number of malignant cells derived from B-lymphocytes and an extensive microenvironment. METHOD: We used Scopus, Web of Science and PubMed databases with search descriptors; “Hodgkin* lymphoma“ and “Hodgkin* disease”. The common bibliometric indicators were applied. RESULTS: We retrieved a total of 18,304 articles within 2002 and 2022. The growth pattern showed a saturated pattern, with the linear and exponential plots coinciding. A s

Burden of Middle-Aged and Elderly Patients With Non-Hodgkin Lymphoma From 1990 to 2021: A Systematic Analysis Based on the Global Burden of Disease 2021.

BACKGROUND: Non-Hodgkin lymphoma (NHL) is increasingly prevalent in middle-aged and elderly populations, contributing to the rising global health burden, particularly in high-income countries. METHODS: This retrospective analysis utilized data from the Global Burden of Disease (GBD) study (1990-2021) to examine temporal trends in age-standardized incidence, mortality, and disability-adjusted life year (DALY) rates of NHL among middle-aged and older adult populations, incorporating age- and sex-stratified comparisons. Furthermore, projections for the subsequent 30 years were generated using Ba

Real-World Outcomes of Allogeneic Stem Cell Transplantation in Relapsed Hodgkin Lymphoma: A Single-Center Experience from India.

BACKGROUND: Relapsed Hodgkin lymphoma (HL) after autologous stem cell transplantation (auto-HSCT) remains a major therapeutic challenge with few curative options. Allogeneic hematopoietic stem cell transplantation (allo-HSCT) may provide durable disease control, yet outcome data from tertiary-care centres in India are limited. OBJECTIVE: To evaluate real-world outcomes of allo-HSCT in patients with chemotherapy-sensitive relapsed HL at a tertiary care centre in North India. METHODS: This retrospective study included patients with chemotherapy-sensitive relapsed HL who underwent allo-HSCT at

Focus on Extremely Late Relapses in Hodgkin Lymphoma: A Single Center Analysis.

Background/Objective: Very late relapses (VLRs) in Hodgkin lymphoma (HL), occurring ≥5 years after initial treatment, have been well-recognized for decades. Recent studies have highlighted the persistent and linear risk of relapse extending well beyond 5 years, while also describing the incidence and risk factors. However, extremely late relapses (EXLRs) occurring ≥20 years after diagnosis remain poorly described and largely limited to case reports. In the present study, we focus on patients experiencing EXLRs after chemotherapy or combined modality therapy, aiming to define their incidence, c

Prognostic Value of the CALLY Index in Diffuse Large B-Cell Lymphoma: Linking Inflammation, Nutrition, and Tumor Biology.

Background/Objectives: Diffuse large B-cell lymphoma (DLBCL) is the most common subtype of non-Hodgkin lymphoma and remains incurable in approximately 30-40% of patients despite advances in immunochemotherapy. Although gene expression profiling has improved risk stratification, there is an ongoing need for non-invasive, cost-effective, and clinically practical biomarkers to identify patients at high risk of treatment resistance or relapse (R/R). Systemic inflammation plays a pivotal role in DLBCL pathogenesis, impacting both tumor progression and treatment response. The C-reactive protein-albu

Response-adapted de-escalation of radiotherapy in pediatric Hodgkin's lymphoma.

Pediatric Hodgkin's lymphoma has a very high cure rate; hence, there is a major focus on minimizing radiation therapy. This study examined the impact of omitting radiotherapy (RT) on patients with rapid early response (RER) and minimizing RT on patients with slow early response (SER). This study included all pediatric Hodgkin's lymphoma patients who were treated with a response-adapted approach (study group). They were compared to a historical cohort who received their treatment with both chemotherapy and classic involved field radiotherapy to all patients regardless of the response (control g
