[Analysis of dermoscopic and histopathological features in misdiagnosed cases of early mycosis fungoides].

Retrospectively, patients who were pathologically diagnosed with early-stage mycosis fungoides (MF) in the Department of Dermatology, Zhongshan Hospital Affiliated to Xiamen University, from October 1, 2023 to April 1, 2024 and had a history of misdiagnosis were enrolled in this study. Their clinical characteristics, dermoscopic manifestations of skin lesions as well as cutaneous histopathological features were analyzed. A total of 10 patients with early-stage MF were included, among whom 8 were males and 2 were females, with a mean age of (30.1±14.3) years. Dermoscopic examination revealed th

Protocol for a mixed-methods modified Delphi study for the development of a core domain set to assess the health-related quality of life of patients with mycosis fungoides and Sézary syndrome in clinical trials.

INTRODUCTION: Cutaneous T cell lymphoma (CTCL) is a group of non-Hodgkin lymphomas that primarily affects the skin and can mimic inflammatory dermatoses. Unlike many skin diseases, CTCL can lead to disabling symptoms, and advanced CTCL can even be fatal. Early studies investigating health-related quality of life (HRQOL) in patients with mycosis fungoides (MF) and Sézary syndrome (SS), common subtypes of CTCL, demonstrated significant impairment across numerous domains. The aim of this current study is to develop a core domain set (CDS) to identify the essential aspects of MF/SS that influence

Efficacy and tolerability of mogamulizumab in mycosis fungoides and Sézary Syndrome: a monocentric retrospective study.

BACKGROUND: Mycosis Fungoides (MF) is a common subtype of primary cutaneous T-cell lymphoma (CTCL), a group of non-Hodgkin lymphomas. The clinical spectrum of MF ranges from isolated cutaneous lesions to widespread involvement of lymph nodes, blood, and skin, as seen in its aggressive variant, Sézary Syndrome (SS). Mogamulizumab, a defucosylated humanized IgG1-κ anti-CCR4 monoclonal antibody approved for relapsed/refractory MF/SS, has demonstrated a favorable safety and efficacy profile in multiple case series. METHODS: This retrospective, monocentric observational study analyzed data from 12

Management of Mycosis Fungoides With Chlormethine Hydrochloride Gel in Combination With Systemic Therapies: A Case Series.

BACKGROUND: Mycosis fungoides (MF) is a subtype of T-cell lymphoma that is characterised by the infiltration of malignant T cells into the skin. Treatment approaches usually include skin-directed therapies for early-stage disease and, additionally, systemic therapies for advanced stages. Chlormethine hydrochloride gel is recommended as a first-line treatment option for adult patients with MF, with previous studies demonstrating its efficacy. Due to the rarity of this disease, available literature on the optimal treatment of MF with chlormethine gel is limited, creating challenges for making in

Laboratory Evaluation of Peripheral Blood Involvement in Mycosis Fungoides and Sézary Syndrome: Evolution of Flow Cytometry and Morphology Quantification and Interpretation.

Background/Objectives: Mycosis fungoides (MF) and Sézary syndrome (SS) are cutaneous T-cell lymphomas (CTCLs) with variable clinical outcomes. Peripheral blood (PB) involvement in MF/SS is an independent predictor of prognosis. Accurate laboratory determination of PB involvement by MF/SS cells, however, is an ongoing challenge. Both flow cytometry (FC) and morphology-based quantification are limited by the overlap of CTCL cells and reactive T-cells. This study looks at the optimization over time of CTCL blood burden evaluation. Methods: This retrospective study reviews CTCL blood assessment at

Inducible Costimulator and Its Ligand Promote Proliferation and Migration of Tumor Cells in Cutaneous T-Cell Lymphoma.

Inducible costimulator (ICOS) is a costimulatory immune checkpoint receptor expressed on activated T-cells, while the ICOS ligand (ICOSL) is expressed on antigen-presenting cells. The ICOS-ICOSL axis promotes the survival of memory and effector T-cells and induces several immune responses. In addition, the ICOS-ICOSL interaction induces cell proliferation, cell survival, and cytokine production. The roles of ICOS and ICOSL in cutaneous T-cell lymphoma (CTCL) are unclear. In this study, we examined the roles of ICOS and ICOSL in CTCL. The tumor cells co-expressed ICOS and ICOSL, and the upregul

Primary Cutaneous Lymphomas in Children: A Case Series Review with Emphasis on Diagnostic Imaging and Multidisciplinary Management.

