Reversal of Cerebral Glucose Hypometabolism in a Patient with COVID and Refractory B-Cell Lymphoma.

Cerebral glucose hypometabolism (CGHM) involves reduced brain glucose uptake on FDG-PET imaging and occurs in neurological and systemic diseases. Its reversibility remains uncertain. We describe a 40-year-old man with reversible global CGHM associated with cryptogenic organizing pneumonia (COP) and COVID-19. His neurocognitive symptoms improved rapidly following steroid-induced hyperglycemia, with FDG-PET demonstrating complete CGHM resolution after two months. His medical background included primary refractory transformed follicular lymphoma diagnosed 30 months earlier, managed with multiple

Real-world safety profile of mosunetuzumab: a pharmacovigilance study based on the food and drug administration adverse event reporting system.

BACKGROUND: Mosunetuzumab is a CD20×CD3 bispecific antibody approved for adult relapsed or refractory follicular lymphoma. Post-marketing evidence on its safety profile remains scarce. This pharmacovigilance study used the Food and Drug Administration Adverse Event Reporting System (FAERS)to characterize real-world adverse event (AE) signals associated with mosunetuzumab. METHODS: We performed disproportionality analysis using reports from the FAERS database spanning from the first quarter of 2004 to the fourth quarter of 2025. The primary analysis set encompassed reports wherein mosunetuzuma

Optimizing CAR-T therapy delivery in the Italian healthcare system: economic and organizational insights in follicular lymphoma.

INTRODUCTION: Follicular Lymphoma (FL) is the second most common lymphoid malignancy, with 20% of patients progressing within 24 months of first-line therapy. Chimeric antigen receptor T-cell (CAR-T) therapies have improved outcomes in relapsed or refractory FL, yet their use requires significant resources and complex care pathways. Although outpatient CAR-T programs have shown promising safety and feasibility, limited evidence exists on their organizational, economic, and ethical implications in the Italian healthcare setting. This study compares the current CAR-T pathway for FL patients with

Comprehensive analysis of mortality risk factors in low-grade B-cell lymphoma.

BACKGROUND: Low-grade B-cell lymphomas (LGBCLs) account for approximately 40% of non-Hodgkin lymphomas with low progression. LGBCL is divided into subgroups, which share common complications. Analyzing prognostic factors and mortality causes could improve patient survival; however, currently available models present limitations in discriminating the cause of death. Therefore, this study aimed to compare the prognostic factors and causes of death, such as secondary malignancies (SMs), aggressive histologic transformation (HT), and infectious complications, including coronavirus disease 2019 (CO

Phase 3 PERSPECTIVE study: Ibrutinib with rituximab for initial treatment of patients with follicular lymphoma.

We report primary analysis results from the Phase 3 PERSPECTIVE study comparing ibrutinib + rituximab versus placebo + rituximab in patients with previously untreated follicular lymphoma requiring treatment per Groupe d'Etude des Lymphomes Folliculaires criteria who were ineligible for chemoimmunotherapy due to age and/or comorbidities. The primary endpoint was investigator-assessed progression-free survival (PFS). Patients were randomly assigned 3:1 to receive ibrutinib (560 mg) or placebo once daily until progression, together with rituximab 375 mg/m2 weekly for 4 weeks, then every 8 weeks f

Mosunetuzumab Successfully Treated Rituximab-Refractory Lymphoma-Associated Protein-Losing Enteropathy With Concomitant Immune Thrombocytopenia in Follicular Lymphoma: A Case Report.

UNLABELLED: Protein-losing enteropathy (PLE) is a rare but potentially severe complication of malignant lymphoma characterized by excessive gastrointestinal loss of plasma proteins, leading to hypoalbuminemia, edema, and serous effusions. We report a case of follicular lymphoma complicated by severe PLE and concomitant immune thrombocytopenia (ITP) without gastrointestinal involvement. A 51-year-old man presented with progressive hypoalbuminemia, refractory ascites, thrombocytopenia, and systemic lymphadenopathy. Initial treatment with rituximab monotherapy was selected due to low tumor burden

Association of age and primary treatment with risk of non-lymphoma-related death and long-term survival outcomes in adult patients with early-stage follicular lymphoma: a population-based analysis.

