Cutaneous Lymphoma International Prognostic Index and Risk Stratification in Advanced Mycosis Fungoides and Sézary Syndrome.

IMPORTANCE: Risk stratification in advanced mycosis fungoides and Sézary syndrome (MF/SS) remains suboptimal given that current TNMB (tumor, node, metastasis, blood) staging has limited prognostic value for clinically relevant outcomes. OBJECTIVE: To determine whether the Cutaneous Lymphoma International Prognostic Index (CLIPI) may be used to predict progression-free survival (PFS) and best objective response (BOR) in patients with advanced MF/SS. DESIGN, SETTING, AND PARTICIPANTS: This international prospective cohort study included patients with stage IIB to IVB MF/SS from the PROCLIPI tr

ESTRO consensus recommendations on total skin electron beam technique - Endorsed by ILROG.

BACKGROUND AND PURPOSE: Total skin electron beam therapy (TSEBT) is an established radiation treatment modality for the management of cutaneous T-cell lymphoma (CTCL). In recent years, there has been an increasing interest in dose de-escalation strategies aimed at lowering treatment-induced side effects whilst maintaining efficacy. This evolution in clinical practice has highlighted the need for consistent technical standards to ensure treatment quality and comparability across medical institutions. A unified set of recommendations for TSEBT in the modern era was sought by the European Society

All-Oral PI3Kδ and HDAC Inhibition with Parsaclisib Plus Chidamide and Step-Down Maintenance in Relapsed or Refractory Peripheral T-Cell Lymphoma: A Phase Ib Trial.

Single-agent targeted therapies for relapsed or refractory peripheral T-cell lymphoma (R/R peripheral T-cell lymphoma [PTCL]) produce modest, short-lived responses, underscoring the need for tolerable, chemotherapy-free combinations. In an open-label, single-arm Phase Ib trial at two tertiary hospitals, we evaluated parsaclisib plus chidamide with step-down maintenance. Adults with R/R PTCL received parsaclisib 10-20 mg once daily plus chidamide 20 mg twice weekly for 8 weeks using 3+3 escalation; patients without progression or unacceptable toxicity continued parsaclisib 2.5 mg once daily plu

The first annotated checklist of the bees of Colombia reveals an overlooked diversity and areas with major data deficiencies.

BACKGROUND: Most existing knowledge about bee biodiversity comes from temperate regions, where diversity is presumed highest-yet substantial data gaps across the tropics confound this pattern. Checklists are critical for documenting species presence, identifying knowledge gaps, and guiding conservation. We present the first comprehensive bee checklist for Colombia, a country in tropical northern South America that serves as a key biogeographic bridge between Central and South America. We hypothesize that Colombia's complex topography, its position at the intersection of multiple biogeographic

Advances in diagnosis and treatment of angioimmunoblastic T-cell lymphoma.

Angioimmunoblastic T-cell lymphoma (AITL) is a peripheral T-cell lymphoma (PTCL) that originates from follicular helper T cells (Tfh) and represents a rare subtype of non-Hodgkin lymphoma (NHL). The disease is characterized by unique morphological, molecular, and clinical features, and exhibits aggressive behavior. Currently, there is no standard treatment protocol, and patients treated with first-line CHOP-like chemotherapy show low remission rates, high recurrence rates, and poor overall prognosis. In recent years, with the in-depth study of the molecular genetic mechanism of AITL, various n

Graft-Versus-Tumour Effect Following Allogeneic Haematopoietic Stem Cell Transplantation in a Paediatric Patient With Mogamulizumab-Treated Primary Cutaneous Peripheral T-Cell Lymphoma.

INTRODUCTION: Primary cutaneous peripheral T-cell lymphoma, not otherwise specified (pcPTCL-NOS), is exceptionally rare in children, and evidence guiding diagnosis and treatment is limited. CASE PRESENTATION: A 10-year-old boy with pcPTCL-NOS refractory to chemotherapy received mogamulizumab prior to allogeneic haematopoietic stem cell transplantation (allo-HSCT). Despite a 60-day washout period, he developed prolonged steroid-refractory acute graft-versus-host disease requiring ruxolitinib and extracorporeal photopheresis. Concurrently, transient skin lesions developed at previous lymphoma s

Systemic Retinoids in Cutaneous T-Cell Lymphoma: A Scoping Review of Clinical Use and Evidence Gaps.

INTRODUCTION: Systemic retinoids are ubiquitously used in cutaneous T-cell lymphoma (CTCL), often in combination with skin-directed or systemic therapies, yet their role within contemporary multimodal treatment strategies has not been comprehensively synthesized. METHODS: We conducted a PRISMA-ScR-guided scoping review of systemic retinoids in CTCL using PubMed, Embase, Scopus, Web of Science, and the Cochrane Library through December 1, 2025. Study-level and treatment arm-level analyses characterized treatment patterns, outcomes, and reporting practices. Reported response rates were descript

Cutaneous T-cell lymphoma emerging during long-term dupilumab therapy in a patient with longstanding atopic diathesis: diagnostic challenges and mechanistic considerations.

