Forty-three patients with cutaneous T-cell lymphomas (mycosis fungoides and the Sezary syndrome) underwent routine staging procedures to assess extent of disease prior to therapy. Evaluation of the liver included physical examination, liver function tests, 99mTc-liver-spleen scans, percutaneous liver biopsy, and peritoneoscopy with multiple liver biopsies. We obtained a monoclonal antibody, A50, after immunizing Biozzi's high responder strain of mice with T cell chronic lymphocytic leukemia (T-CLL) cells. Five cases of adult T cell lymphoma with hyperglobulinemia were reported. Two patients died of extension of the disease despite intensive chemotherapy. Three others are still under treatment. Twenty-two cases of prolymphocytic leukaemia (PLL) have been studied by transmission electron microscopy (TEM); 17 had B-cell surface markers (B-PLL) and five had T-cell characteristics (T-PLL). Isoenzymatic study of leucocytic acid phosphatase under normal conditions identifies 3 isoenzymatic bands, which exhibit a noticeable cell specificity. Antisera against the C57B1 (H-2b) mouse lymphoma, EL4 were prepared in rabbits. After absorption with mouse liver, red cells and thymocytes the antisera appeared to be cytotoxic for a subpopulation of peripheral T cells. The absorbed antisera blocked the immunosuppressor function of Con A-stimulated splenic lymphocytes, but was unreactive against Con A-stimulated and allogeneically primed cytotoxic cells, or helper T cells. Consequently, heteroantiserum against EL4 may provide a useful reagent for the differentiation of cytotoxic from suppressor T-cell subsets.

Results are reported on 724 children and adolescents with acute leukemia-lymphoma. One hundred patients had immunologic marker analysis that defined the major phenotypic groups, i.e., T-cell, B-cell (Burkitt), myeloid, and the most frequent form, non-T, non-T "common" or "undifferentiated" acute leukemia. The development of Sézary syndrome was observed in a patient with mycosis fungoides. The Sézary cells in the peripheral blood were identified as a specific T-cell population with helper cell characteristics. Autoradiographic analysis showed that the Sézary cells obtained from skin biopsies proliferated much more rapidly than those from the peripheral blood. After transformation into a highly malignant T-cell lymphoma (immunoblastic) it came to an abrupt and fatal course of the disease.

Cytology of peripheral blood and lymph node lymphocytes from a group of unselected patients with cutaneous T cell lymphoma (CTCL) was studied by light microscopy. Twenty of 45 patients had circulating lymphocytes with convoluted nuclei recognized in routine Wright-Giemsa-stained peripheral blood smears. Skin infiltrates and tumors characteristic of mycosis fungoides, together with other clinical features typical of Sézary's syndrome (such as erythroderma, itching and Sézary cells in the peripheral blood), were found in a 76-year-old female. Histologically, there was a shift from a well-differentiated to a poorly-differentiated lymphoblastic infiltration indicating the transformation from a low-grade to a high-grade malignant (T-cell) lymphoma.

Adenosine deaminase (ADA) activity was determined in peripheral blood T cells, thymocytes and blasts of 42 acute leukemia patients. An assay that simultaneously detects two differentiation markers found in thymocytes is described. There markers include the membrane receptor for sheep erythrocytes (E) and the nuclear enzyme, terminal deoxynucleotidyl transferase (TdT). Cells are incubated with E, pelleted onto slides and then stained for TdT by an indirect immunofluorescence technique. The E-positive, TdT-positive phenotype is characteristic of the tumor cells in many cases of lymphoblastic lymphoma. Cells of this dual phenotype were not found in the peripheral blood or bone marrow of individuals without neoplastic disease.