In adult patients with early-stage follicular lymphoma (FL), age-related differences in the risk of non-lymphoma-related death (LRD) during the follow-up period after treatment are unclear. We aimed to investigate the interactive effects of age and primary treatment on the risk of non-LRD and survival. We retrospectively analyzed the data of 6030 adult patients with early-stage FL from the Surveillance, Epidemiology, and End Results (SEER) database (2000-2020). Primary therapy involved observation (n = 2509), chemotherapy (CT) alone (n = 1844), radiotherapy alone (RT, n = 1191) or RT and CT (R

Sustained improvement in survival for primary ocular adnexal lymphoma from 2005 to 2024 by model-based period analysis.

PURPOSE: Primary ocular adnexal lymphoma (POAL) is the most common malignancy affecting para-ocular tissues, making prognosis and survival critical concerns. This study analyzed trends in five-year relative survival rates (RSR) to evaluate survival outcomes. METHODS: This retrospective study included patients diagnosed with POAL from the Surveillance, Epidemiology, and End Results database. Five-year RSR was calculated for the periods 2005-2009, 2010-2014, and 2015-2019. Data were stratified by demographic and clinical characteristics. Model-based period analysis was conducted to assess RSR t

Real-world outcomes after CAR T versus standard therapy in third-line or later relapsed or refractory follicular lymphoma in the United States.

BACKGROUND: Relapsed/refractory (R/R) follicular lymphoma (FL) often requires multiple lines of therapy. OBJECTIVE: To compare time to next treatment (TTNT), treatment-free interval (TFI), healthcare resource utilization (HRU), and downstream costs among R/R FL patients treated with CAR T versus non-CAR T therapy. METHODS: Adult R/R FL patients with ≥2 prior lines of therapy were stratified into CAR T and non-CAR T cohorts. The Index Date was CAR T infusion (CAR T) or 3L initiation (non-CAR T). Outcomes included TTNT, TFI, HRU, and costs. Analyses were descriptive, incorporating Kaplan-Meier

RENOIR phase 3 rituximab-lenalidomide vs rituximab as maintenance treatment in relapsed/refractory follicular lymphoma.

Maintenance treatment in older patients with relapsed or refractory (R/R) follicular lymphoma (FL) remains an area of investigation. RENOIR was a multicenter, phase 3, open-label randomized trial conducted by the Fondazione Italiana Linfomi (FIL) in older patients with R/R FL after 1 or 2 previous therapies. Patients achieving partial response or complete response (CR) after 4 to 6 cycles of standard rituximab (R)-based chemotherapy were randomized 1:1 to maintenance with R alone (standard arm) or R plus lenalidomide (R2; experimental arm). The primary end point was 2-year progression-free sur

Digital and Manual Assessment of Intrafollicular Ki-67, MYC, and p53 in Classic Follicular Lymphoma.

Background/Objectives: There remains a need for additional prognostic markers in classic follicular lymphoma (cFL) to identify aggressive disease. Immunohistochemical stains such as Ki-67, MYC, and p53 have shown variable associations with histologic grade and adverse outcomes. In this study, we aimed to assess intrafollicular Ki-67, MYC, and p53 expression in cFL via immunohistochemistry, quantified by both manual and digital methods, and evaluate their relation to histologic grade and clinical outcomes. Methods: We evaluated 37 cases of cFL from 2000 to 2019 and performed immunohistochemistr

CCR7 orchestrates the malignant progression of memory B cells in MYC-overexpressing lymphoma.