Atopic dermatitis and cutaneous T-cell lymphoma can overlap both clinically and histopathologically, potentially delaying recognition of early disease. We report a case of a woman in her 40s with longstanding atopic diathesis who developed patch-stage mycosis fungoides after more than 5 years of stability on dupilumab. Earlier biopsies showed spongiotic dermatitis without evidence of a lymphoproliferative disorder. She later presented with a rapidly progressive, intensely pruritic eruption of erythematous scaly plaques with near-erythroderma, morphologically distinct from her prior dermatitis.

Immunophenotyping of CD8+ T Cells in Classical Hodgkin Lymphoma Reveals a Shift Toward Intermediate Exhaustion Following PD-1 Blockade.

The immune microenvironment of classical Hodgkin lymphoma (cHL) is characterized by rare Hodgkin-Reed-Sternberg (HRS) cells surrounded by dysfunctional lymphocytes. HRS cells exploit the PD-L1/PD-1 axis to suppress antitumor immunity. Although PD1 blockade is effective in some relapsed cHL patients, the mechanisms of resistance remain unclear. We investigated T-cell dynamics in cHL using whole-exome sequencing (WES) of plasma circulating tumor DNA and longitudinal single-cell RNA and T-cell receptor sequencing of peripheral blood lymphocytes from a relapsed patient before and after anti-PD1 th

Regulation of follicular helper T cell subset differentiation in health and disease.

Follicular helper T (TFH) cells play a crucial role in adaptive humoral immunity. They provide help to B cells in the germinal centers (GCs), driving the affinity maturation of antigen-specific antibodies and class switch recombination. TFH cells possess profound heterogeneity in spatial distribution (e.g., GC-TFH, follicular mantle-TFH, circulating TFH, peripheral TFH) and phenotypic characteristics (e.g., TFH1, TFH2, TFH17), which enable TFH cells to respond to diverse immune challenges. This review summarizes the recent progress, including our data, on the TFH regulatory network driven by B

Immune-related adverse events with mogamulizumab in cutaneous T-cell lymphoma: A multicentre study.

BACKGROUND: Mogamulizumab, an anti-CCR4 monoclonal antibody used in advanced mycosis fungoides (MF) and Sezary syndrome (SS), also depletes regulatory T cells, potentially leading to immune-related adverse events (irAEs). Unlike mogamulizumab-associated rash (MAR), other irAEs remain poorly documented to date. OBJECTIVES: To estimate the prevalence of mogamulizumab-associated irAEs (excluding MAR), characterize their clinical profile and assess management strategies and outcome. METHODS: A retrospective nationwide multicentre survey was conducted using the French Study Group on Cutaneous Lym

Advanced-stage cutaneous T-cell lymphomas show distinct immune profiles in erythroderma vs. tumor lesions.

BACKGROUND: Advanced-stage cutaneous T-cell lymphomas (CTCL) typically manifest as either skin tumor formation or erythroderma; however, exact mechanisms of lesion formation remain insufficiently characterized. OBJECTIVE: To better understand mechanisms of erythroderma vs. tumor formation in advanced-stage CTCL. METHODS: We compared molecular profiles of eight erythrodermic CTCL (eCTCL) with seven tumor-stage mycosis fungoides (tMF) samples by using single-cell RNA sequencing combined with T-cell receptor sequencing, and corroborated key findings using spatial transcriptomics. RESULTS: All

Impact of Residual Disease Localization Before Allogeneic Hematopoietic Stem Cell Transplantation on Outcomes in Adult T-Cell Leukemia-Lymphoma.

Allogeneic hematopoietic stem cell transplantation (allo-HCT) is the only curative approach for adult T-cell leukemia-lymphoma (ATL). However, preventing post-transplant relapse remains challenging. While pre-transplant disease status is known to be associated with post-transplant outcomes, the impact of residual disease (RD) localization at the time of allo-HCT has not been well described. In this retrospective study of 33 patients with aggressive ATL who underwent allo-HCT, patients were classified into two risk groups based on RD sites: a low-risk group (no lymph node [LN] or extranodal [EN

Microneedle-Assisted Cell Delivery and Therapy.

STUDY DESIGN: Secondary analysis. OBJECTIVE: To investigate the Fear-Avoidance Beliefs Questionnaire (FABQ) for its ability to predict 6-month outcomes for patients with low back pain (LBP) participating in physical therapy clinical trials. BACKGROUND: Consistent evidence suggests that fear-avoidance beliefs are predictive of short-term outcomes for patients with LBP. However, proposed cut-off scores have not been widely investigated for longer-term outcomes in samples of patients receiving physical therapy. METHODS AND MEASURES: Subjects (n = 160) were participants in 2 separate randomized

Identification and Validation of BATF3 as a Promising Biomarker Gene for Peripheral T-cell Lymphoma.

BACKGROUND: Peripheral T-cell lymphoma (PTCL) is a rare and heterogeneous group of hematological malignancies. Treatment options are limited and often unsatisfactory, leading to a poor prognosis in most subtypes. OBJECTIVE: This study aimed to identify potential biomarker genes for PTCL and to explore the underlying mechanisms by integrating machine learning, Mendelian Randomization (MR), and experimental validation. METHODS: Microarray datasets (GSE6338, GSE14879, and GSE59307) were downloaded from the Gene Expression Omnibus database. Differential expression analysis was conducted to ident