INTRODUCTION: Primary Cutaneous Lymphomas (PCLs) are rare extranodal non-Hodgkin lymphomas that present in the skin without extracutaneous disease at diagnosis; they are exceptionally uncommon in children and frequently mimic benign dermatoses, delaying recognition. We report a case series of three pediatric patients managed at a national referral center. CASE PRESENTATION: Case 1: A 16-year-old with erythrodermic mycosis fungoides (stage IIIA) refractory to multiple systemic and skin-directed therapies who achieved remission after haploidentical allogeneic hematopoietic stem-cell transplanta

Mycosis fungoides and Sézary syndrome.

Mycosis fungoides (MF) and Sézary syndrome (SS) are the most common cutaneous T-cell lymphomas, arising from mature CD4+ memory T cells. Diagnosis is complex and relies on clinicopathological correlation, immunophenotyping and molecular clonality testing, while management is stage-adapted, with potentially curative outcomes achievable only in selected patients through allogeneic haematopoietic stem cell transplantation.

Persistence of sex differences and stable trends in mortality in patients with mycosis fungoides and Sézary syndrome in Spain (1983-2022).

BACKGROUND: Mycosis fungoides (MF) and Sézary syndrome (SS) are the most common subtypes of primary cutaneous lymphomas (PCLs). Despite increasing incidence, data on mortality trends specifically for MF and SS are scarce. This study aimed to analyse mortality trends for these lymphomas in the Spanish population from 1983 to 2022. METHODS: An ecological study was conducted using data from the Spanish National Statistics Institute. Age-standardised mortality rates (ASMRs) were calculated based on the 2013 European standard population. Changes in mortality trends were identified by joinpoint reg

Beyond CD30: Dual-Targeting of Malignant and Regulatory T Cells by Brentuximab Vedotin Remodels the Lymphoma Microenvironment and Overcomes Resistance via BCL2 Inhibition in Mycosis Fungoides.

Mycosis fungoides (MF), the most common subtype of cutaneous T cell lymphoma (CTCL), has a poor prognosis in advanced stages. Brentuximab vedotin (BV), a CD30-targeting antigen-drug conjugate approved for CD30+ MF following prior systemic treatment, still exhibits resistance with unclarified mechanisms. With single-cell RNA analysis on 13 paired tumor samples from 6 CD30+ MF patients, we revealed that BV-induced immunogenic cell death (ICD) in both CD30+ and CD30- malignant T cells while specifically enhanced interferon-α (IFNα) and IFNγ responses in CD30- subsets. BV also directly targeted CD

Mapping malignant T-cell states and immune circuits in Sézary syndrome by single-cell analysis.

BACKGROUND: Sézary syndrome (SS) and leukaemic mycosis fungoides (MF) are aggressive cutaneous T-cell lymphomas (CTCLs) with poor prognosis and limited treatment options. Single-cell RNA sequencing (scRNA-seq) offers an opportunity to dissect malignant heterogeneity and immune dysregulation yet has been underutilized in leukaemic CTCL. OBJECTIVES: To define transcriptional heterogeneity within malignant T cells, identify immunomodulatory signals shaping the tumour microenvironment and uncover actionable vulnerabilities in SS and leukaemic MF. METHODS: We analysed 144,321 peripheral blood mon

A Rare Adverse Effect of Methotrexate Mimicking Disease Progression in Folliculotropic Mycosis Fungoides: A Case Report.

INTRODUCTION: Cutaneous ulceration is an uncommon adverse effect of methotrexate (MTX). This complication has been rarely described in cutaneous T-cell lymphoma and, to date, has not been reported in folliculotropic mycosis fungoides (FMF). CASE REPORT: A 66-year-old Thai man with stage IB FMF achieved excellent clinical improvement with low-dose MTX therapy for 13 months. He subsequently developed disseminated erythematous papules, plaques, and necrotic ulcers on the extremities. Laboratory evaluation revealed mild anemia, and microbial cultures were negative.
