Diffuse large B-cell lymphoma (DLBCL) is the most common histological subtype of canine multicentric lymphoma, whereas epitheliotropism is classically associated with cutaneous T-cell lymphoma. A 9-year-old neutered male Chihuahua was diagnosed with stage IVb multicentric lymphoma and achieved complete remission with a UW-25 L-CHOP protocol. However, an upper lip mass, cytologically consistent with lymphoma, developed during remission. Electrochemotherapy was performed for local control and resulted in marked reduction of the lesion. The dog later developed recurrent peripheral lymphadenopathy

This phase 3 multicenter trial evaluated the efficacy and safety of adding dual epigenetic agents azacitidine and chidamide to CHOP (AC-CHOP) versus CHOP alone in previously untreated peripheral T-cell lymphoma (PTCL). A total of 128 patients were enrolled from 9 centers in China and assigned according to participating centers (AC-CHOP, N = 84; CHOP, N = 44). Azacitidine 200 mg on days 1-2, 100 mg on day 3 and chidamide 20 mg twice weekly were administered in the AC-CHOP arm. The AC-CHOP arm showed numerically higher ORR (60.7% vs. 54.6%) and CR rate (47.6% vs. 36.4%) than the CHOP arm, althou

Orbital apex syndrome is an uncommon neuro-ophthalmologic entity caused by lesions affecting the orbital apex and associated with inflammatory, infectious, vascular, and neoplastic processes. Synchronous tumors represent an exceptional condition defined by the simultaneous coexistence of neoplasms with different histological origins. We present the case of a 45-year-old woman with progressive visual acuity loss, painful ophthalmoplegia, facial hypoesthesia, and a right relative afferent pupillary defect, associated with an oropharyngeal lesion, nasal obstruction, dysphagia, and dysphonia. Adult Epstein-Barr virus-associated hemophagocytic lymphohistiocytosis (EBV-HLH) is a clinically heterogeneous syndrome comprising biologically distinct disease entities. We report three adults with EBV-HLH and describe their diagnostic evaluation and clinical course. We presumed primary EBV infection from anti-VCA IgM positivity, anti-EBNA negativity, and a history of possible saliva-mediated exposure in all three patients. Personal and family histories did not suggest familial HLH (fHLH), and genetic analysis performed in one patient did not identify pathogenic variants of fHLH. No history n

BACKGROUND: Chimeric Antigen Receptor T-cell (CAR T-cell) therapy is a promising treatment option for patients with refractory or relapsed B-cell malignancies. The success of this therapy depends on the efficient collection of lymphocytes (CD3+ T cells) through apheresis. This study analyzed the factors influencing CD3+ cell counts in apheresis products for CAR T-cell therapy.
METHODS: This exploratory analysis evaluated patients enrolled in the registration phase I/II trials of talicabtagene autoleucel at a tertiary care oncology centre in Western India. Patients were enrolled after obtainin

Racial and socioeconomic differences in cancer treatment timing are recognized in solid tumors, but comparable data in aggressive non-Hodgkin lymphoma (NHL) remain limited. Using the National Cancer Database (2004-2022), we conducted a retrospective cohort study of 271 472 patients with diffuse large B-cell lymphoma (DLBCL), Burkitt lymphoma, mantle cell lymphoma (MCL), and peripheral T-cell lymphoma (PTCL) who received multiagent chemotherapy. Time to treatment initiation (TTI) was defined as days from diagnosis to initiation of multiagent chemotherapy. Multivariable logistic regression ident

Allogeneic hematopoietic cell transplantation (allo-HCT) is an effective therapy for selected hematologic diseases. However, graft-versus-host disease (GVHD) limits broader application of allo-HCT. The catecholaminergic sympathetic nervous system modulates immune cells. Beta-2 adrenergic Receptor (β2AR) acts as an immune checkpoint, yet its role in CD8+ T cells remains poorly understood. We used an MHC-mismatched model of acute GVHD to study how β2AR activity on donor CD8+ T cells affects GVHD severity. Host mice receiving β2AR+/+ (WT) CD8+ T cells demonstrated less severe GVHD, lower inflamma

Peripheral T-cell lymphoma (PTCL) is an aggressive malignancy with poor prognosis that requires intensive systemic therapy, highlighting the potential impact of psychosocial factors on disease management and clinical outcomes. In this population-based study using SEER data from 2013 to 2022, we identified 5,919 patients newly diagnosed with PTCL. Propensity score matching (PSM) was applied to control for baseline differences, and survival outcomes were evaluated using Kaplan-Meier analysis and Cox proportional hazards models. Unmarried patients were more likely to be female, younger, and of no

In this single-arm pilot study, a novel regimen of post-transplant cyclophosphamide (PTCy), sirolimus, mycophenolate mofetil (MMF) and RGI-2001 was investigated to explore regulatory T-cell (T-reg) expansion in the setting of haploidentical allogeneic hematopoietic cell transplantation (HCT). RGI-2001 was infused at 100 ug/kg per dose, for six weekly doses. The primary endpoint was feasibility, assessed by the proportion of participants who were able to receive ≥4 doses of RGI-2001. Twenty-five participants enrolled. The primary endpoint was achieved by 80% of participants, deeming the regimen