Follicular lymphoma (FL) is one of the most common types of indolent non-Hodgkin lymphoma that can undergo rapid progression, which is associated with poor patient outcomes. The homing of B cells into aberrant immunological niches within the germinal center (GC) is a prerequisite for disease development. The C-C chemokine receptor type 7 (CCR7), expressed by circulating B cells, mediates their homing and dissemination to draining lymph nodes (drLNs). Here, we identified activated B cells emerging from adjacent GCs in drLNs as the origin of circulating memory B cells in distant LNs. Transcripti

Safety and Efficacy of Mosunetuzumab: Experience in the Hospital Cardinale Giovanni Panico.

BACKGROUND/OBJECTIVE: Follicular lymphoma (FL) is one of the most common indolent B-cell non-Hodgkin lymphomas (NHL) and is characterized by recurrent relapses despite advances in therapy. Bispecific antibodies that redirect T lymphocytes toward malignant B cells represent a major innovation in the treatment of relapsed or refractory disease. Mosunetuzumab is a CD20×CD3 bispecific antibody that induces T-cell mediated cytotoxicity against B-cell malignancies. In this manuscript, we describe the clinical experience with mosunetuzumab in three patients with relapsed or refractory FL treated at t

Serum free fatty acid promotes tumor progression and predicts golidocitinib sensitivity in peripheral T-cell lymphoma.

Peripheral T-cell lymphoma (PTCL) is an aggressive and heterogeneous lymphoma subtype with high chemoresistance and poor prognosis. Common peripheral blood disease biomarkers with therapeutic potential are lacking. In this study, we analyzed the serum metabolic profiles of 557 patients with newly diagnosed PTCL, including 278 extranodal natural killer/T-cell lymphoma (NKTCL), 117 patients with nodal T-follicular helper cell lymphoma (nTFHL), 92 with PTCL, not otherwise specified (PTCL-NOS), 36 with anaplastic lymphoma kinase (ALK)-positive anaplastic large-cell lymphoma (ALCL), and 34 with ALK

Real-world outcomes and toxicities of CAR-T in relapsed/refractory follicular lymphoma: a multicenter cohort study.

Chimeric antigen receptor T-cell therapy has revolutionized the treatment of relapsed/refractory (R/R) follicular lymphoma (FL). Real-world efficacy and toxicity data are needed because clinical trial populations are often unrepresentative. Therefore, we conducted a multicenter retrospective study of R/R FL patients undergoing commercial axicabtagene ciloleucel (axi-cel) or tisagenlecleucel (tisa-cel) between 2021 and 2024. End points included efficacy measures (overall response rate [ORR], complete response rate [CRR], progression-free survival [PFS], and overall survival [OS]) and toxicity (

Bendamustine in combination with gemcitabine and vinorelbine as salvage regimen for relapsed or refractory peripheral T-cell lymphomas: A retrospective single-centre experience.

The prognosis of peripheral T-cell non-Hodgkin lymphomas (PTCL) is dismal, particularly in the relapsed/refractory (r/r) setting, where 3-year overall survival (OS) is 20%-30%. No superior second-line therapy for PTCL has been universally established, and durable remissions rely on consolidation with allogeneic haematopoietic stem cell transplantation (alloHSCT). Enhancing response rates to salvage therapy is, therefore, crucial to increase transplant eligibility. We retrospectively evaluated the efficacy of bendamustine, gemcitabine and vinorelbine combination (BeGeV) in 24 consecutive patien

Extranodal Marginal Zone B-cell Lymphoma Presenting as a Painless Buccal Mass in the Masticator Space.

Extranodal marginal zone B-cell lymphoma (EMZL, mucosa-associated lymphoid tissue type) is an indolent non-Hodgkin lymphoma that only rarely arises in deep facial spaces. Primary masticator-space involvement is particularly uncommon and can mimic benign buccal soft-tissue lesions. We report a 67-year-old woman with a 4-year history of a painless, slowly enlarging left cheek mass. Examination showed a soft, mobile 3-cm buccal swelling with normal overlying skin and no intraoral lesion. Contrast-enhanced computed tomography demonstrated an infiltrative soft-tissue mass measuring 3.1 × 1.5 × 3.5