INTRODUCTION: Chimeric antigen receptor (CAR) T-cell therapy is a standard treatment for several hematological malignancies, yet patient-specific responses and toxicities remain hard to predict. Although multiple covariates and risk scores exist, their prognostic accuracy remains limited.
METHODS: We prospectively measured CAR T-cell kinetics in peripheral blood of 92 patients with B-cell malignancies using multicolor flow cytometry. To explore underlying mechanisms, we developed a mechanistic model of CAR T-cell and tumor cell interactions.
RESULTS: We captured four phases of CAR T-cell kin

PURPOSE: Relapsed/refractory peripheral T-cell lymphoma (R/R PTCL) lacks a uniform salvage standard and outcomes with conventional chemotherapy (CC) remain suboptimal. This meta-analysis compared epigenetic-containing regimens (Epi-Rx) and CC in R/R PTCL.
MATERIALS AND METHODS: We searched MEDLINE, Embase, and the Cochrane Library through May 2025 for prospective or retrospective studies evaluating Epi-Rx (≥1 HDAC, DNMT/hypomethylating, or EZH1/2 inhibitor, alone or in combination) or CC without epigenetic agents. Random-effects models pooled single-arm proportions, with prespecified subgroup

PURPOSE: Pancreatitis in patients with hematologic malignancies, particularly lymphoma, is a challenging complication with heterogeneous etiologies. Large studies with systematic etiological classification and prognostic models are lacking.
METHODS: This retrospective multicenter study (2020-2025) included patients with hematologic malignancies who developed acute pancreatitis. Etiologies were adjudicated using predefined criteria. A nomogram-based prediction model was developed with internal bootstrap validation.
RESULTS: Among 1,705 patients, 31 (1.82%) developed pancreatitis: 19 lymphoma

Although chimeric antigen receptor T-cell (CAR-T) therapy has substantially improved outcomes in diffuse large B-cell lymphoma (DLBCL), disease relapse remains a major clinical challenge. Because treatment-associated immune remodeling evolves dynamically over time, complementary biomarkers capable of longitudinally monitoring immune-cell state changes are needed to evaluate therapeutic response and relapse following CAR-T therapy. In this study, we developed a label-free, high-throughput image cytometry platform for longitudinal, multiparametric profiling of peripheral blood mononuclear cells

BACKGROUND: Pulmonary involvement by peripheral T-cell lymphoma, not otherwise specified (PTCL-NOS), is an exceedingly rare extranodal manifestation with nonspecific radiological and clinical features. Its computed tomography (CT) findings-including ground-glass and consolidative opacities and, in some areas, a crazy-paving appearance-overlap with those of resolving infectious pneumonia and drug-induced lung injury, so that pulmonary lymphoma is readily mistaken for these more common conditions and the diagnosis is often delayed.

CD24 is highly expressed in aggressive B-cell malignancies and represents a novel immunologic vulnerability to overcome resistance to CD19 chimeric antigen receptor T-cell (CAR-T) therapy. We developed a CD24 × CD3 bispecific antibody (CD24 BsAb) and CXCR3-armed CD24 CAR-T cells, both exhibiting potent antitumor activity in lymphoma and leukemia models. CD24 BsAb and CD24 CAR-T overcome CD19 CAR-T resistance driven by antigen loss and apoptosis defects in vitro and in vivo. Beyond direct cytotoxicity, CD24-targeted T cells reprogram macrophages toward an M1-like phenotype and enhance phagocyti

Bispecific antibodies (BsAbs) targeting CD20 × CD3 have achieved remarkable efficacy in relapsed or refractory (R/R) B-cell lymphoma, yet clinical responses remain heterogeneous and predictive biomarkers are lacking. We retrospectively evaluated associations between peripheral immune profiles and outcomes in 40 patients treated with CD20 × CD3 BsAbs and explored BTK inhibitor-associated immune changes using a paired BTK inhibitor cohort, functional assays, mouse models, and single-cell RNA sequencing. Higher baseline CD4+ T-cell counts correlated with superior response rates and prolonged prog

Single-agent targeted therapies for relapsed or refractory peripheral T-cell lymphoma (R/R peripheral T-cell lymphoma [PTCL]) produce modest, short-lived responses, underscoring the need for tolerable, chemotherapy-free combinations. In an open-label, single-arm Phase Ib trial at two tertiary hospitals, we evaluated parsaclisib plus chidamide with step-down maintenance. Adults with R/R PTCL received parsaclisib 10-20 mg once daily plus chidamide 20 mg twice weekly for 8 weeks using 3+3 escalation; patients without progression or unacceptable toxicity continued parsaclisib 2.5 mg once daily plu
